Abstract:
Systemic mast cell activation disease (MCAD, a subclass of mastocytosis), which has a prevalence of around 17% (at least in the German population), is characterized by accumulation of genetically altered dysfunctional mast cells with abnormal release of these cells’ mediators. Since mast cells affect functions in potentially every organ system, often without causing abnormalities in routine laboratory or radiologic testing, this disease has to be considered routinely in the differential diagnosis of patients with chronic multisystem polymorbidity of a generally inflammatory and allergic theme. Pain in its different manifestations is a common symptom in MCAD found in more than three-quarters of the MCAD patients. Because of the specific mast cell-related causes of pain in MCAD it should be treated specifically, if possible, deduced from their putative mast cell mediator-related causes. As yet, there is no official guideline for treatment of MCAD at all. The present review focuses on mast cell mediator-induced acute and chronic pain and the current state of analgesic drug therapy options in MCAD. Due to the high prevalence of MCAD, many physicians are often faced with the issue of pain management in MCAD patients. Hence, our practical guide should contribute to the improvement of patient care.

Abstract:
Angiogenesis is the process by which new blood vessels form from existing vessel networks. In the past three decades, significant progress has been made in our understanding of angiogenesis; progress driven in large part by the increasing realization that blood vessel growth can promote or facilitate disease. By the early 1990s, it had become clear that the recently discovered “vascular endothelial growth factor” (VEGF) was a powerful mediator of angiogenesis. As a result, several groups targeted this molecule as a potential mediator of retinal ischemia-induced neovascularization in disorders such as diabetic retinopathy and retinal vein occlusion. Around this time, it also became clear that increased intraocular VEGF production was not limited to ischemic retinal diseases but was also a feature of choroidal vascular diseases such as neovascular age-related macular degeneration (AMD). Thus, a new therapeutic era emerged, utilizing VEGF blockade for the management of chorioretinal diseases characterized by vascular hyperpermeability and/or neovascularization. In this review, we provide a guide for clinicians on the development of anti-VEGF therapies for intraocular use.

Abstract:
A history of victimization has been linked to the latter development of emotional distress. However, not all victims develop emotional distress in response to victimization, emphasizing the need to identify mediators that can guide intervention, as well as moderators to more targeted preventive actions. Within a developmental psychology framework, we aimed to test two models: (a) the role of adult attachment as a mediator in the relationship between victimization and emotional distress, and (b) the role of household dysfunctions as moderators in the relationship between victimization and emotional distress, in psychiatric patients. Participants were 120 adult psychiatric patients, between ages 20 and 79 years ( M = −47.22, SD = 13.102) that responded to questions assessing household dysfunction in the family of origin, victimization, and adult attachment. Results revealed that adult attachment was a significant mediator in the association between victimization and emotional distress. Parental mental disorder and total household adversity were significant moderators for that same association. These results provide important clues for intervention. The focus on promoting secure adult relationships may contribute not only to the psychosocial adjustment of psychiatric patients but also to a healthier family functioning. Reducing the household dysfunction may provide a protective environment for the development of children, promoting a positive psychosocial adjustment, also preventing the intergenerational transmission of violence, insecure attachment, and emotional distress.

Sphingosine 1-phosphate (S1P), a metabolic product of cell membrane sphingolipids, is bound to extracellular chaperones, is enriched in circulatory fluids, and binds to G protein–coupled S1P receptors (S1PRs) to regulate embryonic development, postnatal organ function, and disease. S1PRs regulate essential processes such as adaptive immune cell trafficking, vascular development, and homeostasis. Moreover, S1PR signaling is a driver of multiple diseases. The past decade has witnessed an exponential growth in this field, in part because of multidisciplinary research focused on this lipid mediator and the application of S1PR-targeted drugs in clinical medicine. This has revealed fundamental principles of lysophospholipid mediator signaling that not only clarify the complex and wide ranging actions of S1P but also guide the development of therapeutics and translational directions in immunological, cardiovascular, neurological, inflammatory, and fibrotic diseases.

