Abstract:

Abstract:
This doctoral thesis deals with design of a new generation of coherence-controlled holographic microscope (CCHM). The microscope is based on off-axis holographic configuration using diffraction grating and allows the use of temporally and spatially incoherent illumination. In the theoretical section a new optical configuration of the microscope is proposed and conditions for different parameters of the microscope and its optical components are derived. The influence of different sources of noise on phase detection sensitivity is studied. In the next section design of experimental setup is described and automatable adjustment procedure is proposed. Last section describes experimental verification of the most important optical parameters of the experimental setup. When compared to previous generation of CCHM, the newly proposed configuration uses infinity-corrected objectives and common microscope condensers, allows more space for the specimens, eliminates the limitation of spectral transmittance and significantly simplifies the adjustment procedure so that automation of this procedure is possible.

Abstract:

This thesis aims to find out how to properly motivate the new generation Y and find out if there are differences in their motivation and job satisfaction from other generations. Using mixed research in the form of a questionnaire survey for employees and in-depth interviews with potential employers will provide a dual view of the issue, which can provide valuable information and serve as an important informative element for the correct motivation of Y generation. Three hypotheses have been identified, which will be subsequently verified in both quantitative and qualitative research. After evaluating the research, it is good news that the outlook of the generation Y and their employers is very similar, and it seems that potential employers are very well prepared for the future generation and are already able to estimate their differentiation from other generations and thus properly motivate them and stimulate them for better work performance.

Abstract:
Předkládaná disertační práce je zaměřena na "heterogenní propojení mobilních zařízení v bezdrátových systémech 5. generace". Navzdory nepochybnému pokroku v rámci navržených komunikačních řešení postrádají mobilní sítě nastupující generace dostatečnou šířku pásma a to hlavně kvůli neefektivnímu využívání rádiového spektra. Vědecký přínos dosažených výsledků dokazuje fakt, že některé z principů představených v této disertační práci byly zahrnuty do celosvětově uznávaného standardu (specifikace) 3GPP Release 12.

Abstract:
The main target of the master´s thesis is reviewing the generation III nuclear reactors in term of the nuclear safety. At first we have to learn some theory of the nuclear safety in order to understand safety systems of the generation III nuclear reactors. Therefore the thesis is divided into two parts. Legislative and technical approaches to nuclear safety are mentioned in the first part. Regulatory bodies, whose task is to supervise nuclear safety in the nuclear power plants, belongs to the legislative approaches. There are defined terms such as defence in depth, redundancy, diversity, etc. There are mentioned methods to assessing nuclear safety – deterministic and probabilistic methods, especially probabilistic methods, for which a simple example is provided. There are also mentioned active and passive safety systems and their significance for nuclear safety and inherent safety too. There is an example of the function of the active and passive safety systems of the EDU nuclear power plant in conclusion of this issue. The second part deals with description of the selected nuclear reactors in context of the construction of the new units of nuclear power plant in Temelín. The nuclear reactors from companies, which applied for the public tender opened by ČEZ, a. s., for the construction of the ETE 3+4. Thus, the nuclear reactor MIR-1200 by ATOMSTROYEXPORT (Russian Federation), the nuclear reactor AP1000 by WESTINGHOUSE (USA) and the nuclear reactor EPR by AREVA (France) are taken into account . Comparison of the generation II and these generation III+ nuclear reactors necessarily belongs to this master´s thesis. These the generation III+ nuclear reactors are compared with the nuclear reactor VVER 440 (EDU) and in particular with the nuclear reactor VVER 1000, which is operated in the nuclear power plant Temelín. The final chapter contains generally appraisal of the whole problem.

Abstract:
This Master’s thesis is focused on the recent 3G mobile networks. The first chapter describes an evolution of standardization 3G and surveys a gradual development of 3G networks in Czech Republic from a point of view of commercially launched technologies and assigned frequency spectrums for single mobile operators. UMTS and CDMA 2000 are briefly approached in next subchapters. The subchapter CDMA 2000 is briefly concentrated on architecture and a physical layer of this technology and characterizes main features of four revises of developmental branch CDMA 2000 1x EV-DO. The subchapter UMTS is especially focused on a comparison of air interfaces TD-CDMA and W-CDMA. New features were implemented to the recent system UMTS for a better support of real-time services. These features increasing a throughput and decreasing a delay are described in a chapter Technology HSPA. This chapter chooses the most important features and describes changes (in a control and architecture) which had to be implemented. First chapter is finished by technology WiMAX. The most recent perspective cell systems are compared with WiMAX in this subchapter and qualitative parameters of WiMAX are shown there. The three different 3G technologies (UMTS-FDD, UMTS-TDD and CDMA 2000) are lunched in Czech Republic. The second chapter briefly compares coverage of three czech mobile operators (Telefónica O2, T-Mobile and U:fon) and then presents results of qualitative parameters of used 3G technologies. The simulation conducted by software IxChariot were focused on to gauge parameters like throughput, delay, jitter, lost data, MLR and DF. The conditions for providing real-time services were specified based on simulations that were practically checked. The lab composed in last chapter should bring closer to students technology UMTS-TDD launched by T-Mobile. The students will measure the qualitative parameters of this technology with the assistance of commonly available tools.

