Intra and Inter Vegetative Compatibility Group (VCG) heterokaryon formation was observed in Fusariurn oxysporum f.sp. cubense (Foc). Using the double-pick method to force heterokaryon formation via hyphal anastomosis, 104 pairwise combinations were done between 100 auxotrophic mutants of Foe representing races 1, 2 and 4 in five VCGs. Genetic variation within the fungus F. oxysporum f.sp. cubense, which causes Fusarium wilt in banana, was examined using RAPD-PCR. RAPDPCR is a method of generating genome specific "fingerprints" which can be compared to determine genetic relatedness. Fusarium oxysporum grown on natural media was believed not to produce mycotoxins of the trichothecene family. Using a defined chemical medium toxin production was investigated for and it was found that trichothecenes were produced. A yeast bioassay using Kluyveromyces fragilis, an organiam sensitive to such trichothecenes as T-2 toxin and verrucarin, was used for detection of toxin in culture filtrates. These were tested for pathogenicity at 20 C and -0.01 MPa water potential in artificially infested, autoclaved field soil. Highly pathogenic, moderately pathogenic, and hypovirulent isolates of both species were identified.
