Full textThis dissertation aims to demonstrate the meaning of freedom in the Christian point of view and the resulting implications of an independent Christian living in addition to demonstrating the relations between faith, Christian freedom, love and service. This work was specifically based on Paul s letter written to the Galatians and, more precisely, on the Gl 5, 1-14 pericope where Paul substantiates the issue of Christian freedom. With this work the author intends to analyze the Christian freedom, delimitating its practice and locating its experience, defining its concept and discussing its consequences and how to live it at community in those days when the slavery imposed by the Roman Empire predominated, in a scientific knowledge construction.
Esta dissertação objetiva demonstrar o significado de liberdade do ponto de vista cristão e as implicações resultantes de um viver cristão independente, além de demonstrar as relações entre fé, liberdade cristã e serviço. Este trabalho se baseou especificamente na carta de Paulo escrita aos gálatas e mais precisamente na perícope de Gl 5, 1-14 onde Paulo fundamenta a questão da liberdade cristã. Com este trabalho a autora pretende analisar a liberdade cristã delimitando a sua prática e localizando sua vivência, definindo seu conceito e discutindo suas consequências e como vivê-la na comunidade naqueles tempos em que predominava a escravidão imposta pelo Império Romano numa construção de conhecimento científico.

Full textEn las últimas décadas, los biomateriales basados en copolímeros segmentados, constituidos por segmentos duros y blandos, han atraído el un creciente interés para una amplia gama de s aplicaciones biomédicas, especialmente como suturas monofilares absorbibles. Se ha demostrado que la poliglicolida es una buena candidata para constituir el segmento duro debido a su biodegradabilidad y a su alta rigidez, mientras que la incorporación de distintos monómeros (por ejemplo, -caprolactona, carbonato de trimetileno o glicolida) en una disposición estadística como segmento blando promueven la flexibilidad del material y mejoran la velocidad de degradación in vitro. Las propiedades de los materiales resultantes pueden ajustarse combinando diferentes proporciones de los segmentos duro y blando, y con una composición específica de monómero. En esta Tesis se presenta el estudio del copolímero segmentado tricomponente comercializado por B.Braun Surgical como sutura absorbible monofilar con el nombre de Monosyn®. Los principales objetivos de este trabajo son: a) la caracterización física completa que incluye el estudio de estabilidad térmica y cinéticas de cristalización, b) el estudio de la degradación y los cambios microestructurales asociados, c) la evaluación de la mejora del uso de la sutura añadiendo componentes con actividad farmacológica, y d) la producción de nanofibras para utilizarlas como agentes reforzantes. Las cristalizaciones isotérmicas y no isotérmicas se siguieron mediante distintas técnicas (microscopía óptica, calorimetría diferencial de barrido y experimentos de difracción de rayos X de bajo ángulo). La composición del segmento blando influyó en la cristalización del copolímero segmentado, como era de esperar por la comparación con los datos publicados del sistema bicomponente similar. La degradación hidrolítica fue llevada a cabo a distintas temperaturas con disoluciones tamponadas en un rango continuo de pH. La morfología de la superficie de las suturas hidrolizadas mostró la formación de fracturas longitudinales y circunferenciales en la zona externa e interna de la sutura, respectivamente. Estas fracturas se asociaron a la existencia de dominios amorfos interfibrilares e interlamelares, tal y como se corroboró mediante los experimentos de difracción de rayos X de bajo ángulo. Para explorar las aplicaciones potenciales de Monosyn® como sutura absorbible monofilar con actividad farmacológica, se incorporaron distintos fármacos. Además, se evaluó el uso de un copolímero amorfo como recubrimiento. El primer grupo de fármacos seleccionados corresponden a la familia de las biguanidas (clorhexidina y polihexametilen biguanida), conocidas por su actividad bactericida. En particular se probó la efectividad de utilizar un recubrimiento y la influencia del tamaño molecular del fármaco en su actividad. El estudio del proceso de carga, el perfil de liberación y la actividad farmacológica se completó considerando también un antibiótico (cloranfenicol) y un agente cicatrizante (captopril). Por último, se prepararon nanofibras del copolímero seleccionado mediante la técnica del electrohilado. Estas nanofibras se cargaron con fármaco y se utilizaron como agentes reforzantes de matrices poliméricas biodegradables. Con el propósito de conseguir unos tejidos apropiados, se probaron distintas composiciones de fibras y diferentes configuraciones en el montaje del sistema de electrohilado.