Abstract:
The question of age and aging is currently gaining in importance. The main reasons for the aging of population are the extension of age of individuals and low birthrate. This master thesis tries to answer the question, how the younger generations see seniors and identify the main stereotypes and myths about age and aging, which are produced in contemporary society. By using a combination of quantitative and qualitative research is presented an image of senior in the eyes of youngest, middle and oldest generations. The empirical part firstly deals with the perception of seniors, depending on age and sex of respondents and secondly the perception of myths about seniors depending on the age of respondents. Statistical data from the questionnaire are supplemented with information based on interviews. The results of the thesis point out that the Czech population perceives seniors rather neutral, between generations are only small differences in the perception of seniors.

Abstract:
Economic reality of these days shows that pension, social, fiscal and educational system (we did not deal with flat politics friendly to young families) suffer from disputable till wrong decisions in last months and years. Many of such a decision seemed not to be effected based on scientific and expert analyses, results and recommendations but based on influence and obstinacy of the lobbyist parties and other groups. In the area of social politics we are missing the real social abstraction (i.e. long-term perspective of the social politics bearing) which could be acceptable by the residents. Pension savings and supplementary insurance act as barter -- we send to you contributions, you invest them into our bonds emitated to balance government debt (caused by this contributions as well) -- you have your profits, we have balance and meet liabilities. Fiscal system with its deductions, discounts and consolidations gives a favour to specific groups prior to the others. Social system with its substitutional durations, early pensions, excess indexation, parent leave duration and other advantages destroys natural development of the society, its revenues and expenditures. Early age educational system cannot ensure a sufficient pre-school education and changes parents into the bribees in bulk in order to find a place for their children (4-years and older). Than prolonged compulsory education causes prolonged entrance to the job market. High schools and universities throw up crowds of graduates without necessary practical skills which cause high amount of a young unemployment. We are the country of phenomenons. In the area of supplementary insurance support to the private pension companies we are the best. In the area of advanced women support for family and occupational life coordination (part-time jobs, flexible labour contracts, huge scale of pre-school day-care service) we are the worst and not only in domestic but also international comparison. We face absence of qualified work-force, human capital (necessary for healthy economic and social progress) in 50 or 60 years. This thesis has had for one's object to confirm or disconfirm the main hypothesis that with combination of defined important instruments and politics (aimed to non-financial benefits and huge flexible labour contracts supply) there is a possibility to achieve the long-term sustainability and balance of the pension account with not big shortage, no direction to destroy the whole economic development and with support to flexible job market. For children the thesis perhaps controversially looks as for human capital but from the economic point of view this is correct. In case of the Czech Republic the government approach does not work, the municipalities approach is absolutely unconceptual which restrains in many regions to achieve the co-operation of work and family life, to achieve the high standard of living of families, both due to unsupported pre-school day-care. We could accept one new solution -- no employers pension funds but employers pre-school day-care in co-operation with government and municipalities. This thesis established a hypothesis at the beginning which has been confirmed. PAYG in case of correct co-operation of important politics remains in long-term period stable (on condition 2 children for 1 mother), is just and effective. State income redistribution has moved to the service provider. Costs of such a day-care have been covered by revocation or reduction of the supplementary pension benefits, cancellation of the second pension pillar, shortening of the parent leave supplement, increasing of the employment and employees activity, increasing of efficiency and productivity and consequently profits of the companies and tradesmen. In combination with the high-quality fiscal politics without distortion and without one-side favouritism and senseless reliefs there has been secured stability of pension account, whole economy and public finance. Lower redistribution has been compensated by higher consumption and investment, government spending etc.

