Ashton-Griffiths, Roger. "'Paul Scott's Hippie' and Hanging On." Thesis, University of East Anglia, 2015. https://ueaeprints.uea.ac.uk/59221/.

Abstract:
This thesis consists of a critical essay examining the evolution of genre and aesthetic in English fiction about South Asia, and a novel about a family of Sri Lankans settled in England. In the essay I question how a contemporary English novelist can place himself amongst the cohort of those earlier writers who have written about South Asia, since the relationship between the former colonists and colonised has changed so greatly. I demonstrate that generic elements were established in the earliest examples of this fiction, which remained notably constant throughout, even up to the late work of Paul Scott. However, I show that the aesthetic attending this Anglo-Indian genre begins with an affirmation of the benefit to the subcontinent which the colonial project might bring and ends with a repudiation of that idea. I seek to break new ground by arguing that the repudiation is so unequivocal in the work of Scott and others that this fiction comes to express an aesthetic of apology. I argue that Scott recognises that it has become possible no longer to write within the Anglo-Indian genre, and that his final novel, Staying On, has evolved into the Commonwealth genre. My novel, Hanging On, is an echo of Scott's last novel. It examines the life of a man who wishes to remain more-or-less British, rather than become independently Sri Lankan, and the legacy of his position as it filters down through the generations of his family in Luton. Cultural difference is now the prime source of conflict, rather than political power, and it aims to demonstrate that cultural hybridity is the paradoxical consequence of the racial rigidity which accompanied the colonial project to its termination.

Abstract:
This Statement of Purpose does not give a history of hip-hop or hip-hop poetry but rather how this particular poet fits into the current phase of hip-hop and performance poetry. In it, I discuss and explain the new pro-working class hip-hop performance poetic. This includes extensive discussion of how rap poetry conveys meaning through sound. Also discussed is the socioeconomic context for the suburban southwestern topics found in the manuscript. This statement is a parallel piece to the manuscript itself in that it explains a brown kid from San Bernardino’s journey of connecting words and music for purposes of personal expression and potentially the betterment of his community. This statement also discusses my aesthetic beliefs and preferences as a sound poet with a background in performance and music.

Abstract : "To make a movie about our lives": In 1981 this was the idea of a group of young film lovers when they put together ideas about their teenage years in the 1970s with a Super-8 camera to release the feature film "Deu pra ti, anos setenta" in Porto Alegre. The film would become a local hit due to its proposition of telling the memoirs of a decade through the eyes of the middle-class youth of that city. In the following year, the same team would release "Coisa na Roda", a feature film in the same aesthetic format that would focus on the survival possibilities of the dreams of four young people who lived in an urban community in Porto Alegre: a flat where everything was shared and discussed. Both works meant the beginning of a new type of discourse to the city: a young and countercultural one, which was eminently affirmative of the urban scene. This dissertation will take into account the fact that both works constitute, together, a possibility of thought about the urban space: a thought that articulated several distinctive ideas, specific to the social and political circumstances of the times where they were produced, but also of the cultural background that its producers carried with them. With the tools available to them, they recreated their lives through video, covering them with meaning while creating an image of Porto Alegre: a city seen through Super-8, open to be signified with memories and quotations and to be both the stage and the object to the dreams of film making.

Mercer, Kevin. "The farm a hippie commune as a countercultural diaspora." Honors in the Major Thesis, University of Central Florida, 2012. http://digital.library.ucf.edu/cdm/ref/collection/ETH/id/587.

Full text
Abstract:
Counterculture history is often divided, with a focus on either the turbulent 1960s or the "back to the land" exodus of the 1970s. A study of Stephen Gaskin and his followers' founding of The Farm, a rural commune near Summertown, Tennessee, provides a unique insight into the commonalities and connections of these two periods. It will be the aim of this thesis to weave the separate narratives of this demographic into one complete idea. The idea that the hippies constituted a counterculture suggests that once that culture went into exile, onto numerous communes, they existed as a diaspora. The Farm's existence as a spiritual commune, with their roots in Haight-Ashbury's short-lived utopian dream, and their continuation and evolution of that dream in Tennessee, make this particular group a model for the diaspora. The Farm, with its larger profile, publishing, and outreach programs, became the preeminent post-Haight-Ashbury commune. The commune was able to preserve the counterculture in exile, while it became a leader in dictating the direction of its progress. The Farm's efforts in midwifery, sustainable living, promotion of vegetarian diets, and outreach in America's inner cities and the Third World all point to a proactive counterculture and the commune's leadership role for the remnants of the counterculture. While the profile of the counterculture has diminished, a shift in American attitudes toward natural childbirth, ecology, and a more earth-friendly diet containing a greater variety of organic and vegetarian options reveal a significant success for their agenda.