Full textA host of endocytic pathways exist at the surface of eukaryotic cells, which lead to the internalization of the bulk of membranes along with membrane proteins, signaling receptors, growth factors, and other cargoes (Smith et al. 2017). For decades, the clathrin-mediated pathway has been the major well characterized endocytic process where clathrin polymerizes along with the associated adaptor proteins to include ligand-bound receptors, leading to membrane bending, membrane scission, and endocytosis (Smith et al. 2017). Recently, multiple alternative mechanisms have been uncovered which facilitate the endocytic uptake of cargo molecules and membrane receptors even in the absence of clathrin machinery (Mayor et al.2014). A model of endocytosis that doesn’t require clathrin but rather sugar-binding galectins and glycolipids has been proposed by my host laboratory (Lakshminarayan et al. 2014). Galectins constitute a family of beta-galactoside–binding lectins, which to date consists of 15 members in mammals. Galectins are broadly distributed in a variety of cells and tissues (Leffler et al. 2004). They are translocated from the cytosol to the extracellular space by a process of non-classical secretion (Hughes 1999). Glycosphingolipids (GSLs) are ubiquitous membrane constituents that are subdivided in neutral or acidic fractions. The term GSLs applies to compounds that contain at least one monosaccharide and a ceramide. Of note, the enzyme UDP-glucose ceramide glucosyltransferase (Ugcg) catalyzes the initial step for the biosynthesis of glycosylceramide-based GSLs.Our current working model, which involves glycolipids and lectins, was termed the GL-Lect hypothesis (Johannes et al. 2016). It is backed up by experimental data as described in Ref. (Lakshminarayan et al. 2014) and can be described as follows:i) Monomeric Gal3 binds to glycoproteinsii) Gal3 then starts to oligomerizeiii) Oligomerized Gal3 has the capacity to bind to glycosphingolipids and this may induce clustering of GSLsiv) Gal3-GSL cluster are inducing the invagination of the plasma membrane to generate tubular endocytic pits from which clathrin-independent carriers (CLICs, which are pre-early endosomes) are generated.Oligomeric Gal3 is indeed able to bind to GSLs and to induce membrane deformation (Lakshminarayan et al. 2014) in a similar way the pathogenic lectin Shiga toxin-B subunit (STxB) does. Therefore, both processes could be summarized under the same hypothesis, the GL-Lect hypothesis, where GL stands for the glycosphingolipids (Gb3 for STxB and gangliosides for Gal3) and Lect summarizes the lectins (STxB, Gal3 and possibly others as well).Understanding if this clathrin-independent but Gal3-dependent internalization mechanism is conserved not only in vitro model systems but in vivo is a main challenge in the field of trafficking.We characterized for the first time that in the gut a new mechanism facilitates endocytic uptake of cargo. This mechanism is driven by Galectin3 and operates in intestinal enterocytes for transcytosis like process and is glycosphingolipid dependent. Indeed, we have found that the lactotransferrin (LTF), a Gal3 cargo that we have identified by Mass spec, strongly required Gal3 and GSLs for its efficient endocytosis and its transcytosis like distribution pattern, respectively. Based on these findings in mouse intestinal epithelium, we established a functional in vivo model system where the newly proposed endocytic mechanism termed in our lab as GL-Lect, was physiologically investigated

Full textWe develop a new mesoscopic approach to crystal plasticity and apply it for the modeling of the homogeneous and heterogeneous nucleation of dislocations in 2D. Plasticity is modeled in the context of geometrically and physically nonlinear elasticity theory, by using Landau-type energy density generating a globally periodic energy landscape. The equivalent energy wells describe atomic configurations which can be mapped on each other by lattice invariant shears. This type of invariance is dictated by the global symmetry group of integer valued invertible matrices GL(2,Z).The resulting model accounts for this tensorial symmetry in the context of nonlinear