El chocolate Trencito es una reconocida barra de chocolate comercializada en Chile por la empresa multinacional Nestlé. El producto se caracteriza por su sabor dulce y textura suave, y quienes lo consumen, identifican la mezcla de sentimientos relacionados a la alegría y a la nostalgia en cada ocasión de consumo. Estos atributos lo hacen un chocolate muy popular entre la población chilena. Nestlé cuenta con una diversificada cartera de productos en diferentes categorías, entre ellas, la categoría de “chocolates” y la de “helados”. Los productos elaborados y distribuidos por esta empresa han logrado un alto nivel de penetración entre sus consumidores, por lo que resulta de gran interés realizar un estudio que abarque las posibilidades de extender un producto tan reconocido como el chocolate Trencito de Nestlé, a otra categoría, como lo es la del helado. Es así como, con el propósito de analizar los aspectos relativos al lanzamiento de una extensión de marca del producto Chocolate Trencito de Nestlé, en esta primera parte de la investigación, se hace relevante compilar los antecedentes necesarios que conformen un análisis situacional, el cual logre reflexionar acerca de los aspectos esenciales en el mercado y su entorno, y en donde se evidencie el potencial de los productos en la categoría de “helados” y “chocolates”, que combinados podrían resultar en la creación de un nuevo producto. En otras palabras, la elaboración de un producto que reúna los aspectos más queridos de un chocolate, y logre plasmarlos en ese nuevo formato: El Helado Trencito

Resumo: Esta dissertação pretende apresentar as análises das obras de Oiticica cronologicamente. Serão apresentadas neste texto análises de obras inéditas como os desenhos de Oiticica para o filme da Cangaceira Eletrônica e o Projeto labirinto para Formigas. Também me concentrarei em eventos de Oiticica dos anos 70 que foram muito pouco estudados, e quase nada se pesquisou sobre eles. Farei uma análise histórica sobre o surgimento da Arte Concreta, da Escola de ULM, Primeira Bienal do Museu de Arte Moderna de São Paulo, a presença de Max Bill no Brasil. Relacionarei a obra de Oiticica com o contexto internacional. Pois, Oiticica estava inserido em um contexto das artes que rumavam para a desmaterialização, o fim das instituições museológicas e das artes tradicionais
Abstract: This dissertation intends to introduce the analysis of the Oiticica's works in a chronological order. It will be presented the analyses of the unpublished works, such as the Oiticica's draws for the film, "Cangaceira Eletrônica" and the "Projeto labirinto para Formigas". I will concentrate in the Oiticica's events of the 70's which were not weU studied - almost nothing was investigated about them. I will make a historical analysis about the rising of the "Arte Concreta" (Concrete Art) from the "Escola de ULM, Primeira Bienal do Museu de Arte Moderna de São Paulo", the presence of Max BiU in Brazil. I will relate the Oiticica's works with the international contexto Because Oiticica was inserted in a context of arts that go towards the desmaterialization, the end of the museum' s institutions and the traditional arts

Full textThe professional experience of Helio de Queiroz Duarte from Rio de Janeiro was rare in his time, he was one of the few architects of his generation to dedicate the same energy and intensity to both the academic career and the job on the drawing board. Appreciated by contemporary architects, but still little studied, Duarte left us as legacy a new paradigm of concept and understanding of the uses and potentiality of schools as public equipment. This heritage still bears fruit nowadays, as can be seen in the projects for the CEUs (Centers for Unified Education). His academic activity was just as impactful, after all, Duarte was one of the founders of the Faculty of Architecture and Urbanism at the University of São Paulo, one of the main creators of the first postgraduate program in the country for this same institucion, as well as for the introduction of the TGI (Final Graduation Interdisciplinary Work), required today by all colleges. The aim of this research is to understand how part of his prodution became a reference for Brazilian modern architecture, which distinguishes and references the production of mainly educational programs, both from the spatial and pedagogical point of view until today. This work intends to measure the influence of his academic activity in the conformation and systematization of the undergraduate and postgraduate education of architecture and urbanism in Brazil. At the same time, it seeks to verify how the complex of Helio Duarte\'s activities collaborated in the consolidation and recognition of the architect and urbanist profession in São Paulo.

