Abstract:
Vi rút viêm gan E (HEV) là một trong những nguyên nhân phổ biến gây viêm gan virus cấp nhưng ít được chẩn đoán. HEV có kích thước nhỏ, không có vỏ ngoài, sợi đơn RNA thuộc chi Hepevirus, họ Hepeviridae và có bốn kiểu gen có thể gây nhiễm trùng cho người. Dịch tễ: nhiễm HEV phân bố trên toàn cầu, với tỉ lệ kiểu gen lưu hành khác nhau giữa các khu vực địa lý, tỉ lệ lưu hành HEV ở các nước đang phát triển (kiểu gen 1,2) cao hơn các nước phát triển (kiểu gen 3,4). HEV có thể lây qua đường tiêu hóa, hiếm hơn là qua đường máu và lây truyền từ mẹ sang con. 
 Biểu hiện lâm sàng: phần lớn viêm gan E cấp có biểu hiện nhẹ và tự giới hạn ở người có miễn dịch bình thường. Tuy nhiên, những bệnh nhân có cơ địa đặc biệt (phụ nữ có thai, có bệnh gan từ trước, suy giảm miễn dịch) có thể bị các biến chứng như suy gan cấp, viêm gan ứ mật hoặc viêm gan E mạn. 
 Chẩn đoán: chẩn đoán nhiễm HEV cấp thường dựa vào sự phát hiện kháng thể anti - HEV IgM trong huyết thanh và sự gia tăng hiệu giá kháng thể anti - HEV IgG (tăng hơn 5 lần sau hơn hai tuần) hoặc phát hiện HEV - RNA trong huyết thanh hoặc trong phân. Nhiễm HEV mạn là sự phát hiện HEV - RNA trong huyết thanh hoặc phân trong thời gian trên 6 tháng. 
 Điều trị: điều trị nhiễm HEV cấp chủ yếu là điều trị hỗ trợ. Điều trị viêm gan E mạn bao gồm giảm liều liệu pháp ức chế miễn dịch và/hoặc điều trị bằng thuốc kháng virus. 
 Dự phòng: sử dụng nước sạch, tránh ăn thịt động vật chưa được nấu chín. Vắc xin phòng HEV đã được phát triển và cấp phép ở Trung Quốc, đã cho thấy hiệu quả cao trong việc dự phòng nhiễm HEV.

Abstract:
Chi lan hài (Paphiopedilum) là chi lan đặc biệt được yêu thích bởi màu sắc đa dạng và cấu trúc hoa độc đáo. Để bảo vệ những loài lan này, việc phát triển các phương pháp nhận diện, phân biệt chúng là vô cùng cần thiết, đặc biệt Paphiopedilum là chi mang nhiều loài có độ tương đồng cao về hình thái thân lá. Bài báo trình bày kết quả phân tích chi tiết hình thái lan hài Đuôi công (P. gratrixianum) có nguồn gốc tại Lào Cai, kết hợp với việc xác định trình tự gen trnH-psbA để nhận diện loài này. Kết quả giải trình tự gen trnH-psbA phân lập từ mẫu P. gratrixianum nghiên cứu đã thu được đoạn gen có kích thước 691 bp. Khi so sánh với các trình tự trên ngân hàng gen NCBI cho thấy trình tự đoạn gen trnH-psbA của P. gratrixianum nghiên cứu có độ tương đồng lên đến 99,32% với gen trnH-psbA của P. gratrixianum phân lập tại Trung Quốc (mã số MV284890.1). Trên sơ đồ phân loại hình cây thiết lập dựa vào trình tự gen trnH-psbA, P. gratrixianum nghiên cứu có quan hệ họ hàng gần gũi với hài Trần liên của Việt Nam (P. tranlienianu - MW794124.1), P. spicerianum (loài đặc hữu của Ấn Độ - NC_502702.1) và P. barbigerum (loài hài được tìm thấy lần đầu ở Trung Quốc - MN153814.1, NC_050870.1).

Abstract:
Loài Sum lông (Adinandra glischroloma Hand.-Mazz. var. hirta (Gagnep.) Kobuski) thuộc chi Dương đồng thu tại Lào Cai, Việt Nam được sử dụng làm vật liệu nghiên cứu. Trong nghiên cứu trước, thành phần hóa học, hoạt tính sinh học của cao chiết từ thân của loài Sum lông đã được chứng minh chứa nhóm chất polyphenol, flavonoid, coumarin và không có khả năng ức chế vi khuẩn kiểm định. Tuy nhiên, chúng lại có hoạt tính chống oxi hóa và ức chế các dòng tế bào ung thư vú, phổi và dạ dày. Trong nghiên cứu này, đặc điểm hình thái và giải phẫu hiển vi lá, thân của loài Sum lông đã được mô tả; đồng thời vùng gen rbcL đã được phân lập, giải trình tự và phân tích sự tiến hóa phân tử. Kết quả phân lập và giải trình tự cho thấy, gen rbcL của loài Sum lông có kích thước 602 bp. Cây phát sinh gen được xây dựng dựa trên 14 trình tự gen rbcL cho thấy các loài phân bố thành 3 nhóm và loài Sum lông phân bố cùng nhóm với các loài thuộc chi Adinandra. Trình tự gen rbcL có thể là ứng cử viên mã vạch DNA lục lạp tiềm năng để nhận biết các loài thuộc chi Dương đồng.

Abstract:
Gen Z has reached the age of 18-24 and will exert an essential impact on the social, economic and political processes in information/digital society. This society attaches great significance to such qualities as Sustainability-Oriented Creativity, Innovation, and Entrepreneurship (CIE), which expand opportunities for networking and creation of communications, give a competitive advantage and contribute to the development of an individual and the society. The purpose of the study is to reveal the possession of generation Z representatives of creative, innovative and entrepreneurial potential, and the presence of a propensity for creative labor. Using the information and communication method we have carried out an analysis based on literature data and our own research such as a Gen Z questionnaire survey and interview data from representatives of the older generation who communicate with young people (Gen Y&X). The completed research shows an aspiration of most of the Centennials for creative labor, proficient use of digital technologies and their desire to work in modern industries, in particular, IT-sphere. The authors of the paper concluded that the formation of the future entrepreneurship activities would be impacted by CIE factors based on horizontal ties and network interaction. It predicts the decreasing role of bureaucracy and the transition of management from organization towards coordination and facilitation of work for professionals, which allows us to assume the possibility of an optimistic forecast for the social development in the medium term. The authors believe that predictions of replacing humans with Artificial Intelligence (AI) in the industrial sector are unlikely. The high potential of the CIE will allow Generation Z to work successfully using all the achievements of digital technologies for the development of the society. This is changing the common perception of the future as a world dominated by robots, god-like elites and marginalized masses.