Purpose The purpose of this paper is to analyse and discuss how sensory cues are preferred in relation to products, service and store image in a retail context and why retailers should consider the importance of congruence in a retail setting. Design/methodology/approach Four qualitative focus group discussions were conducted, following a semi-structured interview guide. A total of 24 participants discussed how and why a lighting department in an IKEA store in Sweden could be regarded as more appealing than the traditional layout. Findings The findings indicate that congruency works as a mediator between a retail setting, sensory cues, products, service and store image. Consumers prefer, compare and categorize sensory cues in relation to the specific product in the department, the service offered by the firm and the store image. Originality/value A model is developed that conceptualizes congruency as a mediator in a retail setting, which provides an opportunity to further explore external influences on congruency in retail settings, both conceptually and empirically.

Abstract:
Regorafenib is used for hepatocellular carcinoma (HCC), but its response does not last long, partly due to chemoresistance acquisition. We performed a clustered regularly interspaced short palindromic repeats (CRISPR)-based loss-of-function genetic screen and aimed to discover molecules involved in regorafenib resistance in HCC. Xenograft tumors established from Cas9-expressing HCC cells with pooled CRISPR kinome libraries were treated with regorafenib or a vehicle. Sequencing analysis identified 31 genes, with the abundance of multiple guide RNAs increased in regorafenib-treated tumors compared to that in vehicle-treated tumors, including 2 paralogues, LATS2 and LATS1, core components of the Hippo signaling pathway. Notably, all eight designed guide RNAs targeting LATS2 increased in regorafenib-treated tumors, suggesting that LATS2 deletion confers regorafenib resistance in HCC cells. LATS2 knockdown significantly mitigated the cytotoxic and proapoptotic effects of regorafenib on HCC cells. LATS2 inhibition stabilized the Hippo signaling mediator YAP, leading to the upregulation of antiapoptotic Bcl-xL and the multidrug resistance transporter ABCB1. Among 12 hepatoma cell lines, the half maximal inhibitory concentration (IC50) values of regorafenib were positively correlated with any of YAP, Bcl-xL and ABCB1 levels. The inhibition of YAP or Bcl-xL in regorafenib-insensitive HCC cells restored their susceptibility to regorafenib. In conclusion, our screen identified the Hippo signaling pathway as the mediator of regorafenib efficacy in HCC.

AbstractObjective:The present study aimed to identify whether discretionary food consumption declined in an intervention focused primarily on promoting fruit and vegetable consumption. We also aimed to identify potential mediators explaining intervention effects on discretionary food consumption.Design:Secondary analysis of data from the ShopSmart study, a randomised controlled trial involving a 6-month intervention promoting fruit and vegetable consumption. Linear regression models examined intervention effects on discretionary food consumption at intervention completion (T2). A half-longitudinal mediator analyses was performed to examine the potential mediating effect of personal and environmental factors on the association between the intervention effects and discretionary food consumption. Indirect (mediated) effects were tested by the product of coefficients method with bootstrapped se using Andrew Hayes’ PROCESS macro for SPSS.Setting:Women were recruited via the Coles FlyBuys loyalty card database in socio-economically disadvantaged suburbs of Melbourne, Australia.Participants:Analyses included 225 women (116 intervention and 109 control).Results:Compared with controls, intervention participants consumed fewer discretionary foods at T2, after adjusting for key confounders (B = −0·194, 95 % CI −0·378, −0·010 servings/d; P = 0·039). While some mediators were associated with the outcome (taste, outcome expectancies, self-efficacy, time constraints), there was no evidence that they mediated intervention effects.Conclusions:The study demonstrated that a behavioural intervention promoting fruit and vegetable consumption among socio-economically disadvantaged participants was effective in reducing discretionary food intake. Although specific mediators were not identified, researchers should continue searching for mechanisms by which interventions have an effect to guide future programme design.

