Abstract:
ABSTRACT Culture is considered the definitive technique for Johne's disease diagnosis, and it is essential for later applications of certain molecular typing techniques. In this study, we have tested four solid media (Herrold's egg yolk medium [HEYM] with sodium pyruvate and mycobactin [HEYMm-SP], HEYM with mycobactin and without sodium pyruvate [HEYMm], Middlebrook 7H11 with mycobactin [Mm], and L�wenstein-Jensen with mycobactin [LJm]) for isolation of Mycobacterium avium subsp. paratuberculosis strains in 319 tissue samples from cattle herds and goat flocks. We have shown that each of the two main groups of M. avium subsp. paratuberculosis (type II and type I/III) has different requirements for growth in the culture media studied. The recommended solid media for isolation of type I/III strains are LJm and Mm, since the combination of both media allowed the recovery of all these strains. The most widespread culture medium, HEYM, is not suitable for the isolation of this group of M. avium subsp. paratuberculosis strains. Regarding the type II strains, HEYMm-SP was the medium where more strains were isolated, but the other three media are also needed in order to recover all type II strains. The incubation period is also related to the strain type. In conclusion, because the type of strain cannot be known in advance of culture, coupled with the fact that cattle and goats can be infected with both groups of strains, we recommend the use of the four solid media and the prolongation of the incubation period to more than 6 months to detect paratuberculous herds/flocks and to determine the true prevalence of the infection.

Abstract:
ABSTRACT The Hey basic helix-loop-helix transcription factors are downstream effectors of Notch signaling in the cardiovascular system. Mice lacking Hey2 develop cardiac hypertrophy, often associated with congenital heart defects, whereas combined Hey1/Hey2 deficiency leads to severe vascular defects and embryonic lethality around embryonic day E9.5. The molecular basis of these disorders is poorly understood, however, since target genes of Hey transcription factors in the affected tissues remain elusive. To identify genes regulated by Hey factors we have generated a conditional Hey1 knockout mouse. This strain was used to generate paired Hey2- and Hey1/2-deficient embryonic stem cell lines. Comparison of these cell lines by microarray analysis identified GATA4 and GATA6 as differentially expressed genes. Loss of Hey1/2 leads to elevated GATA4/6 and ANF mRNA levels in embryoid bodies, while forced expression of Hey factors strongly represses expression of the GATA4 and GATA6 promoter in various cell lines. In addition, the promoter activity of the GATA4/6 target gene ANF was inhibited by Hey1, Hey2, and HeyL. Protein interaction and mutation analyses suggest that repression is due to direct binding of Hey proteins to GATA4 and GATA6, blocking their transcriptional activity. In Hey2-deficient fetal hearts we observed elevated mRNA levels of ANF and CARP. Expression of ANF and Hey2 is normally restricted to the trabecular and compact myocardial layer, respectively. Intriguingly, loss of Hey2 leads to ectopic ANF expression in the compact layer, suggesting a direct role for Hey2 in limiting ANF expression in this cardiac compartment.

Paratuberculosis is an important enteritis of ruminants caused by Mycobacterium avium subsp. paratuberculosis (Map). The disease is officially considered exotic in Brazil, but recent serological surveys and the isolation of the agent suggest it may occur in our herds. The aim of this study was to evaluate three different formulations of Herrold's egg yolk agar with mycobactin J (HEYM) and four faecal culture protocols considering their ability for Map growth as well as cost and ease of application. Three formulations of HEYM were inoculated with two suspensions of Map. Spiked faeces and naturally contaminated faecal samples were treated by the four faecal culture protocols. Centrifugation protocol and HEYM recommended by OIE showed the best results on the recovery of Map.

Abstract:
La paratuberculosis es una enfermedad crónica producida por un bacilo ácidoalcohol resistente (BAAR), el Mycobacterium avium subsp. paratuberculosis (Map). No existe tratamiento ni vacuna aprobada por SENASA para esta enfermedad, por lo que es imprescindible el diagnóstico de los animales positivos para controlar la enfermedad antes de que infecten a otros. Se probó la implementación de un medio de cultivo líquido para el diagnóstico de paratuberculosis en bovinos, modificado a partir del medio Middlebrook M7H9 caldo. Para probar la efectividad del medio se realizó la siembra pareada de muestras de materia fecal y tejidos animales en el medio de cultivo sólido de Herrold con micobactina (HEYM) y en el medio de cultivo líquido modificado a partir del M7H9 caldo. Además, se lograron comparar el tiempo de detección de colonias en el medio sólido con el de detección de BAAR formando grupos en el medio de cultivo líquido en estudio. Se corroboró que hay diferencias significativas entre el medio de cultivo líquido y el medio HEYM, tanto en el tiempo de detección como en la cantidad de muestras positivas detectadas, observando BAAR en el medio líquido o colonias bacterianas en el medio HEYM. Sobre la base de estos resultados se concluye que el medio líquido probado posee una mayor sensibilidad analítica y un tiempo de detección del crecimiento menor con respecto a los del medio HEYM.

