Abstract:

Die hypertrophe Kardiomyopathie ist eine autosomal dominant vererbte Herzmuskelerkrankung. Es kommt zu einer asymmetrischen Hypertrophie insbesondere des Herzmuskels. Symptome sind unspezifisch und reichen von Dyspnoe bis hin zu Synkopen. Gelegentlich ist der plötzliche Herztod die erste Manifestation der Erkrankung. Molekulargenetische Untersuchung des Genomes dieser Patienten zeigten, dass diese Patienten Mutationen im Proteinen des Sarkomers aufwiesen. Hierunter fällt das beta-MHC-Gen und alpha-Tropomysin-Gen. Die ersten identifizierten Krankheitsgene der dilatativen Kardiomyopathie (DCM) kodierten alle für Proteine des Zytoskeletts. Deshalb wurde DCM als Erkrankung des Zytoskeletts bezeichnet. Maize ß-glucosidase is implicated in phytohormone catabolism, disease resistance mechanisms, and also the catabolism of various ß-D-glucosides. The enzyme expressed in maize sporophytes is encoded by a highly polymorphic locus, Glul (chromosome 10). In the present study, maize ß-glucosidase was purified to homogeneity by using differential solubility and chromatography. The enzyme is soluble and synthesized adaptively after germination. The DNA sequence of the gene coding for the OXA-2 beta-lactamase has been completed. The primary amino acid sequence deduced from the DNA sequence was used to study homologies with other beta-lactamases; no good homologies were observed with any class of beta-lactamase. A more detailed analysis has revealed from comparison of both primary and predicted secondary structure, that the OXA-2 beta-lactamase may be more closely related to class A enzymes. The actinomycetes are gram-positive bacteria that produce more than two-thirds of the known biologically active microbial natural products, including many commercially important antibiotics, anti-cancer agents, other pharmacologically useful agents, animal health products and agrochemicals. Abstract:

The OXA-2 beta-lactamase gene was first found on a conjugative plasmid R46 from a clinical isolate of Salmonella typhimurium. To test the expression and secretion of OXA-2 beta-lactamase in Streptomyces lividans a shuttle plasmid (pSU101) was created by fusing an Escherichia coli plasmid (pSU8) carrying the OXA-2 beta-lactamase gene with the S. lividans vector pLJ61. The OXA-2 beta-lactamase gene specified by the hybrid plasmid PSU101 was expressed in S. lividans, although at a lower level than in coli. Almost all the beta-lactamase activity was found in the culture supernatant of S. lividans,
