Partial sequences of the mitochondrial genes 16S rRNA and cytochrome c oxidase subunit I (COI) were used (1) to resolve the internal relationships of the subfamily Tubificinae (Annelida : Clitellata) and (2) to test the existence of cryptic species within the stygobiont oligochaete Troglodrilus galarzai (Giani & Rodriguez, 1988). Phylogenies were estimated using maximum likelihood, Bayesian inference and parsimony. Although trees were incompletely resolved at intergeneric level, a close relationship between Heterochaeta Claparède, 1863 and Troglodrilus Juget, des Châtelliers & Rodriguez was revealed, and the separation of Lophochaeta ignota Štolc, 1886 and Heterochaeta costata Claparède, 1863 from Tubifex Lamarck, 1816 was corroborated by mitochondrial molecular data. Maximum genetic divergence between allopatric populations of T. galarzai was 18% for COI (uncorrected pairwise distance), suggesting cryptic speciation within this nominal species.

Abstract:
Morphological characters for several Palearctic taxa attributed by recent authors to the genus Rhynchelmis Hoffmeister, 1843 were reviewed using new or previously studied museum specimens. The genus Rhynchelmis s. lat. was supported, and the Palearctic subgenus Rhynchelmis (Rhynchelmis) was proposed, based on several probable synapomorphies: longitudinal muscles with lateral margins curled; strong dorsolateral-transverse muscles; vasa deferentia with thickened, glandular epithelium; spermathecal ampullae with anterior, sacciform diverticulum; accessory organ often present in preatrial segment. The Japanese Rhynchelmis orientalis Yamaguchi, 1936 was redescribed using new material, and had none of the abovementioned characters. The separation of other potential subgenera, the Rhynchelmoides and Sutroa groups, is still subject to review of those primarily Nearctic taxa. Attribution by Kaygorodova & Liventseva (2007) of several Lake Baikal species to Pseudorhynchelmis Hrabě (1982) was supported, and Rhynchelmis paraolchonensis Giani & Martinez-Ansemil, 1984 may also belong in Pseudorhynchelmis.

We use a morphological approach focused on characters of the reproductive organs to assess the taxonomic status of thestygobiont oligochaete species Troglodrilus galarzai (Giani and Rodriguez, 1988). After the evaluation of 23 characters,diagnostic differences in the spermathecal vestibule, the penial sac and the penial sheath are observed between northernIberian Peninsula populations and south-eastern France populations. Recent molecular analyses based on mitochondrialgene data have revealed a high genetic variation in T. galarzai. Given that both molecular and morphological data supportthe presence of two species within T. galarzai, we propose the specific status for the French populations under the nameTroglodrilus jugeti n. sp. As a result, the type species of the genus, T. galarzai, is now restricted to the Iberian Peninsula.A possible paleogeographic scenario is hypothesised for the evolution of Troglodrilus during the Para-Tethys regression in the Neogene stage.

Abstract:
In this thesis we shall present work concerning the description of the emergence of solitonic fundamental strings from stable, finite energy, compact D3-branes in a subspace of $AdS_{5}\times S^{5}$ and their subsequent interaction. We work in the planar limit and focus on states of large angular momentum $J$ corresponding to large R-charge in the dual gauge theory. We begin by constructing the full set of boundary giant magnons on $\mathbb{R}\times S^{2}$ attached to the maximal $Z=0$ giant graviton by mapping from the general solution to static sine-Gordon theory on the interval. We then compute the values of the anomalous dimension, $\Delta-J$, of the dual gauge theory operators at finite $J$, examining the behaviour of the leading order corrections when $J$ is large. We then consider the Born-Infeld theory of the giant graviton itself coupled to the background 5-form flux. Constructing BIon spike solutions that correspond to the world volume description of the boundary giant magnons we find a limit amenable to analysis which returns the full range of behaviour exhibited at finite $J$. Finally we produce the open strings on $\mathbb{R}\times S^{2}$ that correspond to the solutions of integrable boundary sine-Gordon theory. Relating the boundary parameters in a way that ensures a given set of string boundary conditions we describe the scattering of giant magnons with non-maximal $Y=0$ giant gravitons and calculate the leading contribution to the associated magnon scattering phase. Our method necessarily describes all integrable scatterings of giant magnons with giant gravitons.

