Full textA procedure for the strengthening of underground structure has been developed as main goal of the present thesis. Two particular case studies, concerning the rebuilding and consolidation of the Risorgimento Gallery in Ancona and the Dry-Dock into the port of Sestri Ponente (GE), has been investigated. A careful assessment of the actual state of conservation of the structure, carried out through an accurate geological and structural survey, has to be the basis for every reconstruction project of deteriorated structures. The degradation of an underground structure is usually characterized by infiltrations through the coating and deterioration of structural materials. For this reason, the aims of a reconstruction project involve the recovery of the waterproofing and the stability of the structure. The operating sequence is divided into four steps, each one with a respective purpose to be accomplished: the 1st step concerns the scarifying of damaged concrete while the 2nd one the renewal of the existing coating and the consolidation of ground behind the structure. On the 3rd step, it is planned to rehabilitate the contact between the coating and the ground behind the structure and to eliminate infiltrations through the current coating using grouting and iterative procedures optimized during the construction. Finally, the 4th step concerns the reconstruction of the final lining using innovative fiber-reinforced materials to ensure durability and strength for the whole service life of the renewed structure. The division into steps of the investigated procedure allows it to be applied to structures having similar conditions to those studied and to optimize the processes according to the purposes to be achieved and to the specific state of conservation of the structure.

Full textDen funktionella kontakten mellan byggnaden och gaturummet påverkar staden och dess attraktivitet. Butiker som är placerade i gallerior och som har sina entréer mot interna stråk skapar en segregation mellan gallerian och gaturummet. Denna segregering innebär att gallerian förlorar en del av sin attraktivitet, vilket kan resultera i en så kallad Greyfield mall. Uttrycket Greyfield mall används för gallerior som förlorat sina besökare. Konkurrensen från nya moderna gallerior samt brist på investeringar är exempel på faktorer som påverkar uppkomsten av Greyfield malls. För att klargöra sambandet mellan byggnaden och gaturumet har ”Space syntax” använts. Space syntax kan förklaras som ordningen mellan de olika rummen. En Space syntax-analys ger ett integrationsvärde som visar hur stadens gator hänger samman. Syftet med detta examensarbete är att undersöka och utveckla metoder för att öka attraktiviteten hos köpcentrum och gallerior som riskerar att utvecklas till, eller redan klassas som Greyfield malls. Förbättringar av dessa platser bidrar till en godare stadsmiljö. Frågeställningarna har inriktats på gatans och byggnadens utformning samt besökarnas rörelsemönster. Frågeställningarna har besvarats genom litteraturstudier inom områdena Space syntax, Shared space och Greyfield malls. Därtill har ett nytt förslag på utformning av gatunätet och byggnaden utförts. Analyser med hjälp av Depth map har gjorts i en fallstudie av Rosengallerian. Rosengallerian är placerad i Huskvarna centrum och har potential att fungera som en samlingspunkt för stadens invånare. Områdets nuvarande utformning gynnar biltrafiken vilket har resulterat i en nedprioritering av gångtrafikanterna. Med hjälp av; Shared space, Boone-gator och axialanalyser med Space syntax har ett nytt förslag för Rosengallerian och dess omgivning tagits fram. Förslaget bygger på att stadens gångtrafikanter istället prioriteras. Gatorna runt gallerian har omvandlats till gångfartsområde, där gatorna har fått en smalare karaktär. Dessa förändringar skapar en attraktivare miljö för gångtrafikanterna kring Rosengallerian. För att öppna upp gallerian har de ursprungliga gångarna inne i gallerian ändrats till två raka tvärgator igenom byggnaden som sammanbundits med den övriga staden. Resultatet av studierna visar att problemet huvudsakligen ligger i byggnadens utformning samt det omkringliggande gatunätets prioriteringar. Ett gatunäts planering och prioriteringar är av vikt för stadens invånare, då biltrafiken förflyttar sig på de gåendes villkor skapas ett centrum där människor kan röra sig fritt. Den fria rörelsen tillsammans med att stadens gator förlängs in i byggnaden medför att invånarna vill återkomma till gallerian. Förslagets förändringar ökar gallerians attraktivitet då människor attraherar andra människor. Slutsatsen är att gallerian skall ses som en enskild byggnad som skall passas in i den omkringliggande befintliga miljön. Gallerian bör vara en naturlig del av stadens rörelsemönster samtidigt som den gärna får markeras arkitektoniskt.

