Abstract:

Abstract:
Abstract:

Abstract:
This article reads Dalila Kerchouche’s Mon père, ce harki (My Father, this Harki) as a postcolonial filiation narrative, which blends memoir and biography, the personal and collective, the past and present. Lack of knowledge and a desire to see for herself the camps her parents and older siblings experienced prompts Kerchouche to adopt an investigative posture characterized by in situ exploration in conjunction with interviews and the consultation of archives. This allows the author to achieve a polyphonic account of the past. At the same time, her family serves as the prism through which she confronts the stigma attached to Harkis (Algerian soldiers hired by the French Army) and examines their unjust treatment in France.

Abstract:
The engagement in the French army during the Algerian war condemned the harkis to a silent existence. Considered as traitors in their country of birth, and undesirable in their country of exile, they strive to remain invisible and silent. In the writings of their descendants, who decided one day to tell the traumatic Hi/story of their community, a character inevitably stands out: the father walled in silence, leading his life alone, for fear of disappointing or not being able to give the best explanations for a “shameful” past. To illustrate the different “configurations” and values of the “entrenched words” that the descendants of harkis inscribe in their literary works, we will base our analysis on the novels Mon père, ce harki (2003) by Dalila Kerchouche, Mohand, le harki (2003) by Hadjila Kemoum and L’Art de perdre (2017) by Alice Zeniter.

Abstract:
In recent decades, female descendants of harkis - indigenous Algerian men employed as auxiliary soldiers in the French army during the Algerian War - have privileged the literary text as a site of reconstruction of silenced familial and collective (hi)stories, and of reconnection with their effaced ancestral heritage. This article examines representations of physical, affective, and imaginative “returns” in two literary works by members of different harki (post)generations: France: Récit d’une enfance (2006) by Zahia Rahmani, a daughter of a harki, and L’Art de perdre (2017) by Alice Zeniter, a harki’s granddaughter. It is argued that these multivocal narratives problematize straightforward understandings of belonging and inheritance to interrogate the reparative potential of return - whether real or imagined, spatial or temporal - and to confront the ongoing effects of (neo)colonial narratives and power structures. Both texts present the notion of return as simultaneously intimate and broad in scope, resulting at once from external pressures and personal necessity, and capable of healing certain wounds while resisting definitive closure. Invoking the frequently gendered role of storytelling in Arabo-Berber societies, the novels also establish dialogues and connections with disparate histories, memories, and literary texts, allowing their protagonists to transcend and deconstruct static, assigned identities. The texts’ filiative and affiliative returns across time and space are shown to reflect Marianne Hirsch’s conception of the existence of vertical and horizontal forms of “postmemory” (2012) and Michael Rothberg’s notion of “rhizomatic networks” (2009). It is argued that it is precisely the notion of return which emerges as a creative organizing principle, allowing Rahmani and Zeniter to negotiate aspects of transgenerational trauma, absence, and loss, while also turning their intimate, self-reflexive quests outwards to critique and rewrite pre-established narratives and to inscribe their texts within current interrogations of commemoration and reparation in postcolonial contexts.

This article proposes a reading of Mehdi Charef’s Le Harki de Meriem [‘Meriem’s Harki’] (1989) and its 2016 edition that focuses on the text’s discursive and paratextual practices employed to represent Franco-Algerian historical and cultural heritage. This non-linear narrative presents a family saga over the course of the twentieth century, revealing in the process key episodes of the mutual and turbulent history of colonial Algeria and postcolonial France. Through a discussion of passages and editorial work that relate the novel with historical events, I will trace patterns of critique that emphasize their concern with history as a source of knowledge. Such a dialectical analysis will in turn provide a reading paradigm that paves the way into a hybrid France that fragments both the French national grand narrative and Franco-Algerian memory.