elasticity with finite stretches and rotations. The activation of the ‘plastic mechanisms’, described in this model by the extended ravines in the energy landscape, is directed by the energy minimization which accounts automatically for the coupling between different slip planes. Such coupling is largely controlled by the saddle points corresponding to the unstable high symmetry phases. Then, we used to simulate the collective nucleation of dislocations with the main goal of quantifing the effects of crystal symmetry and sample orientation in the loading device. Our numerical simulations show that homogeneous nucleation results in the formation of the dislocation patterns which are are characterized by high spatial complexity. We perform a systematic comparison with atomistic simulations, which suggest that our mesoscopic model is capable of capturing the main atomistic effects. The main advantage of the new model is that short range dislocations interaction is accounted automatically, whitout ad hoc phenomenological realtions. Being designed for the study of the evolution of a large number of interacting dislocations, the proposed mesoscopic model opens new possibilities for studying the complexity of plastic flows in crystals associated with the emergence of scale free spatial and temporal correlations

Full textFoi avaliada, no presente estudo, a preval?ncia dos casos de tumores benignos e malignos de gl?ndulas salivares maiores e menores que ocorreram na popula??o atendida no hospital Dr. Luiz Ant?nio Natal/RN, no per?odo de 1989 a 2005, bem como as poss?veis associa??es existentes entre os aspectos cl?nico e patol?gicos exibidos pelos referidos tumores, visando ? obten??o de par?metros indicadores de diagn?stico e/ou progn?stico. Dos prontu?rios dos pacientes foram obtidas todas as informa??es cl?nicas necess?rias para a realiza??o do trabalho. A an?lise dos dados revelou que dos 303 tumores de gl?ndula salivar estudados, a maioria (71%) foram benignos, o mais comum foi adenoma Pleom?rfico. As m?dias de idades para os tumores benignos e malignos foi de 49,2 e 58,5 anos, respectivamente. Diferen?as estatisticamente significativas entre estes tumores foram observadas para as seguintes vari?veis: idade m?dia, o tamanho do tumor e dura??o da doen?a. Em rela??o ao tamanho do tumor, carcinoma mucoepidermoide mostrou-se 1,74 vezes menor que o de outros tumores malignos. Uma associa??o entre o diagn?stico histol?gico e vari?vel consist?ncia do tumor foi observada. Os dados apresentados neste estudo s?o relevantes para a compreens?o das diversas caracter?sticas exibidas por estes tumores, j? que corroboram uma s?rie de estudos anteriores

In the first part of this thesis, we obtain some new results about infinitesimal Cherednik algebras. They have been introduced by Etingof-Gan-Ginzburg in [EGG] as appropriate analogues of the classical Cherednik algebras, corresponding to the reductive groups, rather than the finite ones. Our main result is the realization of those algebras as particular finite W-algebras of associated semisimple Lie algebras with nilpotent 1-block elements. To achieve this, we prove its Poisson counterpart first, which identifies the Poisson infinitesimal Cherednik algebras introduced in [DT] with the Poisson algebras of regular functions on the corresponding Slodowy slices. As a consequence, we obtain some new results about those algebras. We also generalize the classification results of [EGG] from the cases GL, and SP2n to SOl. In the second part of the thesis, we discuss the loop realization of the affine Yangian of gl₁. Similar objects were recently considered in the work of Maulik-Okounkov on the quantum cohomology theory, see [MO]. We present a purely algebraic realization of these algebras by generators and relations. We discuss some families of their representations. A similarity with the representation theory of the quantum toroidal algebra of gl₁ is explained by adapting a recent result of Gautam-Toledano Laredo, see [GTL], to the local setting. We also discuss some aspects of those two algebras such as the degeneration isomorphism, a shuffle presentation, and a geometric construction of the Whittaker vectors.

