Full textThe 67Ga is an important radiopharmaceutical used to identify inflammatory processes in chronic illnesses, diagnosis by image of tumors in soft tissues and the possibility to evaluate the result for therapeutic intervention. In the present work a module of 67Ga processing was developed with the objective to reduce the interventions in the hot cell, in order to avoid oxidation caused by metallic materials, and consuming in hoses of the peristaltic pumps, that release residues that blocked the valves used in the process. With materials such as: acrylic, PVC, PEEK e teflon and they are used vacuum as method (way) of fluid transferences instead of peristaltic pump in the majority of the procedures, with this improvements the system can make shorter the lengths of transference hoses, increasing the yield in the process with less interventions for maintenance and time exposure of the workers, guaranteeing the quality and reducing the time of the processing. using a mobile system for displacement of the processing module making in the cleanness and maintenance of the cell that works with radioactive material. Reducing the time of exposure dose of the workers in compliance with RDC-17 of ANVISA, which ruling the Good Manufacturing Practice Procedures.

Full textRadioisotopes of gallium have been studied and evaluated for medical applications since 1949. Over the past 50 years 67Ga has been widely used in the diagnosis of various diseases, including acute and chronic inflammatory lesions, bacterial or sterile and several types of tumors. In Brazil 30% of clinics that provide services for Nuclear Medicine use 67Ga citrate and the demand for 67Ga at IPEN-CNEN/SP is 37 GBq (1 Ci)/week. The 67Ga presents physical half-life of 3.26 days (78 hours) and decays 100% by electron capture to stable 67Zn. Its decay includes the emission of γ rays with energies of 93.3 keV (37%), 184.6 keV (20.4%), 300.2 keV (16.6%) and 888 keV (26%). In the past 67Ga was produced by the reaction 68Zn (p, 2n) 67Ga at IPEN-CNEN/SP. After irradiation, the target was dissolved in concentrated HCl and the solution percolated through a cationic resin DOWEX 50W-X8, 200-400 mesh, conditioned with 10 mol L -1 HCl. Zinc, nickel and copper were eluted in 10 mol L-1 HCl and 67Ga 3.5 mol L-1 HCl. The final product was obtained as 67Ga citrate. This work presents a new, fast, direct and efficient method for the chemical separation of 67Ga by thermal diffusion (heating of the target) combined with concentrated acetic acid extraction. Purification was performed by ion exchange chromatography. Natural zinc electrodeposition was performed on nickel/copper plates as substrate and the zinc deposits were adherent to the substrate, slightly shiny and uniform. The targets were irradiated with 26 MeV protons and integrated current of 10 μA.h. After irradiation, the targets were heated at 300 °C for 2 hours and placed in contact with concentrated acetic acid for 1 hour. The average yield of extraction of 67Ga was (72 ± 10)%. This solution was evaporated and the residue was taken up in 0.5 mol L-1 NH4OH. The 67Ga was purified on cationic resin Dowex 50WX8 in NH4OH medium. The 67Ga recovery was (98 ± 2)%. This solution was evaporated and taken up in 0.1 mol L-1 HCl. The chemical purity was evaluated by ICP-OES that resulted in (2 ± 1) μg mL-1 of zinc. The concentration of iron, copper and nickel was lower than the detection limits and also than the utilization limits for 67Ga. The radionuclidic purity was greater than (99.9%). This method showed to be suitable to obtain high purity 67Ga in less aggressive chemical conditions than before.