AbstractUsing the case study of Montpellier, this article explores the relationship between local political actors and postcolonial minorities since the end of the Algerian War – particularly, the city's pied-noir, harki, Moroccan and Jewish populations. It examines the discourses used to secure the electoral allegiances of these groups and the myriad ways in which they laid claim to certain civic and political spaces. It employs diverse oral, archival and audio-visual sources to demonstrate how postcolonial minorities have gained important concessions from local authorities and how identity politics has developed under the Fifth Republic, despite France's strong republican tradition.

Abstract:
Alice Zeniter’s 2017 novel The Art of Losing, translated recently by Frank Wynne from French to English, explores how buried histories resurface and haunt generations to come, despite national efforts to ignore, if not minimalize, the enduring impacts of colonialism, independence struggles and exile. Set in contemporary France in the wake of the 2015 terrorist attacks, and loosely inspired by Zeniter’s own family history, the book follows Naïma, a young woman of Algerian decent who grapples with a largely unknown and misconstrued harki heritage. Drawing on Marianne Hirsch’s concept of postmemory, this article investigates intergenerational transmission of memory, trauma, and silence around themes such as war, exile and integration.

Abstract:

Abstract:
Abstract Overexpression of the MYCN proto-oncogene is one of the strongest biomarkers for poor prognosis in neuroblastoma patients. N-Myc, encoded by MYCN, is a highly attractive yet “undruggable” target in neuroblastoma drug discovery. N-Myc is a short-lived protein in normal neuronal progenitor cells; however, in MYCN-amplified neuroblastoma cells, overexpressed N-Myc is stabilized by high levels of Aurora kinase A (Aurora-A). Several Aurora-A allosteric modulators have been developed to disassociate N-Myc binding and promote its degradation. These compounds function through a competitive mechanism against N-Myc, and clinical trial results suggest limited efficacy in high-risk neuroblastoma patients where N-Myc is highly expressed. To develop Aurora-A modulators with greater effects in destabilizing N-Myc, targeted degradation of Aurora-A was investigated in this study. First, we developed a highly selective Aurora-A-binding ligand by chemically modifying ribociclib, which targets CDK4/6. Next, a series of potent Aurora-A/N-Myc degraders were developed using proteolysis targeting chimaera (PROTAC) technology. Our lead compound, HLB-0532259, was found to substantially decrease N-Myc levels in neuroblastoma cells following the degradation of Aurora-A with nanomolar potency. Mechanistic studies suggest that HLB-0532259 binds to Aurora-A in an uncompetitive manner with respect to N-Myc, which contributes to its high potency for degrading the Aurora-A/N-Myc pool. Mass spectrometry analysis demonstrates that HLB-0532259 possesses remarkable proteome-wide selectivity for Aurora-A and N-Myc degradation. Further studies have revealed that HLB-0532259 exhibits nanomolar cytotoxicities against a panel of MYCN-amplified neuroblastoma cells, good pharmacokinetic properties, and favorable therapeutic outcomes in a xenograft neuroblastoma mouse model. Taken together, our data strongly support HLB-0532259 as a promising lead compound for neuroblastoma therapy. Citation Format: Jian Tang, Ramkumar Moorthy, Ozlem Demir, Zachary D. Baker, Jordan A. Abstract:
Human arginase I (hARGI) is an important enzyme involved in the urea cycle; its overexpression has been associated to cardiovascular and cerebrovascular diseases. In the last years, several congeneric sets of hARGI inhibitors have been reported with possible beneficial roles for the cardiovascular system. At the same time, crystallographic data have been reported including hARGI–inhibitor complexes, which can be considered for the design of novel inhibitors. In this work, the structure–activity relationship (SAR) of Cα substituted 2(S)-amino-6-boronohexanoic acid (ABH) derivatives as hARGI inhibitors was studied by using a three-dimensional quantitative structure–activity relationships (3D-QSAR) method. The predictivity of the obtained 3D-QSAR model was demonstrated by using internal and external validation experiments. The best model revealed that the differential hARGI inhibitory activities of the ABH derivatives can be described by using steric and electrostatic fields; the local effects of these fields in the activity are presented. In addition, binding modes of the above-mentioned compounds inside the hARGI binding site were obtained by using molecular docking. It was found that ABH derivatives adopted the same orientation reported for ABH within the hARGI active site, with the substituents at Cα exposed to the solvent with interactions with residues at the entrance of the binding site. The hARGI residues involved in chemical interactions with inhibitors were identified by using an interaction fingerprints (IFPs) analysis.

