Abstract:

This diploma thesis deals with hyaluronan modified by palmitoyl and its gelation. Gels were created from palmitoyl hyaluronan with molecular weight 216 kDa and degree of substitution 11 % in concentrations 15 and 20 g dm-3 in water and concentrations 10, 15, 20 g dm-3 in NaCl and TSB. Also gel from palmitoyl hyaluronan with molecular weight 35 kDa and degree of substitution 10 % in concentrations 20, 30 g dm-3 in NaCl was created. Gels were investigated concerning medical applications. Gels were rigid and had very good properties, which was confirmed by rheology. The physical properties (pH, water content) of gels and stability were investigated. On the grounds of the MTT test, three methods of cell incorporation were suggested. Gels are nontoxic, biocompatible, and biodegradable with nontoxic degradation products and that is why they are excellent aspirants for use in biomedicine.

Abstract:

Předkládaná disertační práce se zabývá charakterizací, zpracováním a gelací rozpustného kolagenu. Literární rešerše shrnuje postavení kolagenu na trhu s transplantáty, jeho aplikace a jednotlivé typy kolagenu. Detailně jsou rozebrány jednotlivé metody charakterizace rozpustného kolagenu a způsoby jeho gelace. Experimentální část je rozdělena na dvě podkapitoly. První podkapitola experimentální části se zabývá rozpouštěním a gelací rozpustného kolagenu v natlakované CO2 atmosféře. Proti jiným podobným metodám, metoda pracuje na poměrně nízkém tlaku (do 0.9MPa) a je bezpečná z pohledu denaturace kolagenu neboť funguje za nízké teploty blízké 0°C. Tímto způsobem mohou být průhledné gely, obsahující velmi tenká vlákna kolagenu, vytvořeny mnohem rychleji než za použití konvenčních metod. Rychlost gelace a transparentnost dává konceptu potenciál v oblasti oftalmologie a měla by přinést podstatné výhody pro injektovatelné produkty, vytváření mikro kuliček a 3D tisk. Druhá část popisuje unikátní charakterizaci rozpustného kolagenu v nativním stavu pomocí metody průtokové frakcionace s víceúhlovým rozptylem světla (AF4-MALS). Získaná molekulová hmotnost (při nejvyšší detekované koncentraci) odpovídá předpovězené hmotnosti a nativní i denaturovaný kolagen lze snadno rozeznat v konformačním grafu. Tato nová metoda poskytuje konzistentní a stabilní výsledky ve srovnání s ostatními zavedenými metodami. Metoda může být použita pro optimalizaci kvality výtěžků během výroby rozpustného kolagenu, nebo pro citlivou detekci denaturace během zpracování kolagenu.

Abstract:

This master's thesis deals with gelation of hydroxyethylcellulose and hyaluronan crosslinked with nontoxic crosslinking agent – citric acid. First of all, the optimalization of gelation process of hydroxyethylcellulose took place. After obtaining the product insolube in the ultrapure deionized water, a reaction mixture of hyaluronan and hydroxyethylcellulose in two different weight ratios were prepared. There was prepared solution containing only hyaluronan as the polymeric part too. Citric acid was used in concentrations of 5 %, 10 % and 20 % (w/w of polymer). These solutions of polymers and citric acid were gelated under the same conditions and the products were compared to each other using Differential Scanning Calorimetry, Thermogravimetry and Fourier Transform Infrared Spectroscopy. The products containing hydroxyethylcellulose and hyaluronan with hydroxyethylcellulose in weight ratio equal to 3 : 7 were considered crosslinked.

Abstract:

The atomization of a gelled boron slurry fuel using two commercially available airblast atomizers was studied at atmospheric pressure in non-reacting flow. The atomization of water was also characterized for comparison. Each atomizer was operated at two different liquid mass flow rates and several air/ fuel ratios. Drop size distribution was measured using a Malvern 2600 HSD Laser Diffraction Particle Sizer, Drop sizes acceptable for use in ramjet combustors could be obtained for the gelled slurry fuel from both atomizers. However, this required air/fuel ratios too high for practical applications. It appears that secondary atomization methods or different types of atomizers will be required to obtain high ramjet combustion efficiencies with these fuels if they are to be used over typical ramjet operating envelopes.