Full textMicrotubules (MTs) are major constituents of the cytoskeleton and are involved in many cellular processes such as the determination of cell architecture, the formation of the mitotic spindle and intracellular trafficking. These tubular structures, composed by tubulin α/β heterodimers assemble and disassemble with a finely regulated dynamics.Stathmin is a cytosolic phosphoprotein that sequesters tubulin in a non polymerizable complex consisting of two tubulin heterodimers per stathmin molecule (T2S complex).The other stathmin family proteins (SCG10, SCLIP, RB3 and its two splice variants RB3 'and RB3 ") can also bind two tubulin heterodimers through their SLD (Stathmin Like Domain), but the different tubulin/SLD complexes display varying stabilities.Based on these observations, current works showed that one chimera protein consisting of the N-terminal domain of stathmin (NS) and of the C-terminal domain of RB3 (CR) forms a T2-SLD complex remarkably stable with tubulin (NS-CR chimera). Molecular simulation experiments in silico suggest that we can increase the affinity of the NS-CR chimera by introducing mutations in the NS subdomain (Jourdain et al., 2004).In parallel to this work, Clement and colleagues found that short peptides derived from the N-terminal region of SLD alone can inhibit tubulin polymerization. Among the N-terminal part of SLD, the most effective peptide is the peptide I19L (IQVKELEKRASGQAFELIL) derived from stathmin. It corresponds to region folded into a "β-hairpin" structure observed in the T2S crystals (Clement et al., 2005). To increase the efficiency of this interaction, different mutations were introduced at the level of the I19L peptide from the stathmin N-terminal domain and tested in vitro. The results showed that the peptides I19L-K4R and I19L-K4R-A10R are the most effective to in inhibit the assembly of MTs.In this work, we introduce these mutations in the N-terminal domains of stathmin and in the NS-CR chimera with the aim of analyzing their efficiencies on MTs disassembly in cancer cell cultures like HeLa, and the impact of the mutation on MTs dynamic and cell cycle.Results from overexpression experiments of WT and stathmin mutant 2 containing the double mutation (K9R-A15R similar to the mutations K4R-A10R for the I19L peptide), suggest that this mutant significantly induces depolymerization of mitotic MTs. In its less phosphorylated state on serine 16; this mutation also reduces MTs dynamics and induces cytotoxicity.We thus suggest that we can ameliorate the binding and efficiency of the N-terminal stathmin part by including this double mutation. This variation has an impact in vitro and in some cellular aspects such as cell proliferation and MTs dynamics. These findings may lead to a targeted therapy for this type of cancer

Full textSyftet med denna uppsats är att undersöka en funktionell svenskundervisning där undervisning om språkliga strukturer integreras i undervisningen kring eget skrivande och analys av egna och andras texter. Fokus ligger på undervisningens relation till kontextuella aspekter av verksamheten, såsom sammanhang som har att göra med praxis i klassrummet, den enskilda skolan och yttre ramar på ett samhälleligt plan. Centrala begrepp är dialogicitet, processer, stödstrukturer, undervisning rörande språkliga strukturer, integrering, erfarenhetspedagogisk ämnessyn och text som socialt beteende. Såväl min analysapparat som den studerade undervisningen har inspirerats av ett sociokulturellt perspektiv på lärande, särskilt nydialogiska och genrepedagogiska teorier.
Studien följer fyra undervisningsförlopp i grundskolans senare år och i gymnasieskolan genom observationer, samtal i fokusgrupper samt intervjuer med lärare och elever.
Resultatet visar att den genomförda undervisningen värderas högt av lärare och elever, och lärarna menar att alla kategorier av elever tjänar på denna typ av integrerad undervisning. Den upplevs som effektiv och engagerande. Vad gäller de centrala begreppen, som nämnts ovan, kan man konstatera att det verkar ha funnits en strävan att implementera dessa i den studerade undervisningen, vilket också har skett i olika grad. En ytlig iscensättning av dessa begrepp i praktiken innebär dock knappast en undervisning som kan anses gestalta forskares teorier kring dessa begrepp. Ett flertal kontextuella aspekter på olika nivåer har identifierats som betydelsefulla i sammanhanget, som exempelvis läromedlens roll; lärarnas kunskaper och syn på lärande, på svenskämnet och på texter; lärarnas behov av forum för diskussioner och reflektion kring svenskundervisningen.

Att leva i ovisshet och att inte veta hur ens framtid kommer att bli kan skapa funderingar som kan leda till olika reaktioner. Sådana funderingar och reaktioner blir ofta synliga i berättelser om det inträffade. En berättelse är en skildring av ett händelseförlopp muntligt eller skriftligt av verkliga eller påhittade händelser. Genom berättandet fångas helheten upp och återskapar upplevelser och känslor. Syfte: Syftet med studien var att få en ökad förståelse för fenomenet ovisshet. Metod: Studien genomfördes som en empirisk studie där studenters livsvärldsberättelser om fenomenet ovisshet utgjorde grunden för resultatet. Materialet har analyserats med kvalitativ innehållsanalys. Resultatet: Resultatet presenterades utefter tre frågor: 1 Vad innebär ovisshet? Tankar om framtiden och att inte ha makt att kunna påverka, 2 När uppkommer ovisshet? När det vardagliga bryts och 3 Vad gör ovisshet med människan? Känslor här och nu, tankar som svävar iväg. Diskussion: En ökad förståelse för vad ovisshet är underlättar för sjuksköterskan i hennes arbete att vårda och förstå den enskilde patientens livsvärld.

