Full textThe Fusarium mycotoxin Deoxynivalenol (DON) leads to numerous adverse health effects in animals and causes serious economic losses. Plants can defend themselves against this toxin by conjugating glucose to DON. The resulting masked mycotoxin deoxynivalenol-3-β-D-glucoside (D3G) is frequently occurring in food and feed. There are major concerns that D3G is hydrolyzed in the digestive tract of mammals, thus increasing the total DON load of an individual. Due to a lack of in vivo data D3G has not been included in the regulatory limits established by the European Commission for DON. Therefore, the aim of our study was to clarify whether orally administered D3G is hydrolyzed in rats and liberated DON is subsequently absorbed. Using a repeated measures design, six Sprague-Dawley rats received water, DON (2.0 mg/kg body weight; bw) and the equimolar amount of D3G (3.1 mg/kg bw) by gavage on days 1, 8 and 15, respectively. After each application, the animals were housed individually for 48 h in metabolic cages to collect urine and feces. The concentrations of D3G, DON, deoxynivalenol-glucuronide (DON-GlcA) and de-epoxydeoxynivalenol (DOM-1) in the excreta were determined by a validated LC-MS/MS based method. After administration of D3G, the masked mycotoxin itself as well as DON, DON-GlcA and DOM-1 were detected in the urine of rats. In total, 3.7 ± 0.7% and 14.9 ± 5.0% of the administered dose were recovered in urine after application of D3G and DON, respectively. Urinary eliminated D3G represented only 0.3 ± 0.1% of the given dose, thus indicating a very low bioavailability of this masked mycotoxin in rats. The majority of administered D3G was recovered as DON and DOM-1 in feces. This study clearly demonstrated that D3G is hydrolyzed in the digestive tract of rats. The liberated DON is metabolized to DOM-1 and DON-GlcA, but only poorly absorbed. Our data indicate that D3G is of considerably lower toxicological relevance than DON in rats.

Full textLegionella pneumophila is a Gram-negative proteobacterium causing Legionnaires’ disease. The results of this study confirm that serine is a carbon source for amino acids and that L. pneumophila is auxotrophic for the amino acids isoleucine, leucine, phenylalanine, tyrosine, histidine, proline, and valine. L. pneumophila uses amino acids of the host cells to incorporate them into proteins. Serine is synthesized de novo. Furthermore, this work demonstrates for the first time that glucose is used for the biosynthesis of amino acids and polyhydroxybutyrate (PHB). The Entner-Doudoroff pathway is identified as the predominant pathway for the catabolism of glucose. In a nutrient-depleted environment, after replication has taken place in A. castellanii, a deletion strain of the Entner-Doudoroff pathway exhibits strongly reduced fitness as compared to the wildtype. The glucoamylase GamA is characterized as the enzyme responsible for the starch and glycogen degrading activity of L. pneumophila for the first time. The putative transcription activator YozG binds to the 5’ region of gamA as well as to his own putative promoter region and regulates GamA activity. Furthermore, arguments are provided to support the hypothesis that YozG also acts as a cis-active element. The biosynthesis of polyhydroxybutyrate (PHB) from glucose starts in the late exponential phase and increases in the stationary growth phase. As an explanation for reduced fitness of the Entner-Doudoroff deletion strain in nutrient-depleted environment, a reduced amount of PHB is proposed. The results of this work prove that L. pneumophila uses not only amino acids but also glucose and the natural glucose polymers starch and glycogen as carbon sources for the biosynthesis of amino acids and PHB and that thereby the fitness of the species is increased.

Full textAim: The aim of this study was to investigate whether oral administration of two milliliters of 30 % glucose have an analgesic effect during vaccination against measles, mumps and rubella in 18-month-old children. Method: The sample group consisted of 18-month-old children who had their 18-month visit at Samariterhemmet or Eriksberg’s healthcare center (intervention group) or Flogsta healthcare center (control group) during the period of the study. The total amount of participants was 24 children; 14 children in the intervention group were observed at Samariterhemmet and Erikbergs healthcare center and 10 children were observed in the control group at Flogsta healthcare center. The study was conducted as a quantitative quasi-experimental study. During the visits the duration of the child’s crying/screaming and the parents’ perception of their child’s pain were observed and noted. Results: When measuring the duration of the child’s crying/screaming, two milliliters of 30 % glucose could not be considered to have any analgesic effect on procedure-related pain. Parents in the study group perceived their children as having more pain than the parents in the control group. Conclusion: Since this study could not show any positive analgesic effect attributed to the administration of 30 % glucose in 18-month-old children this procedure cannot be recommended for this age group. However, this study has its limitations, the low amount of participants should be considered and also the fact that eight out of ten children in the control group received a different type of vaccine (Priorix) than the rest of the sample group (MMR-II). This vaccine has been found to cause less pain than MMR-II.