Abstract:
Background: Managers working with sustainability and environmental issues are receiving higher priority in organizations (Knight & Paterson, 2018) and the environmental issues are today discussed at the top management level in corporations worldwide (Landrum, 2017). In line with the increased interest of the role, literature focusing on the individual sustainability manager, has recognized the need for competencies and motivational drivers in order to take sustainable actions (Wiek et al., 2011; UNESCO, 2017; Rieckmann, 2018). In addition, literature presents a significant positive relationship between motivations of individual managers and firm performance. However, previous literature covering key competencies for sustainable development, as well as individual motivations for working with sustainability, is rather ambiguous. Therefore, the authors identified a gap in the literature and initiated an interest to further investigate the key competencies and individual motivations of a sustainability manager operating in Sweden today. Purpose: The purpose of this study is to investigate the individual sustainability manager operating in Sweden today. The study aims to identify the key competencies required for operating in the role, and to explain corresponding underlying motivations for working with sustainability. Method: The study follows a qualitative and exploratory approach by combining existing literature, theories and empirical data. The empirical data was collected through six semi-structured interviews. The empirical findings were further analyzed following a thematic analysis technique. Conclusion: The authors suggest a proposed framework that illustrates the identified key competencies required for operating as a sustainability manager in Sweden today, as well as the underlying motivations for working with corporate sustainability. The illustration of the proposed framework provides a generalized foundation for understanding the sustainability manager role in terms of key competencies and individual motivations. This framework adds to current literature on the topic of sustainability management and can be used as a means for evaluating current sustainability managers, as well as a means to analyze current education for sustainable development.

Capitalism, nowadays, promotes a global world and a homogenized culture to obtain more profit. It changes the conditions of thousand people lifes and works to reach its objectives. Such process is also happening in Goiânia, what can be noticed by the increase of unemployment, while some commercial and industrial jobs get extinct. So, the recent unemployed person finds himself / herself in a world without structure and he / she cannot find answers in the tradicional sence producer instituions. Some of these recent unemployed people are the group of the researched open marketing workers . When they search explanations for the shaking of their financial and faith situations, they discover, in the symbolic wealth market , religions that promess them solutions. They do not transit uncomfortably, they become walkers among these systems. They make their choices, in a subjective and individualized way, for a model that fits in his interests. Even though there are few exceptions, they question the sense system , but not their faith in God. They still fear Him, keeping with them the historical sense strengh the open marketing workers need so much, even in the day-by-day precariousness.

Abstract:
The hippies are the descendants of the beat generation of the fifties. Both groups showed non-conformism. The hippies tried to create a new type of family based on companionate marriage. They liked life in a community, close to nature. They were against war, but refused to take part in public demonstrations in the streets. They were nonviolent, above all. They rejected western materialism. Eastern philosophy was the only antidote for them. Heyday could only be reached with drugs. The songs of the sixties and seventies tried to awake the American consciousness. At the end of the seventies, the movement faded some hippies became artists, others became contestants and others yuppies. They are the present-day trends of the united-states. They are young urban professionals. They are serious and ambitious. They aspire to glory. They dream of power and money. They like sports and eat only healthy food. They have comfortable incomes. In politics, they are at the same time (in social matters) and conservative (for their business). They work harder than their parents to succeed, (the economical conjuncture is not particularly good for them. ) In the seventies, the American had lost the notion of American dream, after several disillusions: Vietnam, Watergate, energy crisis,. . . , but on the contrary, the yuppies think it is possible. If you really want it badly. Despite the diametrally opposite conceptions, some hippies went through their judgment again and became yuppies

Abstract : The growing presence of outsiders in Samaipata, a tiny town near Santa Cruz de la Sierra, makes that, very often, the noun ?gringo? be used to refer to people, in the same way that he adjective ?gringo? is linked to certain things (gringo parties, gringo places, gringo manias?). Nevertheless, both uses showed to be much more plastic than would be expected at first glance: even when gringo was treated as someone from abroad, identifying this person and establishing what is being considered to be the local (Samaipata, the lowlands, Bolivia, Latin America) depended much more on the circunstances at stake than on any sort of encyclopedic definition. This assumption, however, didn?t imply that the idea of gringo should be treated as a strictly subjective conception, independent from the circulation of other ideas and from the share of some experiences that were going around the town. Moreover: there was a certain ?bad reputation? very widespread ? though not absolutely widespread ? that was reflected in some instituted practices that were being performed by the local authorities in order to undermine eventual ?abuses?practiced by outsiders in town ? tourists and established. The main aim of this dissertation was to understand the reasons that made this label so widespread and why this category, gringo, was so frequently related to some forms of subversion of the local ways of being.