Abstract:
The purpose of our study is to investigate early cellular, molecular, morphological and behavioral changes in humanized amyloid-beta-knock-in (hAbKI) mice. Using seven-month-old homozygous hAbKI mice, we studied behavioral phenotype parameters, including spatial learning and memory (Morris Water Maze), locomotor activity (open field), working memory (Y-maze) and motor coordination (rotarod); mRNA abundance, protein levels, soluble amyloid-beta 40 and 42 levels and regional immunoreactivities of key markers of mitochondrial dynamics, mitochondrial biogenesis, synaptic health, mitophagy and autophagy; mitochondrial function and using transmission electron microscopy & Golgi–Cox staining, we assessed mitochondrial morphology and dendritic spines. Our extensive behavioral analysis revealed that seven-month-old hAbKI mice showed impairments in motor coordination, reduced locomotor and exploration activities, impairments in working memory and spatial learning and memory. Our mRNA and protein analyses revealed the increased expression of mitochondrial-fission genes and reduced expression of mitochondrial-fusion, mitochondrial-biogenesis, synaptic, autophagy and mitophagy genes in seven-month-old hAbKI mice. An immunofluorescence analysis revealed altered immunoreactivities and agreed with the immunoblot results. Transmission-electron-microscopy data revealed increased mitochondrial fragmentation and reduced mitochondrial length in both hippocampal and cortical tissues of seven-month-old hAbKI mice and mitochondrial function defective. A Golgi–Cox-staining analysis revealed reduced dendritic spines in both cerebral cortices and hippocampi of hAbKI mice. Soluble amyloid-beta (1–40 and 1–42) were detected in three-month-old hAbKI mice and progressively increased in seven-month-old mice. These observations suggest that the human amyloid-beta peptide is sufficient to cause behavioral, mitochondrial, synaptic and ultrastructural changes in seven-month-old hAbKI mice. Our study findings also suggest that hAbKI mice might serve as a model for preclinical studies of preventive therapies.

Full text
Abstract:
Background: We estimated the incidence of hospital-acquired respiratory virus infections (HARVIs) by viral species, and we identified risk factors for and outcomes of HARVIs. Methods: We identified a cohort of all inpatient admissions of ≥24 hours duration to University of Michigan hospitals during 3 study years (2017–2018, 2018–2019, and 2019–2020). HARVIs were defined as initial respiratory virus detection (adenovirus, coronaviruses, human metapneumovirus, influenza A and B, parainfluenza viruses, respiratory syncytial virus, or rhinovirus-enterovirus) in a clinical test ordered after the 95th percentile of the virus-specific incubation period. Incidence was calculated as the number of HARVIs per 10,000 patient admission days. Patient demographic and clinical characteristics were assessed as risk factors for HARVI in Cox proportional hazards models of the competing outcomes of HARVIs and hospital discharge. The association between time-varying HARVI status and the competing outcomes of discharge and in-hospital death was estimated in covariate-adjusted Cox-proportional hazards models. All analyses were performed separately for adult patients (aged ≥18 years) and pediatric patients (aged <18 years). Results: The overall incidences of HARVI were 8.5 and 3.0 per 10,000 admission days for pediatric and adult patients, respectively. Rhinovirus was the most common HARVI in both pediatric and adult patients, with incidences of 5.1 and 1.1 infections per 10,000 admission days, respectively. With the exception of influenza A, the incidence of HARVI was higher in pediatric patients than adult patients for all viral species. For adults, congestive heart failure, renal disease, and cancer all increased HARVI risk independent of their associations with extended hospital stays. Risk of HARVI was also elevated for patients admitted September through June relative to July admissions. For pediatric patients, chronic cardiovascular and respiratory conditions, cancer, medical-device dependence, and December admission increased risk of HARVI. Age, sex, and race were not associated with risk of HARVI for children or adults. Inpatient lengths of stay were longer for adults with HARVI compared to those without (range of virus-specific hazard ratios, 0.48– 0.77). However, estimated effects were not statistically significant for human metapneuomovirus, parainfluenza, or adenovirus. Only influenza A was associated with an increased risk of in-hospital death within 30 days of infection for adults. No HARVIs were associated with increased length of stay or risk of death for pediatric patients. Conclusions: The incidence of HARVI varied by viral species and was higher among pediatric patients. HARVIs increased the length of hospital stays for adults but not for pediatric patients.Disclosures: None

