Abstract:

Abstract Motivation Glycine receptors (GlyRs) mediate fast inhibitory neurotransmission in the brain and have been recognized as key pharmacological targets for pain. A large number of chemically diverse compounds that are able to modulate GlyR function both positively and negatively have been reported, which provides useful information for the development of pharmacological strategies and models for the allosteric modulation of these ion channels. Results Based on existing literature, we have collected 218 unique chemical entities with documented modulatory activities at homomeric GlyR-α1 and -α3 and built a database named GRALL. This collection includes agonists, antagonists, positive and negative allosteric modulators and a number of experimentally inactive compounds. Most importantly, for a large fraction of them a structural annotation based on their putative binding site on the receptor is provided. This type of annotation, which is currently missing in other drug banks, along with the availability of cooperativity factors from radioligand displacement experiments are expected to improve the predictivity of in silico methodologies for allosteric drug discovery and boost the development of conformation-based pharmacological approaches. Availability and implementation The GRALL library is distributed as a web-accessible database at the following link: https://ifm.chimie.unistra.fr/grall. For each molecular entry, it provides information on the chemical structure, the ligand-binding site, the direction of modulation, the potency, the 3D molecular structure and quantum-mechanical charges as determined by our in-house pipeline. Abstract:

A high degree of variation in hydrographic conditions is found in the so-called Iroise Sea, within less than 100 km of the west coast of Brittany. Tidal current maximal velocity, especially, ranges there from about 0·5 knot to more than 8 knots (locally, near the island of Ushant), i.e. practically as wide a range as found over the whole of north-west European shelf seas. Pelagic ecosystems accordingly exhibit a high degree of variety, related not only to classical inshore-offshore gradients, but also to the extent of vertical mixing or stratification. Areas where different physical and biological conditions prevail are generally separated by rather clearcut boundaries. The better-known of these is the Ushant thermal front, which runs in summer across the whole entrance to the English Channel, but also extends into the Iroise. In addition, freshwater runoff results in thermohaline stratification, or at least in the existence of thermohaline vertical gradients, in the two major bays of the west coast of Brittany. The relevant area is limited seawards by a thermohaline front, the Iroise inner front (Grail & Le Fèvre, 1967; Le Fèvre & Grall, 1970), beyond which are found the well-mixed waters inshore of the Ushant front. Fig. 1 sums up these hydrographic patterns in the area taken here into consideration.

Abstract This article reopens the question of the relationship between the medieval Welsh version of the Grail narrative, the Historia Peredur vab Efrawc, and the French Conte du Graal of Chrétien de Troyes. It explores the seeming inconsistencies in the Welsh tale’s presentation of the Grail procession, and suggests that the hero’s actions, and in particular his reticence in asking questions about the procession, should be read in the context of medieval Welsh customs and legal tradition. The article concludes with an exploration of the implications of the proposed interpretation for the reading of Historia Peredur as a postcolonial narrative.

Subjek dalam penelitian ini adalah Pembina, Pelatih, Atlet SMK Negeri 1, SMK Negeri 3, SMK Negeri 5, SMK Negeri 7 Kota Bengkulu. Pengumpulan data dan Instrumen penelitian ini berupa kuesioner/angket yang sudah divalidasi oleh ahli. Analisis data dalam penelitian ini menggunakan analisis data kualitatif dan keabsahan data statistik yang menggunakan rumus persentase. Hasil penelitian dalam pembahasan menunjukan bahwa hasil Evaluasi Program Pembinaan Prestasi Olahraga Bola Voli Pada SMK Negeri Di Kota Bengkulu belum seluruhnya maksimal dalam pelaksanaannya dari 4 sekolah yang diteliti hanya 1 sekolah yang menjalankan program pembinaan dengan baik. Hal tersebut dilihat dari segi manajemen pengelolaan sarana dan prasaranan yang sangat minim, pengelolaan dana yang belum tersalurkan menyeluruh untuk kegiatan program pembinaan tersebut, serta kurangnya dukungan dari pihak sekolah. Kurang maksimalnya program latihan yang diberikan pelatih, membuat tujuan pencapaian prestasi puncak tidak dapat terlaksana dengan baik. Kesimpulan dari program pembinaan belum maksimal dari segi prasarana, pengelolaan dana, serta dukungan dari sekolah dari 4 sekolah yang di teliti hanya 25 % sekolah yang sudah melaksanakan program dengan baik.Kata Kunci: Evaluasi, Program Pembinaan Prestasi, Bola VoliABSTRACTThis study aimed to describe the Evaluation of the Achiefment Volley Ball Training Program at Vocational School in Bengkulu City. Abstract:

