Abstract:
The somatic embryogenic potential of leaf explants from greenhouse-grown plants of eight Coffea arabica genotypes was investigated on three different gelose-gelled culture media (Dublin, Pierson, and Yasuda). Four of these genotypes were reactive. Optimal somatic embryogenesis was obtained when the explants were taken from microcutting and cultured on gelrite-gelled Yasuda's medium. Under these culture conditions, somatic embryos and plantlets were obtained in two previously recalcitrant genotypes. The histocytological callus development was found to be identical in responsive or recalcitrant genotypes. Embryogenic cells formed at two successive points during callogenesis and their subsequent development varied according to culture conditions. Cells initiated in 10- to 15-day-old calli either degenerated or developed directly into embryos. Cells initiated in 60-day-old calli became isolated and developed into embryos or their development was arrested. Embryos obtained in these conditions were able to develop into plantlets. Key words: Coffea arabica, genotypic variability, histocytology, in vitro culture, somatic embryogenesis.

Abstract:
AbstractDegradable fiber is widely used to assist gelled acid diversion and reduce acid leak-off in the acid stimulation of carbonate hydrocarbon reservoirs in Sichuan Basin of China. The rheological properties of an acid system will affect the geometry of the acid-etching fracture. However, the effect of fiber on the rheological properties of gelled acid is not yet clear. This paper investigates the rheological properties of gelled acid with various fiber concentrations at different temperatures. The results show that when the temperature is less than the degradable temperature of the fiber, the apparent viscosity of gelled acid rises gradually with an increase in fiber concentration, while the fiber has no significant effect on the viscosity of gelled acid at the degradable temperature. The dissolution process of fiber in gelled acid experiences none-dissolution, surface dissolution, dissolution and fining, and a complete dissolution stage from low to high temperatures, which all have different effects on gelled acid viscosity. The fiber links more gelling agent molecules of gelled acid together to form a quasi-network structure between the fiber and fiber and the fiber and polymer, which results in a rise in the viscosity of gelled acid. The acid system also shows a strong shear thinning property under different temperatures and fiber concentrations. However, the power-law index n of this acid system always maintains a steady average value of about 0.181, while the change pattern of consistency index K is similar to the change in viscosity with varying fiber concentrations and temperatures. The research results are useful for acid fracturing treatment design in carbonate reservoir.

Abstract:
Tamarind gum was obtained from the seeds of Tamarindus indica. It was rich in polysaccharide (79.96%) and protein (13.46%) contents. In this work, the dynamic viscoelastic properties of tamarind gum aqueous solutions were investigated with a Haake Rheometer RS75 as a function of gum concentration and temperature. Four types of sample solution systems: a dilute solution, a concentrated solution, a weak gelled system, and a gelled like system were observed. The effect of concentration showed that the typical shape of the mechanical spectra for the dilute solution occurred for 2.30 wt% and the gelled like behaviour arose for the higher concentration (≥7.05 wt%) as measured at 25°C. While the effect of temperature on the dynamic viscoelastic properties of tamarind gum solution (6.91wt%) showed that the gum solution behaved the weak gelled system (25°C) and subsequently gelled like system (≥30°C).

Abstract:
This diploma thesis deals with hyaluronan modified by palmitoyl and its gelation. Gels were created from palmitoyl hyaluronan with molecular weight 216 kDa and degree of substitution 11 % in concentrations 15 and 20 g dm-3 in water and concentrations 10, 15, 20 g dm-3 in NaCl and TSB. Also gel from palmitoyl hyaluronan with molecular weight 35 kDa and degree of substitution 10 % in concentrations 20, 30 g dm-3 in NaCl was created. Gels were investigated concerning medical applications. Gels were rigid and had very good properties, which was confirmed by rheology. The physical properties (pH, water content) of gels and stability were investigated. On the grounds of the MTT test, three methods of cell incorporation were suggested. Gels are nontoxic, biocompatible, and biodegradable with nontoxic degradation products and that is why they are excellent aspirants for use in biomedicine.