Full textThis work presents the analysis of the enzymatic function of histidine acid phosphatases (HAPs) exemplary studied on two enzymes: the phytase of Klebsiella sp. ASR1 (PhyK) and the glucose-1-phosphatse (AgpE) of Enterobacter cloacae. The AgpE belongs to the HAPs and shows lower catalytic activity for phytate. Besides biochemical analysis the structure of these two enzymes were solved. Binding of phytate and the inhibitor myo-inositolhexa sulfate at the active site of these enzymes were fitted according to the date obtained by co-crystallisations experiments. For the first time two district conformational states could be observed for a glucose-1-phosphatase (AgpE). The open and closed conformations as well as the structures were very similar to the well characterized AppA phytase of E. coli however bothe enzymes showed differences in catalytic efficiency. For this reason substitutions of amino acid within the active site were performed to change substrate spectra and specific activities. The substitutions of only two amino acids strongly changed the substrate spectrum and affected the further phytate hydrolysis by the AgpE. Extensive amino acid substitutions were performed for the phytase of Klebsiella sp. ASR1 as well. In this case phytase mutants with 20% higher specific phytase activities were generated. Besides that, the observed strong substrate-induced inhibitory effect on the phytase activity was lower in these mutants. The biochemical characterizations and high performance ion chromatography analysis (HIPC) of these phytase mutants allowed drawing conclusions of the importance of particular amino acid residues for substrate binding and substrate hydrolysis. Furthermore the combination of Klebsiella phytase mutant and AppA resulted in a more efficient phytate hydrolysis. This aspect offers an interesting possibility for usage of the Klebsiella phytase in livestock breading and animal feeding.

Full textToday there are currently living around 365 000 people with diabetes in Sweden. Measuring of blood sugar can be experienced as painful and difficult with blood glucose monitoring devices found in the market. Using lancets and test strip makes direct contact with blood which increases the risk of infection. To reduce these risks and difficulties a non-invasive method is required. This method can even provide an opportunity for healthy individuals to monitor their blood glucose and prevent the disease.This thesis presents the disease, sequelae and the problem around blood glucose monitoring. The work is based on an earlier study about optic measurement of glucose. By illumine a solution of saline and glucose, with infrared-light from a LED, absorption is carried out by the glucose. The amount of transmitted light is detected and processed by a photodetector which generates a current that is converted to a voltage and amplified. This report examines a modification of the circuit used in the previous study to establish a correlation between the glucose concentration and the output voltage by adding a transimpedance amplifier around the photodetector.The results corroborate that the modification with the transimpedance gives a better correlation between the measured output voltage and glucose concentration. The measurements were compiled and plotted to compare which circuit gave a better correlation between the measured output voltage and glucose concentration. The study showed that the modification with transimpedance amplifier gives a better correlation, though it is not enough for clinical use.

Full textConventional heat-sterilized glucose-containing peritoneal dialysis fluids (PDF) are bioincompatible due to their acidic pH, high glucose concentration and resulting hyperosmolality, and the presence of glucose degradation products (GDP). Alternatives to these solutions are PDL containing amino acids or icodextrin as the osmotic agent. Furthermore, novel glucose-based PDF contain only trace amounts of GDPs due to the sterilisation of glucose at very low pH in a dual-chamber container system. The present study examines the acute and chronic effects of different PDL on human peritoneal mesothelial cells (HPMC). Confluent HPMC were exposed to the different test PDF (glucose containing heat- versus filter-sterilised PDF and conventional versus dual-chambered glucose PDF, 1% amino-acid PDF and icodextrin – all at neutral pH) in an acute (1-4 hours preincubation) and chronic (up to 10 days) cell culture model. Cell viability (MTT assay) and IL-1beta stimulated IL-6 (cell function) release were assessed. Acute and chronic exposure of HPMC to heat-sterilised PDF resulted in a significant reduction of viability and cell function. In contrast, the incubation of filter-sterilised PDF and dual chambered low GDP solution had only minor effects on cell viability and function. Neither viability nor cell function were negatively affected by the amino-acid PDF following acute and chronic exposure. However, incubation with icodextrin resulted in a similar degree of inhibition as compared to incubation with conventional heat-sterilised glucose PDF. In conclusion, removal of GDP from PDF either via filter-sterilisation or manufacture in dual chambered containers helps to conserve viability and function of HPMC in vitro. However, the replacement of the osmotic agent per se does not necessarily result in improved biocompatibility.