Full textThis thesis is devoted to the mod p Langlands program for GL₂. In the first part, I study the Gelfand-Kirillov dimension of the representations π coming from the mod p cohomology of Shimura curves. Let p be a prime number and F a totally real number field unramified at places above p. Let r be a two-dimensional modular mod p representation of the Galois group of F that satisfies the Taylor-Wiles hypothesis and some technical genericity assumptions. For v a fixed place of F above p, we prove that many of the admissible smooth mod p representations of GL₂(Fᵥ) associated to r in the corresponding Hecke-eigenspaces of the mod p cohomology have Gelfand-Kirillov dimension [Fᵥ:Qp]. This builds on and extends the work of Breuil-Herzig-Hu-Morra-Schraen and Hu-Wang, giving a unified proof in all cases (r either semisimple or not at v). In the second part, I study the étale (phi,Gamma)-modules D_A(π) associated to the representations π coming from the mod p cohomology of Shimura curves and compute there ranks. Let K be a finite unramified extension of Qp. For π an admissible smooth mod p representation of GL₂(K) satisfying certain multiplicity-one properties, we compute the rank of the associatedétale étale (phi,Gamma)-modules D_A(π) defined by Breuil-Herzig-Hu-Morra-Schraen, extending their results. In the third part, I study the local-global compatibility properties of the étale (phi,Gamma)-modules D_A(π). For ⍴ any reducible two-dimensional mod p representation of the Galois group of K, we compute explicitly the associated étale (phi,Gamma)-module D_A(⍴) defined by Breuil-Herzig-Hu-Morra-Schraen. Then we let π be an admissible smooth mod p representation of GL₂(K) occurring in some Hecke eigenspaces of the mod p cohomology and ⍴ be its underlying two-dimensional mod p representation of the Galois group of K. Assuming that ⍴ is maximally non-split, we prove under some genericity assumption that the associated étale (phi,Gamma)-module D_A(π) defined by Breuil-Herzig-Hu-Morra-Schraen is isomorphic to D_A(⍴). Full textThis thesis consists of two parts: the first one gives a generalization of fiber spaces constructed above the Bruhat-Tits tree of the group GL(2) over a p-adic field. More precisely we construct a projective tower of spaces over the 1-skeleton of the Bruhat-Tits building of GL(n) over a p-adic field. We show that any cuspidal representation π of GL(n) embeds with multiplicity 1 in the first cohomology space with compact support of k-th floor of the tower, where k is the conductor of π. In the second part we constructed a space W above the barycentric subdivision of the Bruhat-Tits building of GL(n) over a p-adic field. To study the cohomology spaces with compact support of a proper G-simplicial complex X with a rather special equivariant covering, where G is a totally disconnected locally compact group, we show the existence of a spactrale sequence in the category of smooth representations of G that converges to the cohomology with compact support of X. Based on the latter results, we calculate the cohomology with compact support of W as smooth representation of GL(n), and then we show that the level zero cuspidal types of GL(n) appear with finite multiplicity in the cohomology of some finite simplicial complexes constructed in residual level. As a consequence, we show that the cuspidal representations of level 0 of GL(n) appear in the cohomology of W

