Abstract:
In a split N-application system, the objective was to quantify N/15N in gluten and non-gluten proteins after the late application of 30 or 60 kg N, whereby 10% of the third split was applied as 15N. This fertilization was combined with a reduced water supply for 21 days (well-watered (ww); water deficit (wd)). German spring wheat cultivars, Elite wheat Taifun, Quality wheat Monsun and cultivars from the Mediterranean territory, Golia, Gönen, were examined. The protein content in gluten was for 30 kg N, ww, similar for Taifun, Golia, and Gönen, but markedly lower in Monsun (231, 245, 247, 194 mg protein/g DM). The water deficit increased the protein content in the gluten of Golia and Gönen and was higher than that of Taifun and Monsun (297, 257, 249, 202 mg protein/g DM). Fertilization of 60 kg N, ww, did not result in any change in the protein content in gluten and differences between the cultivars were not detectable. The 15N protein in gluten was for 30 kg N, ww, markedly higher in Gönen (2.32 mg 15N protein/g DM), compared to Golia and Monsun (1.93, 1.50 mg 15N protein/g DM), and similar in Taifun (1.64 mg 15N protein/g DM). 15N fertilizer uptake into gluten was stimulated by water deficit for 30 and 60 kg N, leading to significantly increased 15N protein in Golia and Gönen, (2.38, 2.99, 4.34, 5.87 mg 15N protein/g DM). Fertilization of 60 kg N led to a proportional two-time increase in the 15N gluten protein of the four cultivars, in ww and wd plants. Assessed on the basis of 15N fertilizer allocation under wd conditions into gluten proteins, Golia and Gönen have a stronger sink activity, compared to Taifun and Monsun.

Abstract:
Starting with 1981, the objectives of the breeding programmes in the Research Station for Fruit Growing (RSFG) Iaşi, Romania, took into consideration the on-going improvement of the sweet cherry assortment with cultivars having fruit-bearing precocity, with great productions, self-fertile, with decreased trees’ vigour, crown compactness, resistant to anthracnose, monilia, frost and fruit’s cracking, flowering lateness, superior quality of the fruits and ripening time at the extremities of the sweet cherries’ maturation season. As a result of the breeding programmes that took place up till present at RSFG Iaşi, 28 new sweet cherry cultivars, obtained through controlled hybridization, free pollination and conservative clonal selection, were homologated during 1994-2017. The present results refer to a study led during 2015-2017, having as biological material nine sweet cherry cultivars (‘Cetăţuia’, ‘Cătălina’, ‘Golia’, ‘Bucium’, ‘Ştefan’, ‘Iaşirom’, ‘Oana’, ‘Radu’ and ‘Ludovan’) obtained through controlled hybridization. The comparision of the cultivars has been performed versus their genitors ‘Van’ (♀) and ‘Boambe de Cotnari’(♂). There have been taken observations and determinations concerning the trees’ vigour, resistance to frost and anthracnosis, the main growing and fructification phenophases, physical and chemical treats of the fruit. The highest values regarding fruit’s weight (7.5-8.9 g) have been recorded for the ‘Ludovan’, ‘Bucium’, ‘Ştefan’, ‘Iaşirom’ and ‘Golia’ cultivars, while the values of the soluble substance content have been between 15.20-19.25 °Brix, the titratable acidity has been between 0.455-0.764 g malic acid 100 mL-1 of fresh juice and the total content of polyphenols has recorded values between 97.41-574.95 mg GAE 100 mL-1 of fresh juice. The hybridization between ‘Van’ and ‘Boambe de Cotnari’ has allowed the obtaining of valuable cultivars, that got remarked by earliness (‘Cetăţuia’, ‘Cătălina’), decreased trees’ vigour (‘Ştefan’, ‘Golia’), particular quality of the fruits (‘Ludovan’, ‘Iaşirom’, ‘Golia’, ‘Bucium’, ‘Ştefan’, ‘Oana’, ‘Radu’) and resistance to diseases and frost (‘Bucium’, ‘Iaşirom’).

In questo contributo viene esaminato il racconto Golia di Beppe Fenoglio con l’obiettivo di individuare particolari scelte linguistiche e stilistiche finalizzate alla rappresentazione della comunicazione tra italiani e tedeschi. In particolare, si analizza l’uso del tedesco da parte degli italiani e viceversa. Grande risalto, inoltre, è dato ai procedimenti di semplificazione linguistica tipica dei discenti di una lingua sconosciuta. Dopo un confronto con altri testi di argomento simile, l’analisi si chiude con alcune proposte interpretative circa le scelte linguistiche determinate dalla concezione fenogliana della Resistenza.

The studies were conducted at Research Station for Fruit Growing Iași in the years 2018-2020 on six cultivars of sweet cherry tree (‘Regina’, ‘Kordia’, ‘Hudson’, ‘New Star’, ‘Maria’ and ‘Golia’). The paper presents aspect of vegetative pomological parameters under the environmental factors of Romanian Northeastern area. The average of the precipitation for the three years studied was 475 mm, with a deviation of 43.05 mm from normal with a minimum in 2020 (443.6 mm/year). The average values of the area of the trunk section (TCSA) based on the average of three years of study for the six cherry cultivars recorded values between 67.77 cm2 (‘Maria’) and 121.58 cm2 (‘Regina’), with significant differences between cultivars. The crown volume had maximum values for the ‘Golia’ (4.56 m3/tree) and ‘Regina’ (4.53 m3/tree) cultivar and minimum values for the ‘Maria’ (3.78 m3/tree) and ‘Kordia’ with 3.92 m3/tree. The average of annual shoots and their average length recorded insignificant values among the cultivars studied.

