The thymus develops from the ventral portion of 3<sup>rd</sup> pharyngeal pouch endoderm. At E9.5 this region is marked by expression of the transcription factor <i>Rhox4, Rhox4 </i>may therefore have a role in the developing thymic primordia. <i>Rhox4 </i>is a member of the recently described <i>Rhox </i>family of homeobox-containing genes, which are present in 3 clusters on the mouse X chromosome and are primarily expressed in reproductive and extra-embryonic tissues. The expression of <i>Rhox4 </i>in the thymus, in addition to the testis and placenta makes it unique amongst family members.

Quiescent rat thymocytes were stimulated to divide by a variety of agents. One such mitogen was the neurotransmitter acetylcholine which exhibited a biphasic action. Interaction with low affinity nicotinic receptors was linked with an obligatory requirement for magnesium ions whereas combination with high affinity muscarinic receptors induced mitosis only if calcium ions were present in the medium. Natural killer (NK) cells are the major lymphocytes of the innate immune system, but their developmental pathway is not fully defined. It is commonly assumed that all NK cells develop in the bone marrow. In this thesis, I describe a novel thymus-dependent pathway of NK cell development that is specific to those in the thymus and lymph nodes. Microarray analysis revealed TCRγ mRNA expression in NK cells. Genomic and RT-PCR showed that some NK cells have rearranged TCRγ genes while TCRβ and TCRδ genes are in germline order. By techniques of immunohistochemical staining, flow cytometry, and image analysis, the present study demonstrated the following: (1) The ST3 antigen is expressed on the surface of thymocytes of young rats, and decreases in both percentage and intensity with increasing age (after 12 months). (2) The ST3 antigen also is expressed on the surface of thymic nurse cells. Although the majority of lymphoid cells within nurse cells also bear the ST3 antigen, a subpopulation contains lymphocytes that do not. These results suggest that the ST3 antigen may be a useful new marker for cortical lymphocytes,

The thymus is a complex cellular structure made up of several interdependent cell types and is the primary site for T cell development. A population of fetal thymic epithelial cells (TEC), marked by MTS20 and MTS24, when grafted in vivo can generate a functional thymus containing all thymic epithelial cells and is capab,e of supporting T cell differentiation.