Abstract:
The study sought to examine the adoption of digital banking among Gen Z students in Coimbatore City. A survey was administered among 75 Gen Z students who actively used digital banking. The collected data was analyzed using SPSS, employing both the Chi-Square test and T-Test. The Chi-Square test results showed no significant relationship between gender and the type of digital wallet used, nor between age and the wallet used for digital banking. Additionally, the T-Test revealed no significant differences in digital banking usage among Gen Z students based on age. Key words: Digital banking, Gen Z, Mobile banking, Online banking

Tujuan dari penelitian ini adalah untuk mengetahui hubungan SNP (rs867500) pada gen STX1A Exon 10 dengan tingkat kecerdasan intelektual. Penelitian ini merupakan penelitian observasional analitik dengan desain penelitian cross sectional dengan populasi sebesar 368 dan sampel sebesar 36 yang dibagi menjadi dua kelompok, yaitu kelompok IQ diatas rata-rata dan kelompok IQ rata-rata. Untuk mengetahui hubungan SNP (rs867500) gen STX1A Exon 10 dengan tingkat kecerdasan digunakan uji X2 (Chi-square). Hasil menunjukkan tidak ditemukan SNPs pada gen STX1A exon 10. Gen STX1A dapat dikatakan berkontribusi pada kecerdasan bersama gen lainnya, yaitu Gen IGFR2 dan Gen FNBPL1 meskipun tidak terdapat polimorfisme pada gen STX1A exon 10.

In order to avoid the real economy development lags behind and the deterioration of ecological problems in the process of traditional urbanization, China has been promoting city-industry deep integration, but the connection between city-industry integration and green economic growth, especially the spatial effect, has not been systematically explained. Based on the panel data from 2007 to 2018, this paper constructs an evaluation index system and uses the SEEA method to measure city-industry integration (CII) level and green economy growth (GEG) level. Then, by employing spatial Durbin model and intermediary effect model, it further systematically investigates the spatial impact of CII on GEG and the potential mechanism. The study found that: (1) On the whole, CII shows “slow-steady integration” trend, but regional heterogeneity was obvious and accompanied by “slow gap expansion”. GEG experienced “sharp increase” with “polarization” characteristic. (2) CII can directly promote regional GEG (with a marginal effect of 0.689), more effectively than traditional urbanization, and CII has obvious spatial spillover effects on GEG in both “local effect” and “neighboring effect”. (3) Interestingly, technological innovation and consumption structure upgrading are significant mediating mechanisms. (4) The direct positive effect of CII shows the regional imbalance characteristic. Finally, the corresponding policy implications are put forward.

Abstract:
Neonatal exposure of CD-1 mice to diethylstilbestrol (DES) or genistein (GEN) induces uterine adenocarcinoma in aging animals. Uterine carcinogenesis in this model is ovarian dependent because its evolution is blocked by prepubertal ovariectomy. This study seeks to discover novel uterine genes whose expression is altered by such early endocrine disruption via an epigenetic mechanism. Neonatal mice were treated with 1 or 1000 μg/kg DES, 50 mg/kg GEN, or oil (control) on d 1–5. One group of treated mice was killed before puberty on d 19. Others were ovariectomized or left intact, and killed at 6 and 18 months of age. Methylation-sensitive restriction fingerprinting was performed to identify differentially methylated sequences associated with neonatal exposure to DES/GEN. Among 14 candidates, nucleosomal binding protein 1 (Nsbp1), the gene for a nucleosome-core-particle binding protein, was selected for further study because of its central role in chromatin remodeling. In uteri of immature control mice, Nsbp1 promoter CpG island (CGI) was minimally methylated. Once control mice reached puberty, the Nsbp1 CGI became hypermethylated, and gene expression declined further. In contrast, in neonatal DES/GEN-treated mice, the Nsbp1 CGI stayed anomalously hypomethylated, and the gene exhibited persistent overexpression throughout life. However, if neonatal DES/GEN-treated mice were ovariectomized before puberty, the CGI remained minimally to moderately methylated, and gene expression was subdued except in the group treated with 1000 μg/kg DES. Thus, the life reprogramming of uterine Nsbp1 expression by neonatal DES/GEN exposure appears to be mediated by an epigenetic mechanism that interacts with ovarian hormones in adulthood.

With the expansion of Internet and digital media, especially for the social networking sites, there are extreme changes of global hospitality and tourism industry. The current study aims to investigate the intention to use Facebook - an online social network (OSN) for travel planning by examining the effect of Perceived Usefulness, Perceived Ease Of Use and Enjoyment. Secondary data are from the journals, books and other sources of information from websites and published reports of businesses and primary data are from the consultations with 5 experts, 2 group discussions and 4 in-depths structured interviews and survey by questionnaires with 200 Gen-Z users. Findings explain the influence of these perceived values on Gen-Z users’ intention to adopt Facebook as a means of travel planning. The study has demonstrated the TAM model’s applicability to the setting of the usage of Facebook for travel planning in Gen-Z. Furthermore, sampling and study scope limitation can lead to further researches.

A new genus of the millipede family Pseudospirobolellidae, Siliquobolellus gen. nov., is described from limestone mountains in Thailand, based on three new species, viz. Siliquobolellus amicusdraconis gen. et sp. nov. from Uthaithani Province, Siliquobolellus constrictus gen. et sp. nov. from Prachuap Khiri Khan Province and Siliquobolellus prasankokae gen. et sp. nov. (type species) from Lampang Province. The descriptions are based on gonopod morphology and mitochondrial DNA data (COI barcodes). The COI barcodes grouped the three new species in a well-supported Siliquobolellus gen. nov. clade. The mean interspecific COI sequence divergence among the three new species was 12% (range: 8–15%). The mean intergeneric COI sequence divergence between Siliquobolellus gen. nov., Coxobolellus Pimvichai, Enghoff, Panha & Backeljau, 2020, and Pseudospirobolellus Carl, 1912 was 19% (range: 14–23%). Three conspicuous gonopodal synapomorphies differentiate Siliquobolellus gen. nov. from other pseudospirobollellid genera: (1) the telopodital part of the posterior gonopod forms a deep concavity, (2) the telopodite of the anterior gonopod is directed distad and does not reach the tip of the coxal part of the anterior gonopod, and (3) the tip of the anterior gonopod coxa is narrowed, curving mesad. As such, the monophyly of the new genus is well supported by both morphological and mitochondrial DNA data. A distribution map and an identification key to the species are provided.