Abstract:
Abstract Background Healthcare-associated infections (HAI) are major preventable causes of morbidity and mortality. While there are fewer overall HAI in children, there is a greater potential impact in disability-adjusted life years. Healthcare-associated respiratory viral infections (HARVI) are not frequently tracked within institutions, yet the risk for such infections in pediatric hospitals is very high. Recent data demonstrate large inter-hospital variability of HARVI incidence that may depend on various factors including the number of immunocompromised patients in the hospital and the presence of shared rooms. We hypothesize that the burden of healthcare-associated respiratory viral infections and their impact on the length of stay (LOS) is substantial at a large urban pediatric hospital. Methods A cohort of all children with any HARVI admitted to a large urban pediatric hospital between July 2017 and June 2018 were included after obtaining IRB approval. We defined a HARVI as a respiratory infection with an onset of symptoms while the patient was hospitalized meeting three criteria: A positive microbiologic test for one of 8 viruses, presence of symptoms of a respiratory infection, and onset of symptoms after admission beyond the minimum incubation period for each virus. Infections with symptom onset after admission beyond the maximum incubation period were considered definite hospital onset whereas others were considered possible hospital onset. The electronic medical record provided data on demographics, underlying medical conditions, hospital length of stay prior to infection and hospital unit of infection, and consequences and outcome of HARVI. The at-risk population for calculation of the incidence of HARVI was all admitted patient-days at the hospital over this time period. Results Between July 2017 and June 2018 the incidence of HARVI (definite or possible hospital onset) was 1.2 infections per 1,000 admitted patient-days (60% due to rhinovirus/enterovirus, 12% due to respiratory syncytial virus, and 9% due to influenza). Overall, 48% of patients were under 2 years of age, 18% were between 2 and 5 years of age, and 34% were over 5 years of age. Twenty-one percent were immunocompromised and 35% had underlying lung disease. The median length of stay prior to symptom onset was 11 days (IQR 5–36 days) and the median total length of stay was 30 days (IQR 15–82.5 days). Eight individuals had more than one HARVI over this time period. Nineteen percent were transferred to the intensive care unit and 7% died during their hospital admission Conclusion HARVI occurs frequently in a pediatric hospital and often in patients with underlying comorbidities. The risk for HARVI increases substantially with increased length of stay. Such data support the need for tracking HARVI in high-risk institutions.

Abstract:
A túlélés, elkerülés, ellenállás, kiemelés (SERE2) témakörében folyó kiképzéseknek az egész Magyar Honvédségre kiterjedő magas szakmai szintű megvalósulása fokozza az állomány magabiztos feladat-végrehajtását, növelve ezzel a teljes állomány harci morálját. Amennyiben a kialakult harchelyzet lehetővé teszi, akkor katonáink bajba jutása esetén arra dedikált vagy kijelölt erők fogják a kiemelésüket végrehajtani a harci mentés (CR3) vagy a harci kutatás-mentés (CSAR4) módszereivel. A segítség fogadásához szükséges ismereteket a potenciális személyek a SERE-kiképzés során tudják elsajátítani. Abban az esetben, amikor a saját csapatok a kialakult harci helyzet miatt nem tudnak az elszigetelődött katonák segítségére sietni, akkor a jól felépített és végrehajtott SERE-kiképzésnek köszönhetően katonáink képesek lesznek túlélni, miközben elkerülik az ellenséget és végrehajtják a saját csapatokhoz történő visszaszivárgást, vagy fogságba esés esetén ellenállnak a kényszervallatásnak.

