Abstract:
Abstract Leaves and internodes from Stevia rebaudiana Bertoni plants growing in different conditions were used for transformation with two strains of Agrobacterium rhizogenes: ATCC 15384 and LBA 9402. Hairy roots formation was observed and the percentage of the transformed explants depended on the type of explant, time of inoculation and inoculum concentration. Inoculation of explants from ex vitro and in vitro plants with LBA 9402 strain led to higher efficiency of transformation than inoculation with ATCC 15384 strain. Growth rate of hairy roots in liquid culture was assessed under light and dark conditions. It was found that the growth of hairy roots decreased significantly under light conditions. Transformation of hairy roots growing in different culture conditions was confirmed at the molecular level using PCR method with primers constructed against rolB and rolC genes from A. rhizogenes.

Abstract:
The plant Stevia rebaudiana Bertoni (2n=22) is a native of certain regions of South America particularly in Paraguay. It is a short-term plant and needs three months to complete one cycle of vegetative growth and flowering. With days less than 13 hours in tropical countries, stevia plants flower early, resulting in low yield. In addition, the early bud emergence under the short-day length condition causes the sugar content in stevia leaves to reduce drastically, making them no longer valuable for commercial use. The stevia accessions available in Malaysia today are limited in number and poor in performance, emphasizing the necessity of varietal improvement programs. Assessment of genetic variability, diversity and intrarelationships is an essential step for such breeding programs. This study aims to evaluate agronomic performance of among 21 stevia accessions, namely SRBA-1 to SRBA-21, collected from different origins and to reveal genetic diversity utilizing 52 novel microsatellites. Evaluation of agronomic traits revealed wide range of variation in leaf weight, dry leaf weight, plant height, number of branches, stevioside; rebaudioside A (Reb A), rebaudioside C (Reb C) and total steviol glycosides (TSG). The total genetic diversity detected among the accessions through amplification of the 43 polymorphic microsatellites showed that almost all markers had deviation from Hardy–Weinberg equilibrium (Ho>He). Three distinct heterotic groups were identified among the accessions based on their agronomic performance and molecular characteristics. Crosses among different accessions coming from different heterotic groups can be further used to produce potential stevia variety for plantation in Malaysia.

Abstract:
The purpose of this Master’s Report was to determine an appropriate preservation strategy for a particular set of buildings and their accompanying art from the Modern Architecture Movement. The research question was: What type of strategy is best suited for the preservation of installed art created for Modern style buildings? The study analyzed preservation strategies afforded to Modern art and architecture during rehabilitation of the buildings. The case studies are Modern Movement office or bank buildings with art commissioned for the space by the architects or owners. An analysis of the main case study’s preservation strategies looks at all the actions taken and proposed to protect, not only the material fabric of the art, but the primary interior space. The main case study was the American National Bank building in Austin, Texas designed by Kuehne, Brooks and Barr Architects with a mural by Seymour Fogel. The secondary case studies were: Harry Bertoia sculpture + Manufacturers Trust Building, New York City, Pietro Belluschi mural + Equitable Building, Portland, Oregon, Richard Lippold sculpture + Inland Steel Building, Chicago, and Roger Darricarrere dalle de verre + Columbia Savings Buildings, Los Angeles. After study and analysis, the preservation strategies were categorized in four categories: in situ conservation, removal, recreation/replacement and demolition/destruction. It was concluded that there is not a general approach for these projects, and each should be analyzed through various factors (Design Intent, Intrinsic Value, Collaboration and Context) to determine the appropriate intervention.<br>text