Nanopartículas magnéticas (NPMs) têm sido alvo de inúmeras investigações por seu grande potencial de aplicação nos mais diferentes campos tecnológicos. Dentre tantos, elas destacam-se na área biomédica, seja no diagnóstico ou tratamento de diversas doenças. Neste trabalho foram sintetizadas nanopartículas magnéticas (NPMs) de óxido de ferro (magnetita) pelo método de coprecipitação de íons Fe2+ e Fe3+ em meio alcalino. O objetivo central foi estudar as características morfológicas, estruturais, magnéticas e o comportamento biológico desses compostos em células cancerígenas, visando futuras aplicações biomédicas. Inicialmente as nanopartículas magnéticas foram avaliadas em função dos parâmetros físico-químicos que influenciam diretamente as características finais do produto (pH, molaridade, temperatura e tipo de base) para se observar as melhores condições de síntese e a influência de cada um nas características do produto. As nanopartículas foram caracterizadas por difratometria de raios-X, microscopia eletrônica de varredura, potencial zeta e magnetometria. Desse primeiro estudo concluiu-se que a coprecipitação produz partículas com polidispersão de tamanhos alta e que os parâmetros de síntese influenciam drasticamente as propriedades dos materiais, no entanto, todas as amostras exibiam características magnéticas. Depois de estabelecida esta etapa, as NPMs foram submetidas à transfecção em cultura celular de carcinoma cervical humano (células HeLa) e a testes biológicos como coloração com Azul da Prússia e hematoxilina-eosina, ensaio de MTT e ensaio de apoptose para averiguação da citotoxidade. A principal observação vinda desses resultados foi que as nanopartículas magnéticas sintetizadas, salvo algumas adaptações de síntese...
Magnetic nanoparticles (MNPs) have been the subject of numerous investigations because of its great potential application in many different fields of technology. Among many, they stand out in the biomedical area, either in diagnosis or treatment of various diseases. In this work were synthesized magnetic nanoparticles (MNPs) of iron oxide (magnetite) by the coprecipitation method of Fe2+ and Fe3+ in an alkaline medium. The main objective was to study the morphological, structural, magnetic and biological behavior of these compounds in cancer cells, in order to future biomedical applications. Initially the magnetic nanoparticles were evaluated against the physical and chemical parameters that directly influence the final characteristics of product (pH, molarity, temperature and type of base) to observe the best synthesis conditions and influence of each characteristics. The nanoparticles were characterized by X-ray diffraction, sccaning electron microscopy, zeta potential and magnetometry. In this first study showed that coprecipitation produces particles with high polydispersity of sizes and that the synthesis parameters dramatically influence the properties of materials, however all samples exhibited magnetic characteristics. After you make this step, the MNP were subjected to transfection in cell culture of human cervical carcinoma (HeLa) and biological tests such as staining with Prussian blue and hematoxylin-eosin, MTT assay and apoptosis assay to investigate cytotoxicity. The main observation was that these results coming magnetic nanoparticles synthesized, except for some adjustments in short, constitute a class of nanocomposites with enormous potential for therapeutic and / or diagnosis. The work brings together information ranging from the synthesis of nanoparticles to their behavior inside the cells, emphasizing the best conditions for each procedure

The species of the genus Marsdenia, Apocynaceae, are widely used in folk medicine of several countries. In Brazil is found several species belonging to this genus. The in vitro antioxidant, anticoagulant and antiproliferative activities were evaluated to aqueous extracts of stalk, leaf and root of Marsdenia megalantha. In the total antioxidant capacity assay (expressed as ascorbic acid equivalents) the stalk extract showed 76.0 mg/g, while leaf and root extracts 141.3 mg/g and 57.0 mg/g, respectively. The stalk and leaf extracts showed chelating activity around 40% at 1.5 mg/mL, while root extract, at the same concentration showed, 17%. Only the leaf extract showed a significant ability in superoxide scavenging (80% at 0.8 mg/mL). Any extract was able in scavenge hydroxyl, as well anticoagulant activity. The antiproliferative activity of the extracts was evaluated against HeLa tumor cell line. The extracts inhibited in a dose-dependent manner the cell growth. However, the leaf extract showed 80% of inhibition at 1.0 mg/mL, while stalk and root extracts inhibited 63% and 30%, respectively. To assess the mechanism of cell death caused by the leaf extract in HeLa, was performed flow cytometry and western blot. The results show that leaf extract induces cell death by apoptosis through an activation caspase-independent pathway. These data indicate that stalk and leaf extracts obtained have potential to be used as antioxidants and anticancer drugs
As esp?cies do g?nero Marsdenia, Apocynaceae, s?o bastante utilizadas na medicina popular de v?rios pa?ses. No Brasil s?o encontradas v?rias esp?cies pertencentes a esse g?nero. As atividades antioxidante, anticoagulante e antiproliferativa foram avaliadas para os extratos de caule, folha e raiz de Marsdenia megalantha. Na capacidade antioxidante total (expressa como equivalente de ?cido asc?rbico), o extrato do caule apresentou 76,0 mg/g, enquanto os extratos da folha e raiz apresentaram, respectivamente, 14,3 mg/g e 57,0 mg/g. Os extratos do caule e da folha mostraram habilidade quelante em torno de 40% na concentra??o de 1,5 mg/mL, enquanto o extrato da raiz, na mesma concentra??o, apresentou 17%. Apenas o extrato da folha apresentou uma capacidade significante em sequestrar radicais super?xidos (80% em 0,8 mg/mL). Nenhum extrato mostrou capacidade em seq?estrar radicais hidroxila, bem como atividade anticoagulante. A atividade antiproliferativa dos extratos foi avaliada contra a linhagem tumoral HeLa. Os extratos inibiram, de forma dose-dependente, o crescimento celular. Entretanto, o extrato da folha foi capaz de inibir em 80% a prolifera??o celular na concentra??o de 1,0 mg/mL, enquanto os extratos de caule e raiz inibiram 63% e 30%, respectivamente. Para avaliar o mecanismo de morte celular causada pelo extrato da folha nas c?lulas HeLa, foi realizada citometria de fluxo e western blot. Os resultados mostraram que o extrato da folha induz morte celular por apoptose atrav?s de uma via de ativa??o independente de caspase. Estes resultados indicam que os extratos de caule e folha obtidos t?m potencial para serem futuramente utilizados como f?rmacos antioxidantes e antic?ncer