Tesis de grado de maestría del programa de Maestría en Ciencias Agropecuarias y Recursos Naturales de la UAEM, dentro de la linea de investigación de biotecnología.<br>Durante los últimos años el cáncer y sus diversos tipos han surgido como una de las principales causas de mortalidad en el mundo, de acuerdo con las recientes estadísticas del Global Burden of Disease Cancer Collaboration, en 2017, hubo 24.5 millones de incidencias de cáncer en todo el mundo, de las cuales 9.6 millones se traducen en muertes causadas por el mismo (Fitzmaurice et al., 2019); Aunado a esto, hay evidencia de que en México durante el periodo de 2011-2016, aproximadamente 50% de las muertes observadas fueron por tumores malignos en la población de 0-17 años, donde se tiene como segundo lugar de importancia el cáncer de encéfalo y otras partes del sistema nervioso central, entre las cinco principales causas de mortalidad por neoplasias malignas. Asimismo, en este sector de la población, son más comunes los tumores cerebrales que no se extienden fuera del cerebro o a la médula espinal, dentro de los que se destacan está el neuroblastoma, el tumor sólido extracraneal más frecuente durante la infancia (Instituto Nacional de Estadística y Geografía [INEGI], 2018; Szewczyk, 2018); Por otro lado, con lo que respecta al tratamiento del neuroblastoma, el uso de métodos convencionales para erradicarlo, basados en quimioterapia, radioterapia y cirugía, han mostrado efectos secundarios que influyen sobre la calidad de vida del paciente durante el tratamiento y una vez finalizados causando daños irreversibles (Secretaría de Salud., 2010); debido a esto es necesaria la búsqueda de alternativas que pueden ser de origen natural y a su vez tener mayor éxito para erradicarlo. Con la finalidad de mejorar lo anterior, se ha iniciado el aprovechamiento de recursos naturales que puedan atacar a las células cancerosas con éxito, ya que como lo menciona Khusroa et al., (2018) dentro de los agentes anticancerígenos descritos en las terapias dirigidas, los péptidos de los venenos animales como el alacrán. han creado un enorme interés para usarse como modelos para diseñar nuevos fármacos anti-cáncer eficaces, ya que se ha demostrado que el veneno de alacrán de algunas especies pueden inhibir el crecimiento de varios tipos de líneas celulares cancerosas, y sólo unas pocas toxinas son responsables de estos efectos anticancerígenos ejerciendo su acción por tres diferentes mecanismos: 1) el bloqueo de un canal iónico específico (Jäger et al., 2004); 2) inhibir la invasión de células de cáncer de unión a un sitio específico (diferente de un canal de iones) en la membrana plasmática (Deshane et al., 2003); 3) la activación de vías intracelulares que inducen apoptosis (Gupta et al., 2010). De acuerdo a lo anterior existen reportes donde el veneno de alacrán es empleado para inducir muerte en células cancerosas in vitro e in vivo, por lo tanto, la opción que se propuso en esta investigación fue el uso de la fracción CII-6, una toxina que constituye el veneno de alacrán, de la especie Centruroides limpidus, dicha fracción, es una neurotoxina que inhibe la sinapsis neuronal mediante el bloqueo de canales iónicos, específicamente los de sodio; Anteriormente, la fracción CII-6 ha demostrado la capacidad de inducir muerte celular en líneas celulares cancerosas, dentro de las cuales se incluyen adenocarcinoma de cérvix uterino (HeLa) y carcinoma hepatocelular (HuH-7), (Rosas-Aguilar, 2016; Desales-Salazar, 2016); Por lo tanto, considerando que el neuroblastoma es un tumor que se origina a partir de células derivadas de la cresta neural, que son precursoras del sistema nervioso simpático y que a su vez presenta una sobreexpresión de los canales de sodio (Hernández-Plata, 2012), se sugiere que el efecto citotóxico de la fracción CII-6 sobre la viabilidad de estas células sea mayor a lo que se ha reportado anteriormente. Para su evaluación, se empleó el plásmido pDream2.1/MCS/CII-6, en el que fue ensamblada la secuencia del gen CII-6 (Contreras-Ortiz et al., 2016), cuya secuencia genética de 316 pares de bases esta reportada en la base de datos GenBank con número de acceso AF491132 (National Center for Biotechnology Information [NCBI], 2018); El plásmido fue transfectado en la línea celular de Fibroblastos Embrionarios de Ratón (NIH/3T3), células que fungieron como 10 productoras de la fracción CII-6; una vez identificada la transcripción de la fracción CII-6 en estas, el medio de las células productoras denominado como “condicionado”, fue recolectado, filtrado y aplicado como tratamiento a la línea celular de Neuroblastoma Humano (SH-SY5Y) con la finalidad de evaluar el efecto de la fracción CII-6 sobre la viabilidad celular. Por otro lado, también se analizaron las ventajas, desventajas y sus posibles mejoras a largo plazo. El gen CII-6, se transcribe de forma correcta al ser transfectado en la línea celular NIH/3T3 de fibroblastos embrionarios de ratón y mediante ensayos de viabilidad se determinó que no ejerce un efecto sobre la viabilidad de células no cancerosas. En las líneas celulares de glioblastoma (U-87) y neuroblastoma (SH-SY5Y) humano la subfracción CII-6 induce un efecto de reducción de viabilidad. El tratamiento mostro una mayor eficiencia cuando se administró en una concentración del 25% con un periodo de incubación de 24 horas en la línea celular SH-SY5Y. Por otro lado, en la línea celular U-87 se observó un mayor efecto inducido por la subfracción CII-6 en comparación con el observado en la línea celular SH-SY5Y en 72 horas de incubación, administrando a una concentración del 100% la subfracción CII-6.<br>SIAE-UAEM Registro 6224/2020CIB