Full textGrowth conditions, influenced the growth rate and the interactions of the bacterium with nonimmune G. mellonella larvae. In general, X. nematophilus cells grown under aerobic conditions were more susceptible to the nonself defences of G. mellonella larvae than those grown under less than ideal conditions, resulting in increased clearance of the bacteria from the hemolymph (blood) of the insects. Clearance of the bacteria from the hemolymph of the insect was positively correlated with culture condition, culture age, and attachment to insect hemocytes in vitro.
Isolates of X. nematophilus produced flagella and fimbriae when grown under microaerobic and aerobic conditions. The type of fimbriae produced was influenced by culture conditions. The injection of both flagella and fimbriae in picogram quantities into nonimmune G. mellonella caused an increase in total hemocyte counts within these insect larvae. The injection of fimbrial and flagellar antigens into G. mellonella larvae caused changes in the hemocyte types found in circulation in the insect's hemolymph. (Abstract shortened by UMI.)

Full textThe involvement of apolipophorin-III in the immune responses of G. mellonella larvae to lipoteichoic acids, surface components of Gram-positive bacteria, was examined. Lipoteichoic acids from Bacillus subtilis, Enterococcus hirae and Streptococcus pyogenes caused a dose- and time-dependent drop in the total counts of circulating hemocytes and a partial or complete depletion of plasmatocytes depending on the species of lipoteichoic acid. All lipoteichoic acids tested activated phenoloxidase in vitro; however, in vivo, only B. subtilis lipoteichoic acid elevated the phenoloxidase activity while the other two suppressed it. Binding of apolipophorin-III to lipoteichoic acids was demonstrated. Apolipophorin-III prevented the complete depletion of plasmatocytes and depressed the activation of phenoloxidase by lipoteichoic acid from B. subtilis. The concentration of apolipophorin-III in hemolymph two hours post injections of lipopolysaccharides or lipoteichoic acids into larvae of G. mellonella did not change with respect to control insects that received phosphate-buffered saline. The concentration of apolipophorin-III in hemolymph at the end of the feeding larval stage was 8--12 mg/mL of hemolymph. Apolipophorin-III was present in significant amounts in the prepupal, pupal and adult stages. The protein was detected immunologically in hemocyte lysates, plasma and fat body. Non-denaturing polyacrylamide gels and immunoblots of fresh hemolymph suggested that apolipophorin-III is associated with a 77 kDa protein.

Full textGalleria mellonella es un modelo animal utilizado extensamente como alternativa para investigar la virulencia y patogenicidad bacteriana in vivo. También es apropiado para estudiar la eficacia y toxicidad de compuestos. Las larvas tienen un tamaño manejable, son económicas de adquirir y reproducir, presentan un bajo riesgo biológico, y son más aceptadas éticamente. Además, tienen un sistema inmunológico innato muy similar al de los mamíferos. Utilizamos G. mellonella para desarrollar un modelo animal de infección y toxicidad estandarizado y reproducible. Pseudomonas aeruginosa, un patógeno oportunista, que infectando emplea ribonucleótido reductasa (RNR), catalizando la reducción de ribonucleótidos a desoxirribonucleótidos y proporcionando así las moléculas precursoras necesarias para la síntesis de ADN. Desarrollamos un vector sin promotor con bioluminiscencia, el cual se utilizó para construir fusiones con los promotores de los genes RNR. Además, optimizamos un protocolo de extracción de ARN bacteriano para facilitar el estudio de los niveles transcripcionales de genes in vivo. Debido a la multiresistencia emergente de Staphylococcus aureus, se probó la toxicidad y eficacia antimicrobiana de nuevos derivados del ácido oleanólico y maslínico en G. mellonella. De los catorce derivados probados, dos tenían menos toxicidad y más eficacia in vivo que in vitro. G. mellonella se usó para determinar la toxicidad de nanopartículas y estrategias terapéuticas. Mycolicibacterium brumae no fue tóxica para las larvas y los resultados se correlacionaron con los obtenidos con ratones. Las nanopartículas causaron efectos tóxicos en las larvas detectados por la medición de la dosis letal y la proliferación de hemocitos, entre otros indicadores. Debido a la amplia aplicabilidad de G. mellonella, se necesitan nuevas metodologías para maximizar su potencial. Además del protocolo de extracción de ARN previamente mencionado, también se optimizó otro de aclaramiento. Las larvas fueron inyectadas con nanopartículas, fijadas con paraformaldehído, permeabilizadas con metanol y aclaradas con alcohol bencílico y benzoato de bencilo.