Full textThis thesis is devoted to the construction of simple types for the reductive group GL(m, D), where m is a positive integer and D a finite dimensional division algebra whose center is a nonarchimedean local field. The underlying aim of this work is the explicit description of the set of irreducible smooth complex representations of GL(m, D) whose inertial support is reduced to one element. In a first stage, we produce, for each simple stratum of the matrix algebra M(m,D), a set of simple characters, related to those constructed by Bushnell and Kutzko in the split case by a transfert property. Those characters fulfill some remarkable properties, as an intertwining formula and a nondegeneracy property, allowing to build their Heisenberg representation defined on a certain compact open subgroup of GL(m, D). This construction is based on a unramified base change process, which allows us to make use of the results of Bushnell and Kutzko. In a second stage, when the underlying hereditary order of the stratum is principal, we build for each simple character corresponding to it an extension of its Heisenberg representation without reducing the intertwining (such an extension is called a beta-extension). Full textObesity is one of the leading causes of health care issues all over the world. A large slice of the worldwide population develops co-morbidities as a consequence of this condition, such as type 2 diabetes mellitus (T2DM), atherosclerosis, fatty liver, cardiovascular diseases and even some cancers, conditions that are associated with reduced life expectancy. The inefficiency of current therapies that aim to treat obesity leads to further investigation on novel therapeutic targets for this condition, including modulation of white adipose tissue (WAT). In WAT, glucose and lipid metabolism converge into triglyceride/ glycerol + free fatty acid (TG/ (GL + FFA)) cycle, which is considered a futile cycle since it doesn’t generate products while consuming energy substrates. The decreased activity of this cycle in obesity, decreases substrates oxidation and increases free fatty acids (FFA) storage in WAT. If coupled to increased mitochondrial β-oxidation, modulation of TG/ (GL+FFA) cycling provides an attractive therapeutic target to stimulate fat mobilization and prevent WAT dysfunction. Currently there are no known strategies that simultaneously stimulate both these processes. Chenodeoxycholic acid (CDCA) is a bile acid known for its anti-obesity effects; however the mechanism of action is still controversial. CDCA activates both peroxisome proliferator activating receptor (PPAR) α and PPARγ, regulators of the transcription of key metabolic genes involved in the TG/ ( GL+FFA) cycle, mitochondrial oxidative capacity and FFA oxidation. As such, we hypothesized that CDCA mediates the coupling of futile cycles with energy expenditure in WAT, increasing simultaneously futile acid activity and mitochondrial substrates oxidation. This work focused on evaluating the metabolic alterations of TG/ (GL+FFA) cycle, in culture 3T3-L1 cells exposed to CDCA. Specifically, we addressed if CDCA supplementation alters the TG/ (GL+FFA) cycle activity, if this was related to improved metabolic status and if mitochondrial metabolism was involved. Cultured 3T3-L1 cells exposed to CDCA exhibit decreased lipid accumulation and an increase in uncoupling protein-1 (UCP-1) content, which in turn leads to a decrease in reactive oxygen species (ROS) generation. CDCA also leads to an increase in glyceroneogenesis, suggesting an increase activity in TG/ (GL+FFA) cycle activity. Indeed, these results show that administration of CDCA seems to be an attractive therapy to obesity and T2DM. However, a question still remains, what leads to a reduction in triglycerides (TG) accumulation? Based on our results CDCA up-regulates genes involved in β-oxidation and mitochondrial biogenesis, but there are no differences in fatty acid oxidation between control and cell exposed to CDCA. A possible explanation is that lipids are being exported to the medium. However this is an in vitro study, it would be interesting understand the destination of this lipids in vivo. Finally the possible occurrence of metabolic compartmentalization opens new perspectives in futures works.