The specific aim was to evaluate if the evolution in the hierarchy of Generic Change Indicators is associated with the quality of adaptive responses. Methodologically, the research was based on an intensive case study design. The List presents an ideal of what happens when psychotherapy is successful. The List is composed by 19 indicators disposed hierarchically, which relate primarily to how the patient sees his or her problem, his or her willingness to face it and his or her expectations for psychotherapy and psychotherapist. Adaptive efficacy comprises the responses of the individual in the face of difficulties and vicissitudes of life. The more adequate a set of responses is, the more the adaptation of the subject will be considered effective. The videos and the transcriptions of the sessions of a brief psychodynamic psychotherapy process were evaluated with the GCIL and with the Scale of Adaptation (EDAO-R). According to the GCIL 14 episodes of change were identified into the 11 sessions of the psychotherapy. Results pointed to evolution in the hierarchy of indicators of change. The initial sessions comprised indicators of lower hierarchy and final sessions encompassed indicators of higher hierarchy. The results also showed that there was an evolution in the patient s adaptive efficacy, from very slightly adequate to adequate in the A-R area and from very slightly adequate to slightly adequate in the Pr area. We suggest that the progress in the hierarchy of the indicators showed that the patient began to offer more adequate responses, which resulted mainly in changes in the A-R area. It was possible to conclude in this study the association between the two measures. Some common factors to different therapeutic approaches probably contributed to the changes that the patient achieved and these are relate to the patient (she actively participated in the process, was aware of her difficulties and showed motivation to work on the problems), to the therapist (interventions used during the sessions) and to the relationship between therapist and patient (therapeutic alliance). Further researches must be pursued, involving a larger number of psychotherapies, including unsuccessful and dropout processes.<br>A presente pesquisa, de natureza explorat?ria, buscou combinar o crit?rio da efic?cia adaptativa com os da Lista de Indicadores Gen?ricos de Mudan?a (LIGM), para se compreender melhor como as pessoas mudam em psicoterapia. O objetivo mais espec?fico foi avaliar se existe rela??o entre a evolu??o na hierarquia dos Indicadores Gen?ricos de Mudan?a e a qualidade das respostas adaptativas. Utilizou-se o delineamento de estudo de caso intensivo. A LIGM refere-se a uma sequ?ncia ideal do que se espera que ocorra, se a psicoterapia obtiver ?xito. Ela ? composta por 19 indicadores dispostos de modo hier?rquico, que se referem prioritariamente ? forma como o paciente encara o seu problema, sua disposi??o para enfrent?-lo e as expectativas em rela??o ? psicoterapia e ao psicoterapeuta. A efic?cia adaptativa engloba as respostas dadas pelo indiv?duo frente ?s dificuldades e vicissitudes da vida. Quanto mais um conjunto de respostas ? adequado, mais eficaz ser? sua adapta??o. Para tanto, os v?deos e as transcri??es das sess?es de um processo de psicoterapia breve psicodin?mica de paciente adulta que foi atendida por psicoterapeuta experiente foram avaliadas com a LIGM e com a Escala Diagn?stica Adaptativa Operacionalizada Redefinida (EDAO-R). De acordo com a LIGM foram encontrados 14 epis?dios de mudan?a, distribu?dos em 11 sess?es. Observou-se evolu??o dos indicadores de mudan?a, em que as sess?es iniciais englobaram indicadores de menor hierarquia e as sess?es finais abarcaram indicadores de maior hierarquia. Os resultados tamb?m mostraram que houve evolu??o na efic?cia adaptativa da paciente, de pouqu?ssimo adequada para adequada, no setor Afetivo-Relacional (A-R, que diz respeito aos relacionamentos intra e interpessoais) e de pouqu?ssimo adequada para pouco adequada no setor Produtividade (Pr, que envolve trabalho ou principal ocupa??o que a pessoa exerce). Sugere-se que o progresso na hierarquia dos indicadores demonstrou que a paciente passou a oferecer respostas mais adequadas, que ocasionaram principalmente em mudan?as no setor A-R. Alguns fatores provavelmente contribu?ram para as mudan?as alcan?adas e estes dizem respeito ? paciente (participou ativamente do processo, tinha consci?ncia de suas dificuldades e apresentou motiva??o para trabalhar nos problemas), ? terapeuta (interven??es utilizadas durante as sess?es) e ? rela??o estabelecida entre ambas (alian?a terap?utica). Foi poss?vel concluir que neste estudo houve associa??o entre as duas medidas, que podem ser usadas para auxiliar o terapeuta na avalia??o de progresso do paciente e na compreens?o das vari?veis que podem facilitar ou limitar a mudan?a do paciente. Sugere-se a realiza??o de novas pesquisas, que compreendam maior n?mero de casos e que contem com processos considerados mal sucedidos e em que houve abandono por parte do paciente.

