Fossil fuel is a rare commodity and the combustion of this fuel results in negative environmental effects. This paper evaluates and validates the electronics needed to run intelligent algorithms to lower the fuel consumption for commercial vessels. This is done by integrating advanced fuel saving functions into an electronic device that controls the fuel injection of large diesel engines, as known as a diesel engine governor. The control system is classified as a safety critical system. This means that the electronics needs to be designed for fail safe operation. To allow for future research a

The cystic fibrosis transmembrane conductance regulator (CFTR) is a plasma membrane chloride channel expressed in epithelial tissues. While protein kinase A (PKA) is the main activator of CFTR, patch clamp data have suggested a permissive role for PKC in modulating PKA responses. To clarify the role of PKC in CFTR regulation we used PCR mutagenesis to construct a CFTR mutant (9CA) in which the serines or threonines in nine consensus sequences for PKC phosphorylation were replaced by alanines. Iron is an essential nutrient for many biochemical functions within the body. However, its flexible redox potential is a "double edged" sword that makes it both essential for life but also renders it potentially toxic. As iron excretion is limited, iron absorption is tightly regulated. Systemic iron homeostasis is controlled through the hepatic hormone hepcidin. Iron and other stimuli control hepcidin transcription. When produced hepcidin circulates through out the body and binds to the cellular iron exporter ferroportin (FPN1) expressed on duodenal enterocytes, macrophages, and hepatocytes

By impeding the expression of the tumor suppressor p53, Bmi1 is able to prevent apoptosis and senescence. c-Myc, a prominent oncogene, cooperates with Bmi1 to stimulate cellular transformation and tumorigenesis. Further investigation of the basic biological interplay between Bmi1 and c-Myc is crucial for our understanding of their tumorigenic ability. In my project I demonstrated that c-Myc and Bmi1 directly interact with each other and form nuclear foci. Overexpression of Max,

Abstract:
Aircraft noise is one of the most controversial environmental concerns in the aviation industry, partly due to the difficulty in harmonizing countries' regulation regarding this issue. The purpose of this thesis is to analyze the ways in which aircraft noise is regulated at the national and international levels, and to compare the legislative responses to aircraft noise issues in Europe and North America. Each of the four main chapters of the thesis takes into consideration a different aspect of the problem. The first chapter describes the objective and subjective ways of measuring airc

Abstract:
Limited competition among the few fuel retailers in rural and remote locations results in fuel pricing problems, such as high rural fuel prices and rural-urban fuel price differentials. Thus, the regulation of fuel retailing activities in these locations is expected to resolve these problems, and thus contribute to a rural development policy. These problems are of concern to governments and rural communities, primarily because high fuel prices generate significant multiplier effects on the cost of rural living and economic activities. Therefore, the purpose of this study was to investigate the

Abstract:
Interferon regulatory factors (IRFs) is a family of transcriptional regulators that are essential for cell differentiation and growth, oncogenesis and the regulation of immunity. IRF1 and IRF8 play a crucial role in immunity by regulating the differentiation of the cells of myeloid lineage, particularly the development of immune cells including macrophages, NK and dendritic cells. Both transcription factors are essential for the resistance to viral and bacterial infections. We optimized the chromatin immunoprecipitation (ChIP) technique to be used with IRF8 and IRF1 antibodies in a J774 line

Many forms of cancer express activated Stat3, a pleiotropic transcription factor that is implicated in pro-tumourigenic signaling in a variety of cancer cell lines, revealing that it may be important for tumour onset, progression, and malignancy. Yet, the role of Stat3 in the context of tumourigenesis in vivo is still unclear. Downstream of an activated form of ErbB2, we have demonstrated that Stat3 is dispensable for in vivo tumour initiation but is crucial for metastasis. Conversely, the loss of Stat3 in a novel inducible model of PyVMT-driven tumourigenesis revealed that Stat3 is necessa

Intracranial gliomas are the most common primary brain tumors. Despite advances in medicine, patient outcomes have not changed significantly over the last century. The invasive behavior of these cells is the main reason for treatment failure. Cancer invasion and cell motility are topics under intense investigation in cancer research. However, they are less studied in gliomas and the literature describing the molecular mechanisms which govern glioma cells motility is lagging. We have recently reported Down Regulated in Renal cell carcinoma (DRR) as a novel modulator of glioma invasion and migrat

Myelin is an evolutionarily late acquisition of the vertebrate nervous system which speeds electrochemical signaling in mature nerve fibers by providing insulation in the form of a lipid-rich multilammelar sheath. Proteolipid Protein (PLP) is the most abundant protein in mature mammalian central nervous system myelin where it serves as a structural component in addition to other yet undefined roles. It is coordinately regulated at the transcriptional level with other myelin genes such as Myelin Basic Protein (MBP). The major components of MBP transcriptional regulation have been defined using

Biochemical regulatory networks are involved in many essential processes of life, such as maintaining homeostatic conditions, cell-cycle division, and responding to environmental stimuli. Such networks contain a staggering degree of complexity, inhibiting our ability to understand how they function. Two major challenges in quantitative biology are addressed in this thesis. First, the problem of identifying regulatory models from observational data. Second, characterizing how regulatory networks evolve. Making use of a simple regulatory model that is based on piecewise linear