Os insetos respondem à infecção através da montagem de reações imunes do tipo celular e humoral. Os reguladores primários dessas respostas são células chamadas hemócitos, os quais medeiam importantes respostas celulares, incluindo a fagocitose, encapsulamento, nodulação, e também segregam fatores humorais, tais como opsoninas, fatores de melanização e peptídeos antimicrobianos. Os hemócitos circulam ao longo da hemocele (cavidade corporal do inseto) pelo fluxo rápido da hemolinfa (sangue), além disso, partes desses hemócitos também existem como células sésseis que estão associados aos tecidos. As larvas de Galleria mellonella são uma alternativa viável para os modelos tradicionais dos mamíferos para estudar a eficácia de drogas antimicrobianas e a patogênese de microrganismos in vivo. No entanto, apesar da sua importância como um modelo de infecção, aspectos biológicos sobre as células do sistema imunológico, tais como a densidade e dinâmica dos hemócitos das larvas são mal compreendidos. No presente trabalho, investigamos a resposta imune celular dos hemócitos circulantes das larvas de G. mellonella contra diferentes cepas da bactéria Gram-negativa Actinobacillus pleuropneumoniae: baixa virulência (780), alta virulência (1022), e cepas de referência do sorotipo 8 (R8). Os hemócitos foram classificados com base no seu tamanho, morfologia, coloração e seus papeis na resposta imune, incluindo cinco tipos: prohemócitos, plasmatócitos, granulócitos, esferulócitos, e oenocitóides. Contagem total de hemócitos, contagem diferencial de hemócitos, atividade dos fagolisossomos, resposta autofágica, viabilidade celular, e a ativação da caspase-3 (como indicador de apoptose) foram determinados em hemócitos circulantes provenientes de larvas desafiadas e controle. Demostramos pela primeira vez no modelo de G. mellonella que os plasmatócitos e granulócitos ativam suas respostas autofágicas através da formação dos autofagossomos após o contato com A. pleuropneumoniae. Além disso, nossos dados demonstram que a imunidade celular do presente modelo de infecção muda dependendo do grau de virulência das cepas bacterianas.
Insects respond to infection by mounting cellular and humoral immune reactions. The primary regulators of these immune responses are cells called hemocytes, which mediate important cellular immune responses including phagocytosis, encapsulation, nodulation and also secrete immune factors such as opsonins, melanization factors and antimicrobial peptides. Hemocytes circulate through the hemocoel (body cavity) by the swift flow of hemolymph (blood), and part of these hemocytes population are sessile and are attached to tissues. Larvae of Galleria mellonella is a widely used factitious host as a viable alternative to traditional mammalian models to study the efficacy of antimicrobial drugs and the microbial pathogenesis in vivo. However, despite their importance as an infection model, biological aspects about the immune cells, such as density and hemocyte dynamic of larvae are poorly understood. In the present study, we investigated the cellular immune response of hemocytes from G. mellonella larvae against three strains of the gram-negative bacterium Actinobacillus pleuropneumoniae: low virulent (780), high virulent (1022), and the serotype 8 reference strain (R8). Five types of larval hemocytes, prohemocytes, plasmatocytes, granulocytes, oenocytoids, and spherulocytes, were distinguished according to size, morphology, detection by molecular probes, dye-staining properties, and their role in the immune response. Total hemocyte count, differential hemocyte count, lysosome activity, autophagic response, cell viability, and caspase-3 activation were determined in circulating hemocytes of naïve and infected larvae. Granulocytes and plasmatocytes were the major hemocyte types involved in the cellular defense against A. pleuropneumoniae; these hemocytes activated phagolysosome activities associated with an autophagic response against the bacteria. Moreover, our results showed that apoptosis in circulating hemocytes after exposure to virulent bacterial strains was related to an excessive autophagic cell death response induced by stress and subsequent caspase-3 activation.