From 1994 - 2016, the Research Station for Fruit Growing (RSFG) Iași approved 28 cherry cultivars, of which three were early cultivars (Cetăţuia, Cătălina and Amaris), 20 were middle maturation cultivars (Maria, Golia, Ştefan, Bucium, Iaşirom, Tereza, Lucia, Radu, Oana, Alexus, Andreiaş, Ludovan, Cociuvaş, Iosifan, Paulică, Mihailis, Muşatini, Elaiaşi, Amar Maxut, and Amar Galata), and five had late maturation (Andante, Marina, Croma, Margonia, and George). Van, as the most widespread cultivar in Romanian orchards, was used as a control. The sweet cherry cultivars were created through direct hybridisation or free pollination. Maria Cultivar is the first Romanian self-fertile cultivar. The locally approved bitter cherry biotypes are Amar Maxut with black fruit, Amar Galata with double-coloured fruit, and Amaris with dark red fruit. Regarding the tree’s vigour, cultivars Amaris, Tereza, Ştefan, and Golia recorded weak vigour, in comparison with the other cultivars, which had medium vigour. Bitter-tasting cherry cultivars have semi-firm pulp with semi-adherence to the stone, and the fruit weight was under 7 g. All 28 cultivars presented high precocity, productivity, fruit quality, and resistance to frost, drought, and diseases.

Objetivou-se neste trabalho avaliar o enraizamento, a brotação e o desenvolvimento de raízes de diferentes porta-enxertos de videira em condições de campo. Estacas lenhosas dos porta-enxertos '420 A', 'Golia', '5C', '8B', 'RR101-14', 'SO4', '99R', 'Kober 5BB', 'IAC 766', 'IAC 572', 'IAC 571-6', 'Ripária do Traviú' e 'Rupestris du Lot' foram colocadas em canteiro de terra, sem tratamento prévio. O delineamento foi em blocos ao acaso, com cinco repetições e vinte estacas por parcela, com as estacas dispostas em espaçamento de 12 x 5cm. As estacas foram colocadas para enraizar no início de julho e removidas no final de setembro para as avaliações. A porcentagem de estacas enraizadas variou de 79% para 'Ripária do Traviú' a 99% para o 'RR101-14'. Quanto à brotação, o 'Ripária do Traviú' apresentou 47%, 'IAC 571-6', 'IAC 572', '420 A', 'Rupestris du Lot', 'Kober', 'IAC 766', '8B', '5C', apresentaram de 76 a 89% e 'Golia', 'SO4', '99R' e 'RR 101-14' mais de 90%. 'IAC 572' e 'IAC 571-6' apresentaram o menor número de raízes por estaca, no entanto, foram as que apresentaram raízes mais desenvolvidas, seguidas pelo '5C' e 'Rupestris du Lot'. 'Kober 5BB' e 'Ripária do Traviú' apresentaram as raízes menos desenvolvidas. As demais variedades apresentaram valores intermediários. Concluiu-se que, entre todos os porta-enxertos, o 'Ripária do Traviú' apresentou os menores índices de enraizamento e brotação das estacas, nas condições de campo.

Em Monte Alegre do Sul, SP, efetuou-se um experimento de longa duração para observar o comportamento do cultivar IAC 138-22 (Máximo) de uvas para vinho, sobre cinco porta-enxertos: RR 101-14, 'Golia', "Ripária do Traviú', IAC 572 e IAC 313. Em oito colheitas sucessivas anuais, de 1974 a 1981, analisaram-se as produções médias de uvas por planta. Os resultados indicaram que o IAC 572' foi o melhor porta-enxerto para o IAC 138-22 'Máximo'. O IAC 313 superou o IAC 572 em um dos anos estudados. Todos os outros porta-enxertos apresentaram desempenho inferior.

Last five year statistics show that av-eragely 8900 theft incidents reported in a yearat police which is 1036 or 11.7 percent out of allcrimes adjudicated. As for sentence type andlength, 80 percent of defendants were im-posed imprisonment. It reveals that sentencepolicy has been very strict towards defendants.

Abstract:
The study in this paper was carried out as a result of the observation of pollution phenomena and foaming effects associated with anthropogenic activities, including street cleaning activity. The processes of dust binding used in order to reduce PM10 and PM 2.5 pollution has been proven to be inefficient, and even contributing to pollution with particulate matter. Our results suggest that the use of dust binders must be integrated in a technique that includes methods of removing agglomerated particle structures resulting from the process of coagulation or flocculation. These are the conclusions of the investigations carried out by spectroscopic methods (FTIR, SEM-EDX) on samples collected from the streets of Iasi on 10 March 2021, and on samples collected from the surface of the Precinct Wall of the historical monument Golia—Iasi Monastery Ensemble (Romania). On the later samples, coloristic analysis was also performed. The alert for investigation was given by the foaming waters that were leaking on the streets. The phenomenon was observed after the streets had been washed by specialized vehicles. Analyses revealed compounds used as dust binders and coagulant type (aluminum sulfate, sodium aluminate and their derivatives, plus anti-skid chemicals such as calcium chlorine and magnesium chlorine), as well as organic compounds included in aggregate type structures, and they showed contamination of the Golia Precinct Wall. The results show that the dust binders or coagulants used as such, or embedded in various products intended for the cleaning process of streets or other outdoor public places, must be subject to regulation. Otherwise, there is a risk of adding more pollutants during an operation with the opposite purpose. The migration of these pollutants on the studied building offers an image on how both our health and all constructions and equipment exposed in the open air are affected.