A obesidade é um dos principais problemas de saúde pública a nível mundial. A grande maioria da população obesa desenvolve patologias associadas que consequentemente levam à diminuição da esperança média de vida. Estas incluem diabetes tipo 2 (T2DM), aterosclerose, esteatose hepática, doenças cardiovasculares e ainda alguns tipos de cancro, Dado que as estratégias atuais são ainda pouco eficazes, no que se refere ao tratamento da obesidade, é urgente que se investiguem novas terapias, nomeadamente a modulação do tecido adiposo branco (WAT). No WAT, as vias metabólicas da glicose e dos lípidos convergem no ciclo dos glicerolípidos/ácidos gordos livres (TG/ (GL+FFA)), sendo este considerado um ciclo fútil dado que não gera produtos enquanto consome substratos energéticos. A diminuição da atividade deste ciclo em situações de obesidade e diabetes diminui a oxidação de substratos e aumenta o armazenamento de ácidos gordos (FFA) no WAT. Assim, a estimulação deste ciclo fútil surge como um potencial alvo terapêutico. Porém, a oxidação de substratos implica a estimulação simultânea do ciclo fútil e da função mitocondrial sendo que, até à data, não existe um mecanismo capaz de acoplar estes processos. O ácido quenodesoxicólico (CDCA) é um ácido biliar conhecido pelos seus efeitos anti-obesidade, no entanto, o seu mecanismo de ação é ainda controverso. O CDCA activa dois reguladores da transcrição de genes metabólicos envolvidos no ciclo TG/ (GL+FFA), na capacidade oxidativa mitocondrial e na β-oxidação, o PPARα e o PPARγ. Assim, a nossa hipótese propõe que o CDCA medeia o acoplamento do ciclo fútil com o gasto energético no WAT, aumentando simultaneamente as taxas de atividade do ciclo fútil e de oxidação mitocondrial de substratos. Este trabalho foca-se na avaliação das alterações metabólicas no ciclo TG/ (GL+FFA), na linha celular 3T3-L1, induzidas pelo CDCA. Especificamente, avaliou-se se a exposição ao CDCA altera a atividade deste ciclo, se essa alteração está relacionada com uma melhoria no perfil metabólico e ainda se o metabolismo mitocondrial está envolvido. As células expostas ao CDCA demonstraram uma diminuição na acumulação de lípidos e um aumento no conteúdo de proteína desacopladora-1 (UCP-1), o que por sua vez diminui a produção de espécies reactivas de oxigénio (ROS). Adicionalmente, a exposição ao CDCA induziu um aumento na gliceroneógenese, sugerindo, assim, um aumento na atividade do ciclo TG/ (GL+FFA). Dessa forma, a administração de CDCA parece ser uma estratégia terapêutica atrativa no combate à obesidade e à T2DM. No entanto, uma questão mantém-se, o que leva à redução da acumulação dos triglicerídeos (TG)? Com base nos resultados obtidos, o CDCA aumenta a expressão dos genes envolvidos na β-oxidação e na biogénese mitocondrial, mas não existem diferenças na oxidação dos FFA entre o controlo e as células expostas ao CDCA. Uma possível explicação é que os lípidos estejam a ser exportados para o meio. No entanto, este é um estudo in vitro, logo seria interessante perceber o destino destes lípidos in vivo. Finalmente a possível ocorrência de compartimentalização metabólica abre novas perspectivas para trabalhos futuros.

Full textEsta pesquisa teve como objetivo geral analisar as políticas educativas de jovens e adultos no Brasil, seus contornos na transição do milênio, entre 2000 e 2015, bem como suas relações com a Agenda Globalmente Estruturada para a Educação (AGEE), e os tensionamentos promovidos no âmbito da sociedade civil. Preconizada por Roger Dale (2010), a AGEE possibilita compreender os impactos da globalização na reconfiguração do papel do Estado, e como os Organismos Internacionais (OI) se tornam representantes dessa agenda para induzir e padronizar as políticas públicas em âmbito transnacional. Partimos da hipótese de que a política internacional, que vem se