Abstract:
In the 1940s, 50s and 60s the two youth movements of the beats and the hippies emerged in the United States. Disagreeing with the progressive positivism of the previous generations these youths were part of a counter culture that adhered to an ideal of living in the present. Alan Wilson Watts (1915–1973), most known for his popularization of eastern philosophy and religion in general and Zen-Buddhism in particular, became one of the most influential and well-read advocates of the new movements. Drawing upon eastern philosophy and religion as well as modern psychology, Watts challenged the western world view by rejecting the dualism of self and environment, arguing that man is one with God and nature and claiming that nothing exists but the present experience here and now. Based on Watts’ view of man and reality, and in the context of the emerging youth movements, this study examines Watts’ criticism of western culture and society as well as his view on happiness and the possibility of a better life for the individual living in the west. The study shows that, according to Watts, the root of the problem facing western society lies in man’s incapability of distinguishing concept from reality, preferring abstract ideas and symbols to the experience present in everyday life. As a result, man perceives himself as separate from nature, waging war on his environment in a futile attempt at finding happiness by constantly striving for ever greater achievements of which the main goal always seems to lie somewhere in the future but never in the present. His ideas inspired the youth movements, although they moved in a different direction than the one he would have wanted. Watts, suggesting that an altered understanding of oneself as one with the universe is central in maintaining a thriving society in union with nature, claims that the essence of happiness consists of this understanding here in the present rather than the pursuit and obtaining of a desired goal.

Yang, Chao. "Solidification behaviour and hipping induced surface modification in Ti4522XD castings." Thesis, University of Birmingham, 2012. http://etheses.bham.ac.uk//id/eprint/7614/.

Full text
Abstract:
The solidification behaviour of Ti45Al2Mn2Nb1B (at.%) has been studied together with its response to HIPing (Hot Isostatic Pressing) in order to understand the mechanism of grain refinement in castings and to understand the influence of surface changes occurring during HIPping on the properties of HIPped net shape cast turbine blades. Samples which had been rapidly cooled from near the melting point from a Bridgman furnace, where a thermal gradient was imposed, have been used to understand the grain refinement mechanism and the details of the solidification sequence. In addition the structure of powder samples, which have been gas-atomised and hence very rapidly cooled have also been used to further the understanding of solidification and of the role of borides. It has been shown that borides themselves play an important role in grain refinement. It has been shown that HIPping results in the formation of a surface which is caused by oxidation from the oxygen present in the argon used in the HIP. The details of the chemistry and microstructure of the surface layers have been shown to be influenced by oxygen partial pressure, by HIPping time and HIPping temperature. Conventional HIPping conditions lead to a surface which contains a γ-layer which does not appear to downgrade either the tensile properties or fatigue properties of the samples and may slightly improve the corrosion resistance. Further work is required to produce net shape castings, which have properties comparable with conventionally cast Ti4522XD, but the present work shows that these could then be HIPped without the γ-layer contained surface causing any downgrading in properties.

Abstract:
“Bluesbewegung” is the name of a youth movement that arose in the late 1960’s in the GDR. This movement encouraged the cultural transfer which brought American blues music into the GDR. My study wants to reconstruct this process beginning with the motives for being interested in Blues music, continuing with the transfer of Blues music throughout the media (especially in radio broadcasting), and finally, ending with the GDR-specific appropriation of Blues within and outside of the youth scene

Abstract:

