Abstract:
One hundred nine HIV seronegative gay (HSG) males from the Montreal gay community, drawn from downtown health clinics, advertisements in gay friendly newspapers, and referrals from colleagues, were examined with respect to how they were coping with the HIV and AIDS epidemic (July 3, 1981 to August, 1998). Participants were asked to complete both a detailed demographic questionnaire and the Ways of Coping Questionnaire. Coping theory and coping research relevant to the research at hand are discussed in detail. Results indicate that HSG males in Montreal are coping most frequently with the HIV a

The primary aim of this thesis was to examine whether HIV -1 specific T cell responses in HIV -1 exposed seronegative (HESN) subjects could be firstly detected and if so, conferred protection against productive infection from HIV -1. Cultured IFN-y ELISpot found 38.7-60% of HESN subjects had detectable HIV-1 specific T cell responses. HIV -1 specific T cell responses could be titrated, were typically mediated by CD4+ T cells and tended to map to previously defined, promiscuous epitopes. The aim of this thesis was firstly to investigate the possibility of local antibody in urine samples from HEPS individuals to evaluate the theory that such individuals may develop a protective local anti-HIV-l antibody response, detectable in urine. Since elicitation of mucosal immunity is important in preventing viral infection by sexual transmission, development of a vaccine which is effective for the induction of mucosal immune responses may provide protection to mucosal surfaces against HIV-1 transmission. Objective: Serpins are serine protease inhibitors that are involved in a wide variety of biological functions in nature. They are known to regulate inflammation processes as well as provide host defense against microorganisms. Recent evidence has associated many types of mucosal serpins with a protective phenotype against HIV infection in women. Our hypothesis is that serpins with known antiviral activity against HIV-1 are correlated with protection in a group of HIV exposed seronegative individuals (HIV-resistant) from the Pumwani sex worker cohort. Abstract:
Abstract Introduction HIV serodiscordant couples are presenting more often to their doctors as the epidemic continues to spread. There have been various theories as to why some couples remain discordant, but none have yielded a conclusive answer as yet. The negative partners in discordant couples have previously been ignored. However, more of them are now presenting with psychosocial problems of their own. They are increasingly being recognized as ‘hidden’ patients and as potential resources. Since the discovery of HIV-1, the production of an effective prophylactic or therapeutic vaccine remains elusive. An effective vaccine must be able to elicit a potent humoral and cellular immune response. Neutralizing antibodies target the envelope glycoproteins on the surface of HIV-1 virions thereby preventing viral entry. Unfortunately, to date only a handful of neutralizing antibodies have been identified that are capable of neutralizing different viral strains within diverse subtypes, and none have been isolated from HIV-1 subtype C infected patients. The innate immune system is at the interface between the host's immune system and the initial contact with HIV. The innate immune system has been linked to the reduced susceptibility of HIV-exposed seronegative (HESN) female commercial sex workers in Kenya by a number of studies. Understanding natural mechanisms of protection against HIV infection and disease progression are key priorities for informing vaccine and microbicide design. The research presented in this thesis aimed to characterize mechanisms of defence in HIV-exposed seronegative (HESN) individuals, who naturally resist infection by HIV, and HIV-controllers, who are HIV-infected, but suppress viral replication in the absence of treatment. Previous studies have linked resistance to HIV infection with low basal levels of gene transcription and reduced production of inflammatory mediators, suggesting an o

Abstract:
The mucosal antibody repertoire of the cervical mucosa in commercial sex workers from Nairobi, Kenya, who are highly sexually exposed to human immune deficiency virus type-1 (HIV-1) but remain persistently IgG seronegative (HEPS), may represent a novel source of broadly neutralizing monoclonal antibodies (mAbs) against HIV-1.