Abstract:
The biological properties of the sweet cherry (Prunus avium L.), as well as the physico-chemical characteristics of the fruit were studied for 3 years within RSFG Iași (2018-2020). The investigations were carried out at the Sârca Research Base on six varieties of sweet cherries. These included following the phenological stages (flowering period and fruit ripening period), but also the particularities of the fruit in terms of physical characteristics and biochemical composition. The highest weight of the fruit was recorded in 2020 in the ‘Regina’ cultivar. ‘Regina’ is characterized by an average flowering period, lasting between April 11th and 28th. The earliest cultivar both in terms of flowering and fruit ripening is the ‘Golia’ cultivar. All other varieties have an average fruit ripening period (June 15th-30th). The soluble dry matter content was between 11.29 (‘Hudson’) and 18.61 (‘Maria’). The researches carried out and the results obtained show that the studied varieties presented good results of adaptability in the pedoclimatic conditions in the area of Romania.

The development of the Oyu Tolgoi copper-gold mine, located in Mongolia's South Gobi province, promises to rebuild the nation after two decades of economic and social instabilities following the 1990 revolution. While the company promotes the mine as the teleological solution to Mongolia's development, the state and public remain ambivalent, as concerns about a resource curse and Dutch Disease loom. In this paper, I argue that Oyu Tolgoi remains contested due to tensions between corporate and state actors as well as public concerns about the potential negative political, economic, and environmental effects of mining. Debates over the Oyu Tolgoi investment agreement negotiations and the immediate repercussions of the agreement signing reveal how the dual teleologies of building mineral nations crystallize in the neologism “Mine-golia.” This paper begins to fill a gap in the literature on mineral nations which privileges the role of the state, leaving how corporations engage in nation-building underexamined.

Mudas oriundas de estacas enraizadas de 17 porta-enxertos de videira ('SO4', 'Teléki 8B', 'Harmony', 'Golia', 'Riparia de Traviú', 'Paulsen', '420 A', '99R', '5C', 'RR101-14', 'Kober 5BB', 'Rupestris Du Lot', 'IAC 313 Tropical', 'IAC 766 Campinas', 'IAC 572 Jales', 'VR 043-43' e 'IAC 571-6 Jundiaí') foram levadas ao campo no fim da primavera, em condições subtropicais. Após quatro meses, avaliou-se a severidade da antracnose e ferrugem da videira nas folhas das brotações dos porta-enxertos e após cinco meses o vigor dos mesmos. No inverno seguinte, foi realizada a enxertia por garfagem da variedade 'BRS Violeta' e após 60 dias, avaliaram-se a porcentagem de brotação, o diâmetro e o comprimento médio do enxerto. Nos porta-enxertos 'IAC 572 Jales' e 'IAC 313 Tropical' apresentaram menor infecção foliar a antracnose e ferrugem. O porta-enxerto 'IAC 572 Jales' proporcionou maior desempenho em campo, previamente à operação de enxertia, enquanto os porta-enxertos 'SO4', 'Harmony', 'Paulsen' e 'IAC 766 Campinas' promoveram maior vigor ao desenvolvimento do enxerto 'BRS Violeta'.

The idea of writing this first article about the notes made on the cult books from the heritage of the Metropolitanate of Moldavia and Bucovina, came at the moment when as a museographer of the young Metropolitan Museum of Iași, I had to inventory the mobile cultural heritage, which had just been transferred from the concentration warehouse organized by the Metropolis, at the Golia Monastery during the communist period (1980s). On this occasion, I encountered a rich fund of old books (3355 copies), many of them presenting various notes, unique in their own way. These notes are very precious, because they have the merit of bringing information about events, which at the time of their writing had an important value for the person or community in which the book was located, about the identity of the participants in these events and sometimes about the actual circulation of the book. They were made in different areas/places of the books by clerics or laymen with handwriting, in the Romanian language, with the Cyrillic alphabet and later, with the transition and the Latin alphabet. The historical value of these cult books, which the Metropolitanate of Moldavia and Bucovina owns, is doubled by the notes made during the time by the readers and their owners, who, by making these notations, found themselves in oblivion.

In Italy picture postcards were disseminated during the Giolitti era and throughout the Great War, thus becoming a concrete medium with a pervasive potential. Postcards influenced mass culture by constructing a collective imagination, they promoted the education of adults as well as of young people. As postcards reflected the shape of cultural industry by reproducing large scale images, they stemmed from a complex productive process that became a language of its own, starting from the artistic originality of the image to the printed form, reaching a wider audience both in terms of visual education and as swift communicative tool. In 1909 in Corriere dei Piccoli (1908-1995), the most prominent magazine for children in Italy at the time, Paola Carrara Lombroso (1871-1954) began the project for Bibliotechine per le scuole rurali (Libraries for rural schools), destined to a success until halfway through the Twentieth century. The young readers of the magazine took part with initiative by colouring and selling postcards created by Italian artists. The sum raised was used to donate books to rural schools. This research wishes to analyse a sample of postcards illustrated by well-known artists such as Mussino, Golia, Altara, Bologna and Gugù in order to shed light on the way childhood was represented. Toys, books, expressions and clothes are visual evidence of the multiple perspectives that adults adopted to reach children through entertainment. The artist’s gaze met child readers as they became protagonists and at the same time owning that message by modelling their thought and imagination.

