Abstract:
Saccharomyces cerevisiae Hansen GAL1 (galactokinase) generates galactose-1-phosphate; GAL7 (galactose-1-phosphate uridylyltransferase) transfers UDP between galactose or glucose and their respective sugar-1-phosphate conjugates, and both are essential on galactose. Aspergillus nidulans ANID_04957 has 41% amino acid sequence identity with GAL1; ANID_06182 has 50% sequence identity with GAL7. The names Aspergillus nidulans GalE (galactokinase) and GalD (galactose-1-phosphate uridylyltransferase) are consistent with prior studies. Complemented galDΔ:ScGAL7 and galEΔ:ScGAL1 strains had wild-type phenotype, demonstrating functional homology. The galD5 and galE9 alleles were truncated. Strains galDΔ and galD5 were impaired on minimal medium containing 1% galactose (MM-Gal) at pH 7.5 and did not grow on MM-Gal pH 4.5. Strains galEΔ and galE9 grew on MM-Gal at both pH levels. Strains galDΔ and galEΔ produced wild-type conidiophores on minimal medium containing 1% glucose (MM-Glu) but few spores; for both, sporulation was lower on MM-Gal pH 7.5. GalD-GFP (green fluorescent protein) and GalE-GFP were cytosolic and upregulated on MM-Gal, consistent with quantitative real-time polymerase chain reaction. Galactofuranose immunolocalization in galDΔ resembled wild type on MM-Glu but was reduced on MM-Gal. The galEΔ strains had immunolocalizable Galf on all these media. Strains galDΔ and galEΔ were more sensitive to calcofluor, caspofungin, and itraconazole on MM-Gal. Neither galD nor galE is essential on galactose at high pH, implying additional routes for galactose metabolism in Aspergillus. Aspergillus galactose metabolism is more complex than that of S. cerevisiae.

The effect and law of photocatalytic degradation of reactive gaudy red X-3B dye were investigate and the applied process parameters were determined.Using self-synthesized immobilized Fe3+- TiO2 photocatalyst,self-made catalytic oxidation reactor was made to research on reactive gaudy red X-3B dye wastewater by using light source of 20W UV sterilization light.The effects of immobilized Fe3+- TiO2 catalyst content、pH value、Fe3+doping content and adding trace H2O2 on photocatalytic degradation were discussed.The experimental result show that reactive gaudy red X-3B dye wastewater decolorization rate and the removal rate of COD are more than 99% and 90% when the initial reactive gaudy red X-3B dye wastewater concentration was 50mg/L,pH value was 3,input amount of catalyst was 10g/L,Fe3+doping content was 0.5% and the dosage of H2O2 was 0.6mL/L(3%).Immobilized Fe3+- TiO2 photocatalyst can remove the color and persistent organic pollutants in reactive gaudy red X-3B dye wastewater effectively.

Abstract:
Transmission electron microscopy (TEM) has been used to investigate the damage produced following high temperature (350˚C) Xe implantation into [100] Si at fluencies (>1x1016 Xe ions/cm2) and energy (250keV) which produce a buried amorphous layer; and the defect structures produced following thermal anneals of 400˚C, 600˚C or 800˚C for 30 minutes. Analysis of these samples yielded results which suggested that the Xe gas contained within the amorphous layers was swept by the amorphous/crystalline interfaces during solid phase epitaxial re-crystallisation (SPEG) into large bubbles elongated along a direction perpendicular to the interfaces. In order to further investigate this sweeping effect, buried amorphous layers were produced in Si by implanting Li at liquid nitrogen temperature and post implanting the layers with 1x1016 He ions/cm2. Contrary to the spherical bubbles produced under similar conditions in crystalline Si, irregular shaped bubbles were formed in the amorphous layer. Results from in-situ TEM studies showed that these bubbles are mobile at temperatures lower than expected in crystalline Si. Thus, upon reaching the moving interfaces between amorphous and crystalline Si, the bubbles are forced back into the amorphous material which ultimately results in coalescence of the gas into larger bubbles once the two interfaces combine. In addition, microtwins have been shown to form in regions of the re-crystallising layer where there exists an excess of interstitial-type defects. This is contrary to previous microtwin nucleation models which suggested that microtwins are either formed on [111] planes or on bubbles
L'endommagement induit par implantation de xénon dans le silicium a été étudié par microscopie électronique à transmission (MET). Les implantations réalisées à 350°C, à une énergie de 250keV et pour des fluences supérieures à >1x1016 Xe ions/cm2 conduisent à la formation d'une couche amorphe enterrée. Les observations effectuées sur les échantillons recuits montrent la présence d'une rangée de grandes cavités allongées dans la direction perpendiculaire à l'interface. Ceci suggère que, lors de la recristallisation du silicium, le déplacement simultané des deux interfaces entraîne le déplacement du gaz jusqu'à son confinement dans de larges bulles. Afin de mieux appréhender les mécanismes qui conduisent au mouvement des bulles, de l'hélium à faible dose a été implanté dans du silicium préalablement amorphisé par implantation de Li à basse température. L'implantation d'hélium dans le silicium amorphe conduit à la formation de bulles de forme irrégulière. Ce résultat diffère du silicium cristallin où des bulles sphériques sont obtenues pour des conditions d'implantation identiques. Les expériences réalisées ‘in situ’ dans le MET montrent clairement d'une part que les bulles sont poussées par l'interface, et d'autre part la nucléation de micromacles. Il a été mis en évidence au cours de cette étude que les bulles sont mobiles à plus basse température dans le silicium amorphe que dans la phase cristalline. Lors de la recristallisation, les bulles se trouvent alors confinées dans le matériel amorphe, ce qui résulte en leur coalescence et à la formation de larges bulles une fois que les deux fronts de recristallisation se sont rejoints. De plus, il a été établi que la formation de micromacles dans la région recristallisée est liée à un excès de défauts de type interstitiels dans la zone amorphe. Ce résultat est contraire aux modèles de la littérature qui suggèrent que les micromacles se forment soit sur des plans {111} soit sur les bulles