O objetivo central foi estudar as características morfológicas, estruturais, magnéticas e o comportamento biológico desses compostos em células cancerígenas, visando futuras aplicações biomédicas. Inicialmente as nanopartículas magnéticas foram avaliadas em função dos parâmetros físico-químicos que influenciam diretamente as características finais do produto (pH, molaridade, temperatura e tipo de base) para se observar as melhores condições de síntese e a influência de cada um nas características do produto. As nanopartículas foram caracterizadas por difratometria de raios-X, microscopia eletrônica de varredura, potencial zeta e magnetometria. Desse primeiro estudo concluiu-se que a coprecipitação produz partículas com polidispersão de tamanhos alta e que os parâmetros de síntese influenciam drasticamente as propriedades dos materiais, no entanto, todas as amostras exibiam características magnéticas. Depois de estabelecida esta etapa, as NPMs foram submetidas à transfecção em cultura celular de carcinoma cervical humano (células HeLa) e a testes biológicos como coloração com Azul da Prússia e hematoxilina-eosina, ensaio de MTT e ensaio de apoptose para averiguação da citotoxidade. Abstract: Magnetic nanoparticles (MNPs) have been the subject of numerous investigations because of its great potential application in many different fields of technology. Among many, they stand out in the biomedical area, either in diagnosis or treatment of various diseases. In this work were synthesized magnetic nanoparticles (MNPs) of iron oxide (magnetite) by the coprecipitation method of Fe2+ and Fe3+ in an alkaline medium. The main objective was to study the morphological, structural, magnetic and biological behavior of these compounds in cancer cells, in order to future biomedical applications. Initially the magnetic nanoparticles were evaluated against the physical and chemical parameters that directly influence the final characteristics of product (pH, molarity, temperature and type of base) to observe the best synthesis conditions and influence of each characteristics. The nanoparticles were characterized by X-ray diffraction, sccaning electron microscopy, zeta potential and magnetometry. In this first study showed that coprecipitation produces particles with high polydispersity of sizes and that the synthesis parameters dramatically influence the properties of materials, however all samples exhibited magnetic characteristics. After you make this step, the MNP were subjected to transfection in cell culture of human cervical carcinoma (HeLa) and biological tests such as staining with Prussian blue and hematoxylin-eosin, MTT assay and apoptosis assay to investigate cytotoxicity. The main observation was that these results coming magnetic nanoparticles synthesized, except for some adjustments in short, constitute a class of nanocomposites with enormous potential for therapeutic and / or diagnosis. The work brings together information ranging from the synthesis of nanoparticles to their behavior inside the cells, emphasizing the best conditions for each procedure

Full textThe purpose of this master thesis is to investigate the residential area Holma, Sweden, its preconditions and how it is experienced by the people that reside and work there, and to analyze and discuss this from theoretical viewpoints regarding space, place, urbanity and neighborhood. Furthermore, the aim is to identify resources for the promotion of community and sustainable improvement. The work is written from a social constructionist perspective in which one premise is that people, through their actions form meaning and that this behaviour itself is influenced by social structures. The empirical work consists of ethnographic studies that are further analyzed through the previously mentioned theoretical basis. The study culminates in discussions about the importance of civic engagement and long-term perspective in the implementation of the initiatives. A major strong point is that people should have the opportunity to identify and have influence over their place, as thorough knowledge of a place is to be found through its people. The thesis shows how the social resources that operate in the field should be better used for development of a strong neighborhood and community.