Liver disease accounts for millions of deaths worldwide annually being a major cause of global morbidity. Hepatotoxic insults elicit a multilayered response involving tissue damage, inflammation, scar formation, and tissue regeneration. Liver cell populations act coordinately to maintain tissue homeostasis and providing a barrier to external aggressors. However, upon hepatic damage, this tight regulation is disrupted, leading to liver pathology which spans from simple steatosis to cirrhosis. Inflammation is a hallmark of liver pathology, where macrophages and endothelial cells are pivotal players in promoting and sustaining disease progression. Understanding the drivers and mediators of these interactions will provide valuable information on what may contribute to liver resilience against disease. Here, we summarize the current knowledge on the role of macrophages and liver sinusoidal endothelial cells (LSEC) in homeostasis and liver pathology. Moreover, we discuss the expanding body of evidence on cell-to-cell communication between these two cell compartments and present triggering receptor expressed on myeloid cells-2 (Trem-2) as a plausible mediator of this cellular interlink. This review consolidates relevant knowledge that might be useful to guide the pursue of successful therapeutic targets and pharmacological strategies for controlling liver pathogenesis.

Purpose The purpose of this study is to identify the role of mediators in supporting value co-creation for vulnerable consumers in a service context. The authors propose that in transformative services, the roles of actor mediators facilitate control and empowerment for the vulnerable consumer – labelling these transformative service mediators (TSMs). Design/methodology/approach The authors develop a theoretical framework for the activities of mediators in value co-creation considering the interrelationships of vulnerability, structure and agency. The authors then use Prahalad and Ramaswamy’s DART (Dialogue, Access, Risk Assessment and Transparency) model as the integrating framework to describe the TSM roles in the context of the foster care service ecosystem. Findings The authors introduce a future research agenda regarding TSM roles in transformational service experiences and value co-creation with vulnerable consumers. Service researchers and providers are encouraged to explore effective training and motivation of TSMs. Research limitations/implications Understanding value co-creation for vulnerable consumers is an emerging area in service research. The TSM concept introduces a new approach to explore how value co-creation and transformative outcomes can be enhanced in service contexts where consumers experience vulnerability. Practical implications This paper presents an agenda for future research. The outcomes of future research based on TSM roles may help guide service providers in identifying opportunities for enhancing well-being and reducing vulnerability in service delivery. Originality/value This paper suggests that exploring the role of TSMs in the service process offers new insights into reducing vulnerability in service relationships.

Five study guides prepared for two productions of I, Claudia by Kristen Thomson will serve as a case study to determine the nature and orientations of the tools provided by theatre companies to pedagogues, who in turn teach students how to understand and read theatrical performance. More specifically, this case study will seek to explain how processes typical of “cultural mediation” help students accumulate what Pierre Bourdieu names “cultural” or “social capitals”, both necessary when training future citizens. This thesis will thus serve as a first critique of the workings, strengths and failings of a sociological mechanism typical of English Canadian theatre designed to help teachers mediate theatre and create more informed audience members.

Abstract:
The thesis presents a critique of the nature of 13 to 15 year-old students' images of futures. Arguments are made for their importance to the individual community and culture and their value and use in a science learning setting.This study of the nature of futures images is based upon data collected from small samples in two school settings between 1990 and 1999 using a guided fantasy approach. The diversity of student hopes and fears for futures is categorised using several research tools developed from the futures literature. The results identify a range of futures scenarios ranging from highly utopic to highly dystopic and themes including human interactions, the natural and built environment and the nature and use of futuristic technologies.The importance of student images of futures is discussed from the personal perspective of the students and then considered in the broader context of society and Western culture. The data are examined through a number of theoretical frames including psychology, history and the futures field of enquiry. The consensus seems to be that expectations of the future are inseparable from human nature and influential in determining the viability of cultures.The value of student images of futures in science learning is explored through a case study of one of my own Year 9 classes. In teaching this class I used the technique of futures imaging integrated into a critical futures teaching approach utilising a constructivist planning model. I found that students' futures images revealed a broader aspect of student worldviews than is usual in science learning environments, and were valuable in exposing student prior knowledge, interests and concerns. Student learning seemed to be enhanced using this futures oriented approach.