Abstract:
This master thesis aims to compare consumer behavior of generation Y (people born in the period 1982-2002) in the Czech Republic and Hong Kong, based on questionnaire data. Special attention is paid to corporate social responsibility and protection of the environment, which begin to play an important role in consumer behavior. The differences between nations are examined through the questionnaire survey, which was filled out by 155 respondents. For a deeper understanding and identification of individual motifs, consumers were conducted semi-structured interviews that focused on environmental problems and corporate social responsibility. Hong Kong and the Czech Republic have different level of individualism and uncertainty avoidance, except these two cultural dissimilarities, there were no significant differences between these two nations. Environmental protection plays a larger role in the consumer behavior of Generation Y than corporate social responsibility for both nations. Corporate social responsibility is more important for young women than for young men.

The diploma thesis deals with some aspects of the life of the Czech generation Y, the first generation growing up in a free period after the Velvet Revolution of 1989. The thesis focuses on university students. The introduction focuses on defining the concept of generation in the Czech environment as well as in the world, as well as the issue of generations and discussions on the subject both at the media and at the professional level. The research section is based on interviews with twelve members of this generation. The interviews are oriented on the specifics that distinguish the Czech generation Y from the young people in other parts of the world, especially history of the Czechoslovakia between years 1948 and 1989. The interviews drew on their opinion and outlook on the development of the republic before 1989 by opting for family, education, peers, and other factors that could influence them. The conversations also focused on the current attitude towards politics, ideologies and values. Narrators also looked for generational specifics and characteristics related to generational determination, such as technology, social networks, anti-communism... Starting point. Voluntary simplicity is a form of lifestyle. Its typical feature is selection intensity in all areas of human life. Severity of voluntary simplicity lies in self-control of consumer behavior in the daily confrontation with the media pressure, which is oriented at a high rate of consumption. The task of this thesis was to find out if people raised in voluntarily simple families keep living this way, or if they turned away from this lifestyle and joined the high consumption society. Methods and results. Research included 40 respondents who met the requirement that voluntary simplicity was part of their upbringing. Addressed were people from alternatively living groups (preppers, ecovillages, macrobiotics, minimalists, forums dedicated to maternal education and child care, and other groups possessing forums on an alternative way of life of voluntary simplicity). The investigation was conducted by a questionnaire which was distributed electronically among respondents. We found that 13 out of 40 respondents (32,5 %) fully met our stated profile of voluntary simplicity. Relative frequency of responses from all respondents that meet the requirements of modest behavior amounted to 74,3 %. Hypothesis H1 predicted that the influence of upbringing in a voluntary simplicity will make respondents prefer...

Abstract:
The master's thesis "Over-indebtedness of Young Generation in the Czech Republic" deals with over-indebtedness of individuals at the age of 18 to 26 years. Over-indebtedness is a general social problem as all age groups get overburdened with debt. However, it is important to focus on the issue of young people's over-indebtedness directly because the young generation represents people who are or will be active in the general working process. The authoress of the thesis points out the young generation's over-indebtedness and provides an overview of development of this issue in 2008 - 2015. She also examines the most frequent causes which she has found out using questionnaires and data collected. In her work, the authoress also outlines a gradual development of possible indebtedness, which primarily includes loans, from both banks and other institutions, followed by a court procedure and execution of the judgement. A possible solution to over-indebtedness then consists in filing a petition with a court to permit a personal discharge from debts. The work aims at finding out and examining development of the young generation's indebtedness in the Czech Republic, analysing causes of over-indebtedness, describing the changes of the causes in time and at analysing the process of discharge from debts, which...

Abstract:
Lipophosphonoxins are small synthetic antimicrobials targeting the cytoplasmic membrane of bacteria. This thesis focuses on comparison of three lipophosphonoxins which differ in the number of carbons in its modules and in antimicrobial and hemolytic activity. The most promising candidate compound is lipophosphonoxin 7072 showing good antimicrobial activity as well as low hemolytic activity. Other two tested lipophosphonoxins are 7070 displaying high hemolytic activity and weakly antibacterial lipophosphonoxin 7107. The pore-forming activity of lipophosphonoxins is investigated using model membranes as well as living bacteria Staphylococcus aureus and Escherichia coli. The results show that small difference in structure can fundamentally affect the activity of these molecules. Lipophosphonoxins 7072 a 7070 display equal antibacterial activity against tested bacteria by forming pores in the bacterial membrane. Bacteria rapidly die of loss of membrane potential caused by lipophosphonoxins. The high hemolytic activity of the compound 7070 is probably related with its preference for uncharged membranes. The weak antimicrobial activity of 7107 is caused by its capability to form only small pores and its incapability to overcome and disrupt the outer membrane. Key words: antimicrobial agents,...