This dissertation investigates the role that eros in general, and philosophical eros in particular, plays in the search for the eidos of the beautiful in Plato’s Hippias Major. It defends the claim that noesis of the eidos of the beautiful can only be accomplished within the life of philosophical eros, that is, within the life of eros which is directed toward the good. As such this dissertation aims both to provide an interpretive key to the Hippias Major, allowing us to read the dialogue in a rich and novel way, and also to make the claim that the Hippias Major presents us with a picture of the interrelation between eros, philosophy, and beauty, and about how these three elements manifest themselves in human life. As such, some continuities and parallels can be found between it and the other two dialogues which deal most explicitly with beauty and eros, the Phaedrus and Symposium. The first five chapters interpret a particular section of the Hippias Major according to role the eros plays within it, attempting to show that eros, both in general and in its unique manifestation as philosophical eros, is a crucial mediating term for any comprehensive understanding of any section of the dialogue, and therefore of the dialogue as a whole. In each of these five chapters, I will articulate the role that eros plays within the search for obtaining a noetic glance at the eidos of the beautiful. The first chapter demonstrates how Socrates’s philosophical eros gives birth to the question about the beautiful itself within the context of a discussion about sophistry and money. The second chapter shows how Socrates’s philosophical engagement with Hippias’s definitions of the eidos of the beautiful generates a dialectic of ascent, allowing Hippias to expand his understanding of what counts as beautiful in a trajectory that mirrors Diotima’s ascent in the Symposium. The third chapter articulates the erotic significance of Socrates’s claim that the eidos of the beautiful inheres in being and not appearances. The fourth chapter gauges the erotic significance of Socrates’s and Hippias’s claim that the beautiful is good, and the good beautiful. The fifth chapter interprets the comic and tragic aspects of the dialogue in terms of philosophical eros, its rejection and fulfillment. The sixth chapter will take stock of the overall interpretation of the Hippias Major developed in the first five chapters, and will present the overarching view about the relationship between the contemplation of beauty, on the one hand, and desire for possession of beauty and moral concern, on the other, which one can glean from the character and action of Socrates in Hippias Major. It will bring this view into a conversation with the notion of “liking devoid of interest” which is found in Kant’s Critique of Judgment. The conclusion of this dissertation will underscore the principle claim, that the philosophical search for the eidos of the beautiful can neither be separated from the eros which beauty inspires in a human being, nor can it be accomplished without one’s eros benig directed toward the good, and that this philosophical search is marked by suffering and possible tragedy

Zhang, Kun. "The microstructure and properties of hipped powder Ti alloys." Thesis, University of Birmingham, 2010. http://etheses.bham.ac.uk//id/eprint/856/.

Full text
Abstract:
In the present study, the effect of hot isostatic pressing (HIPping) variables such as HIPping temperature, HIPping cycle and powder particle size, on the microstructure and mechanical properties of HIPped samples of two Ti alloys have been assessed. Powders of the most commonly used (α + β) alloy, Ti-6Al-4V and one specific beta alloy, Ti-25V-15Cr-2Al-0.2C wt% (burn resistant titanium alloy, BuRTi) were studied. The Ti-6Al-4V powder was made by the plasma rotating electrode process (PREP). BuRTi powders, which were made both by gas atomisation and by PREP were HIPped to investigate the influence of the initial structure of the powder on the microstructure and associated mechanical properties of the HIPped alloy. The PREP Ti-6Al-4V powder was shown to be fully martensitic in the as-atomised condition. The gas-atomised and PREP powders of BuRTi showed very different as-atomised structures, but in both cases the structure was, as expected single phase beta, with the carbon retained insolution. The individual particles of gas-atomised BuRTi powder were always polycrystalline, although there was a significant scatter in grain sizes within different particles. In contrast the individual particles in the PREP powder were either coarse grained polycrystals or single crystals. These differences led to significant differences in the microstructures and properties of HIPped samples. It was found that HIPping of Ti-6Al-4V samples resulted in the formation of equiaxed regions and lath-like microstructure. The small equiaxed regions are formed by recrystallisation which occurs at original particle boundaries where most of the deformation occurs during HIPping; the lath-like microstructure is formed by simply tempering the (less deformed) original alpha prime martensite within the central part of original particles. Among the three HIPping temperatures used, samples machined from powder HIPped at 930°C exhibited a better balance of properties than those HIPped at 880°C or 1020°C. The fatigue properties of samples HIPped at 930°C, made using different HIPping procedures were compared. It was found that samples which contain the as-HIPped surface, which were made using a new HIPping procedure, have better fatigue properties than samples with as-HIPped, machined or electro-polished surfaces which were produced by conventional HIPping . The properties of optimally HIPped Ti-6Al-4V samples are as good as or better than ingot-route samples. In the case of BuRTi the original single crystals or coarse grained polycrystals in the PREP powder are retained after HIPping and limited grain growth occurs in the gas-atomised samples. The tensile strength is comparable for the gas-atomised and PREP samples, but samples tested to failure showed a significant scatter in ductility (a larger scatter in the PREP powder samples) and all fracture surfaces contained large circular fracture initiation sites, with larger sites associated with lower ductility. Initiation occurs in the centre of these circular regions in large grains or in adjacent grains which have similar orientations and the failed region expands symmetrically in powder samples where no texture is expected. The fatigue properties of the PREP samples are much lower whereas the fatigue properties of the gas atomised samples are better than those of samples from ingot route. This behaviour is associated with obvious facetted failure sites in the PREP powder samples where it is suggested that the coarser microstructure has allowed persistent slip to occur leading to localised deformation and to premature failure. These observations are discussed in terms of the potential of net shape HIPping for the production of engineering components and in this context the fact that a new HIPping schedule has been developed during this study, where the fatigue properties of samples containing an as-HIPped surface are excellent, is very significant.