While interagency collaboration among Children’s Aid Societies and violence against women (VAW) agencies have been mandated by the Ontario Ministry of Community and Social Service, little is known about these local collaboration agreements. This study seeks to explore how the Ottawa CAS/VAW Collaboration Agreement mediates the work of VAW agencies to protect women and children. Using a purposive sample, a total of eight VAW informants were interviewed. Smith (1999) argued that people’s everyday experiences are organized, often unknowingly, by the actions of people located outside the local setting and that this organization is textually-mediated. This study used institutional ethnography and the listening guide approach to critically examine the collaboration process. The results explore the narratives and standpoints as they relate to the informants’ understanding of the agreement and their descriptions of doing collaboration. This study also adopts the mapping technique developed by institutional ethnographers to map social relations. The findings indicate that informants differed in their familiarity and knowledge of the contents of the collaboration agreement – only two informants indicated that they have reviewed and read the document. The findings also show that although most informants were able to describe positive experiences of collaboration, most also described negative experiences. This study concludes that the collaboration agreement has made VAW workers’ work with women more focused and specific to helping women address Children Aid Society’s concerns. The findings demonstrate that the collaboration agreement requires an update and further research is required to evaluate these collaborations and whether they improve outcomes for women and their children.

Our world is made up by millions of colors. Everywhere we go we see a variation of colors that describes the environment. Artists use colors to give an emotion to the viewer and give a deeper meaning to the art. Companies have different colors in their logos and on their websites. For example, a red colored logo can give a feeling of passion, energy and power. But used in a different situation, a red color can implicate dangerand death. The research about colors goes very deep, but there is still alot that has not been tested or discovered. This study will examine whether companies can use colors to mediate the right feeling to their customers. The method to do this included a lot of theoretical background and knowledge about colors. When data was collected a mid-fi prototype of a website was created. This mid-fi website was then transformed into three different high-fi prototypes. The three prototypes had the same content but what put them apart was their color schemes. The tests was done face to face or over the internet. The test person had to answer a short questionnaire were the person described the feelings that appeared when using the three different prototypes. After the test a short interview with each test person took place were some expressions from the test was discussed. The result for the first and second website showed a correlation between the two websites. The users had similar answers on these websites, but they also had some comments that separated the websites. The second website was in the end the most popular website which supported the hypothesis.The conclusion from the study is that colors are a very powerful tool to use if a company wants to attract more customers. This was a quite small study and to draw the conclusion that the right colors actually gives more clicks on a website, a larger and longer study must be made. This is so that the study can compare previous years sales with the year werethey introduced the new color scheme.