Resumo: Na primeira parte da tese, os efeitos das contribuições interatômicas de três corpos na equação de estado do 4 He são investigadas. Um potencial recente de dois corpos junto com o potencial de três corpos de Cohen e Murrell são utilizados para descrever o sistema de átomos de hélio. A dispersão de triplo dipolo e as energias de troca são estimadas sujeitas só a incertezas estatísticas. Uma extensão ao método de difusão com Monte Carlo é empregado para calcular diferenças de energias muito pequenas. Os resultados mostram como as contribuições de três corpos afetam a energia do estado fundamental, e as densidades de equilíbrio, solidificação e liquefação. Na investigação dos sistemas de hélio, diversos potenciais ab initio de dois corpos foram propostos na literatura. Nesta segunda parte da tese realizamos uma análise cuidadosa de como estes potenciais descrevem as propriedades do sistemas de 4 He usando a extensão proposta ao método de difusão com Monte Carlo. Esta extensão nos permitiu calcular com grande precisão pequenas diferenças de energia associadas a diferentes potenciais como função da densidade do sistema. Com estes resultados foi possível acompanhar cuidadosamente como a descrição das propriedades do sistema mudam com os potenciais de dois corpos considerados. Da nossa análise identificamos o potencial que em conjunto com interações de três corpos foi capaz de fornecer a melhor descrição dos sistemas de hélio
Abstract: In the first part of the thesis, the effect of three-body interatomic contributions in the equation of state of 4 He are investigated. A recent two-body potential together with the Cohen and Murrell three-body potential are applied to describe the sistem of helium atoms. The triple dipole dispersion and exchange energies are evaluated subjected only to statistical uncertainties. An extension of the diffusion Monte Carlo method is applied in order to compute very small energies differences. The results show how the three-body contributions affects the ground-state energy, the equilibrium, melting and freezing densities. In the investigation of the helium systems several ab initio two-body potentials have been propoused in the literature. In the second part of the thesis, we performed a carefull analysis of how these potentials describe the properties of the 4 He systems by using the proposed extension to the diffusion Monte Carlo Method. This extension have allowed the calculation of small energies differences, due to the different potentials, as a function of the system density with great accuracy. With our results it is possible to carefully follow how properties of the sistem change with the two-body potentials considered. From our analises we identify the potential that together with three-body interactions is able to give the best description of the helium systems

Resumo: Bacteriófagos são vírus que infectam bactérias e arqueias, representando as entidades biológicas mais abundantes do mundo e influenciando de maneira marcante populações naturais de seus hospedeiros. A terapia com bacteriófagos, que representa uma das primeiras formas modernas de combate a infecções bacterianas, foi recentemente redescoberta e vem sendo reavaliada seguindo metodologias atuais quanto à sua viabilidade terapêutica. Para completar a caracterização dos fagos de nossa coleção, sequenciamos completamente o genoma da maior parte destes, através da metodologia multiplex pair-ended utilizando a plataforma Illumina. Visando contribuir para verificação da viabilidade terapêutica de bacteriófagos testamos os efeitos protetor e terapêutico dos fagos Shfl1, Saen1v2 e Saen1v4, pertencentes à coleção de nosso laboratório, em modelos biológicos relevantes. O fago Shfl1, lítico contra Shigella flexneri, foi testado em ensaio de invasão em células HeLa. A redução de bactérias intracelulares foi mensurada independentemente através de plaqueamento e citometria de fluxo, além da observação direta por microscopia de fluorescência. O fago Saen1v2, lítico contra Salmonella Typhimurium, foi estudado quanto à biodistribuição e meia-vida em modelo murino, e uma variante viral com maior persistência in vivo foi selecionada. Essa variante, denominada Saen1v2p5, e o fago Saen1v4, lítico contra Salmonella Typhi, foram testados em modelo murino de infecção tifoide, contra seus respectivos hospedeiros. Encontramos similaridade genômica a fagos conhecidos, como T4, T7, T1 entre outros, em maior ou menor grau. Obtivemos um efeito protetor e terapêutico contra Shigella flexneri utilizando o fago Shfl1 em ensaio de invasão em cultura de células HeLa, verificado por todas as metodologias empregadas. Não verificamos efeito antimicrobiano in vivo do fago Saen1v2p5 em modelo murino de infecção por Salmonella Typhimurium. Por outro lado, observamos efeito terapêutico e protetor dose dependente utilizando o fago Saen1v4 em modelo murino de infecção por Salmonella Typhi. O sucesso obtido com baixas multiplicidades de infecção sugere um possível efeito indireto ou estimulação imune inespecífica
Abstract: Bacteriophages are viruses that infect Bacteria and Achaea, representing the most abundant biological entities in the world and markedly influencing natural host populations. Phage therapy, which represents one of the first modern ways to fight bacterial infections, was recently rediscovered and is being re-evaluated according to current methodologies regarding its therapeutic viability. In order to complete phage characterization in our collection, we sequenced completely the genomes of most of these, through the multiplex pair-ended methodology using the Illumina platform. Aiming to contribute to the therapeutic viability verification of bacteriophages we tested phage protective and therapeutic effects of Shfl1, Saen1v2 and Saen1v4, which belong to our collection, in biologically relevant models. Phage Shfl1, lytic against Shigella flexneri, was tested in a HeLa invasion assay. Intracellular bacteria reduction was measured independently through plating and flow cytometry, besides direct observation through fluorescent microscopy. Phage Saen1v2, lytic against Salmonella Typhimurium, was studied about its bio-distribution and half-life in murine model, and a viral variant with longer in vivo persistence was selected. This variant, denominated Saen1v2p5, and phage Saen1v4, lytic against Salmonella Typhi, were tested in murine typhoid model, against their respective hosts. Genomic similarity to known phages such as T4, T7, T1 among others, was found, in various degrees. We obtained both protective and therapeutic effect against Shigella flexneri using phage Shfl1 in the HeLa invasion assay, through all methodologies utilized. We could not verify in vivo antimicrobial effect of phage Saen1v2p5 in the murine model of Salmonella Typhimurium infection. On the other hand, we observed both therapeutic and protective dose dependent effect using phage Saen1v4 in Salmonella Typhi murine infection model. The success obtained with low multiplicities of infection may suggest a possible indirect effect or unspecific immune stimulation