Accurately estimating left atrial (LA) volume with Doppler echocardiography remains challenging. Using angiography for validation, Marino et al. (Marino P, Prioli AM, Destro G, LoSchiavo I, Golia G, and Zardini P. Am Heart J 127: 886–898, 1994) determined LA volume throughout the cardiac cycle by integrating the velocity-time integrals of Doppler transmitral and pulmonary venous flow, assuming constant mitral valve and pulmonary vein areas. However, this LA volume determination method has never been compared with three-dimensional LA volume data from cardiac MRI, the gold standard for cardiac chamber volume measurement. Previously, we determined that the effective mitral valve area is not constant but varies as a function of time. Therefore, we sought to determine whether the effective pulmonary vein area (EPVA) might be time varying as well and also assessed Marino's method for estimating LA volume. We imaged 10 normal subjects using cardiac MRI and concomitant transthoracic Doppler echocardiography. LA and left ventricular (LV) volumes were measured by MRI, transmitral and pulmonary vein flows were measured by Doppler echocardiography, and time dependence was synchronized via the electrocardiogram. LA volume, estimated using Marino's method, was compared with the MRI measurements. Differences were observed, and the discrepancy between the echocardiographic and MRI methods was used to predict EPVA as a function of time. EPVA was also directly measured from short-axis MRI images and was found to be time varying in concordance with predicted values. We conclude that because EPVA and LA volume time dependence are in phase, LA filling in systole and LV filling in diastole are both facilitated. Application to subjects in select pathophysiological states is in progress.

This research was carried out as a pot experiment to investigate the effects of 4 different drought stresses (FC-100%, 75%, 50%, 25%) created through field capacity (FC) on some morphological and physiological traits of bread wheat cultivars at the heading and ripening stages. In the study, 8 bread wheat varieties (Kate A1, Karahan 99, Tosunbey, Golia, Alpu 2001, Sultan 95, Konya 2002 and Eser) were used as material. The experiment was set up with 3 replications according to the split-plot experimental design, with the varieties main plots and drought stress applications sub plots. In the study, characteristics such as flag leaf area, flag leaf angle, chlorophyll content, stomata number, leaf water loss rate, relative water content, root length and root dry weight were investigated. It was determined that the effect of drought stress applications on all the examined traits was statistically significant. In drought stress applications, flag leaf area, flag leaf angle, chlorophyll content, leaf water loss rate, relative water content, stomata number, root length and root dry weight were respectively 4.01-14.02 cm<sup>2</sup>, 20.00-40.83<sup>o</sup>, 36.87-49.07 SPAD, 13.68-17.89%, 48.56%-72.61%, 8.01-10.95, 28.92-77.79 cm and 146.04-473.46 mg. As a result, it was determined that as drought stress increased, flag leaf area, flag leaf angle, chlorophyll content, relative water content, root length and root dry weight decreased, while leaf water loss rate and stomata number increased. In addition, it was noted that drought-resistant varieties had higher values in terms of chlorophyll content, relative water content, root length and root dry weight, while drought-sensitive varieties had higher values in terms of flag leaf angle and leaf water loss rate.

Namun buah ini memiliki karakteristik cepat membusuk sehingga diperlukan pengolahan menjadi produk olahan yang memiliki daya simpan lebih lama seperti dengan mengolahnya menjadi nata. Prinsip suatu bahan dapat dibuat menjadi nata adalah kandungan karbohidrat yang memadai pada bahan tersebut sehingga sebelum dilakukan pengolahan tentunya perlu dianalisis kandungan glukosa pada buah salak golla-golla. Penelitian ini bertujuan untuk menentukan kadar karbohidrat (glukosa) yang terdapat pada buah salak Golla-golla sebagai analisis awal untuk bahan baku pembuatan nata. Prosedur pada penelitian ini terdiri atas tiga tahapan utama yaitu penentuan jenis sampel, analisis kualitatif karbohidrat melalui pengujian benedict dan analisis kuantitatif karbohidrat dengan metode DNS. Hasil penelitian menunjukkan bahwa pada analisis kualitatif dengan uji benedict menunjukkan bahwa teerdapat kandungan glukosa pada buah salak Golla - golla yang ditunjukkan dengan terbentuknya endapan merah bata sedangkan pada analisis kuantitatif dengan metode DNS diperoleh bahwa kadar karbohidrat (glukosa) yang terdapat pada buah salak Golla - golla sebesar 2,11 % sehingga dapat digunakan sebagai bahan baku pembuatan nata dengan melakukan penambahan glukosa berdasarkan kadar karbohidrat (glukosa) awal yang terkandung pada buah tersebut.