The subject of this thesis consists in an analysis of an experiment that took place over the course of two years, at the kindergarten level, in association with « Philosoph’art », an organization from Lyon. In this research, we interrogate the status of innovation of this experience and we focus on the posture of the quizmaster during workshops with philosophical content as well as artistic projects. We closely examine what sets apart such experiences from those already in use in philosophy with children and artistic practices. On pourrait dire que le projet de cette thèse consiste à comprendre l’articulation dans l’espace, du tourisme et de la communication dans des situations qui mettent en tension un sujet individuel, dévoilant toute la singularité de son désir dans des récits ou des pratiques et des acteurs collectifs engagés dans les politiques touristiques, les médias et les institutions du tourisme.Notre réflexion s’organise ainsi en suivant les modalités de l’articulation entre ces dimensions réelle, symbolique et imaginaire du tourisme. Dans le champ des SIC et particulièrement grâce à des modèles permettant de formaliser la médiation touristique, notre approche conceptuelle permet d’appréhender les significations du tourisme. Nous modéliserons les composantes, les liens et interrelations de la médiation touristique pour proposer une approche théorique de ce que l’on peut appeler la signification touristique de l’espace.En premier lieu, nous abordons les définitions, un état de la recherche en tourisme et le lexique du tourisme par l’étymologie et l’analyse lexicale des termes les plus fréquents : « tour », « tourisme », « touriste », « loisir », « villégiature », « vacances » et « voyage ».En second lieu, nous proposons de poser les bases du champ de la médiation touristique et de montrer que la pratique sociale du tourisme est une médiation. Nous présenterons, commenterons et analyserons, en nous fondant sur un schéma de la médiation touristique, les différentes relations de communication en jeu dans la médiation touristique. Puis nous présenterons les structures de la médiation touristique pour fonder « une théorie critique » de la médiation touristique.Par la suite, nous approfondirons le concept en présentant les opérateurs, les acteurs et les politiques de l’organisation du tourisme, en nous fondant sur l’analyse d’exemples et d’études concernant principalement la médiation et les médiateurs dans le cas de la France pour comprendre comment pratiques et scènes de tourisme s’organisent. Par la suite, nous décrirons et analyserons le système d’information touristique en deux points. Le premier concerne l’énonciation de l’espace produit par le touriste, les touristes et les médiateurs du tourisme (énonciation de la parole du guide par exemple) au travers des récits pluri et multimédias. L’autre concerne la mise en œuvre des stratégies par les destinateurs (opérateurs et acteurs de l’organisation du tourisme privés et publics) au travers des médias du tourisme (généralistes, spécifiques du tourisme et guide touristiques). It may be said that this thesis aims at understanding the spatial articulation between tourism and communication in such situations as those in which an individual subject – expressing the whole peculiarity of his or her desire through telling or practices – and collective actors are brought together.My reflection thus organises itself in accordance with the modalities of the articulation between the three dimensions of tourism: the practical dimension, the symbolic dimension and the imaginary dimension.Tourist activity consists in travelling and in visiting a site, a place or a destination; tourist mediation means the range of activities that are organised around tourism or those that are associated to it. Tourist mediation concerns the agents and the practices in the field of tourism. In this context, I wish to show that tourist mediation structures the perceptions, the interpretations and the practises of space.I address the definitions; give an overview of academic research on tourism and address the notions of tourism through the etymology of its main words (tour, tourism, and tourist) and through the differences that exist between tourism and other notions similar to it such as leisure, vacations, holiday and travelling. These notions are necessary to the comprehending of tourist mediation.I intend to lay the foundations of the field of tourist mediation and to demonstrate that the social practise of tourism is a form of mediation. I will present the tourist objects that structure the tourist practises of space. The structures of tourist mediation will then be laid; this will enable me to craft a “critical theory” of tourist mediation. With a view to understanding how the practises and the scenes of tourism organise, I will subsequently widen the concept by presenting the operators, the agents and the policies of the tourist organisation; the examples and the studies of the latter are mainly concerned with mediation and mediators in the case of France. I situate tourist mediation in relation to the experience of otherness and to the relationship to the others, to wanderlust, to the tourist imagination and to cultural imagination too. I then describe and give a two-point analysis of the tourist information system. The first point is concerned with the enunciation of space produced by the tourist, the tourists and by the tourist mediators (the enunciation of the guide’s discourse for instance) through various multimedia telling. The second point is concerned with the enunciation, through the tourist media (non-specialised or tourism media, tourist guides), by the speakers (e.g. operators and private or public agents of the tourist organisation) of the communication strategies and of the tourist descriptive signing. Sonification with musical characteristics can engage users, and this dynamic carries value as a mediator between data and human perception, analysis, and interpretation. A user engagement study has been designed to measure engagement levels from conditions within primarily melodic, rhythmic, and chordal contexts. This paper reports findings from the melodic portion of the study, and states the challenges of using musical characteristics in sonifications via the perspective of form and function – a long standing debate in Human-Computer Interaction. These results can guide the design of more complex sonifications of multivariable data suitable for real life use.