consolidando no Brasil, integra uma agenda globalmente estruturada para a educação, que envolve agendas globais como a Educação para Todos (EPT), os Objetivos do Desenvolvimento do Milênio (ODM), os Objetivos para o Desenvolvimento Sustentável (ODS) e as que contemplam a EJA, como as Confinteas. Como suporte teórico, operamos com as categorias globalização, política educativa e suas relações com o Estado na formulação das políticas públicas, com base na sociologia política (IANNI, 1994; 1997), (HELD, 1984; 2002) e (TORRES, 2001; 2003) e na teoria política (GRAMSCI, 1978; 2001), (DUSSEL, 1977; 1993). Os fundamentos epistemológicos do Realismo Crítico (BASKAR,1978) são tomados como referência para a construção das análises que fomos produzindo no decorrer da investigação, sustentando assim metodologia mista que envolveu: pesquisa documental; recolha de dados oficiais do Instituto Brasileiro de Geografia e Estatística (IBGE), da Pesquisa Nacional de Amostra Populacional (Pnad) e Instituto Nacional de Estatística e Pesquisa Anísio Teixeira (INEP), bem como;a utilização da ferramenta NVivo 11 co mo ferramenta qualitativa de análise de dados.No processo investigativo descrevemos e buscamos analisar criticamente os relatórios do Banco Mundial (BM), da Organização para a Cooperação do Desenvolvimento Econômico (OCDE), da EPT, ODM, ODS e V e VI Confinteas. Em âmbito nacional, os documentos específicos da Educação de Jovens e Adultos e os relatórios dos Encontros Nacionais de educação e Jovens e Adultos (ENEJA) estiveram em foco. As análises realizadas nos levam a afirmar a incidência da lógica neoliberal expressa nas orientações dos OI sobre as políticas educativas de EJA que vêm se desenvolvendo no Brasil, no período estudado, através de mecanismos, processos e eventos que evidenciam uma reconfiguração do papel do Estado voltado para responder às demandas do mercado em detrimento das demandas da sociedade civil. As políticas de redução da pobreza, de promoção da alfabetização, de focalização nos grupos mais vulneráveis, desenvolvimento econômico e a ênfase na educação e aprendizagem ao longo da vida, dentre outras categorias, incidem expressamente na formulação das políticas educativas de EJA, desafiando a capacidade de tensionamento da sociedade civil, em específico os Fóruns de EJA, na mudança dos rumos das políticas educativas. Reafirma-se, assim, a submissão do Estado brasileiro a uma Agenda Globalmente Estruturada para a Educação.

Full textEste estudo teve por objetivo avaliar o efeito da laserterapia no funcionamento da gl?ndula tire?ide, atrav?s da mensura??o dos n?veis hormonais de triiodotironina (T3) e tiroxina (T4) no soro sang??neo de coelhos. Para tanto, foram utilizados 24 coelhos da ra?a Nova Zel?ndia, machos, distribu?dos aleatoriamente em tr?s grupos: o grupo controle (animais n?o irradiados), grupo EI (laser aplicado sobre o alv?olo p?s-extra??o do incisivo inferior) e grupo EII (laser aplicado sobre a regi?o da gl?ndula tire?ide). Todos os animais foram submetidos ? exodontia do incisivo inferior esquerdo, criando uma condi??o cl?nica inicial de igualdade entre os grupos. Os animais dos grupos experimentais foram irradiados, com o laser GaAlAs (λ=830 nm, 40mW), durante 13 dias, a cada 48 horas, com uma dose de 120 J/cm? por sess?o. O protocolo de irradia??o foi iniciado imediatamente ap?s o procedimento cir?rgico nos animais. As dosagens hormonais de T3 e T4 foram realizadas nos tr?s grupos antes da exodontia e no ?ltimo dia de observa??o do experimento (15? dia). Os resultados obtidos n?o demonstraram diferen?as estatisticamente significativas nos valores de T3 e T4 pr? e p?s-aplica??o do laser entre os grupos estudados. Conclui-se que a laserterapia, no protocolo de irradia??o utilizado neste estudo, n?o alterou os n?veis hormonais de T3 e T4 circulantes no soro de coelhos.