Le nom de Stefan Heym est associé à la controverse politique depuis son enfance. Jeune juif, il doit quitter sa ville natale pour la Tchécoslovaquie, puis les Etats-Unis pour échapper aux Nazis. Officier de l'armée américaine, il participe au débarquement en Normandie. L'arrivée au pouvoir de Mac-Carthy, l'oblige à fuir à nouveau vers l'Allemagne de l'Est, où il jouera à plein son rôle d'intellectuel, toujours fidèle à ses convictions. En choisissant d'entrer en politique en 1994 sous la bannière du PDS, Heym devient, tout d'abord, l'archétype d'une certaine prise de parole, peu commune au regard de la tradition intellectuelle allemande. Stefan Heym a en effet toujours évolué aux frontières de la politique et de la morale cherchant à les concilier plutôt qu'à les opposer. Devenir représentant du peuple à la suite d'élections démocratiques offrait à Heym une tribune privilégiée pour délivrer son message civique. Cet exemple nous permet de réfléchir à la double logique qui soustend l'espace public : logique intellectuelle et logique politique, construites chacune autour de leurs propres règles du jeu. Mais paradoxalement, sa sortie du politique s'avère tout aussi problématique. L'intérêt du " cas Heym " réside également dans la manière dont le passé d'un homme va se retrouver au cœur d'un processus d'instrumentalisation complexe. Sa vie est certes suffisamment riche pour constituer un réservoir d'images exemplaires ou répulsives, mais les usages stratégiques sont ici tels - entre promotion et stigmatisation - qu'ils finissent par poser la question des véritables enjeux d'une mise en scène aussi sophistiquée. On peut se demander si, à travers la vie de Heym, l'Allemagne réunifiée ne se prête pas à un vaste travail d'introspection collective
Stefan Heym's name has been associated with political controversy since his early youth. As a young Jew, he had to flee his hometown to Czechoslovakia and finally to the United States to escape the Nazi régime. As an officer in the U. S. Army, he took part in the Normandy D-Day operations. Mac Carthy's arrival in office urged him to leave the US for the GDR, where he played his role as an intellectujal woleheartedly, always true to his convictions. By choosing to embrace politics in 1994 for the PDS party, Heym became first and foremost the archetype of a certain voice unusual in the German intellectual tradition. Indeed Stefan Heym always moved along boundaries between politics and ethics, trying to combine them both rather tan to oppose them. Becoming a member of parliament elected by citizens in a democratic election was to offer him a privileged rostrum to deliver his civic message. This example makes it possible to ponder over the twofold logic wich underlies the public sphere : intellectual logic and the political one, both being constructed on their own rules. Paradoxically, résigning from politics was equally problematic. The interest of the " Heym case " lies also in the way a man's past is brought into the heart of a complex instrumentalization process. His life is rich enough to provide a wide range of images - be they exemplary or repulsive -, but here strategic uses are such-between promotion and stigmatisation - that they lead us to ask the question of the real stakes of such a sophisticated staging. Through Stefan Heym's figure, we can wonder if reunified Germany doesn't indulge in a far-reaching bout of mass soul-searching

Abstract:
Earlier studies of Stefan Heym have tended to mention the ‘cult of personality’ only in passing, but the investigation of megalomaniac, and usually historical, figures is a focal point of both his literary and journalistic output. Viewed in its broader sense, the cult of personality can be found throughout his works, and not solely in those which confront the legacy of Stalinism. This study charts the progression of Heym’s investigation into charismatic attraction and deliberate image cultivation. It first examines the pieces of his youth, and those in which he recognises the phenomenon within American society and post-war communism. Attention is then turned to demystification of the Stalin cult through the veiled criticism contained within his historical fiction written in the GDR, in which, crucially, pre-Stalinist forbears are identified. Heym’s approach to the problem is also problem is also examined in texts published after reunification and thus devoid of self-censorship. Heym’s focus was not restricted to charismatic performance on the political level alone; the social level of personal interaction represents a significant aspect of his fiction. Thus the function of ‘minor characters’ is given considerable attention, both as followers or creators of leader figures and in the context of the individual’s role as an agent of history. Against this background, Heym’s understanding of the writer’s role in society is considered and the question raised as to whether he himself developed a ‘cult’ of the dissident intellectual or literary figure. Much use is made of previously unexplored material from Heym’s personal archive in Cambridge University, while the archive itself is examined as a literary form and as an essential part of Heym’s own self-presentation.