Introduction: GOLCA is a potential first-in-class CELMoDTM agent for the treatment of lymphoma, purposefully designed to target the proteins Ikaros/Aiolos to produce dual immunomodulatory and anti-tumor activity, with preferential distribution to lymphoid organs. Pharmacodynamic data in pts with R/R FL show that, combined with RTX, the immunomodulatory effects of GOLCA are significantly increased (Chavez et al, EHA 2024, #1132). GOLCA as monotherapy (mono) and + RTX has been shown to be well-tolerated and effective in pts with R/R DLBCL (Chavez et al, ASH 2023, #4496) and R/R FL (Chavez et al, EHA 2024, #1132). Here, we report 30 mo follow up of pts with R/R FL who received GOLCA mono, and updated safety/efficacy of GOLCA + RTX from the ongoing Phase 1/2 CC-99282-NHL-001 study (NCT03930953). Methods: Pts with R/R FL with ≥ 2 prior lines of therapy (L) received GOLCA mono at different dosing schedules during part A (dose exploration) and GOLCA ± RTX at 0.2 mg or 0.4 mg days (d) 1-14 every 28 d (q28d) in part B (dose expansion). Total treatment duration with GOLCA was up to 2 years or until progressive disease or unacceptable toxicity, whichever occurred first. The efficacy-evaluable population consisted of pts completing ≥ 1 cycle of GOLCA and having baseline tumor assessment and ≥ 1 post-baseline tumor assessment or clinical progression or disease-related death. Results: As of May 31, 2024, 63 pts with R/R FL have enrolled: GOLCA mono (n = 19) at various dosing schedules and GOLCA + RTX at 0.2 mg (n = 22) or 0.4 mg (n = 22) on d1-14 q28d schedule. The median time from diagnosis to first dose of GOLCA was 68 mo (range, 23-136) and 59 mo (range, 15-309) in GOLCA mono and + RTX cohorts, respectively. The median number of prior therapies received was 4 (range, 2-11), and 3 (range, 1-12) in GOLCA mono and + RTX cohorts, respectively. In the GOLCA mono and + RTX cohorts, 9/19 (47%) and 13/43 (30%) pts had received prior lenalidomide (len)-based therapy, with 5/9 and 7/13 having len-refractory disease, respectively. In the GOLCA + RTX cohort, 10/43 (23%) had prior CAR T cell therapy and 6/43 (14%) had prior anti-CD20xCD3 bispecific antibody with or without CAR T cell therapy. In the GOLCA + RTX-treated safety population (n = 43), the incidence of treatment emergent adverse events (TEAEs) was similar across 0.2 and 0.4 mg dose levels (DL). The most frequent grade (G) 3+ TEAEs were neutropenia (44%) and anemia (12%). The incidence of febrile neutropenia was 7%. Serious adverse events occurred in 30% of pts, with febrile neutropenia and pneumonia being the most common, in 2 pts (5%) each, as well as pulmonary embolism in 1 pt. Neutropenia led to GOLCA dose interruption in 4 pts (9%) and dose reduction in 1 pt (2%). No pts discontinued treatment because of GOLCA-related TEAEs. Non-hematological TEAEs (e.g., GI toxicity, rash, fatigue) were all low-grade (except 1 pt with G3 fatigue). There were no new safety signals identified for GOLCA mono (part A) at the 30-mo follow up period. In the efficacy-evaluable population, at a median follow up of 6 mo (range 3-18), the ORR and CRR in the GOLCA + RTX cohort (n = 29) were 57% and 29% at 0.2 mg dose (n = 14) and 93% and 67% at 0.4 mg dose (n = 15), respectively. The median time to response was 2 mo (first response assessment). At the 0.4 mg DL of GOLCA + RTX, consistent responses were seen in pts at 2L/3L and 4L+, including in those with prior exposure to len-based therapies and/or T-cell-redirecting therapy. At the 0.4 mg DL, 12/14 (86%) pts remained in response at the time of data cut-off. In pts receiving GOLCA mono across different dose schedules in part A (n = 12), ORR and CRR were 67% and 42%, respectively. Responses were durable, with median DOR of 22 mo (range, 2-51) at a median follow up of 30 mo. 33% of pts (4/12) have completed 2 yrs of protocol defined GOLCA treatment and remain in response at the 1 yr post-treatment follow-up period, without requiring any intervening treatment. Conclusions: GOLCA 0.4 mg + RTX was well tolerated and highly active in heavily pre-treated pts with R/R FL, including those with prior exposure to len-based therapies and/or T-cell-redirecting therapies. Treatment with fixed-duration GOLCA mono led to durable responses including a subset of pts remaining treatment free, without progression at extended follow up. These data further support development of GOLCA + RTX as a fixed-duration, chemotherapy-free, outpatient treatment option in pts with R/R FL, and also pts with newly diagnosed advanced stage FL (NCT06425302).

