Abstract:

Pusat Peragaan Ilmu Pengetahuan dan Teknologi (PP-IPTEK) adalah science center atau wahana pembudayaan Iptek non formal untuk masyarakat umum khususnya peserta didik melalui media alat peraga sentuh-mainkan (hands-on interactive) dan pendidikan sains (educational program). PP-IPTEK berlokasi di Taman Mini Indonesia Indah (TMII), status kelembagaannya adalah satuan kerja yang berada dilingkungan Kementerian Riset, Teknologi dan Pendidikan Tinggi serta manajemen keuangannya menerapkan pengelolaan keuangan yang fleksibel dengan menonjolkan produktivitas, efisiensi dan efektivitas atau dikenal dengan istilah Badan Layanan Umum. Upaya PP-IPTEK dalam membudayakan Iptek belum optimal karena dihadapkan pada kondisi keterbatasan kapasitas serta beban kerja yang belum ditunjang oleh struktur organisasi yang ada saat ini. PP-IPTEK sebagai enterprise the goverment perlu melakukan upaya pembaharuan penataan organisasi dengan memasukkan unsur pengembangan layanan untuk meningkatkan koordinasi dan kemitraan strategis agar PP-IPTEK senantiasa dapat mengakomodasi perkembangan Iptek, kebutuhan masyarakat serta pembudayaan Iptek yang merata keseluruh wilayah di Indonesia. Penataan organisasi menggunakan analisa konfigurasi organisasi dari teori HenryMintzberg. Hasil analisis bahwa struktur organisasi model machine bureaucrazi dari Henry Mintzberg dapat diimplementasikan untuk penataan organisasi PP-IPTEK yang baru, mengingat model tersebut dianggap cocok dan relevan dengan kultur birokrasi di Indonesia serta dapat mengakomodasi peningkatan pengembangan layanan, penguatan kerjasama dan kemitraan strategis dengan seluruh pemangku kepentingan (stakeholders) PP-IPTEK.

Abstract:

Comparison of isolation methods for microbial pathogens is complicated by the variable interference caused by the competitive microflora present in test samples such as foods. In principle, using measured amounts of a standard competitor in a defined surrogate food matrix might control the effect of variable interference. This possibility was investigated using Listeria monocytogenes and enrichment broths belonging to the acriflavine-nalidixate selective agent class. Triplicate test sample sets were prepared. Each set consisted of suspensions of variable levels of the standard competitor, Enterococcus faecium strain 111 (≈10 to 109 CFU/25 g), mixed with a low constant level (10 to 100 CFU/25 g) of L. monocytogenes. These test samples were enriched at 30°C for 48 h in different selective media and streaked onto selective isolation agars. The input CFU ratio (E. faecium/L. monocytogenes) that permitted a 50% end point L. monocytogenes recovery was 2.2 × 106 or higher for the Food and Drug Administration one-step enrichments and 0.8 × 106 for the International Standards Organization (ISO) two-step enrichment. These and other results show that this evaluation method is feasible with this class of enrichments. Interestingly, L. monocytogenes could be detected in enrichment cultures at high-input E. faecium/L. monocytogenes ratios even when the enriched samples were plated onto nonselective media. The pinpoint colonies of L. monocytogenes embedded in a confluent lawn of E. faecium 111 were detectable by their contrasting coloration in Henry obliquely transmitted illumination.

Abstract:

Castanea henryi is an important woody grain tree species native to China. The objective of the current study was to find the suitable plant growth regulators (PGRs) and the optimal concentrations for direct organogenesis by using axillary shoots and cotyledonary nodes. Seeds were collected from the field, sterilized, and germinated in vitro. Axillary shoots and cotyledonary nodes of 3-week-old seedlings were used as explants. To find the suitable PGR for adventitious shoot induction, 0.5 mg·L–1 6-benzylaminopurine (6-BA), 0.1 mg·L–1 indole-3-acetic acid (IAA), 0.1 mg·L–1 2,4-dichlorophenoxyacetic acid (2,4-D), or 0.1 mg·L–1 1-naphthaleneacetic acid (NAA) was supplemented to Murashige and Skoog (MS) medium containing 0.65% agar and 3% sucrose. A high induction percentage of adventitious shoots (85.67%) was obtained from cotyledonary nodes supplemented with 0.1 mg·L–1 2,4-D. The type of explant influenced shoot proliferation rates and quality. Apical explants produced more and longer shoots than nodal segments. For shoot multiplication, 1 mg·L–1 6-BA + 0.05 mg·L–1 indole-3-butyric acid (IBA) supplemented with MS medium produced 12.33 and 6.25 shoots per explant, respectively, from apical and nodal explants. For shoot elongation and strengthening, 2 mg·L–1 6-BA + 0.05 mg·L–1 IBA supplemented with MS medium was the best combination, producing shoots with a mean length of 3.50 cm, a diameter of 0.46 cm, and about eight leaves per shoot. The greatest rooting of 76.70% and 11.33 roots per shoot was achieved when cultured in MS medium supplemented with 3.5% perlite + 1.5 mg·L–1 IBA. For acclimatization of the rooted plantlets in the greenhouse, a survival rate of 80% was achieved. This protocol—from multiplication to acclimation—is helpful to realize mass propagation of high-quality trees of chinquapin for increasing production and nut quality.