Full textDesenvolve uma tese sobre pontos em comum entre o pensamento de Martin Heidegger e a invenção dos trabalhos de Helio Oiticica. Esses nexos se estabelecem no tocante à questão do espaço e se dão, especialmente, nas propostas de Helio Oiticica intituladas Penetráveis. Na execução desses trabalhos, há uma confluência de pensamentos e de questões longamente analisadas e desenvolvidas em textos de Martin Heidegger, a saber, o lugar do homem no mundo, a experiência como caminho para a descoberta ou o desencobrimento do mundo tal como ele se dá, bem como o caminhar como experiência que propicia essa descoberta. Entende-se a ação obstinada de Helio Oiticica em construir lugares para criação do espaço, criação da experiência, de possibilitar um habitar poético, como uma tentativa de despertar o homem de uma espécie de sono cotidiano e colocá-lo no acontecimento das coisas, no mundo. Desenvolve-se este trabalho em três capítulos, nos quais se trata cronologicamente a produção de Helio Oiticica, a partir de sua experiência com o Grupo Neoconcreto, até o momento em que realiza os Penetráveis e os Projetos Ambientais que contam com a participação deles. Atendo-se mais demoradamente a essa série de trabalhos, tanto aos Penetráveis construídos em escala humana quanto às maquetes, aponta-se especialmente para a busca do espaço no trabalho do artista, mostrando como sua obra foi libertando-se, ganhando asas e encaminhando-se no e para o espaço. Com a construção desses lugares de experimentação, o artista cria espaços de possibilidade de reencontro do homem com o seu próprio, trazendo para sua obra uma prática de pensamento que se dá no caminhar, no estar, no viver e no experimentar fenomenológico.

Full textEste projeto tem como objetivo a formulação de um plano de marketing para 2020 visando o aumento da notoriedade do estabelecimento Hello Bubble Waffle por meio de aplicações de social media. Hello Bubble Waffle é um estabelecimento de sobremesas localizado em Lisboa, que se dedica exclusivamente a produzir uma sobremesa típica de Hong Kong - egg waffle no local e a vendê-la diretamente a consumidor final. O presente projeto é desenvolvida na área de planeamento de marketing em torno de 3 temas: planeamento de marketing, marketing de social media e word-of-mouth e notoriedade de marca. Com a utilização da combinação de métodos quantitativo e qualitativo, realizaram-se entrevistas semi-estruturadas face-a-face e um questionário online, visando recolher informações para fundamentar o plano de marketing. Com base na pesquisa realizada, foi obtida informação sobre: 1) a notoriedade atual e a notoriedade desejada da HBW; 2) as características do público alvo da HBW; 3) as aplicações de social media eficazes para lançar atividades de marketing; 4) os formatos de mensagens publicadas em social media que capturam a atenção; e 5) o efeito de word-of-mouth para a notoriedade de marca. Com todos os dados primários e secundários, formularam-se no final as estratégias de marketing mix de 4P (produto, preço, distribuição e comunicação), com ênfase na comunicação, a fim de aumentar a notoriedade de marca do estabelecimento Hello Bubble Waffle com recurso às aplicações de social media.
This project is aimed at formulating a marketing plan for 2020 to increase awareness through social media applications for a dessert shop in Lisbon - Hello Bubble Waffle (HBW), which focuses its operation totally on producing a typical dessert from Hong Kong - egg waffle on site and selling directly to final customers. The present project is developed in the marketing planning area around three topics: (1) marketing planning, (2) social media and word-of-mouth marketing, and (3) brand awareness. Both qualitative and quantitative approaches were used through semi-structured face-to-face interviews and online questionnaire, in order to collect information to better inform the marketing plan. Based on the research carried out, it was mainly collected information about 1) actual brand awareness and intended awareness of HBW; 2) characteristics of HBW's target public; 3) effective social media applications to launch marketing activities; 4) the formats of messages posted on social media which catch attention; 5) the effect of word-of-mouth to increase brand awareness. Based on both primary and secondary data, the strategies of 4P's marketing mix (product, price, place and promotion) were formulated, with emphasis on promotion, in order to increase brand awareness of Hello Bubble Waffle through social media applications.