Indiana Jones and the Last Crusade, a popular motion picture, offers a modem version of a Quest for the Holy Grail. Although this grail legend is new, a survey of medieval through nineteenth-century stories of heroic quests for a grail reveals that grail legends have always differed from each other in significant ways. The grail itself has been identified in some legends as a cup or chalice, and in others as a dish, platter, book, stone, or, possibly, a reliquary. Also profoundly different are the ways in which legends describe the purposes effects of a quest for the grail. What these diverse legends have in common, however, is their association of a quest for the grail with a hero's attempt to reverse the evils that endanger a particular society. This essay traces various grail legends to determine how these popular tales, including the film version, present man's quest for transcendence, and moral and spiritual renewal.

Abstract:

Abstract The topic of this paper revolves around the links that unite the motif of the Waste Land with the wound of the Fisher King in the Conte du Graal. These links are based, on the one hand, on ancestral beliefs that connect the land’s fecundity with the goodness of the king, while his faults are punished with its sterility, and on the other, with Augustinism. The ills of the Grail’s family constitute a deserved castigation derived from a chain of sins, originating in the previous generation with the commission of a sin of origin. The punishment embraces all the members of the family, and, in the very same way that Original sin is the cause of both mortality and the earth’s aridity, its punishment presupposes the king’s impotence and the kingdom’s barrenness, Perceval’s matricide, and his silence in front of the Grail. The awaited irruption of Perceval in the Fisher King’s domain signified for the Grail’s family the same as that of redemption for mankind. Perceval thus redeems his lineage, and with it, the earth, in the line of the old belief that linked the king’s sins with the earth’s sterility, in conformity with the pattern of the Fall, Punishment and Redemption.

Abstract:

Gene related to anergy in lymphocytes (GRAIL) is an E3 ubiquitin ligase that regulates energy in T-lymphocytes. Whereas, the relevance of GRAIL to T lymphocyte function is well established, the role of this protein in other cell types remains unknown. Given that GRAIL is abundant in the liver, we investigated the potential function of GRAIL in nutrient metabolism by generating mice in which the expression of GRAIL is reduced specifically in the liver. Adenovirus-mediated transfer of a short hairpin RNA specific for GRAIL mRNA markedly reduced the amounts of GRAIL mRNA and protein in the liver. Blood glucose levels of the mice with hepatic GRAIL deficiency did not differ from those of control animals in the fasted or fed states. However, these mice manifested glucose intolerance in association with a normal increase in plasma insulin levels during glucose challenge. The mice also manifested an increase in the serum concentration of free fatty acids, whereas the serum levels of cholesterol and triglyceride were unchanged. The hepatic abundance of mRNAs for glucose-6-phosphatase, catalytic (a key enzyme in hepatic glucose production) and for sterol regulatory element-binding transcription factor 1 (an important transcriptional regulator of lipogenesis) was increased in the mice with hepatic GRAIL deficiency, possibly contributing to the metabolic abnormalities of these animals. Our results thus demonstrate that GRAIL in the liver is essential for maintenance of normal glucose and lipid metabolism in living animals.