Abstract:

Penelitian ini bertujuan untuk mengevaluasi pelaksanaan (implementasi) Kurikulum 2013 di SMK Negeri 1 Lintau Buo dengan menggunakan Evaluasi Logic Model yang mencakup 4 aspek yaitu; evaluasi input, evaluasi aktivitas, evaluasi output dan evaluasi outcomes. Pendekatan penelitian yang digunakan adalah pendekatan gabungan/mixed dengan metode sequential explanatory (urutan pembuktian), penelitian ini dilakukan dengan melakukan penelitian kuantitatif terlebih dahulu, kemudian melanjutkan penelitian dengan metode kualitatif untuk menemukan kesimpulan. Teknik pengambilan sampel menggunakan Tabel Nomogram Henry King, sehingga jumlah guru yang dilibatkan dalam penelitian ini adalah 78 orang. Pada penelitian ini, peneliti menemukan bahwa implementasi Kurikulum 2013 di SMK Negeri 1 Lintau Buo masih tergolong cukup dengan rata-rata skor adalah 3,84 dan TCR adalah 75,52%. Hasil evaluasi implementasi Kurikulum 2013 pada aspek input adalah baik dengan rata-rata skor adalah 3,96 dan TCR adalah 80,28%, pada aspek aktivitas adalah cukup dengan rata-rata skor adalah 3,70 dan TCR adalah 74,06%, pada aspek output implementasi Kurikulum 2013 adalah cukup dengan rata-rata TCR adalah 3.96 dan rata-rata indikator adalah 73,62%, selanjutnya pada aspek outcomes adalah cukup dengan rata-rata skor adalah 3,73 dan TCR adalah 74,13 %. Implikasi penelitian ini adalah adanya perlunya peningkatan dan perbaikan pada aspek aktivitas, input, dan outcomes, meski komponen evaluasi aktivitas telah menunjukkan pelaksanaan Kurikulum 2013 yang baik. Penulis merekomendasikan kepada kepala sekolah dan pengawas agar secara berkesinambungan mengawasi dan mendampingi pelaksanaan implementasi Kurikulum 2013 oleh guru, guru juga harus mampu meningkatkan kemampuan dan wawasan serta menciptakan hubungan yang harmonis dan saling mendukung demi terlaksananya Kurikulum 2013 di SMK Negeri 1 Lintau Buo

Abstract:

Extensive blighting associated with root rot was seen in a 0.5-ha field of flowering corn (Zea mays inbred line ‘Tx714B’) at the Texas Agricultural Experiment Station in Weslaco, TX (Hidalgo County) in October of 2006. All plants were affected and most of the leaves of each plant were dead. Plants subsequently failed to produce any seed. Primary and secondary roots of blighted plants had extensive, dark brown necrotic lesions. Symptomatic root samples from 11 plants were surface disinfested with 10% bleach for 1 min, followed by two rinses with sterile distilled water, and plated onto water agar plus streptomycin. Fungal growth was transferred to one-fifth-strength potato dextrose agar plus streptomycin (PDAS). A dematiaceous hyphomycete was isolated from 45% of plated root segments from eight plants. The isolates had characteristics consistent with Exserohilum pedicellatum (Henry) Leonard & Suggs: straight, elongated conidia 57 to 75 μm long × 23 to 25 μm wide, with 4 to 6 pseudosepta, a protuberant hilum, and a prominent pedicel-like extension at the base (2). Four isolates were evaluated for pathogenicity. Conidia were scraped from PDAS plates, suspended in 100 ml of water, and mixed with 2.9 kg of autoclaved 1:1 (v/v) sand/sandy loam soil to obtain a density of 150 to 475 conidia per g of soil. Soil was placed in 20- × 4-cm-diameter plastic Cone-Tainer cells (Stuewe & Sons, Corvallis, OR) and a corn seed (‘Tx714B’) was planted 1.5 cm deep in each. Seeds were previously disinfested 2 min with 10% commercial NaOCl. Plants were kept in a growth chamber at 26°C for 4 weeks. There were six replicates per isolate and the experiment was repeated once. All four isolates caused a dark brown root necrosis covering 16 to 29% of the nodal roots and 43 to 83% of the seminal roots. Necrosis did not occur on roots from noninfested control containers. The pathogen was consistently reisolated from symptomatic roots, fulfilling Koch's postulates. This fungus is also pathogenic to sorghum (1), which is grown in rotation with corn in this field, increasing the chances that the fungus will persist.