Introduction: Compounds inducing cereblon-mediated degradation of Ikaros and Aiolos (I/A), like lenalidomide (LEN) and avadomide, have demonstrated antitumor activity in non-Hodgkin lymphoma (NHL), with significant but limited cell-autonomous activity in diffuse large B-cell lymphoma (DLBCL) cell lines. Their pharmacodynamic activities also include T-cell activation, increased macrophage tracking, and increased regulatory T lymphocytes (T-regs) in peripheral blood (PB) [Carpio C et al Blood 2020]. High immune infiltration in tumors at baseline (BL) was correlated with improved outcome (Risueño A et al Blood 2020). Golcadomide (GOLCA) is a potential first-in-class cereblon E3 ligase modulatory drug (CELMoD™) that, compared with immunomodulatory (IMiD) agents like LEN, has shown more profound cell autonomous activity and immune stimulation in preclinical DLBCL models (Lopez-Girona A et al Hematological Oncology, 2021). Its potent I/A degradation, prolonged exposure, and high tissue penetration predicted superior GOLCA activity vs IMiDs, including in tumors with low immune infiltration. In the CC-99282-NHL-001 study, GOLCA ± rituximab showed a manageable, predictable safety profile characterized by an on-target toxicity of neutropenia and promising efficacy in relapsed/refractory NHL in heavily pretreated pts (Chavez JC et al Blood 2023). Here, we describe pt immune profiles, their modulation by GOLCA and association with objective clinical response in the CC-99282-NHL-001 DLBCL biomarker cohort. Methods: Cell intrinsic and immune-mediated activity of GOLCA in evaluable pts with DLBCL enrolled in the study were assessed by RNA-Seq, whole genome sequencing, immune-fluorescence and flow cytometry (FC) in screening and on-treatment biopsies, and by FC in PB samples. Results: I/A degradation was necessary but not always sufficient for GOLCA activity. We observed deep I/A degradation in biopsies and PB samples from all evaluable pts independent of response, suggesting additional tumor-related features influence response to GOLCA. In contrast to IMiDs, efficacy of GOLCA (best overall response and progression-free survival) was independent of BL tumor microenvironment (TME) content. Despite this, BL CD4 naive T cells, T follicular helper and CD8 T cell levels were higher in responders while mono/macrophages were lower. GOLCA sensitivity analysis was run separately in immune infiltration (TME+) vs high B-cell content (TME−) tumors to explore immune mediated and tumor intrinsic mechanisms. Low leukocyte migration and low chemokine signaling were enriched in TME− responders. TME+ tumors showed distinct expression levels of genes involved in immune activation and trafficking (↑AIRE, ↑CLDN12, ↑IQGAP3, ↓MADD, ↓UBE7). GOLCA responders showed a significant increase in pathways of chemokine and cytokine receptor binding and chemokine signaling. Interferon (IFN)-stimulating genes and other proteins involved in immunoregulatory/inflammatory processes (IFI6, OAS3, CCL3L etc) were upregulated in responders, with a modest increase in pathways like tumor necrosis factor alpha and IFNα/β. GOLCA-induced I/A degradation led to increased natural killer- (NK) and T-cell activation with dose/schedule-dependent T-reg increase and decrease in naive T cells in PB. Response was associated with increased PD-1, potentially due to increased T-cell activation. GOLCA responders showed a significant decrease in B-cell content and increase in NK and dendritic cells, macrophage and T-cell subsets. In these responders, there was positive correlation of T-regs between PB at Cycle 1 Day 15 and on-treatment biopsies. Other T-cell populations like naive and memory T cells showed anti-correlation, suggesting peripheral changes may be due to trafficking. In non-responders, these changes were less pronounced. Conclusion: These results show GOLCA fulfills its design intent, causing deep and prolonged I/A degradation in NHL lesions resulting in BL TME-independent activity, compared with IMiDs like LEN which are TME-dependent. BL immune profiles and their modulation on treatment link to treatment effect and imply an immune contribution to GOLCA efficacy. These data support broader activity of GOLCA than IMiDs in NHL, with high potential for combination with anti-lymphoma agents acting on tumor B cells and/or TME.

Background: Diffuse large B cell lymphoma (DLBCL) is a heterogeneous disease of malignant B cells with accelerated growth kinetics and known to suppress immunity in the tumor microenvironment (TME) through a variety of mechanisms. Understanding the mechanisms of this immunosuppressive TME is critical to developing additional effective therapies. Golcadomide (golca), a CRBN modulator which degrades transcription factors Aiolos and Ikaros, is being developed for the treatment of R/R DLBCL in combination with CD20 targeting T cell engagers (TCE). Herein, we utilized a novel in vivo disease model to characterize the effects of golca on immune TME by combining multiplex immunofluorescence (mIF) and spatial transcriptomics (ST) to analyze the interaction and function of cells in TME to support the combination strategy for golca and TCE. Methods: We developed an in vivo mouse model of DLBCL using eµ-myc mice backcrossed with hCRBN mice by adoptive transfer of eµ-myc/hCRBN splenocytes into recipient hCRBN mice. Mice were treated with the vehicle or golca from day 5 to 8. On day 8, spleen was harvested and analyzed for ST using Visium 10X system as well as by mIF. In the ST data, cell fractions were inferred following spot deconvolution with BayesPrism, spots were clustered using gene expression with Seurat, and gene-set scores were obtained with ssGSEA. In a subsequent set of experiments, mice were treated with isotype or TCE alone or in combination with golca. In combination studies, efficacy was measured as survival benefits. Additionally, peripheral blood and splenic tissue were harvested and assessed by mIF and flow cytometry in satellite cohorts to measure pharmacodynamic effects of the treatments. Results: Eµ-myc/hCRBN transplanted mice developed large B cell lymphoma like disease in spleen. Spleens from diseased mice revealed that vehicle treated spleens contained increased tumor cells compared to golca treated spleens. These observations were qualified with H&amp;E and mIF staining. ST analysis further identified 14 clusters based on spot-level gene expression data. These clusters were evaluated in the context of the cell fractions obtained by spot deconvolution. Two clusters of interest were prioritized for further analysis; cluster 5 (C5) was enriched for cycling B cells and eµ-myc tumor cells and cluster 6 (C6) was enriched for cytolytic T and NK cells. The relative fraction of C5 is the highest in vehicle group and is decreased in response to golca treatment, validating prior observations of direct anti-tumor activity of golca. Conversely, C6 abundance is enhanced in mice treated with golca compared to vehicle controls, indicating increased levels of CD8+ T cells through proliferation or trafficking. We next examined pathways more abundantly expressed in tumor rich areas, which included myc targets and G2M checkpoint signaling. The expression of genes associated with these pathways were decreased in golca treated mice compared to vehicle. The reduction of the tumor-associated pathway expression patterns by golca suggests a reversion of disease toward a normal state. Based on these observations, we examined the ability of golca to enhance TCE effects with a murine CD20xCD3 TCE in our in vivo model. Flow cytometric analysis of day 11 peripheral blood and spleens demonstrated that both the TCE and golca alone and in combination decreased naive T-cells and increased central/effector memory populations compared to isotype/vehicle controls. mIF performed on diseased spleens demonstrated increased abundance of T cells and caspase 3 expression, decreased Pax5+ tumor cells in the combination arm compared to single agents. The combination treatment also decreased splenic hypertrophy compared to single agents and vehicle controls. Finally, compared to isotype control, the TCE monotherapy extended survival of the mice by an additional week (21 days vs 14 days) with the combination of golca/TCE further extending survival by two additional days. Conclusion: We utilized a hCRBN/eµ-myc mouse model to demonstrate that the combination of golca with a CD20XCD3 TCE can enhance T-cell activation, cytotoxic capacity and extend survival compared to single agents. ST and mIF analysis of immune components within the TME provide mechanistic evidence of the dual mechanism of action for golca (direct anti-tumor activity and enhanced immune activity) further supporting this combination in on-going clinical trials.