Abstract:

NK cells (natural killer cells) being a part of the innate immune system have been shown to be involved in immunoregulation of autoimmune diseases. Previously we have shown that HINT1/Hsp70 treatment induced regulatory NK cells ameliorating experimental autoimmune encephalomyelitis (EAE) course and CD4+ T cells proliferation. NK cells were isolated from mice treated with HINT1/Hsp70 and co-cultured with proteolipid protein (PLP)-stimulated CD4+ T cells isolated from EAE mice. Cell proliferation was assessed by thymidine uptake, cytotoxicity by lactate dehydrogenase (LDH) release assay and fluorescence activated cell sorting (FACS) analysis, protein expression by Western blot, mRNA by quantitative RT-PCR. Gene related to anergy in lymphocytes (GRAIL) expression was downregulated by specific siRNA and GRAIL overexpression was induced by pcDNA-GRAIL transfection. HINT1/Hsp70 pretreatment of EAE SJL/J mice ameliorated EAE course, suppressed PLP-induced T cell proliferation by enhancing T cell expression of GRAIL as GRAIL downregulation restored T cell proliferation. HINT1/Hsp70 treatment induced immunoregulatory NK cells which inhibited PLP-stimulated T cell proliferation not depending on T cell necrosis and apoptosis. This immunoregulatory NK cell function depended on NK cell expression of GRAIL as GRAIL downregulation diminished inhibition of NK cell suppression of T cell proliferation. Similarly GRAIL overexpression in NK cells induced their regulatory function. HINT1/Hsp70 treatment generated regulatory NK cells characterized by expression of GRAIL.

Abstract:

Designing the front grill with the focus on analyzing the flow from outside to the inside of a passenger car bonnet is very important in the automotive industry. This study was done to increase performance of the flow through the analysis on the effect of aerodynamic flow through the front grill by designing different cases of frontal areas of the front grill. Velocity at 34 m/s at steady condition were done to obtain the flow structure around a passenger car as well as the front grill of a passenger car. Analysis was taken inside the wind tunnel as the boundary condition. The grid generation was based on the tetrahedral unstructured meshes. Result obtain was compared with past experiment data. It was found that an appropriate design of frontal area of front grill can improved the stability of a car and the heat surrounding inside the car bonnet. Finally, the aerodynamics of the most suitable design of front grill was introduced and analyzed.

Penelitianini bertujuan untuk mengetahui tingkat kepadatan lalat, faktor-faktor yang mempengaruhi dan upayapengendaliannya.Metode yang digunakan dalampenelitian ini adalah metode deskriptif.pengukuran kepadatan lalatsecara langsung maupun wawancara dengan masyarakat di lapangan dilakukan untuk mencapai tujuanpenelitian.Hasil penelitian menunjukan bahwa kepadatan lalat rata-rata di Pasar Kota Banjarnegara yaitu 10ekor/block grill. Masing-masing lokasi di pasar Kota Banjarnegara mempunyai kepadatan lalat rata-rata yangberbeda-beda, yaitu 4 ekor/block grill di los buah, los sayuran, dan los daging. 2 ekor/block grill di los ikan, 20ekor/block grill di tempat pembuangan sampah(TPS), dan 3 ekor/block grill di tempat jajanan terbuka. Los yangmemiliki tingkat kepadatan lalattertinggi terdapat di TPS yaitu 20 ekor/block grill. Faktor-faktor yangmempengaruhi tingkat kepadatan lalat di pasar Kota Banjarnegara adalah buruknya kondisi sanitasi, lokasi TPSyang tidak sesuai, kondisi fisik seperti suhu, kelembaban, pencahayaan, upaya yang dapat dilakukan untukmengatasi masalah tersebut salah satunya adalah dengan memperbaiki kondisi sanitasi di lingkungan pasar KotaBanjarnegara.

