The synthesis of the requisite protected steroidal core unit 13 en route to secosterol 1, a cytotoxic constituent of the soft coral Gersemia fruticosa, is described. The conversion of the intermediate precursor 3 into 13 succeeds through oxidative side chain degradation, deoxygenation of C-12 via reductive desulfurization and cleavage of the C-ring moiety by ozonolysis.

Abstract:

The minor metabolites of both Pacific and Atlantic Ocean specimens of the cold water soft coral Gersemia rubiformis have been investigated. Pacific coast animals yielded two new pseudopterane diterpenoids, isogersemolides A (11) and B (12), and four new cembrane diterpenoids, isoepilophodiones A (13), B (14), and C (15) and rubifol (16). Atlantic coast animals yielded 3-acetoxy-β-cubebene (17). The proposed structures of all the new metabolites are based on spectroscopic analysis and chemical interconversions.

Responses of delicate soft corals to mechanical injuries inflicted by bottom fishing (e.g., trawling, dredging) are not known. Effects of mechanical disturbance on the soft coral Gersemia rubiformis (Ehrenberg, 1834) were examined by experimentally simulating disturbances caused by bottom fishing. Eight colonies were collected in the Bay of Fundy and maintained in individual aquaria.