Abstract:
The Hypothetical Equivalents and Inequivalents Method (HEIM) has been developed to support decision making in multiattribute problems where one decision maker is making the decision. In this paper HEIM is modified to support group decision making in multiattribute problems, resulting in the Group Hypothetical Equivalents and Inequivalents Method (G-HEIM). Instead of aggregating attribute weights or overall alternative values from each individual as is common in other group decision methods, G-HEIM operates by aggregating individual preferences. It is recognized that in group decision making, common preferences among group members can rarely be guaranteed, unless individual freedom is greatly limited. G-HEIM instead allows individuals to freely express preferences over a number of hypothetical alternatives and then explores the level of conflict or differences from the aggregated group preferences. The relationship between the level of conflicting preferences and the usability of the resulting decision is also directly studied using the G-HEIM. An automotive selection example is used to illustrate the approach.

Günümüz dünyasının iletişim ve ulaşım imkânlarıyla gelişen seyri, yalnızca düşünce ve ürünlerin değil aynı zamanda uluslararası nüfus hareketlerinin de yoğunlaşmasını beraberinde getirmektedir. Tarihsel olarak çeşitli göç hareketlerinden etkilenen Türkiye, bugün eş zamanlı olarak hem göç veren kaynak ülke hem göç alan hedef ülke hem de göçlere geçiş güzergâhı olarak transit ülke pozisyonundadır. Bununla birlikte, Türkiye ekonomisindeki göçmen iş gücünün durumu, çeşitli risk ve fırsatların birlikte ele alınmasını zorunlu kılmaktadır. Zira göçmenlerin çoğunlukla tarım, sanayi ve hizmet sektörlerinde kayıt dışı istihdama yönelmesi, yerel halkın istihdam olanaklarının daralmasına; söz konusu sektörlerde ücret düşüşleri ve iş kayıplarının yaşanmasına sebep olmaktadır. Diğer taraftan, ekonomik ve kültürel sermaye sahibi göçmenlerin iş yaşamına dâhil oluşu ve gerçekleştirdikleri yatırımlar, ülke ekonomisine katma değer oluşturmanın yanında hem yeni istihdam alanlarının geliştirilmesi hem de nitelikli iş gücü ihtiyacının karşılanmasına yönelik önemli katkı sağlamaktadır. Bu kapsamda elinizdeki metinde, ülkemizdeki göçmen iş gücüne ilişkin verilerden hareketle, entegrasyonun gerçekleşmesinde belirleyici bir parametre olan nitelikli göçmenlerin çalışma yaşamına uyumları risk ve fırsatlar temelinde irdelenerek somut politika önerilerine yer verilecektir.

Bu politika notu, ev sahipliği ve kiracılığın değişen yapısını istatistiki veriler üzerinden analiz etmeyi ve temel bulgular ışığında kısa, orta ve uzun vadede uygulanabilecek eylemler, stratejiler ve politikalar önermeyi amaçlamaktadır. Bu kapsamda raporda kiracılığın değişen anlamı, dünya ve Türkiye’de konut piyasasında, konut fiyatlarında ve ev sahipliği oranındaki değişim, kentsel dönüşümün konut piyasası ve kiracılar üzerindeki etkileri irdelenmekte, genel çıkarımlar yapılmakta ve öneriler sunulmaktadır. Raporun temel bulguları, küresel konut piyasasındaki ve Türkiye’deki eğilimlerin benzer istikamette olduğu, kiracıların motivasyonunun ve buna bağlı olarak ihtiyaç ve sorunlarının farklılaştığı, Türkiye’de ailelerin %6’sına tekabül eden yaklaşık 1,5 milyon ailenin hem yoksulluk sınırının altında hem de ikamet ettiği evde kiracı konumunda yaşadığı, kentsel dönüşümün kiracıları ve hassas kesimleri çoğunlukla olumsuz etkilediği yönündedir. Bu sebeple konut politikalarının odağında hem yoksul hem kiracı olan 1,5 milyon aile yer almalıdır. Rapordaki öneriler destekleme ve düzenlemeye ilişkin acil müdahale ve eylemler, kiralanabilir konut stokunu artırmaya yönelik kısa vadeli çözümler ve stratejiler, hassas kesimleri ev sahibi yapma ve konut arzının yapısını değiştirme gibi orta ve uzun vadeli politikalar şeklinde özetlenebilir.