Full textProcesses that involve sensitization are extremely important for several areas of knowledge, including biology, chemistry and medicine. The application of sensitization in medicine is especially important, in face of an alternative modality of cancer treatment called Photodynamic Therapy (PDT). One of the lines of basic research important for the evolution of PDT is the development of new photosensitizers (PS) with defined composition, that absorb in the therapeutical window (600-800nm) and that present greater efficiency in the induction of apoptosis. Positively charged PS with the proper lipophilic/hidrophilic balance permeate membranes and are attracted by the negative potential of mitochondria, which is an organelle that has central roles in the control of cell life and death. In this work the influence of the functional groups in the activity of PS was carried out, through the functionalization of protoporphyrin IX (Pp IX). Three different synthetic routes were used. In route I (scheme 14), Hematoporphyrin IX (Hp IX), was used as departure compound and it was modified with amino and amide groups in hydroxyls and carboxyls, respectively. We found low yield in the synthetic (20%) and purification steps. However 1 pure PS was obtained. In route II (scheme 15), Pp IX was used as departure compound, which was modified in the vinilic groups with amino groups. We obtained 4 compounds, with global yield superior than 50%, but it was observed that an elimination reaction occurs that hinders the quarternization of the amino groups located in position α to the porphyrin ring. Into route III (scheme 16), the carboxilic groups of Pp IX had been transformed into chloride acid, which was substituted by bi- functional groups, primary amine in one side and another group in the other. This route made it possible to obtain 7 compounds, including quaternary ammonium compounds with global yield superior than 70%. Two derivatives of chlorophyllin, that present QIV band with large extinction coefficient in 650nm, also were synthesized. All compounds were characterized structurally through the electronic and vibrational spectra (UV-vis and fluorescence, IR), RMN of 1H and 13C (1D and 2D) and mass spectrometry. This series of compounds allowed us to study the relation between the chemical structure of the Fs with its photoactivity. The photophysical properties were characterized by emission and absorption spectra, laser flash photolysis, fluorescence emission in the visible and NIR regions (generation of singlet oxygen). These determinations indicated that the photophysical properties of the PS were not modified in the synthetic process. The formation of aggregates were characterized in aqueous solution and the balance between aggregate and monomer species was dislocated in the direction of the formation of monomers in the presence of CTAB, SDS micelles and fetal serum. It was observed, that the disaggregating efficient is larger with micelles that have the opposite charges to that of the PS. The Octanol water partition coefficient (logPo/a), was determined as a function of pH. logPo/a values between -1 and 1 were observed for charged PS in the pH range of 3-10. The incorporation of these compounds in liposomes, followed the expected profile considering the charge, logPo/a and the amphifilic nature. Generically, the photoinduced cell death efficiency followed the profile of incorporation of the PS in HeLa cells. Comparative studies of cytolocalization were carried out by co-incubation of the cells with porphyrins and Rhodamine 123 (Rd), which localizes in mitochondria. The localization in cytoplasmic organelles was determined through fluorescence confocal microscopy images. The overlapping emission of the Rd was of 80% and 31% with the cationic PpNpNI and the anionic PpNetPO3 compounds, respectively, proving the mitochondrial localization of the positive PS.