In this paper, whānau Māori highlight how a Kaupapa Māori-centred intervention (the Harti Hauora Tamariki tool, hereafter Harti tool) has improved interactions with health services. The Harti tool is undergoing a randomised control trial (RCT) at Waikato Hospital in New Zealand. As part of the RCT, the authors engaged in a series of qualitative interviews with whānau members of tamariki Māori (children aged 0–5 years) admitted to Waikato Hospital’s paediatric ward. Whānau who met at least one criteria for New Zealand’s domains of deprivation were included. Using a Kaupapa Māori approach to the study, participants shared their views on barriers and facilitators to accessing health resources and primary care services. The interviews conducted highlight how the Harti tool, when administered in a culturally appropriate and respectful manner that prioritised relationship-building, enabled better connection to healthcare services. Prevalent in our analysis were connections to wider determinants of health and ways to reduce existing health inequities. To conclude the paper, how the Harti tool has enhanced feelings of being in control of health, with the potential to reduce the likelihood of a hospital readmission, is highlighted.

Abstract:
The right to life is one of the fundamental rights. There are many moral and legal debates on the right to life of embryos and fetuses. Aside from the debates on whether the embryo/fetus can be considered a human and person, determining the moment when the fetus acquired the legal capacity and personality is important for the legal protection of the embryo/fetus. In addition to the crimes and abortion, with the technological developments in biology, the violations of the right to life of the fetus have increased. Especially with the implementation of assisted reproductive treatment methods, new debates arose about the right to life of IVF embryos. Besides that, genetic diagnostic test applications, spare and research embryos have revealed the importance of determining the legal status of the embryo/fetus. The legal status of the embryo and fetus within the national legislation is still unclear. In the decisions given by the ECHR, it hasn’t reached a definite judgment about when the right to life begins. ECHR, in most of its decisions, didn’t discuss the right to life of the embryo/fetus. When the court found the issue worth discussing, it concluded that the fetus didn’t have the right to life before birth.

Abstract:
Aim: This study aimed to investigate the polymorphisms in genes related to meat production, including growth hormone receptor (GHR) and diacylglycerol acyltransferase 1 (DGAT1) genes, in different breeds of sheep, including Barki, Najdi, and Harri. Materials and Methods: Blood samples were collected from 75 randomly selected healthy Barki, Najdi, and Harri breeds of sheep, with 25 samples per breed. GHR and DGAT1 genes were identified using a single nucleotide polymorphism assay followed by digestion with the restriction enzyme Alu1. Results: The analysis of the GHR gene sequence showed nucleotide substitutions at nt 69 in exon 10 (c.69 G > A); this mutation is considered a transition mutation. The sequences of detected SNPs in the GHR gene in the different sheep breeds were submitted to the GenBank database with accession numbers MG906773 to MG906781. The substitutions at exon 10 (c.69 G > A) results in an alteration to the amino acid (p. Lysine > Arginine). At c.69, the A allele frequency was 0.61, 0.59, and 0.54, while the G allele frequency was 0.39, 0.41, and 0.46, for Barki, Najdi, and Harri breeds, respectively. The genotype AG at nt 69 locus had the highest frequency in the Najdi and Harri sheep. The frequency of AG was 0.62, 0.61, and 0.64, while the frequency of AA was 0.30, 0.28, and 0.22, for Barki, Najdi, and Harri sheep, respectively. After digestion with the restriction enzyme AluI, the DGAT1 locus had two genotypes, CC and CT. The highest frequency, 0.88, was found for allele C, which was detected in Barki breed. The lowest frequency, 0.75, for the same allele was found for Harri. Conclusion: The detected CT genotype may explain the moderate intramuscular fat content and muscle marbling in the Barki sheep breed.