Two commercially available atomizers were tested for their ability to atomize a gelled boron slurry fuel. Particle size distributions were measured in non-reacting flow using a Malvern 2600 HSD Laser Diffraction Particle Sizer. A sub-scale ramjet combustor was designed and fabricated which utilized a sudden expansion inlet dump together with inlet air swirl for flame stabilization. An airblast atomizer produced sufficiently small particles for good combustion, but at the cost of a high pressure drop across the atomizer, making it impractical for use in a slurry fueled ramjet. Sustained steady combination of the slurry fuel was not achieved using the airblast atomizer. A whistle type ultrasonic atomizer als produced sufficiently small particles and at a much lower pressure drop across the atomizer. Sustained stable combustion was achieved using the ultrasonic atomizer which yielded a combustion efficiency of 76% at 96 psia and an equivalence ratio of 0.78.

Além de uma crescente importância econômica da produço de petróleo contendo parafina dos recursos águas ultra-profundas, as preocupações coma deposição e gelificação de parafina desempenha uma funcão significativa nesta questão. óleos parafínicos gelificados frequentemente exibem comportamento reológico análoga para fuidos com tensão de cedência. Existe uma tensão crítica, abaixo do qual nenhum fluxo ocorre, mas acima doqual a viscosidade de uma sistema gelificada diminui drasticamente. Assim, para reinício de escoamento de óleo cru gelificado, uma pressão maior do que a pressão de funcionamento normal deve ser aplicada para exceder a tensão de cedência do fluido na parede, esta pressão mínima exegida deve ser determinada. Neste estudo para explorar o conceito de pressão mínima, fluidos com tensão de cedência foram investigados inteiramente. A investigação foi iniciada com o fluido de tensão de cedência simples e, em seguida, avanou com um tixotrpico usando reômetro e configuração experimental para escoamento de fluido em tubo. Em ambos, os casos a condio não deslizamento foi atingido, o comportamento de fluidos com tensão de cedência foi examinado em detalhe, os resultados de experimentos de reometria e transporte de fluido em tubos foram verificados. Com a colocação de todos juntos, a pressão mínima exigida ou em outra palavra a tensão crítica para partida de escoamento de fluidos com tensão de cedência foi descoberta.
Besides an increasing economic importance of producing wax-containing crude oil from ultra deep water resources, concerns about wax deposition and gelation play a significant role in this area. Gelled waxy crude oils often exhibit rheological behavior analogous to yield stress fluids. There is a critical or range of stresses, below which no flow occurs, but above which the viscosity of a gelled system decreases drastically. Thus, in order to restart flow of a gelled crude, a pressure larger than the typical operating pressure should be applied to exceed the yield stress of the fluid at wall, this required minimum pressure should be determined. In this study to explore the minimum pressure notion, yield stress fluids were investigated comprehensively. The investigation was initiated with simpler yield stress fluid and then advanced with thixotropic one using rheometer and designed experimental setup for fluid flow into tube. In both cases no-slip condition was achieved, the behavior of yield stress fluids was screened in detail, the outcomes of fluid transport in tube and rheometric tests were combined and cross checked. Finally with putting all together, the required minimum pressure or in other words, the critical shear stress needed to startup the flow of yield stress fluids was discovered.