In der vorliegenden Arbeit wird ein neuer Ansatz vorgestellt, um Lipiddomänen, die Bindungsorte peripherer und integraler Membranproteine darstellen können, zu charakterisieren. Insbesondere wurde die Analyse der Fluoreszenzlebenszeiten von NBD-markierten Lipidanaloga benutzt, um Lipiddomänen in Giant unilamellar vesicles (GUV) und darauf aufbauend, in der Plasmamembran von Säugerzellen zu untersuchen. Das typische Zeitfenster von Fluoreszenzlebenszeiten im Bereich von Nanosekunden ermöglicht es, auch sehr kurzlebige Lipiddomänen nachzuweisen. Mit Hilfe des Fluorescence lifetime imaging (FLIM) wurden für die liquid disordered (ld) und liquid ordered (lo) Domänen in GUV jeweils spezifische Werte für das Abklingen der Fluoreszenz gemessen. Sogar die Existenz von submikroskopischen Domänen in GUV konnte nachgewiesen werden. Die Fluoreszenzlebenszeit des Lipidanalogs C6-NBD-PC zeigte in der Plasmamembran von Säugerzellen eine breite Verteilung. Dies legt in Übereinstimmung mit FLIM-Experimenten an aus der Plasmamembran von HeLa-Zellen gewonnenen Giant vesicles nahe, dass in der Plasmamembran von Zellen eine Vielzahl verschiedener submikroskopischer Lipiddomänen existiert. Darauf aufbauend wurde die Fluoreszenzmikroskopie an GUV angewendet, um die Bindung von fluoreszenzmarkiertem alpha-Synuclein an mittels FLIM charakterisierte Lipiddomänen zu untersuchen. Die Experimente zeigten, dass das Protein mit hoher Affinität an negativ geladene Phospholipide unter der Vorraussetzung bindet, dass diese sich in ld Domänen befinden. Im Gegensatz dazu erfolgt keine Bindung wenn diese Lipide in lo Domänen lokalisiert sind. Im Vergleich zum wildtypischen alpha-Synuclein zeigte die Variante A30P eine geringere Affinität zur Membran, während die E46K-Variante eine stärkere Bindung zeigte. Dies deutet darauf hin, dass bei den erblichen Formen des Morbus Parkinson eine veränderte Assoziation des alpha-Synucleins mit der Membran eine Rolle spielen kann.
In the present study a novel approach to characterize lipid domains, which may provide binding sites for peripheral or integral membrane proteins, is demonstrated. In particular, analysis of fluorescence lifetimes of NBD-labeled lipid analogues was used to study lipid domains in Giant unilamellar vesicles (GUV) and – based on the GUV results – in the plasma membrane of mammalian cells. As fluorescence decays in a few nanoseconds it is possible to to detect also very short-lived lipid domains. Fluorescence Lifetime Imaging (FLIM) revealed that the fluorescence decay of NBD-lipid analogues showed domain dependent decay times in the liquid disordered (ld) and the liquid ordered (lo) phase of GUV. Even the existence of submicroscopic domains in lipid membranes could be detected by FLIM. A broad distribution of the fluorescence lifetime was found for C6-NBD-PC inserted in the plasma membrane of mammalian cells. In agreement with FLIM studies on lipid domain forming Giant vesicles derived from the plasma membrane of HeLa-cells this may suggest that a variety of submicroscopic lipid domains exists in the plasma membrane of intact mammalian cells. Based on that, fluorescence microscopy was used on GUV to study the binding of fluorescently labeled alpha-synuclein at lipid domains previously characterized by FLIM. The experiments suggested that alpha-synuclein binds with high affinity to negatively charged phospholipids, when they are embedded in a ld as opposed to a lo environment. When compared with wildtype alpha-synuclein, the disease-causing alpha-synuclein variant A30P bound less efficiently to anionic phospholipids, while the variant E46K showed enhanced binding. This suggests that an altered association of alpha-synuclein with membranes may play a role in the inherited forms of Parkinson’s disease.