Abstract:
This practice-led thesis situates the experimental novella Gaudy Bauble within the context of interdisciplinary approaches to experimentation which cross the arts, humanities, literature and sciences. The novella and thesis develop a queer avant-garde poetics and writing methodology that I have called transliteracy. Transliteracy builds on my situated and embodied writing practice as a queer identified novelist and nonnative English speaker. I have mobilised the perceived 'otherness' of English to produce narratively and linguistically experimental prose fictions (Waidner, 2010, 2011). Transliteracy develops this practice by sharing agency (the capacity to influence the narrative) across assemblages of human and nonhuman, fictional and real, material and semiotic 'actors', to use the philosopher of science Bruno Latour's (1987, 1999) term for participants in action and process. Transliteracy has allowed me to subvert normative versions of authorship, intentionality, causality, and process in Gaudy Bauble, and to produce a radically subverted version of a plot that is intelligible and captivating to the reader. Gaudy Bauble inaugurates a genre I have called agential realist fiction, which is original in its genre-bending, gender-bending, interdisciplinary and queer avant-garde orientation. The practice was further shaped according to a generative constraint, which dictated that the most marginal actors on and beyond the page were made relevant for the plot. This conceptual apparatus is also reflected in the novella's narrative as a 'not quite' detective story: Gaudy Bauble stages what happens if previously inconsequential actors are allowed to become effectual, rather than actions located within a conventional protagonist. Enacting an "insurrection of subjugated knowledges" (Foucault, 1980, p. 81) in fiction, Gaudy Bauble stages a landscape of reversed power relations, a locally subverted surface of emergence in fiction, where radically nonnormative phenomena and imaginaries can come into being. The thesis connects transliteracy to a wider political LGBTQI+ project and agenda.

La dégradation de la qualité de l’eau de la Seine à Paris et dans sa banlieue depuis la deuxième moitié du XIXe siècle a fait l’objet de plusieurs études. Rares sont les études qui confrontent les perceptions de médecins, d’ingénieurs, d’hygiénistes ou d’administrateurs sur la qualité des eaux des rivières et l’évolution de leur état. Cette confrontation permet d’analyser sur la longue durée l’efficacité des réponses apportées pour améliorer la qualité, ce que nous avons réalisé ici pour la ville de Versailles et ses eaux. La première partie du mémoire décrit les perceptions de la qualité des eaux des rivières par des experts entre 1854 et 1964 sur la base de l’analyse de revues d’hygiène et d’assainissement urbain, montrant l’évolution des indicateurs de qualité et de la réglementation. Versailles a pour particularité d’être, au XIXe siècle, essentiellement alimentée par de l’eau de Seine grâce à la machine de Marly, par le biais d’un Service dépendant directement de l’Etat, conséquences de son passé royal. Mais la qualité de l’eau du fleuve se dégradant à cause des rejets des égouts de Paris, les différentes administrations, locales et nationales, doivent résoudre la question d’approvisionnement en eau alimentaire et recourent à l’aide d’experts pour s’assurer de sa qualité. Les analyses chimiques et bactériologiques effectuées sur les eaux conduisent à l’abandon de l’eau de la Seine en 1894 au profit des eaux de la nappe de Marly-Croissy. En même temps, Versailles rejetait ses eaux usées dans des cours d’eau à faible débit : le ru de Marivel et le ru de Gally. Epidémies et plaintes accompagnent la dégradation de leur état. Les nombreuses expertises menées témoignent de l’intérêt pour la qualité des eaux de surface et en même temps de l’absence de la volonté politique et des limites techniques pour l’améliorer, conduisant à leur sacrifice pendant plus de 100 ans
The Seine River pollution both in Paris and its suburbs has been already the subject of several studies. Few studies compare perceptions that physicians, engineers, hygienists or managers might have of the quality of water resources, and the evolution of their state. This confrontation makes it possible to analyze over the long term the effectiveness of the responses made to improve quality. We have achieved this here for the city of Versailles and its waters. The first part of this work describes perceptions of river water quality by experts between 1854 and 1964 based on an analysis of hygiene and urban sanitation reviews, showing the evolution of quality indicators and of regulations. In the 19th century, Versailles has the peculiarity of being essentially fed by water from the Seine thanks to the Marly machine, through a service directly dependent on the State, consequences of its royal past. But as the water quality of the Seine River deteriorates because of the discharges from the Paris sewers, the various administrations, local and national, have to solve the question of water supply and asked for the help of experts to ensure its quality. The chemical and bacteriological analyzes carried out on waters lead to stopping the use of the Seine waters in 1894 in favor of the Marly-Croissy water table. At the same time, Versailles rejected its wastewater into two low-flowing streams: Marivel and Gally. Epidemics and complaints accompany the deterioration of their state. The numerous experiments carried out show the interest in the quality of surface water and at the same time the absence of the political will and the technical limits to improve it, leading to their sacrifice for more than 100 years