Abstract:

The scope of this masters thesis is the preparation of a model mucin system and its utilization as an artificial model of the native mucus system. The creation of this model system, according to several designed methods was a part of experimental part of the thesis. The preparation of mucin system comprised of physical and chemical methods of hydrogel formation, screening and characterisation of the various physical conditions of the mucin properties on its molecular level, and the preparation of sorbent with sorption surface containing mucin. Methods of light scattering, namely dynamic light scattering (DLS), used for mucin particles size change monitoring, and electroforetic light scattering (ELS), used for Zeta potential change monitoring, were used for the screening of the impact of physical factors on the properties of mucin.For the characterisation of impact of the temperature on changes in mucin sctructure was, apart from monitoring of light scattering, used also a diferential scanning calorimetry (DSC), which registered temperature value, at which mucin thermal denaturation occurs. In the next part of the thesis we subdued the created sorption surfaces to various physical-chemical analyses, which task is the characterisation and projection of surface and confirmation of mucin presence.Substancial part in monitoring and characterisation of changes in surface sctructure of sorption surface was accomplished by Fourier transform infrared spectroscopy (FTIR). Scanning electron microscophy (SEM) was used for the final, more detailed, projection of the mucin enriched, sorbent surface structure. Suggested methods of mucin hydrogel, didnt prove sufficient results for the possibility of application of hydrogel as a artificial model of real mucus system, but the sorbent application was indicated as a suitable alternative and an instrument for the further mucin behaviour research and possibly subsequent bacterial adhesion, which represents the first step in the formation of the bacterial biofilm.

Johannes Gelbke (1846 Radeberg – 1903 Buffalo/ USA) „Unter den vielen tüchtigen Musikern, welche die Stadt Buffalo beherbergt, ist Herr Johannes Gelbke sicherlich der allertüchtigsten einer.“ So beginnt in dem Buch „Die Geschichte der Deutschen in Buffalo und Erie County, N.Y., mit Biografien und Illustrationen hervorragender Deutsch-Amerikaner, welche zur Entwickelung der Stadt Buffalo beigetragen haben“ der Beitrag über Johannes Gelbke. Geboren und aufgewachsen in Radeberg, erhielt er seine musikalische Ausbildung am „Gymnasium zum Heiligen Kreuz Dresden“ (Kreuzschule), seine ersten Kompositionen entstanden dort. Er wird Musiklehrer in Leipzig und wirkt als Sänger, Chorleiter und Gesangsvereins-Dirigent, ab 1869 auch in Wurzen. 1882 komponierte er sein sog. Heimatlied: „Horch, die alten Eichen rauschen“ (Text: Emil Schimpke). Im November 1882 wanderte Gelbke nach Buffalo / USA aus, wo er bis zu seinem Tode 1903 als Dirigent, Komponist und Chorleiter gewaltiger Chöre arbeitete. Nach Deutschland ist er nie zurückgekehrt.

Mit multiaxialen Gelegen auf Basis der Kettenwirktechnik stehen hervorragende textile Halbzeuge für die Weiterverarbeitung als Verstärkungskomponente in Faser-Kunststoff-Verbunden zur Verfügung. Die bisherige Konfiguration der für die Herstellung dieser Textilien verwendeten Nähwirkmaschinen führt verfahrensbedingt zu einem unsymmetrischen Produktaufbau mit üblicherweise nur einer Fadenlage in Gelegelängsrichtung und ebenso zu Einschränkungen bei der Anordnung des Bindefadens im Textil. Durch die Erweiterung des Nähwirkprozesses wird es möglich, Nähwirkstoffe mit einer beliebigen Abfolge der Einzellagen herzustellen, zum Beispiel in symmetrischer Anordnung. Die neuen Varianten der Lagenanordnung und der Bindungskonstruktion bilden den Ausgangspunkt für die Produktentwicklung am Beispiel zweier Anwendungen aus den Bereichen der Faser-Kunststoff-Verbunde und des textilbewehrten Betons. Hier wird deutlich, dass über die Herstellung symmetrischer Gelege hinaus der Einsatz des erweiterten Wirkprozesses die Eigenschaften der Gelege und der Endprodukte vorteilhaft beeinflussen kann. Aus den untersuchten Beispielen und grundsätzlichen Betrachtungen leitet sich ab, unter welchen maschinentechnischen Voraussetzungen der Einsatz des erweiterten Wirkprozesses sinnvoll ist. Es wird ein Konzept entwickelt, auf dessen Grundlage Nähwirkstoffe mit variabler Lagenanordnung auf Nähwirkmaschinen gefertigt werden können
Multiaxial multi-ply fabrics made by warp knitting are excellently suited for the application in fiber reinforced composites. The usual configuration of the stitch-bonding machines, which are used to produce these fabrics, necessarily leads to composite laminates with an asymmetric layer arrangement and only one layer of yarns in the zero degree direction of the fabric. The variability of patterning with the binding yarn is also limited. By completing the stitch-bonding process with an additional work step it is possible to produce stitch-bonded fabrics without any restrictions concerning the arrangement of the individual layers in the fabric, for example with a symmetric composition. This is the basis for the development of two exemplary products in the fields of textile reinforced plastics and textile reinforced concrete. It can be shown that the application of the extended stitch-bonding process is advantageous beyond the layer arrangement, positively affecting the mechanical properties of the fabric and the composite. From these examples, conclusions are drawn regarding the configuration of future stitch-bonding machines