Abstract:
In this work, the efficiency of the LUMBA UVES pipeline for processing of spectroscopic observations is tested by use on 23 high-resolution spectra of the open star cluster M67. An abundance trend discovered by Gavel et al. (2019) concerning iron abundances of giant and sub-giant stars of that cluster is examined. An initial run for a set of ”Gaia FGK benchmark stars”, as described in Blanco-Cuaresma et al. (2014) and Heiter et al. (2015) helps inspect the structure, method and output of the pipeline. Through Python language programming, processes are greatly automatized and the pipeline is run for a total of 460 weak and strong iron lines of our 23-star sample. The line fitting and efficiency of the pipeline is appreciated by statistically analyzing the results and looking into individual discrepant ones. The abundance trend is reproduced while using FeI lines, unlike runs using FeII lines. Trends in abundance over line strength plots also hint at bias through the Gaia-Eso Survey (GES) microturbulence relation. Using internal Data Release 6 (iDR6) and LUMBA-derived starting parameters, log(g) - Teff plots of our sample stars agree with a previously established 4.3 Gyr cluster age. An alternate run is performed for those stars, using LUMBA-derived starting parameters. The choice of starting parameters does impact abundance derivation, but is not the primary source of persistent systematic discrepancies.

This chapter examines the Panj Piare story in the works of Giani Gian Singh, specifically Panth Prakash (1880) and Tavarikh Guru Khalsa (1892–1921), and it situates this examination in the context of Gian Singh’s historical environment. This study of the Panj Piare narrative in these texts provides us with insight into the messages that Gian Singh was encoding throughout his prolific career, messages that were very much aligned with the trajectory that was plotted by the Sikh intellectual movements of the late nineteenth and early twentieth centuries, the Singh Sabha and the Tat Khalsa.

Abstract:
This chapter discusses Sacrifice's spiritual education. Fascist mysticism promotes a number of values, including love of family, loyalty to friends, fairness in sports competitions, solidarity with the poor, love for Italy, voluntary service, and physical exercise. These values, however, can be considered “secondary.” There are three primary values in revolutionary Fascism. The first value is the courage one demonstrates by accepting or provoking fights with the enemy. The second value is sacrifice: fighting even though one knows he cannot win. The third value is honor, which Sacrifice comrades consider a consequence of courage and sacrifice. The greater the willingness of Sacrifice comrades to fight against a stronger enemy, the greater the honor. The chapter then describes the books that the Lenintown and Mussolinia militia members read. Abstract:
The discovery of the giant magnetoresistance effect in metallic multilayers and alloys has led to extensive theoretical and experimental investigations of this effect for the past few decades. Recently, we discovered that Cu/CoFe giant magneto-resistance layers exhibit a giant magnetothermal resistance effect (GMTR), in which the thermal conductivity shows a “giant” change under a magnetic field. In the present study, we adopted the Fuchs-So¨ndheimer extension of the semiclassical Boltzmann theory to predict the in-plane giant magneto-resistance (GMR) and giant magnetothermal resistance (GMTR) ratios in the Cu/CoFe multilayer structure. The present model has been implemented in an attempt to explain the measured differences in the giant magnetoresistance and giant magnetothermal resistance ratios, which have previously been attributed to the different mechanisms for charge and heat transport in the Cu/CoFe multilayer structure.