Durant el desenvolupament dels organismes multicel·lulars els fenòmens de transducció de senyals estan íntimament lligats als mecanismes de comunicació cèl·lula-cèl·lula. En el marc d'un teixit epitelial, la formació i manteniment de l'arquitectura cel·lular i dels contactes que estableixen les cèl·lules són processos altament regulats. Una de les vies de senyalització més coneguda, i alhora estudiada, és la via de Ras/Raf/MAPK. L'activació d'aquesta via es dóna per lligands externs a la cèl·lula i s'interpreta per factors de membrana i citosòlics que transduiran el senyal intracel·lular cap al nucli. Aquesta via està altament conservada en tots els organismes eucariotes i s'usa reiteradament durant el desenvolupament en processos tan importants com la diferenciació cel·lular, la proliferació, l'apoptosi o la supervivència cel·lular. Per això, és indispensable entendre com es modula la via de Ras/Raf/MAPK, ja que segons la durada, magnitud i compartimentalització d'aquest senyal es pot donar una resposta cel·lular específica.<br/> Per tal d'identificar nous factors implicats en la regulació de la via Ras/Raf/MAPK es va dissenyar un screening genètic a Drosophila utilitzant com a model la determinació del fotoreceptor R7 durant el desenvolupament de l'ull. Per l'especificació d'aquest fotoreceptor es requereix de l'activació de dos receptors tirosina quinasa, Sevenless (Sev) i Epidermal growth factor receptor (Egfr). Una activació constitutiva del receptor Sev es manifesta en un increment en el nombre de fotoreceptors R7 en l'omatidi, i l'ull esdevé rugós. L'screening genètic per guany de funció en aquest fons sensibilitzat es basava en la cerca de factors que suprimissin aquest fenotip d'ull rugós i per extensió, factors que serien reguladors negatius de la via. D'entre els resultats que es van obtenir es va identificar un gen, center divider (cdi), la funció del qual està vinculada a la reorganització del citoesquelet d'actina cortical.<br/> Cdi és una serina/treonina quinasa encarregada de fosforilar per inactivar el factor despolimeritzador dels filaments d'actina, la ADF/Cofilina. En la present Tesi Doctoral s'ha estudiat el patró d'expressió i la funció del gen cdi en el desenvolupament de l'ull de Drosophila. Un guany de funció d'aquest gen és capaç de promoure un acúmul d'actina polimeritzada en el grup de cèl·lules de l'omatidi. També s'ha demostrat que tant el guany com la pèrdua de funció de cdi alteren la localització en l'eix apico-basal de factors integrals de les unions adherents i dels receptors i lligands propis de la via de Sev. A banda d'altres estudis fets en Drosophila per caracteritzar la funció de cdi i entendre el mecanisme d'acció en la regulació de la via de Sev, s'ha emprat el model de cèl·lules en cultiu de mamífers per analitzar el paper que té TESK1, l'homòleg de cdi en mamífers, en la regulació de la via Ras/Raf/MAPK. Localized actin polymerization is required to constrict the apical surface of epithelial cells of the eye imaginal disc to maintain the refined arrangement of the developing ommatidia. The identity of each photoreceptor cell within the epithelium is determined by cell-to-cell contacts involving signal transduction events. The R7 photoreceptor cell requires the activity of the Sevenless RTK to adopt a proper cell fate. We performed an EP screen for negative regulators of this inductive process, and we identified the serine/threonine kinase Center divider (cdi) as a suppressor of the phenotype caused by an activated Sevenless receptor. Cdi is homologous to the human testis-specific kinase 1 (TESK1), a member of the LIM kinases involved in cytoskeleton control through ADF/cofilin phosphorylation. We have analyzed the effects of gain- and loss-of-function of cdi and found alterations in actin organization and in the adherens junctions proteins DE-cadherin and &#946;-catenin, as well as in Sevenless apical localization. Interference with the function of the ADF/cofilin phosphatase Slingshot (ssh), which antagonizes Cdi, also results in a suppression of signaling triggered by the Sevenless RTK. The results of this first chapter of the Thesis reveal a critical interplay between the localization of molecules involved in epithelial organization and signal transduction.<br/>In the second chapter of this Thesis we describe the role of the mammalian homolog of cdi, testis-specific kinase 1 (TESK1), in PC12 neuronal cell differentiation and fibroblast proliferation. In addition, we show data indicating that TESK1 interferes with both the magnitude and duration of the ERK activation profile. </I>

A fotoisomerização transcis das moléculas azoaromáticas ocorre quando estas moléculas fotocromáticas são expostas à irradiação de luz com comprimento de onda apropriado. A incorporação desses azocompostos em matrizes poliméricas tem originado materiais com potenciais aplicações em diversos campos como óptica não linear e sistemas de armazenamento óptico. Esse trabalho é dedicado à preparação e caracterização óptica de filmes de ORMOSIL (silicatos organicamente modificados) dopados com os azocorantes Methyl Red (MR) e Methyl Orange (MO). Estes materiais híbridos combinam as vantagens do processo sol-gel, com características específicas de polímeros orgânicos, permitindo o processamento de filmes finos, sem fraturas e fissuras. As amostras foram preparadas via processo sol-gel, a partir do alcóxido 3-glicidoxipropiltrimetoxissilano (GPTMS) dopado com MR e MO. A matriz foi depositada em forma de filmes finos sobre lâminas de vidro pela técnica de revestimento por imersão (dip coating). Para as caracterizações ópticas dos filmes foram realizadas medidas de espectroscopia de absorção UV/Vis, que permitiu a identificação das bandas de absorção e sua variação quando expostas a um feixe de luz. Os filmes mostraram mudanças reversíveis no seu espectro de UV/Vis de absorção relacionada com a fotoisomerização de trans-cis induzida pela luz quando expostos a um LED branco de alta intensidade. O efeito fotocrômico apresentado pelas amostras foi analisado em diferentes irradiâncias do feixe de excitação, onde foi possível a determinação das constantes de tempo para cada irradiância. Também foi analisado o fotocromismo na presença de bombeio óptico em diferentes temperaturas, o que permitiu a determinação da energia de ativação do processo de fotoisomerização cis-trans(em ausência de luz) (91,4 kJ/mol para a amostra dopada com MR e 82,6 kJ/mol para a amostra dopada com MO).<br>The reversible photoinduced trans  cis isomerization of azo dye-doped compounds has been extensively studied in the last decades. The trans  cis photoisomerization of azodye molecules occurs when these photochromic molecules are photo-selected by linearly polarized light of appropriate wavelength. The incorporation of these azo compounds in polymeric matrices has given rise to materials with potential applications in various fields as nonlinear optics and optical storage systems. This work is dedicated to the preparation and optical characterization of ORMOSIL (Organic Modified Silicates) films doped with Methyl Red (MR) and Methyl Orange (MO) azo dyes. These hybrids materials combine the advantages of sol-gel process with specifics characteristics of organic polymers, allowing the preparation of thin films without fractures or cracks. ORMOSIL samples were prepared by sol-gel process derived from 3-Glycidoxypropyltrimethoxysilane (GPTS) alkoxide doped with MR and MO. MR and MO-doped GPTS-derived thin films were deposited onto glass slides by dip-coating technique. In order to characterize optically, UV/Vis absorption spectroscopy measurements were performed, which allowed the identification of the absorption bands, and its variation when exposed to a light beam. Films samples showed reversible changes in its UV-Vis absorption spectrum related to the trans-cis photoisomerization induced by light when exposed to a cold white LED. The photochromic effect of the samples was analyzed at different light intensities of the excitation beam, where it was possible to determine the time constants for each irradiance. It was also analyzed in the presence of optical pumping at different temperatures, allowing the determination of the activation energy of the cis-trans photoisomerization process (in the dark) (91.4 kJ/mol for the MR-doped and 82.6 kJ/mol for the MO-doped GPTMS-derived thin films).