Increasing reports on the incidental ingestion of metallic bristles from barbeque grill cleaning brushes have been reported. We sought to describe the clinical presentation and grilling habits of patients presenting after ingesting metallic bristles in an attempt to identify risk factors. We performed a chart review of six patients with documented enteric injury from metallic bristles. Subjects were contacted and administered a survey focused on the events surrounding the bristle ingestion. We arranged for in-home visits to inspect the grill and grill brush whenever possible. Of the six subjects identified, three (50%) were male, five (83%) were white, and they ranged in age from 18 to 65 years (mean 42.5). All complained of abdominal pain. All bristles were identified by CT scan. Three patients underwent laparoscopic enterorrhaphy, and two underwent laparotomy. The remaining patients did not require intervention. None had replaced their grill brush in at least two years. Surgeon's awareness of this unusual injury is important to identify and manage this problem. Alternative methods to clean the grill should be sought and grill brushes should be replaced at least every two years.

When the Grail emerged unobtrusively for the first time shortly after 1180 in The Story of the Grail authored by Chrétien de Troyes it was hardly noticeable. It emerged in the context, viewed from certain angles, of a table ritual on the occasion of a feast presented by the Fisher King to young Perceval whom he invited to share his table. When it appeared before the two table guests it did not stop in front of them but disappeared into a side chamber. It was accompanied by a plate, a serving dish on which to carve meat. But it was also preceded by a bleeding lance whose presence at the feast seemed wholly out of context and inexplicable. The rather cryptic description of the grail, replete with hidden meanings and ambiguities, started a myth, more evocative than anything the Middle Ages had ever known. The Grail theme remained in evidence throughout the Middle Ages, then disappeared from the 16th to the 19th century, then sprang into life vividly once more in the form of Wagnerian opera as Parzifal. Today all manifestations of the Grail are egregiously misrepresented as if it had always been the Holy Grail. It behooves us to take another look at its earliest manifestation before all the transformations and Christianisations to which it was subjected by western culture1. In fact, the original Grail had nothing at all in common with Christianity.

Abstract:

The automatic fish grill is a tool that is very helpful for food vendors who are processed in a roasting manner. For now, conventional automatic grills that are already on the market have very expensive prices so that many traders still use manual fish grills which are less efficient in the process of doing them. The purpose of making this automatic fish grill design is to help human work to save more time and energy. The automatic fish roasting box is designed with heat absorbers in each layer, equipped with two racks, each measuring 50cm and also equipped with 2 250 watt heaters. From the test results using different types of carp with different weights, it shows that the automatic fish grill works well and gets a 100% percentage result with evenly cooked fish. The economical level of automatic fish grill has a good value, the whole design of automatic fish grill uses good quality materials and materials. For now, automatic roasters on the market have very expensive prices, this difference in price has made many traders not take full advantage of the technology. Because of this problem, researchers created an automatic fish grill based on a temperature controller, which is targeted at an inexpensive price, but can still be used safely and with maximum results. Thus, using a temperature controller-based automatic fish grill design can help simplify work to make it more efficient and safer.

Abstract Rainfall is a driving factor of climate in the tropics and needs to be properly represented within a climate model. This study customizes the precipitation processes over the tropical regions of eastern Africa and the Indian Ocean using the International Centre for Theoretical Physics (ICTP) Regional Climate Model (RegCM3). The convective schemes of Grell with closures Arakawa–Schubert (Grell–AS)/Fritch–Chappel (Grell–FC) and Massachusetts Institute of Technology–Emanuel (MIT–EMAN) were compared to determine the most realistic spatial distribution of rainfall and partitioning of convective/stratiform rainfall when compared to observations from the Tropical Rainfall Measuring Mission (TRMM). Both Grell–AS and Grell–FC underpredicted convective rainfall rates over land, while over the ocean Grell–FC (Grell–AS) over- (under-) estimates convective rainfall. MIT–EMAN provides the most realistic pardoning and spatial distribution of convective rainfall despite the tendency for overestimating total rainfall. MIT–EMAN was used to further customize the subgrid explicit moisture scheme (SUBEX). Sensitivity tests were performed on the gridbox relative humidity threshold for cloudiness (RHmin) and the autoconversion scale factor (Cacs). An RHmin value of 60% (RHmin-60) reduced the amount of total rainfall over five heterogeneous rainfall regions in eastern Africa, with most of the reduction coming from the convective rainfall. Then, Cacs sensitivity tests improved upon the total rainfall amounts and convective stratiform partitioning compared to RHmin-60. Based upon all sensitivity simulations performed, the combination of the MIT–EMAN convective scheme, RHmin-60, and halving the model default value (0.4) of Cacs provided the most realistic simulation in terms of spatial distribution, convective partition, rainfall totals, and temperature bias when compared to observations.