Full textIn recent decades, nuclear medicine has been used as diagnostic agent for disease and for the treatment of cancer. Part of this success is related to the constant research and development of new radiopharmaceuticals. Thiosemicarbazone and their derivatives have proven to be promising agent for the treatment of tumors, both in its organic form or as organo-metallic complexes. Also, they can to form complexes with metal radioisotopes giving radiopharmaceuticals for diagnosis and therapy. In this work we prepared complex of benzyl-5-hydroxy-3-methyl-5-phenyl-4,5-dihydro-1H-pyrazol-1-carboditionato (H2bdtc) with radioisotopes [99mTc]technetium (67/68Ga)gallium and (111In)indium and the radiochemical purity, Log P and stability in the presence of L-cysteine, L-histidine, human serum albumin (HSA) and human blood plasma were assessed; also were assessed the in vitro uptake rate of radiopharmaceuticals in murine melanoma cells B16F10 and TM1M, besides the evaluation of ex vivo uptake and in vivo using C57Bl/6 mice inoculated with both tumor lines. With [99mT] technetium two different complexes were obtained, depending on the concentration of the PBS in the solution, and one of them was had its structure to confirm as [[99mTc]O(bdtc)(Hbdtc)] from the standard rhenium complex [ReO(bdtc)(Hbdtc)], the other [99mTc] echnetium complex as well as (67/68Ga)gallium and (111In)indium not have characterized the structure. The labeling efficiency of compleos was higher than 90%, with log P higher than 1 for the complexes [[99mTc]O(bdtc)(Hbdtc)], [[99mTc]-bdtc] and [(67/68Ga)-bdtc] and 0.9 to [111In-bdtc]. All the complexes showed good stability in the presence of L-cysteine and L-histidine, especially in the first hour of incubation, but not in the presence of HSA and plasma. The uptake in vitro complexes in B16F10 and TM1M cells varied between 0.6% and 1.8%, and in ex vivo biodistribution studies was obesrvada intense and persistent liver uptake and spleen, exceeding 90%, and tumor uptake in changing from 0.2% to 3%, while in vivo imaging was not possible to observe a properly uptake in tumors, not allowing to use these molecules as a diagnostic agent. The results indicate that the thiosemicarbazone derivative complex can give complexes with different metals, but new derivatives should be prepared to try to improve performance in biological systems. Este trabalho tem como objetivo investigar a concentração de impurezas radioativas gama emissoras presentes nas soluções dos radiofármacos produzidos no Instituto de Pesquisas Energéticas e Nucleares - IPEN em São Paulo. Para que este radiofármaco possa ser utilizado adequadamente, sua qualidade deve ser avaliada de acordo com os procedimentos estabelecidos de acordo com os \"Requisitos Gerais para a Competência de Laboratórios de Teste e Calibração\", ISO / IEC 17025: 2005 e pelas \"Boas Práticas de Fabricação\" (BPF), controladas pela ANVISA (Agência Nacional de Vigilância Sanitária), no Brasil. Para determinar a atividade, dos radiofármacos das impurezas gama emissoras, foi utilizado um espectrômetro gama de alta resolução em duas distâncias fonte-detector; uma de 18 cm e outra de 1,7 cm. Para a distância de 18 cm, o espectrômetro HPGe foi calibrado com energias gama entre 81 kev e 1408 kev, medindo ampolas seladas de 60Co, 133Ba, 137Cs e 152Eu, padronizadas no Laboratório de Metrologia Nuclear (LMN) do IPEN. Para impurezas com baixas atividades, utilizou a distância fontedetector de 1,7 cm. A esta distância, o efeito soma em cascata é muito elevado, tornando difícil a medição das ampolas de calibração padrão, com isso, a curva de eficiência do espectrômetro foi obtida por um código de simulação de Monte Carlo, desenvolvido no IPEN. Neste código, todos os detalhes do sistema de detecção são modelados e as curvas de resposta para raios X e raios gama são calculadas pelo código de transporte de radiação MCNPX. Os espectros gama foram analisados pelo programa Alpino, que aplica o método de integração numérica da área sob os fotopicos de absorção total. Para as impurezas gama emissoras não detectadas visualmente, os limites de detecção foram calculados a partir da taxa de contagem de fundo, sob a área do pico de interesse. As soluções radioativas analisadas foram 67Ga,99Mo, 99mTc, 111In, 131I, 153Sm, 177Lu e 201Tl. Os resultados da relação entre a atividade do radionuclídeo em análise e as impurezas identificadas apresentaram acordo com os certificados de análise dos fabricantes, assim como, com as especificações da ANVISA.