Full textEl retículo endoplásmico (RE) y la mitocondria son organelos celulares que entablan una continua y dinámica conversación que permite a la célula responder coordinadamente a diversas situaciones fisiopatológicas. Datos previos de nuestro Laboratorio mostraron que ambos organelos incrementan los puntos de contacto físico durante la fase temprana del estrés de RE, permitiendo una mayor transferencia de Ca2+ desde el RE hacia la mitocondria y estimulando así el metabolismo de este último organelo y favoreciendo la sobrevida celular. El presente proyecto estudió la participación de proteína quinasa A (PKA) y caveolina-1 (Cav1) como agentes reguladores de esta interacción física y funcional entre el RE y la mitocondria. Como modelo de estudio se utilizaron células HeLa tratadas con tunicamicina como agente inductor de estrés de RE por 4 h. Primero, se investigó la participación de PKA en la fase temprana del estrés de RE. Esta quinasa se activó ante condiciones de estrés, medido por la fosforilación de DRP1 en Ser637 por Western blot y concomitante elongación mitocondrial (microscopía confocal). El inhibidor específico de PKA H89 previno estos cambios, comprobando que ambos son dependientes de PKA. Posteriormente, se estudió la participación de PKA en el acoplamiento RE-mitocondria a través de la medición de proximidad (microscopía confocal), contactos físicos (microscopía electrónica), transferencia de Ca2+ (microscopía de fluorescencia) y bioenergética mitocondrial (tasa de consumo de oxígeno). El papel de PKA en estos procesos se evaluó por modulación farmacológica con H89. Los resultados mostraron que la activación de PKA es necesaria para inducir la formación de contactos RE-mitocondria, y de esta forma, gatillar la respuesta adaptativa frente a estrés de RE. Luego se investigó el papel de Cav1 sobre la interface RE-mitocondria. La expresión de Cav1 abolió completamente la respuesta adaptativa frente a estrés de RE temprano, evidenciado mediante proximidad entre organelos, transferencia de Ca2+ y respiración mitocondrial. Además de alterar esta plasticidad organelar frente a condiciones de estrés, la expresión de Cav1 disminuyó significativamente la bioenergética mitocondrial basal, junto con inducir un remodelado basal de la interface RE-mitocondria medido por purificación de las membranas del RE asociadas a mitocondria (MAM). Finalmente, se determinó que la expresión de Cav1 antagonizó la señalización de PKA, impidiendo que ella fosforile a DRP1 y promueva la elongación mitocondrial en respuesta a estrés de RE. Más aún, mediante fraccionamiento subcelular, se estableció que Cav1 afectó la localización subcelular de PKA, evitando que se relocalice a las membranas microsomales en respuesta al estrés de RE. De este modo, Cav1 actúa como un regulador negativo de PKA, previniendo su activación frente a estrés de RE, evitando así la respuesta adaptativa celular
The endoplasmic reticulum (ER) and mitochondria are two organelles that continuously communicate between one another. This organelle crosstalk allows mitochondria and ER to coordinate responses to a variety of physiological and pathophysiological situations. Data from a previous work show that during the early phase of ER stress, both organelles increase their contact sites, augmenting thereby calcium transfer from ER to mitochondria. This response boosts mitochondrial bioenergetics and ultimately promotes cell survival. Here, we sought to study the role of Protein Kinase A (PKA) and Caveolin-1 (Cav1) as regulators of such organelle crosstalk. As an experimental model, HeLa cells were treated with tunicamycin to induce ER stress. First, we observed that PKA was activated during early stages of ER stress, as assessed by increased phosphorylation of DRP1 at the inhibitory site Ser637 (Western blot), and that this event was followed by mitochondrial elongation (confocal microscopy). The specific PKA inhibitor H89 prevented these changes, thus confirming that they were due to PKA and not another kinase. Subsequently, we studied ER-mitochondria coupling by evaluating organelle proximity (confocal microscopy), physical contact sites (electron microscopy), Ca2+ transfer (fluorescence microscopy) and mitochondrial bioenergetics (oxygen consumption rate). PKA was again involved in these processes by pharmacological modulation using the inhibitor H89. Our results show that PKA activation is necessary to induce the formation of ERmitochondria contacts and to trigger the adaptive metabolic responses to ER stress. Next, we investigated the role of Cav1 as a modulator of the ER-mitochondria interface. Cav1 overexpression completely abolished the early adaptive response to ER stress, as assessed by evaluating the proximity between organelles, calcium transfer and mitochondrial respiration. In addition to altering organelle plasticity in response to stress conditions, Cav1 overexpression severely decreased baseline mitochondrial bioenergetics and induced basal remodelling of the ER-mitochondria interface, as determined by purification and analysis of mitochondria-associated ER membranes. Finally, we determined that Cav1 overexpression antagonizes PKA signalling, preventing PKA-dependent DRP1 phosphorylation and mitochondrial elongation in response to ER stress. Moreover, subcellular fractionation revealed that Cav1 affected PKA localization, prevented redistribution to microsomal membranes in response to ER stress, and altered the pattern of DRP1 phosphorylation. Thus, Cav1 functions as a negative regulator of PKA that precludes PKA activation during early ER stress and thereby prevents the adaptive cellular response

Full textThis study aims to examine how teachers by educating in accordance to the kiwimethod can develop students' reading skills. Literature related to how children learn to read in general, and how students develop reading skills by the kiwimethod are presented. Empirical data consist of qualitative interviews with two teachers, who have extensive experience of the kiwimethod. Additional observations have also been implemented. The results have been presented in various themes. The investigation revealed that its effective to teach reading strategies, by viewing and thinking aloud, in other words to “model” what good readers do, when they create understanding of a text. A successful way of teaching readingstrategies also seems to be, by discussing and dissecting texts and images together. It was also found that students, who were taught, in accordance to the kiwimethod become good at writing, got a well-developed language, and experienced reading in a positive way. Working with guided reading however requires, that students receive training in working accordance to the method, which means that students need to learn strategies, that will help them to work independently.
