This thesis is an investigation into effect of elevated levels of hepatocyte growth factor (HGF) on immune responses. High levels of HGF are found in cancer such as in MPM patient serum and pleural fluid. However, little is known about the consequence of high HGF levels on the development and function of dendritic cells and contribution of these effects to tumour immune evasion. A pre-treatment in vitro model system was applied to study the effects of high HGF concentrations on the development of dendritic cells (DC) from monocytes. Glycosaminoglycans (GAGs) such as heparin, heparan sulfate (HS), chondroitin sulfate (CS) and dermatan sulfate (DS) are sulfated polysaccharides that exist on animal cell surfaces and in the extracellular matrix. Some of these fragments were then engineered based on structural information in order to produce an antagonist of HGF/SF-Met signalling. An effective way to treat liver disease is often liver surgery, such as liver resection for cancers and liver transplant for failure. Bone formation and remodeling require continuous generation of osteoprogenitors from bone marrow stromal cells (MSC), which are regulated by local growth factors and hormones with putative roles in mesenchymal proliferation and differentiation. Hepatocyte growth factor (HGF) and its receptor c-Met are widely expressed in MSC and are thought to play a key role in the interactions between cells. 1,25-dihydroxyvitamin D (1,25OHD) is the most active metabolite of vitamin D. 1,25OHD binds to its nuclear/membrane vitamin D receptor (VDR) and generates appropriate biological responses. Many of the anabolic effects of growth hormone (GH) are indirect, occurring through GH-stimulated production of insulin-like growth factor-I (IGF-I) by the liver. As well as being GH regulated, plasma IGF-I concentrations have been demonstrated to be dependent upon protein nutrition, with low protein diets being associated with reduced plasma IGF-I concentrations. This effect cannot be reversed by GH, suggesting that liver sensitivity to GH is impaired. To investigate the mechanisms through which protein supply affects GH sensitivity, primary cultures of ovine hepatocytes were grown in defined

Membrane rafts are small plasma membrane domains that contain high levels of cholesterol and sphingolipids. They have been implicated in processes as diverse as signal transduction, endocytosis and cholesterol trafficking. Traditional methods for the biochemical preparation of lipid rafts involve the extraction of membranes with nonionic detergents followed by the separation of a low-density, detergent-resistant membrane fraction on density gradients. Abstract:
The overall objective of this research was to understand the mechanims by which growth hormone (GH) regulates insulin-like growth factor-I (IGF-I) gene expression in liver. Previous studies have suggested that GH regulation of IGF-I gene expression in liver is mediated by binding of the transcription factor signal transducer and activator of transcription (STAT) 5 to four binding sites located distantly from the IGF-I promoter. The first specific objective of this research was to determine whether additional STAT5 binding sites were involved in GH stimulation of IGF-I gene expression in liver.