Pertarungan antara Daud dan Goliat merupakan sebuah metafora yang digunakan untuk menggambarkan konfrontasi yang tak seimbang antara kekuatan yang kecil dengan kekuatan yang besar. Pertarungan ini memperlihatkan keberanian dan ketahanan Daud ditengah ketidakadilan fisik dan ketidakseimbangan kekuatan. Akibatnya, Goliat digambarkan sebagai sosok yang kuat karena memiliki badan besar, sedangkan Daud sebaliknya. Stereotipe ini memengaruhi persepsi terhadap laki-laki yang bertubuh besar. Penelitian ini berusaha membahas stigma maskulinitas yang dialami oleh laki-laki yang memiliki badan besar. Maskulinitas, bahkan hypermaskulinitas, yang terkadang dilekatkan pada citra diri laki-laki berbadan besar yang digambarkan dengan kesan agresif dan opresif. Laki-laki berbadan besar, akibat dari standar patriarki menjadi sebuah kekerasan gender yang ikut melumpuhkan laki-laki. Berangkat dari kritik pascakolonial yang mengedepankan hubungan antara pihak dominasi dan subordinasi, tulisan ini berusaha menelisik dan mempertanyakan ulang posisi pembaca dalam melihat tokoh Goliat yang terkena stigma maskulinitas dalam pertempurannya dengan Daud. Melalui analisis teks, penulis berusaha merekonstruksi pembacaan 1 Samuel 17 dengan menguraikan pertempuran Goliat yang merupakan jawara dari bangsa Filistin, dengan Daud yang mewakili bangsa Israel. Artikel ini mendapati bahwa badan besar yang dimiliki oleh Goliat bukan lah menjadi kekuatannya, melainkan sumber kelemahannya. Dengan demikian, artikel ini meruntuhkan stigma hypermaskulinitas yang melekat pada laki-laki yang berbadan besar. Kesadaran akan kekerasan yang tidak memandang gender, menghadirkan pembacaan inklusif yang sarat akan makna baru bagi Goliat, pembaca dan konteks masa kini.