AbstractMutations affecting constitutive splice donor sites (5′ss) are among the most frequent genetic defects that disrupt the normal splicing process. Pre-mRNAsplicing requires the correct identification of a number of cis-acting elements in an ordered fashion. By disrupting the complementarity of the 5′ss with the endogenous small nuclear RNA U1 (U1 snRNA), the key component of the spliceosomal U1 ribonucleoprotein, 5′ss mutations may result in exon skipping, intron retention or activation of cryptic splice sites. Engineered modification of the U1 snRNA seemed to be a logical method to overcome the effect of those mutations. In fact, over the last years, a number of in vitro studies on the use of those modified U1 snRNAs to correct a variety of splicing defects have demonstrated the feasibility of this approach. Furthermore, recent reports on its applicability in vivo are adding up to the principle that engineered modification of U1 snRNAs represents a valuable approach and prompting further studies to demonstrate the clinical translatability of this strategy.Here, we outline the design and generation of U1 snRNAs with different degrees of complementarity to mutated 5′ss. Using the HGSNAT gene as an example, we describe the methods for a proper evaluation of their efficacyin vitro, taking advantage of our experience to share a number of tips on how to design U1 snRNA molecules for splicing rescue.

Abstract C2 toxin is binary in structure and consists of a binding component (C211, 100 kDa) and an enzyme component (C2I, 50 kDa) (Aktories et al. 1992). Both components are separated proteins. The gene of the enzyme compo¬ nent has been cloned recently (Fujii et al. 1996; EMBL Data Library D63903). C2II has to be activated by trypsin treatment to release an 75 kDa active fragment. C2II binds to a membrane receptor (not identified), which is present on all cell types studied so far. Binding of C2II induces a binding site for C21. The toxin is taken-up by receptor-mediated endocytosis (Simpson 1989) (see Fig. 1). In artificial membranes, C2II induces cation selective and voltage-dependent channels (Schmid et al. 1994).

DNA molekülünde taşınan bilginin protein moleküllerine çevrilmesine gen ifadesi (ekspresyonu) denir. Gen ifadesi hem prokaryotik hem de ökaryotik organizmalarda değişen çevre koşullarına yanıt olarak düzenlenmektedir. Gen ifadesinin düzenlenmesi, her bir hücrenin işlevsel bütünlüğünü sürdürebilmesi için bir zorunluluktur. Bir hücre tipik olarak genlerinin yalnızca bir kısmını ifade etmektedir. Hücrede tüm genler her zaman ifade edilmez. Bazı genler sürekli ve düzenli olarak ifade edilirken (housekeeping genler), bazı RNA ve proteinler, sadece çalıştıkları özelleşmiş hücrelerde yüksek miktarda eksprese edilir. Gen ifadesinin kontrol mekanizması oldukça karmaşıktır. Gen ifadesi kontrol edilerek genin ne zaman, nerede ve ne kadar ürün oluşturacağı belirlenmektedir. Gen ifadesi, DNA'dan RNA'ya ve proteine giden yoldaki birçok adımda kontrol edilmektedir. Bu adımlar, transkripsiyonal kontrol, RNA’nın işlenme kontrolü, RNA taşınımı ve lokalizasyonunun kontrolü, translasyonel kontrol, mRNA'nın yıkım kontrolü, protein yıkımının kontrolü ve protein aktivitesinin kontrolüdür. Ökaryot genlerin ifadesinin kontrolünde, RNA transkripsiyonunun başlangıç aşaması en önemli kontrol noktasıdır. Genlerin başlangıç dizisinin upstream bölgesinde bulunan çok sayıda dizi gen ekspresyonundan ve transkripsiyonun modülasyonundan sorumludur. Bu diziler promotör diziler ve düzenleyici (regülatör) elemanlardan oluşmaktadır. Genlerin ifadesi, transkripsiyon faktörleri (düzenleyicileri) olarak bilinen bir grup proteinin, DNA üzerindeki özgül dizileri (cis-regülatör diziler) tanıyıp bağlanması ile kontrol edilmektedir. Transkripsiyon faktörleri, genom boyunca dağılmış olan promotör ya da güçlendirici (enhancer) gibi cis-regülatör dizilere bağlanmaktadır. Bu bağlanma, hangi genlerin hangi hızda transkribe olacağını belirleyen bir dizi reaksiyonu harekete geçirmektedir. Her genin transkripsiyonu, kendi cis-düzenleyici dizisi tarafından kontrol edilmektedir. Son yıllarda, kodlayıcı olmayan RNA'ların gen ekspresyonunu düzenlemede yaygın rolleri bulunduğu gösterilmiştir. Ökaryotlarda önemli bir translasyon kontrolü türü, mRNA'lara bağlanan ve protein çıktısını azaltan küçük RNA'larla gerçekleşmektedir.

PCR-RFLP of three mtDNA regions ( srRNA gene ,irRNA gene and intergenic COI – COII region) were used to investigated the distribution of northern and southern Apis cerana populations of 89 colonies from Prachuap Khiri Khan and Chumphon provinces. Three, four and eight haplotypes were obtained from DraI digestion of PCR-amplified 410 bp srRNA gene, 755bp IrRNA gene and 1710 bp intergenic COI-COII region,respectively. These three mtDNA regions generated 11 composit haplotypes. Twelve composite haplotypes were generated when samples from Yunnan and Hanoi were included. A UPGMA phenogram based on genetic distance allocated A.cerana in these province into 2 distinct group: northern and southern. Their distribution areas had overlapped in Amphur Bang Sapan (Prachuap Khiri Khan ). Bang Sapan Noi (Prachuap Khiri Khan). Tha Sae (Chumphon) and Pa Thiu (Chumphon). Only one type of intermediate haplotype. BAB was found in the contact zone with low frequency indicated that northern and southern populations of bees in Thailand were colonized by separate population. The northern population of A.cerana in Thailand might be colonized by bee from Vietnam. Notably,private haplotype of all amplified regions were found from one sample in Prachuap Khiri Khan. The composite haplotype,CED,was extremely different from all samples having morphological similarity. It was suspected to be the other species. Further study is needed to be carried out to clarify their actual taxonomic status. Microsatellite DNA analysis of 4 geographic samples (1. Central, 2. South, 3. Prachuap Khiri Khan and 4.Chumphon ) was performed by using A.mellifera microsatellite primer. Three microsatellite loci (A28, A107, and A113) showed polymorphic. PCR products of loci A28 and A107 were very difficult to accurately score because of their stutter bands nature. The heterozygosities of A.cerana were estimated from microsatellite loci A113 was 0.451 - 0.550. The analysis of geographic differentiation indicated no differentiation of four geographics of A.cerana . Therefore, crossed mating of male bees in the contact zone (Prachuap Khiri Khan and Chumphon) might occurred.

