The 5 year survival rate can be as high as 80% for cases diagnosed early, but the asymptomatic nature of the disease means that it is most frequently detected in the later stages. This thesis describes attempts to investigate the regulation of the Candida albicans hyphal-specific gene HYR1 by a functional dissection of the HYR1 promoter, protein localisation studies and analysis of HYR1 expression in C. albicans morphological mutants. In this thesis I show that macrophages (both cell lines and primary cultures) can be transfected by recombinant adenoviruses expressing -galactosidase (A-gal/Av1Bng), that the macrophages become activated by the transfection process and can be easily manipulated to localise to inflamed glomeruli after direct injection into the renal artery of rats with an experimentally induced glomerular inflammation caused by nephrotoxic nephritis. The injection of transfected macrophages reduces the severity of injury in this model of glomerulonephritis as shown by a reduction in the degree of albuminuria.

As part of the EUROFAN Transcript Consortium, northern analysis was used to study the expression of 200 yeast orphan genes. Five transient growth conditions were analysed, including glucose upshift, stationary phase, nitrogen starvation, osmotic shock, and heat shock. Around 72% of the genes analysed were expressed at low level, about 12% were expressed at a medium level, and 3.5% at a high level relative to the ACT1 mRNA. Fatty acids and glucose are fuels of major importance to mammalian tissues, and their uptake is controlled by two key proteins, lipoprotein lipase (LPL), and in insulin-sensitive tissues, the glucose transporter GLUT4. To study its role in bone metabolism I examined a transgenic mouse with a targeted deletion of the col1A1 first intron (col-Int). The role of tumour suppressor genes, in particular p53 and DCC, was investigated in human ovarian cancer. p53 codes for a DNA-binding nuclear phosphoprotein whose expression, in the wild-type form, is essential in the control of the cell cycle. Loss of function of this gene by mutation allows unrestrained cell proliferation to occur. The function of the DCC gene is not yet fully understood.