Small RNA are very important post-transcriptional regulators in both, bacteria and eukaryotes. One of such sRNA is GraL, encoded in the greA leader region and conserved among enteric bacteria. Here, we conducted a bioinformatics search for GraL’s targets in trans and validated our findings in vivo by constructing fusions of probable targets with lacZ and measuring their activity when GraL was overexpressed. Only one target's activity (nudE) decreased under those conditions and was thus selected for further analysis. In the absence of GraL and greA, the nudE::lacZ fusion's β-galactosidase activity was increased. However, a similar effect was also visible in the strain deleted only for greA. Furthermore, overproduction of GreA alone increased the nudE::lacZ fusion’s activity as well. This suggests existence of complex regulatory loop-like interactions between GreA, GraL and nudE mRNA. To further dissect this relationship, we performed in vitro EMSA experiments employing GraL and nudE mRNA. However, stable GraL-nudE complexes were not detected, even though the detectable amount of unbound GraL decreased as increasing amounts of nudE mRNA were added. Interestingly, GraL is being bound by Hfq, but nudE easily displaces it. We also conducted a search for genes that are synthetic lethal when deleted along with GraL. This revealed 40 genes that are rendered essential by GraL deletion, however, they are involved in many different cellular processes and no clear correlation was found. The obtained data suggest that GraL's mechanism of action is non-canonical, unique and requires further research.

Accidental ingestion of a grill brush wire bristle is a rare event. Retrieval rarely requires surgical intervention as the wire typically causes symptoms above the gastro-oesophageal junction and can frequently be removed endoscopically. There are few reported cases of gastrointestinal injury due to ingestion of wire bristles lodging past the gastro-oesophageal junction in adults. We present four cases of wire brush bristle ingestion that required operative intervention. Our case series illustrates how the commonly used wire grill brush may cause a serious injury. This diagnosis should be considered in patients who present with abdominal pain, non-specific symptoms and normal labs, with liner radio-opaque imaging findings and a history of grill use. Education as to the dangers of grill wire brushes to clean grills should be provided commercially.

Lalat tidak dapat diberantas habis tetapi dapat dikendalikan sampai dengan batas yang tidak membahayakan atau menimbulkan masalah bagi kesehatan masyarakat. Cara yang paling mudah dan cepat untuk mengukur tingkat kepadatan lalat yaitu dengan menggunakan fly grill. Fly grill merupakan alat berupa potongan kayu yang disusun untuk melakukan survei kepadatan lalat. Tujuan penelitian ini adalah untuk mengetahui efektivitas warna kuning fly grill dalam menarik lalat. Metode: Penelitian ini dilakukan di TPS Pasar Jepang. Metode dalam penelitian ini menggunakan metode eksperimen dengan desain Posttest Only Design. Sampel yang digunakan dalam penelitian ini menggunakan teknik sampling jenuh yang artinya semua anggota populasi dijadikan sampel. Analisa data dalam penelitian ini menggunakan uji One Way Annova. Hasil: Persentase lalat yang hinggap pada warna kuning muda sebanyak 15,16%, warna kuning kenari sebanyak 46,87% dan warna kuning tua sebanyak 37,96%. Berdasarkan uji statistik menunjukan terdapat perbedaan efektivitas warna kuning fly grill dalam menarik lalat (p=0.0001). Kesimpulan: Fly grill warna kuning kenari efektif dalam menarik lalat karena mampu menarik lalat untuk hinggap sebanyak 46,87%.