การผสมข้ามสายพันธุ์ (hybridization) เป็นการผสมพันธุ์ระหว่างสิ่งมีชีวิตที่มีความใกล้ชิดกันเชิงวิวัฒนาการที่มีโครงสร้างทางพันธุกรรมของประชากรหรือสปีชีส์ที่แตกต่างกัน ทั้งนี้ลูกผสม (hybrids) ที่เกิดขึ้นอาจทำให้เกิดการถ่ายเทเคลื่อนย้ายของยีน (gene flow) ระหว่างประชากรหรือสปีชีส์ได้หากลูกผสมดังกล่าวสามารถอยู่รอดและสืบพันธุ์ได้ ซึ่งสามารถตรวจสอบได้โดยใช้เทคนิคทางด้านอณูชีววิทยา อึ่งน้ำเต้า (Microhyla fissipes) อึ่งข้างดำ (M. heymonsi) และอึ่งลายเลอะ (M. butleri) จัดเป็นสัตว์สะเทินน้ำสะเทินบกที่อยู่ในสกุลเดียวกัน มีขนาดลำตัวใกล้เคียงกันและมีการกระจายอยู่ในทุกภาคของประเทศไทย จากการสำรวจเบื้องต้นพบว่าอึ่ง 2 หรือ 3 ชนิด มีการใช้พื้นที่อยู่อาศัยร่วมกัน และที่สำคัญด้วยธรรมชาติของอึ่งที่มีการปฏิสนธิภายนอกร่างกายเป็นการเพิ่มความเสี่ยงในการเกิดการผสมข้ามสายพันธุ์ได้ การศึกษาครั้งนี้จึงสนใจตรวจสอบความหลากหลายทางพันธุกรรมของยีน COI และประเมินความเป็นไปได้ในการเกิดการผสมข้ามสายพันธุ์ระหว่างอึ่งทั้งสามชนิดในธรรมชาติ โดยนำอึ่งน้ำเต้าจำนวน 48 ตัว อึ่งข้างดำจำนวน 43 ตัวและอึ่งลายเลอะจำนวน 9 ตัว ที่เก็บมาจากสวนสัตว์เปิดเขาเขียว จังหวัดชลบุรี มาตรวจหาและเปรียบเทียบลำดับนิวคลีโอไทด์ของยีน COI ในไมโทคอนเดรียลดีเอ็นเอที่สกัดมาจากเนื้อเยื่อตับของอึ่งทั้งสามชนิด และเพิ่มปริมาณยีน COI โดยใช้เทคนิคพีซีอาร์ ผลการศึกษาพบว่าผลิตภัณฑ์พีซีอาร์ของอึ่งทั้งสามชนิดจำนวน 72 ตัวอย่างให้ผล sequencing ชัดเจนและน่าเชื่อถือ โดยลำดับนิวคลีโอไทด์ที่ได้มีความยาว 677 คู่เบส จากการวิเคราะห์ลำดับนิวคลีโอไทด์ด้วยโปรแกรม DnaSP พบจำนวนแฮพโพลไทป์ที่แตกต่างกันจำนวน 36 แฮพโพลไทป์ ที่มีความแปรผันทางพันธุกรรมจำนวน 189 (27.92%) ตำแหน่ง มีค่าความหลากหลายของแฮพโพลไทป์ และค่าความหลากหลายของนิวคลีโอไทด์ค่อนข้างสูง โดยเฉลี่ยเท่ากับ 0.973 ± 0.006 และ 0.10891 ± 0.00525 ตามลำดับ ระยะห่างทางพันธุกรรมระหว่างประชากรของอึ่งทั้งสามชนิดอยู่ระหว่าง 0.000 ถึง 0.223 แสดงว่าประชากรของอึ่งทั้งสามชนิดมีความแตกต่างทางพันธุกรรมของยีน COI ค่อนข้างสูง ซึ่งเหมาะที่จะนำมาใช้เป็นเครื่องหมายดีเอ็นเอเพื่อใช้ในการจำแนกชนิดของอึ่งทั้งสามชนิด นอกจากนี้จากการศึกษาความสัมพันธ์เชิงวิวัฒนาการยังพบว่าอึ่งน้ำเต้า อึ่งข้างดำ และอึ่งลายเลอะมีความสัมพันธ์เชิงวิวัฒนาการเป็นแบบ monophyletic group การตรวจพบการเกิด gene flow ระหว่างประชากรของอึ่งข้างดำและอึ่งน้ำเต้า และระหว่างประชากรของอึ่งลายเลอะและอึ่งน้ำเต้า แสดงว่าอึ่งทั้งสองชนิดนั้นสามารถผสมข้ามสายพันธุ์กันได้ในธรรมชาติ

In Colombia, Venezuela, Ecuador, Brazil and Honduras, the tomato borer, Neoleucinodes elegantalis, is the most important fruit-related plague of the Solanaceae family. A suitable molecular characterization using a DNA barcoding system is necessary to clarify different issues inside the taxonomy of Neoleucinodes genus. Additionally, other DNA sequences used for molecular identification and phylogenetics studies, can be implemented to obtain a better understanding of the genetic variability across different animal groups and allows to acquire a enhanced description of the population s genetic variation. The main objectives of this study are: 1. Evaluate the performance of DNA barcoding sequences (COI gen and 18S rDNA gene), in the genetic characterization of populations of N. elegantalis, collected in different wild and cultivated solanaceous plants in Colombia and Ecuador. 2. Determination of possible haplotypes related with each population belonging to this species. 3.Identification of geographical patterns associated with the distribution of this insect.