Kirwan, M. A. Q. "Diamond machining in 5 wt% Y2O3 sinter hipped silicon nitride." Thesis, University of Surrey, 1992. http://epubs.surrey.ac.uk/843045/.

Full text
Abstract:
A collaborative research project was set up to study peripheral diamond wheel machining damage in silicon nitride ceramics. The objective of the work to be carried out at the University of Surrey was to study the nature and depth of machining damage in 3 point flexural rupture test bars made from 5 wt% Y2O3 sintered hot isostatically pressed silicon nitride. The bars were machined to three surface finishes. The work carried out at Rolls-Royce by Mr R Quinn concentrated on the effects the machining damage had on the fracture strengths of the test bars. Work at the University of Surrey has identified in detail the nature and depth of machining damage in "coarse" 0. 4mum centre line average roughness surfaces. Deep grooves up to 2mum depth, 18mum width are found to have been superimposed on the general surface roughness by singularly large diamonds in the 350 grit diamond wheel. Sub-surface median cracks normal to the machining direction were clearly identified in bar cross-sections using oblique, diffuse "penumbra" illumination in an optical microscope, an as yet undocumented technique. Cross-section views of the machined surface and sub-surface were made possible by the very difficult and delicate technique of producing sandwich cross-sections of the machined bars. An analysis of machining-induced median cracks has not been carried out in such detail before. Semi-elliptical in shape the median cracks extend from 6mum - 45mum below the machined surface, and range from 19mum to 101mum in length parallel to the machining direction. They initiate at the focal point of a tributary system of microcracks at an average depth of 4mum - 5mum below the machined surface. It is believed that the median cracks initiate at the plastic/elastic boundary of a plastically deformed surface layer. Therefore a residual compressive layer, formed by the overlap of localised residual stresses from multi-particle contact events. and bound by an underlying tensile field, is thought to have an average depth of 4mum - 5mum. A very innovative technique was used to reveal sub-surface deformation, where TEM X-ray microdiffraction spots were distorted by mechanical damage in the ceramic grain structure. The "arcing" or "streaking" of the diffraction spots tended to disappear at a depth of 4mum - 5mum below the machined surface. This is further evidence of the existence of a thin layer in residual compression, which has an average depth of 4mum - 5mum. This technique is not known to have been used before. Fine diamond machining with a 600 grit wheel produced a centre line average roughness of 0.01/0.02 mum. However, evidence of machining damage is still present in the form of "remnant tracks" which lie parallel to the machining direction and consist of material pull-out. They are remnants of machining damage under grinding grooves introduced in previous machining stages. Single point Vickers pyramid diamond scratches were implemented at different loads on a polished surface. The morphology of the grooves and material fragmentation and the sub-surface median cracks were examined. Many features were found to resemble the deformation/fracture formed under a deep grinding groove in the coarse machined surface. Work carried out at Rolls-Royce by R Quinn showed that an increase in the quality of surface finish is accompanied by an increase in the mean strength and Weibull modulus of the machined bars. Furthermore a distinct anisotropy in the fracture strengths parallel and normal to the "coarse" machining direction is evidence of anisotropy in machining damage formed by a peripheral diamond grinding wheel. X-ray diffraction tests carried out at the CEGB by P E J Flewitt showed that machining damage produces a long range biaxial residual compressive field with the highest component acting normal to the machining direction. These results are consistent with the nature of machining damage identified at the University of Surrey, namely the strength-controlling median cracks which lie parallel to the machining direction and the residual compressive stress which exists as a thin 4mum - 5mum layer below the machined surface. Processing flaws were discovered in the as-hipped billets received for the project. Their elemental composition and likely origin were examined. A three dimensional "cellular network" flaw ranging from 400mum to 2.1mm in size (in different production batches) is believed to have been formed as a result of flocculation clustering during processing. Clusters of 1mum - 3mum metallic particles were also identified. They range from 5mum - 45mum in size. The contaminant particles are steel and were introduced as a result of the original ceramic powder ball milling process which employed a steel ball mill.