Fly Grill adalah alat berupa potongan kayu yang disusun untuk melakukan survey kepadatan lalat. Lalat merupakan salah satu jenis serangga pengganggu dan dapat menyebarkan penyakit terhadap kesehatan manusia. Keputusan Menteri Kesehatan Republik Indonesia Nomor : 519/MENKES/SK/VI/2008 tentang pedoman penyelenggaraan pasar sehat dalam bidang pengendalian vektor dan binatang penular penyakit yang memerlukan adanya koordinasi pengendalian vektor salah satunya adalah lalat. Tujuan penelitian ini adalah untuk melihat efektivitas Fly Grill modifikasi dalam mengurangi kepadatan lalat di tempat penjualan daging di Pasar Sukaramai Kota Medan Tahun 2018. Penelitian ini merupakan penelitian eksperimen semu (quasi experiment) yaitu kegiatan percobaan yang bertujuan untuk melihat pengaruh yang ditimbulkan sebagai akibat adanya perlakuan dengan mengabaikan beberapa faktor pengganggu. Sampel penelitian adalah seluruh populasi yaitu setiap sudut yang mewakili tempat penjualan daging di Pasar Sukaramai Kota Medan sebanyak 2 sudut tempat penjualan daging dan diulangi selama 3 hari. Data diperoleh langsung melalui pengukuran kepadatan lalat dan observasi. Hasil penelitian menunjukkan pengukuran kepadatan lalat dengan Fly Grill yang sudah dimodifikasi dengan lem A lebih efektif yaitu (0,028) < 0,05 untuk mengurangi kepadatan lalat di Pasar Sukaramai Kota Medan karena didapatkan rata-rata yang telah melebihi baku mutu sesuai dengan Peraturan Menteri Kesehatan RI No. 50 Tahun 2017 Tentang Standar Baku Mutu Kesehatan Lingkungan Dan Persyaratan Kesehatan Untuk Vektor Dan Binatang Pembawa Penyakit Serta Pengendaliannya. Oleh karena itu disarankan untuk menggunakan Fly Grill dalam mengukur kepadatan lalat serta perlu adanya pemberantasan dan pengendalian lalat di Pasar Sukaramai Kota Medan dengan memodifikasi Fly Grill karena dari hasil penelitian yang didapatkan telah melebihi 2 ekor per blok grill.

The majority of gastrointestinal (GI) foreign bodies (FBs) discovered in adults are the result of intentional ingestion, most of which are found in patients with a preexisting psychiatric illness, with substance abuse disorders, or in people seeking secondary gain. No similar case of internal injuries following unintentional ingestion of a barbecue grill cleaning brush bristle has been reported. A 58-year-old Caucasian male with no significant history presented with complaint of halitosis, not improving after oral care and dental hygiene measures. He denied any other symptoms. After ruling out oral causes of halitosis, an esophagogastroduodenoscopy was performed, which revealed a black wire-shaped metallic FB embedded in the duodenum. The FB was identified as a silver metallic flexible wire resembling the bristle of a barbecue grill cleaning brush. The halitosis resolved completely within 3 weeks of the removal of the FB. To the best of our knowledge, this is the first case of duodenal impaction of a barbecue grill cleaning brush bristle atypically presenting with halitosis. Physicians’ awareness of this potential injury from grill cleaning brush bristles would lead to a quick diagnosis after a focused history. Also avid grillers must be made aware of this potential hazard and should be encouraged to carefully examine the barbecue grill surface for any remnant bristle of the grill cleaning brush. Appropriate history taking, considering GI causes as potentially inducing halitosis, and an awareness of this entity among providers are important to facilitate prompt diagnoses and prevent major adverse outcomes.