Introduction Up to 40% of pts with diffuse large B-cell lymphoma (DLBCL) relapse after first-line chemo-immunotherapy with rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone (R-CHOP). CELMoD agents such as GOLCA potently degrade target proteins Ikaros/Aiolos, resulting in antiproliferative, apoptotic, and immunomodulatory activity. GOLCA preferentially distributes to tissues and lymphoid organs. In a phase 1 study, GOLCA demonstrated a manageable safety profile with promising clinical activity as monotherapy in relapsed/refractory non-Hodgkin lymphoma (Michot et al. Blood 2021). CC-220-DLBCL-001 (NCT04884035) is an ongoing open-label, multicenter, dose escalation and expansion trial to assess safety and preliminary efficacy of CELMoD agents + R-CHOP for untreated a-BCL. GOLCA demonstrated a manageable safety profile in the dose escalation phase (Munoz et al. ICML 2023 abstract 438); we report combined results from the GOLCA dose escalation and expansion phases. Methods Eligible pts were ≥ 18 years, had untreated a-BCL with measurable disease (Lugano 2014), ECOG performance status ≤ 2 and International Prognostic Index (IPI) score 0-5 (2-5 in dose expansion). Pts were treated with R-CHOP plus GOLCA in 21-day (D) cycles (C) for up to 6C, or until disease progression/unacceptable toxicity/study withdrawal/physician decision. During dose escalation, pts received GOLCA at dose levels (DL): 0.2 mg D1-7 (DL-1), 0.4 mg D1-7 (DL1), and 0.4 mg D1-10 (DL2). DL2 met the dose-limiting toxicity threshold and was not continued in the expansion phase. During dose expansion, pts were randomized 1:1 to R-CHOP plus GOLCA at DL-1 or DL1. The primary endpoint for the expansion phase was safety of GOLCA at the recommended phase 2 dose (DL1) (based on adverse events [AEs]); secondary endpoints included overall response rate (ORR) and complete metabolic response (CMR) rate (assessed at end of treatment [EoT] by PET-CT [Lugano 2014]). Circulating tumor DNA (ctDNA) was measured at C1D1, C2D1, C3D1, and EoT using the PhasED-Seq assay. Results Across escalation and expansion, 78 pts were treated (DL-1, n = 35; DL1, n = 37; DL2, n = 6). Median age was 63.0 years, 56.4% were male, most had high-intermediate/high IPI (3-5) score at diagnosis (64.1%), Ann Arbor stage III-IV disease (83.3%), and germinal center B cell as cell of origin (51.3%); 83.3% pts had DLBCL histology. At data extraction (May 25, 2023), across escalation and expansion, 38 pts had treatment ongoing, 31 completed treatment, and 35 continued to follow-up. A total of 3 pts discontinued due to AEs, 1 each at DL-1, DL1, and DL2. In the safety population (n = 78), median GOLCA relative dose intensity (RDI) was 99.4%; 82.1% of pts received RDI ≥ 85%. RDI for CHOP components was &amp;gt; 90%. A total of 73 (93.6%) pts had ≥ 1 treatment-emergent AE (TEAE); 70 (89.7%) pts had ≥ 1 TEAE considered related to GOLCA; 65 (83.3%) pts had ≥ 1 grade 3/4 TEAE, 62 (79.5%) pts had ≥ 1 grade 3/4 TEAE considered related to GOLCA. The most frequently occurring grade 3/4 TEAEs were neutropenia (76.9%) and thrombocytopenia (32.1%) ( Table). A total of 30 (38.5%) pts had serious TEAEs, most frequently febrile neutropenia (preferred term: 11 pts, 14.1%); 12 (15.4%) pts had serious TEAEs of infection or infestation (system organ class); 6 pts (7.7%) had venous thromboembolisms considered related to GOLCA (4 deep vein thrombosis, 1 superficial thrombosis, 1 pulmonary embolism). Treatment-related AEs leading to GOLCA dose reduction or discontinuation occurred in 11 (14.1%) pts and 6 (7.7%) pts, respectively. After a median follow-up time of 4.1 months (range 0.3-12.3) and among all treated pts with EoT response available, 21/25 had CMR; together, 14/14 pts who received GOLCA 0.4 mg (DL1 and DL2) achieved CMR at EoT ( Figure). Among efficacy-evaluable pts (n = 56), ORR was 91.1% (95% CI 80.4-97.0). Pts treated with GOLCA at DL-1 and DL1 who were ctDNA high-risk (≥ 2.5 log hGE/mL) responded to study treatment (CMR-PMR) and had early decreases in ctDNA (Kaplan et al. ASH 2023). Conclusions GOLCA demonstrated a manageable safety profile, showed good combinability with R-CHOP, and did not compromise delivery of curative treatment. Efficacy results and substantial ctDNA decrease indicate robust clinical activity in frontline DLBCL, with a high CMR rate at EoT, particularly among patients treated at DL1. Abstract Chronic Lymphocytic Leukemia (CLL) is an adult leukemia characterized by the accumulation of incompetent B lymphocytes expressing CD19, CD20, CD23, and CD5. Current therapies include chemotherapy and combination with targeted therapies such as ibrutinib (Bruton’s tyrosine kinase inhibitor), venetoclax (BCL2 inhibitor), and obinutuzumab (anti-CD20). The tumor microenvironment promotes CLL development and drug resistance. Golcadomide (GOLCA, CC-99282) is an oral cereblon E3 ligase modulator (CELMoD®) agent with immunomodulatory and tumor cell-autonomous activities under clinical investigation for R/R non-Hodgkin lymphomas and CLL/SLL. Here we investigated the effect of GOLCA on inhibiting proliferation and inducing apoptosis of CLL preclinical models by degrading Ikaros and Aiolos. We also studied its combination with other anti-CLL agents. A panel of 10 CLL cell lines and 15 primary CLL patient samples were used to assess efficacy of GOLCA alone or in combination with ibrutinib (Ibru), venetoclax (Ven), and obinutuzumab (Obi). Primary CLL cells were grown ex vivo in a co-culture system with CD40L expressing fibroblasts designed to mimic the CLL lymph node microenvironment. Substrate degradation, viability, cell cycle and immunophenotyping were assessed by flow cytometry. GOLCA showed potent in vitro antiproliferative activity on 6 out of 10 CLL cell lines, including those with high-risk features, with IC50 of 1-20 nM. It also inhibited CLL stimulated cell proliferation and induced apoptosis in all evaluated patient samples in the co-culture system, with low to sub-nanomolar IC50, independent of IGHV mutation status and other chromosomal characteristics. Cell-cycle analysis confirmed inhibition of proliferation in the 5 patient samples evaluated after 3 days of treatment with GOLCA, with a dose-dependent decrease in the fraction of cells in the S phase. GOLCA also demonstrated degradation of the proximal substrates Ikaros and Aiolos in tumor and T cells from CLL patient samples. In combination with Obi, Ven, and Ibru, GOLCA led to synergistic or additive tumor cell toxicity in most patient samples evaluated, suggesting potential clinical benefit of these targeted agent combinations. Additive tumor toxicity was observed in two out of four patient samples when GOLCA was combined with Ibru. We have previously demonstrated that GOLCA induces immune activation in T cells from healthy donor PBMCs. In co-cultures of CLL PBMCs and CD40L-expressing fibroblasts, GOLCA increased T cell numbers and activation characteristics. Ven inhibited this immune activation, while Obi or Ibru maintained the immune activation. Collectively, these findings suggest that GOLCA, alone or in combination with targeted agents, may produce clinical benefit in CLL patients. This research aims to analyze the determination of cost of production using the full costing method as the basis for setting selling prices at the micro business Golla Kambu Teppo Mama Rina. The focus of this research is on the cost of production using the full costing method and the selling price. Data collection techniques were conducted using triangulation techniques, namely observation, interviews, and documentation. Data analysis was carried out using qualitative descriptive analysis. The results of this study indicate that the calculation of the cost of production based on the full costing method results in higher costs compared to the calculation by Golla Kambu Teppo Mama Rina. The cost of production obtained according to Golla Kambu's business is IDR 29.778 per package, while that obtained by the researcher is IDR 30.407 per package. The selling price determined by Golla Kambu Teppo Mama Rina is IDR 32,000 per package, whereas that obtained by the researcher is IDR 36.487 per package. This difference occurs because Golla Kambu Teppo Mama Rina does not account for all costs (factory overhead costs) incurred in the production process in detail