Abstract:
The main motivation for the research is a verification of applicability of the fitness test as a predictor for specific alpine skiing performance of the Czech national team of U14 and U16 categories. We want to verify if the results from the fitness test correspond with points for the performance in alpine skiing disciplines (slalom, giant slalom, super giant slalom). In total, 42 men and women (U14 and U16 categories), members of the Czech national team, participated in the research. Participants were grouped by gender (women n=21, age 14.23 (± 1.04), men n = 21, age 14.19 (± 1.07)). Fitness test consisted of six individual tests and was used as a tool to examine the level of motor abilities (jumping over the Swedish bench, shuttle run 4 × 10m, standing long jump, hurdles agility run - boomerang test, twist test, 20m shuttle run - beep test). Points for the performance in alpine skiing show the best result from a single competition transferred to the points. In our research we were looking for relationship between the points from three different disciplines (slalom, giant slalom, super giant slalom) and the results from the fitness test. In the men category we found high correlation (at the significance level of p ˂ 0.05) between super giant slalom and standing long jump (r = -0.73; r² = 0.53), between super giant slalom and boomerang test (r = 0.62, r² = 0.38) and between super giant slalom and shuttle run 4 × 10m (r = 0.61, r² = 0.37). Small correlation was found between slalom and box jump test (r = -0.31, r² = 0.1), between slalom and twist test (r = -0.33, r² = 0.11) and between super giant slalom and twist test (r = -0.34, r² = 0.12). In women category we did not found high correlation (at the sig-nificance level of p ˂ 0.05). Small correlation was found between slalom and box jump (r = -0.31, r² = 0.1), between giant slalom and twist test (r = 0.01, r² = 0.0001), between super giant slalom and standing long jump (r = -0.03, r² = 0.0009) and between slalom and shuttle run 4 × 10m (r = -0.05, r² = 0.0025). Twist test had very small correlation in both men and women categories. We can say that it is not relevant for prediction of alpine skiing per-formance. Based on different significance of correlations between men and women (super giant slalom and standing long jump; slalom/super giant slalom and shuttle run 4 × 10m) it might be necessary to adjust fitness testing as a predictor of alpine skiing performance according to a gender.

Abstract:
Giant platelet syndrome are characterized morphologically by many giant platelets associated with several functional abnormalities in the peripheral blood. However, the mechanism of large platelet production has not yet been clarified. In 1981, we reported acase with Bernard-Soulier syndrome(BSS) in whom giant platelets were considered to be formed by fusion of two or three platelets in the circulating blood. We examined the ultrastructure of platelets and megakaryocytes in another case with BSS (29 year-old female) and a case with May-Hegglin anomaly (31 year-old male). Whole blood and bone marrow specimens were fixed with glutaraldehyde-osmium solution. Thin sections were prepared and stained with uranyl acetate and lead cytrate. Membrane systems of platelets and megakaryocytes in a case with BSS was investigated by staining of surface coating with ruthenium red.In a case with BSS, most platelets were very large and similar in morphology to those in formerly reported case. Giant platelets contained several-fold increased number of α-granules and mitochondria. Typical dense bodies were also observed. Contents of ATP/ADP, platelet factor-4(PF-4), B-thromboglobulin(B-TG) and platelet factor-3 availability(PF-3) were increased. Disorganization of microtubules was recognized. Some giant platelet contained membrane systems similar to demarcation membranes(DM) in megakaryocytes, characteristically. In mature megakaryocytes, areas divided by DM similar in size to those in normal megakaryocytes were observed. Several of these areas appeared to fuse together to form the giant platelets containing many granules and remnants of DM. In a case with May-Hegglin anomaly, typical Dohle’s bodies were shown in neutrophilic granulocytes. Giant platelets in this case also contained large number of α-granules and some of them contained membrane systems similar to DM. Areas similar in morphology to these giant platelets were clearly noted in the cytoplasm of mature megakaryocytes.In these cases, most giant platelets in the peripheral blood may be formed in the cytoplasm of megakaryocytes by fusion of several areas divided by DM, each of which may become normal sized platelets in normal megakaryocytes.

Peripheral giant cell granuloma (PGCG) is a relatively Common reactive exophytic lesion of the oral cavity. The influence of hormones has been suggested as contributory factor in PGCG development and predominance of these lesions in young females as well as some previously reported pregnancy related cases support this belief. It has been observed that majority of lesions present in the 4th decade of life, when hormonal changes are more pronounced. Cailluette and Mattar in their study found that peripheral giant cell granuloma are under the influence of the ovarian hormones. However Chambers and Spector suggested peripheral giant cell granuloma to be enhanced by pregnancy rather than being pregnancy dependent. The responsiveness of gingiva to these hormones along with the immunosuppressive actions of the hormones contributes to the growth of the lesion. Clinically, PGCGs may present as polypoid or nodular lesions, predominantly bluish red with a smooth shiny or mamillated surface.This poster will review the literature available on the association of Massive Peripheral Giant Cell Granuloma With Pregnancy with focus on possible causes of PGCG during pregnancy.