In Brazil, accidents with scorpions are considered of medical importance, not only by the high incidence, but also for the potentiality of the venom from some species in determining severe clinical conditions. Tityus stigmurus is a widely distributed scorpion species in Northeastern Brazil and known to cause severe human envenomations, inducing pain, hyposthesia, edema, erythema, paresthesia, headaches and vomiting. The present study uses a transcriptomic approach to characterize the molecular repertoire from the non-stimulated venom gland of Tityus stigmurus scorpion. A cDNA library was constructed and 540 clones were sequenced and grouped into 37 clusters, with more than one EST (expressed sequence tag) and 116 singlets. Forty-one percent of ESTs belong to recognized toxin-coding sequences, with antimicrobial toxins (AMP-like) the most abundant transcripts, followed by alfa KTx- like, beta KTx-like, beta NaTx-like and alfa NaTx-like. Our analysis indicated that 34% include other possible venom molecules , whose transcripts correspond to anionic peptides, hypothetical secreted peptides, metalloproteinases, cystein-rich peptides and lectins. Fifteen percent of ESTs are similar to cellular transcripts. Sequences without good matches corresponded to 11%. This investigation provides the first global view of cDNAs from Tityus stigmurus. This approach enables characterization of a large number of venom gland component molecules, which belong either to known or atypical types of venom peptides and proteins from the Buthidae family
No Brasil, acidentes com escorpi?es s?o considerados de import?ncia m?dicosanit?ria, n?o somente por sua incid?ncia, mas tamb?m pela potencialidade do veneno de algumas esp?cies em determinar quadros cl?nicos graves. Tityus stigmurus ? uma esp?cie de escorpi?o amplamente distribu?da na regi?o Nordeste do Brasil sendo conhecida por causar graves envenenamentos humanos, cujos sintomas incluem: dor, hipoestesia, edema, eritema, parestesia, cefal?ia e v?mitos. A composi??o qu?mica do veneno do escorpi?o T. stigmurus ainda n?o foi bem avaliada, havendo uma car?ncia na literatura de estudos com enfoque na elucida??o do repert?rio molecular dos componentes deste veneno. Neste trabalho, realizamos uma abordagem transcript?mica para caracterizar o repert?rio molecular da gl?ndula de veneno n?o-estimulada do escorpi?o Tityus stigmurus. Para tanto, uma biblioteca de cDNA foi constru?da e, ? partir dela, 540 genes foram clonados e agrupados em 37 clusters com mais de um EST (expressed sequence tag) e 116 singlets (somente um EST). Do total de transcritos, 41% pertencem a sequ?ncias de toxinas conhecidas, sendo os pept?deos antimicrobianos (AMPs) os transcritos mais abundantes, seguido por α-KTX, β- KTX, β-NaTx e α-NaTx, respectivamente. Nossa an?lise revelou ainda que 34% s?o "poss?veis toxinas", cujos transcritos correspondem a pept?deos ani?nicos, pept?deos hipot?ticos secretados, metaloproteases, pept?deos ricos em ciste?na e lectinas. Quinze por cento dos ESTs s?o semelhantes a transcritos celulares. Transcritos sem similaridade com outras sequ?ncias do GenBank, foram chamadas de no hit e correspondem a 11% do total. Este trabalho apresenta a primeira vis?o global de cDNAs das gl?ndulas venenosas do escorpi?o Tityus stigmurus. Esta abordagem permite a caracteriza??o de um grande n?mero de mol?culas componentes da gl?ndula de veneno, que pertencem a tipos conhecidos ou at?picos de pept?deos e prote?nas do veneno da fam?lia Buthidae

Full textThis thesis is a study of finite dimensional representations of the Lie superalgebra gl(m,n). In the first chapter we recall some results on these Lie superalgebra. In the second chapter we study the simple representations of gl(2.2). These modules can be obtained as quotient of some induced modules, the knowledge of the composition series of these modules allow us to compute an explicit finite character forumula for simple modules. In the third chapter we look at representations of a quantum deformation of the universal enveloping algebra of gl(m,n). We first recall the construction of crystal bases for the direct factors of a tensor power of the standard representation. We show by weakening the definition of crystal, that there exist crystal bases for non-semisimple modules, and we give a way to construct them. The fourth chapter focuses on the understanding of the maximaly atypical block of the category of finite dimensional representations of gl(2.2). Knowing the full subcategory of projective maximally atypical modules allows us to reconstruct the category. First, we study the projective indecomposable modules, and we compute their Loewy series. We then study their morphisms. Finally we make a conjecture on the composition of those morphisms