Abstract:
The most common question that scholars ask regarding the concept of techne in the Plato dialogues is if it is possible to understand arete, morale or the good, as a techne, that is, if arete kan be viewed as a knowledge, an ability or a skill. Is arete something that can be taught? In this essay I will explore this question from a reversed viewpoint, and try the claim that techne always in itself is an excercise of arete, inasmuch as the goal of a techne has to be the good, or ”what is best”, in order for it to be a techne for Plato at all.

Zhang, Qinqin. "The microstructure and properties of powder HIPped nickel-based superalloy CM247LC." Thesis, University of Birmingham, 2011. http://etheses.bham.ac.uk//id/eprint/1467/.

Full text
Abstract:
The response of powder of the Nickel-based superalloy CM247LC to a range of Hot Isostatic Pressing (HIPping) conditions and post-HIP heat treatments has been investigated as a preliminary step in the assessment of net shape HIPping as a possible process-route for component production. A HIPping pressure of 150MPa was used at temperatures of 1100°C, 1200°C, 1260°C and 1320°C. 1260°C as-HIPped specimens had the best tensile and HCF fatigue properties at both room temperature and 750 °C; the influence of post-HIP solution treatment conditions and of subsequent ageing on the properties of samples which had been HIPped at 1260 °C was thus investigated. A solution treatment at 1100 °C for 1h followed by forced air cooling and ageing at 870 °C for 16h resulted in the optimum tensile, high cycle fatigue at both room temperature and 750 °C. The optimized microstructure consists of a ‘necklace’ structure of coarse γ′ particles along the grain boundaries; cuboidal γ′ distributed inside grains with hyper-fine γ′ precipitated in γ channels; and fine carbides homogeneously disperse in the matrix. The properties of samples with this microstructure were comparable with those of cast, directionally solidified CM247LC. Solution treatment at 1260 °C gives better creep resistance at 760 °C/ 350MPa. It was also concluded that with gas-atomised powder, solution treatment must be carried out below the HIP temperature to minimise porosity. Analytical scanning electron microscopy showed that Hf-rich inclusions, some of which had alumina cores, initiated failure in almost all tensile samples and in all fatigue samples. It was shown that this type of inclusion can be avoided by removal of Hf from the alloy; but the removal of Hf – which is added to the alloy to improve castings – degrade the creep properties over the temperature range of 700 °C to 950 °C and the stress range of 150 MPa to 550 MPa. Further work is required to assess the influence on the tensile and fatigue properties.

Abstract:
The von Hippel-Lindau (VHL) disease is a hereditary cancer syndrome that is transmitted in an autosomal dominant manner. The disease is characterized by the formation of highly angiogenic tumors in many organs but the main causes of mortality are renal cell carcinomas and hemangioblastomas. Mutations in the VHL protein are responsible for the pathogenesis of the disease. VHL associates with elongin Band C to form the VBC complex. The cullin 2 protein (CUL2) and ring box protein 1 (RBX1) also associate with the VBC complex to form an E3 ubiquitin ligase involved in the ubiquitination and subsequent degradation of the hypoxia inducible transcription factor (HIF2alpha). Mutations in VHL that abrogate its E3 ligase activity lead to increased levels ofHIF2alpha and the subsequent accumulation of pro-proliferative and pro-angiogenic HIF2alpha target genes. VHL also has an important function in the regulation of extracellular matrix (ECM) assembly which is independent of its HIF2alpha regulation pathway. VHL's regulation of ECM assembly was shown to have important consequences for tumor angiogenesis and cell invasion. It was shown to be necessary for the proper assembly of a fibronectin matrix and was most recently found to interact with collagen IV alpha 2 (COL4A2). The aim of this thesis is to further characterize the VHL-COL4A2 interaction. VHL was shown to interact directly and specifically to COL4A2 and is necessary for proper COL4A2 matrix assembly. The association of VHL with COL4A2 appears to be independent of its functions as an E3 ubiquitn ligase and CUL2 was identified as part of the VBC complex that associates with collagen IV (COL4). Furthermore, a strategy to identify the binding site of VHL on COL4A2 has been employed and is in progress. These experiments represent the beginning of investigations into the novel interaction between VHL and COL4A2.