การผสมข้ามสายพันธุ์ (hybridization) เป็นการผสมพันธุ์ระหว่างสิ่งมีชีวิตที่มีความใกล้ชิดกันทางวิวัฒนาการที่มีโครงสร้างพันธุกรรมของประชากรหรือสปีชีส์ที่แตกต่างกัน ทั้งนี้ลูกผสม (hybrids) ที่เกิดขึ้นอาจทำให้เกิดการถ่ายเทเคลื่อนย้ายของยีน (gene flow) ระหว่างประชากรหรือสปีชีส์ได้หากลูกผสมสามารถอยู่รอดและสืบพันธุ์ได้ อึ่งน้ำเต้า (Microhyla fissipes) อึ่งข้างดำ (M.heymonsi) และอึ่งลายเลอะ (M.butleri) เป็นสัตว์สะเทินน้ำสะเทินบกที่อยู่ในสกุลเดียวกัน มีขนาดลำตัวใกล้เคียงกันและมีการกระจายอยู่ในทุกภาคของประเทศไทย จากการสำรวจเบื้องต้นพบว่าอึ่ง 2 หรือ 3 ชนิด มีการใช้พื้นที่อยู่อาศัยร่วมกัน และที่สำคัญด้วยธรรมชาติของอึ่งที่มีการปฏิสนธิแบบภายนอกร่างกายเป็นการเพิ่มความเสี่ยงในการเกิดการผสมข้ามสายพันธุ์ได้ การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อตรวจสอบสอบความหลากหลายทางพันธุกรรมและประเมินความเป็นไปได้ในการเกิดการผสมข้ามสายพันธุ์ระหว่างประชากรของอึ่งน้ำเต้า อึ่งข้างดำ และอึ่งลายเลอะในพื้นที่อำเภอเวียงสา จังหวัดน่าน โดยทำการเก็บตัวอย่างอึ่งทั้งสามชนิดและตรวจสอบการเกิด gene flow โดยเทคนิคทางด้านอณูชีววิทยา ผลการสำรวจและเก็บตัวอย่างพบว่ามีอึ่งน้ำเต้าเพียงสปีชีส์เดียวเท่านั้นที่สามารถเก็บตัวอย่างมาได้ จำนวนทั้งหมด 25 ตัว จากสองพื้นที่ในอำเภอเวียงสา คือ ที่ตำบลไหล่น่าน (n=9) และที่ตำบลส้าน (n=16) ผลการเพิ่มปริมาณยีน COI ในไมโทคอนเดรียลดีเอ็นเอด้วยเทคนิคพีซีอาร์และการทำ DNA sequencing พบว่าอึ่งน้ำเต้าทั้ง 25 ตัวอย่างให้ผลิตภัณฑ์พีซีอาร์และผล sequencing ชัดเจนและน่าเชื่อถือ โดยลำดับนิวคลีโอไทด์ที่ได้มีความยาว 678 คู่เบส จากการวิเคราะห์ลำดับนิวคลีโอไทด์ด้วยโปรแกรม Dnasp พบจำนวน haplotype ที่แตกต่างกันจำนวน 7 haplotype ที่มีความแปรผันทางพันธุกรรมจำนวน 6 (0.88%) ตำแหน่ง และมีค่าความหลากหลายของ haplotype (hd) และค่าความหลากหลายของนิวคลีโอไทด์ (π) ค่อนข้างต่ำ โดยเฉลี่ย hd=0.627 ±0.102 และ π = 0.00111±0.00024 โดยมีระยะห่างทางพันธุกรรมระหว่างประชากร อยู่ระหว่าง 0.000 ถึง 0.003 แสดงว่าประชากรอึ่งน้ำเต้าจากอำเภอเวียงสา จังหวัดน่าน มีความแตกต่างทางพันธุกรรมของยีน COI ค่อนข้างต่ำ นอกจากนี้จากการศึกษาความสัมพันธ์ทางวิวัฒนาการยังพบว่าอึ่งน้ำเต้ามีความสัมพันธ์เชิงวิวัฒนาการเป็นแบบ monophyletic group แต่เนื่องจากการศึกษาครั้งนี้ไม่ประสบความสำเร็จในการเก็บตัวอย่างอึ่งข้างดำและอึ่งลายเลอะจากพื้นที่ในอำเภอเวียงสา จังหวัดน่านได้เลย ดังนั้นจึงไม่สามารถประเมินความเป็นไปได้ในการผสมข้ามสายพันธุ์ระหว่างอึ่งทั้งสามชนิดในพื้นที่ดังกล่าวได้

Abstract:
Den här rapporten sammanställer flera års forskning runt civila insatspersoner (CIP) – frivilliga som samarbetar med kommunal räddningstjänst och larmas på exempelvis hjärtstopp, mindre bränder, trafikolyckor och drunkning. Rapporten ska ses som lite av en handbok i att införa CIP – från rekrytering till upprätthållande – vilka är framgångsfaktorerna och hur undviker man fallgroparna? Dessutom jämförs CIP i landsbygdsmiljö och stadsmiljö. Rapporten täcker aspekter av organisation, utbildning, lagstiftning och IT-stöd för utlarmning och koordinering av CIP. En rad behov och förslag på lösningar ges. Ett exempel på införande av CIP införande i en räddningstjänst (Räddningstjänsten Östra Götaland) visar på hur man kan arbeta. Dessutom visar rapporten på effekter av CIP såsom kortade responstider och en ökad känsla av trygghet och samhörighet i lokalsamhället. Den visar också att CIP är villiga att bidra även till större händelser, kriser och till ett civilt försvar.

Review question / Objective: The aim of this meta-analysis was to get an updated opinion, which was about the role of the single nucleotide polymorphism (SNP) rs4973768 in the SLC4A7 gene played in the incident of breast cancer. Eligibility criteria: The included criteria were formulated for this meta-analysis as following:(1) studies with both case and control groups;(2) studies assessing the relation between the SLC4A7 rs4973768 polymorphism and sensibility to breast cancer;(3) studies with sufficient information such as genotype frequency for results of odds ratios (ORs) and 95% confidence intervals (95% CIs); and (4) full-text articles of studies with human subjects. Studies meeting any one of the criteria were determined unqualified for this meta-analysis as following:(1) conference abstracts, comments, reviews, case reports, or editorials;(2) inadequate data for OR calculation;(3) no control group; and (4) animal studies.