Thesis (MA (Afrikaans and Dutch))—University of Stellenbosch, 2005.
The aim of this study is to examine the relationship between eroticism, violence and death as it occurs in Jeanne Goosen's short story collection, ' n Gelyke kans. The research is based upon the hypothesis that these stories express in a particular manner the transgressive role of eroticism in breaking through social conventions and barriers. As a possible framework for discussion reference is made in the first place to George Bataille's theories on the subversive nature of eroticism, Julia Kristeva's semiotic and pre-Oedipal theories, and Mikhail Bakhtin's notion of the carnivalesque. Subsequently a reading strategy was designed by means of an in-depth analysis of ten stories from the collection in accordance with general formalistic principles. The conclusion is reached that Goosen with non-judgmental sympathy exposes man's inborn, continuous search for the illusion of perfect happiness. This state of bliss is sometimes achieved by the violent "incorporation" of the beloved/desired “other", and the final outcome is a concomitant and inevitable decline into an unwholesome, destructive, and fatal erotic power play.

Yes
Waxy crude oils are pumped hot but upon power cut, pumping stops, the oil cools leading below the wax appearance temperature to precipitation of the wax and the formation of a gel throughout the pipe. In such a situation, what is the minimum pressure required to restart flow, not to merely deform the gel or break it? This paper provides a solution to this problem using microscopic observations under controlled cooling conditions and rheological data conducted in constant stress mode under controlled temperature and cooling conditions and restart experiments in laboratory pipelines replicating the rheometric conditions and deviations from them to inform large diameter operation in the field. Three important findings derive from the experimental data collected: (i) A fragmentation stress , rather than the static stress that precedes it, is found to be the more accurate predictor of flow re-start pressures; (ii) Waxy crude oils gels exhibit true yield stress and yielding process but also show flow on application of the slightest stress below yielding; (iii) This flow, in the elastic region, is jagged rather than continuous suggesting a consolidation process of the crystals and their agglomerates forming the gel. Abstract:

La thèse a été réalisée en cotutelle entre le Centre de biophysique moléculaire, CNRS UPR 4301, conventionné avec l'Université d'Orléans et l'Université TUM de Munich.
La gelée royale est produite dans les glandes des abeilles nourricières Apis mellifera par digestion partielle de pollen et de nectar. La gelée royale joue un rôle clé dans la reproduction, la croissance, l'immunité et l'organisation sociale des abeilles. Elle constitue également une source potentielle de nombreuses molécules à vocation pharmaceutique.
Le candidat a analysé des gelées royales d'origines botaniques et géographiques bien définies. Des fractions de peptides et de protéines ont été isolées à l'aide de multiples techniques physico-chimiques. Ces fractions se sont avérées présenter des propriétés antimicrobiennes. Le candidat a également examiné les possibilités d'application pharmaceutique des fractions de gelée royale qu'il a isolées.
