Resumo: Ao principiante no estudo da moderna Geometria Algébrica se requer uma bagagem de técnicas, tais como: categorias abelianas, funtores, feixes, cohomologias, etc. Neste trabalho, procuramos apresentar algumas destas técnicas a este público. Embora o texto seja de nível introdutório, os conceitos são apresentados com um nível de generalização superior a exigida na maioria das aplicações. A teoria de cohomologias, por exemplo, é tratada no contexto das categorias abelianas, ao invés da forma algébrica, mais simples e, em geral, utilizada nas aplicações. Por outro lado, com este enfoque, ganhamos a apresentação dos delta-funtores. Os feixes são, por um lado, os coeficientes locais para cálculo de invariantes chamados cohomologias, por outro lado, uma ferramenta para expressar propriedades geométricas. Esta ferramenta é mais relevante aos estudos de propriedades de espaços com uma estrutura rígida (tais como espaços analíticos, variedades algébricas etc.) do que invariantes topo lógicos, mas abrange ambos casos. Incluímos uma rápida explanação sobre feixes coerentes. Essencialmente, a última parte do texto deseja apresentar diversos métodos básicos de cálculos de cohomologias com coeficientes num feixe, incluindo o método de Cech. Abstract: For any beginner who will study the modero Algebraic Geometry, it is required some amount of techniques such as abelian categories, functors, sheaves, cohomologies, etc. In this work, we are going to represent some ofthem. Although the text is introductory, the conceptions represented are more general than needed in most applications. For instance, the cohomologies are treated in the context of abelian categories instead of the simpler algebraic approach normally used in applications. On the other hand, this allows us to introduce the delta-functors. The sheaves are the local coefficients for invariants named cohomologies. On the other hand, they are some way to express many geometric properties. This tool is more adequate in studying the properties ofthe spaces equipped with some rigid structure (such as analytic spaces, algebraic varieties, etc.) than that of topological invariants. Nevertheless, it involves both cases. There is also some brief exposition of coherent sheaves.Essentially, the last part ofthe text is devoted to difIerent basic methods of calculating cohomologies with coefficients in sheaves, including the Cech one.This work was supported by FAPESP

Full textIn this thesis, we introduce two new approaches to compute the Persistent Homology (PH) of a sequence of simplicial complexes. The basic idea is to simplify the complexes of the input sequence by using special types of collapses (strong and edge collapse) and to compute the PH of an induced sequence of smaller size that has the same PH as the initial one.Our first approach uses strong collapse which is introduced by J. Barmak and E. Miniam [DCG (2012)]. Strong collapse comprises of removal of special vertices called \textit{dominated} vertices from a simplicial complex.Our approach with strong collapse has several salient features that distinguishes it from previous work. It is not limited to filtrations (i.e. sequences of nested simplicial subcomplexes) but works for othertypes of sequences like towers and zigzags. To strong collapse a simplicial complex, we only need to store the maximal simplices of the complex, not the full set of all its simplices, which saves a lot ofspace and time. Moreover, the complexes in the sequence can be strong collapsed independently and in parallel.In the case of flag complexes strong collapse can be performed over the $1$-skeleton of the complex and the resulting complex is also a flag complex. We show that if we restrict the class of simplicial complexes to flag complexes, we can achieve decisive improvement in terms of time and space complexities with respect to previous work. When we strong collapse the complexes in a flag tower, we obtain a reduced sequence that is also a flag tower we call the coreflag tower. We then convert the core flag tower to an equivalent filtration to compute its PH. Here again, we only use the 1-skeletons of the complexes. The resulting method is simple and extremelyefficient. We extend the notions of dominated vertex to a simplex of any dimension. Domination of edges appear to be very powerful and we study it in the case of flag complexes in more detail. We show that edge collapse (removal of dominated edges) in a flag complex can be performed using only the 1-skeleton of the complex as well. Furthermore, the residual complex is a flag complex as well. Next we show that, similar to the case of strong collapses, we can use edge collapses to reduce a flag filtration F to a smaller flag filtration F^c with the same persistence. Here again, we only use the 1-skeletons of the complexes. As a result and as demonstrated by numerous experiments on publicly available data sets, our approaches are extremely fast and memory efficient in practice. In particular the method using edge collapse performs the best among all known methods including the strong collapse approach. Finally, we can compromizebetween precision and time by choosing the number of simplicial complexes of the sequence we strong collapse

Full textSimplicial homology is a useful tool to access important information about the topology of wireless networks such as : coverage and connectivity. In this thesis, we model the wireless network as a random deployment of cells. Firstly, we introduce an algorithm to construct the Cech complex, which describes exactly the topology of the network. Then, the Cech complex is used in further applications. The first application is to save transmission power for wireless networks. This application not only maximizes the coverage of the network but also minimizes its transmission power. At the same time, the coverage and the transmission power are optimized. The second application is to balance the traffic load in wireless networks. This application controls the transmission power of each cell in the network, always under the coverage constraint. With the controlled transmission power, the users are redirected to connect to the lower traffic load cells. Consequentially, the balanced traffic load is obtained for the network

Full textGiven a contact 3-manifold (Y; α), let HF(Y ) and \ECH(Y; ) be the associated Heegaard Floer and, respectively, embedded contact homologies. In a series of papers Colin, Ghiggini and Honda proved that there exists a chain map that induces an isomorphism : HF(Y ) → ECH(Y; α) in homology. Given a knot K in Y , in [13] a hat embedded contact knot homology ECK(K; Y; α) is defined and an isomorphism with the hat Heegaard Floer knot homology HFK(K; Y ) is conjectured. These two homologies can be defined as first pages of spectral sequences arising from filtrations induced by K on chain complexes for ECH(Y; α) and HF(Y ). The aim of this thesis is to provide some evidences about the veracity of this conjecture. We define a full ECK homology and we generalize the definitions of ECK and ECK to any link. We compute then the Euler characteristics of these homologies for knots and links in homology three-spheres (endowed with a suitable contact form) and we prove that in S3 the ECK homology is a categorification of the multivariable Alexander polynomial. This fact, together with a well known analogous result in HFK, implies that the conjecture is true at the level of Euler characteristics in S3. Finally we show that, up to chain homotopies, the chain map Φ preserves the knot filtrations. This can be considered as a first step of a proof of the conjecture for fibered knots

Full textIn this thesis, we study homologous recombination at the single molecule level. We ¯rst introduce the physico-chemical characteristics of RecA and hRad51, the main proteins involved in homologous recombination in E. Coli bacteria and in humans. Then, we present some experiments on homologous recombination perfor- med with magnetic tweezers. We ¯rst study RecA-mediated strand exchange. We then address the study of the human analog of RecA, the protein hRad51. The ki- netics of the polymerisation and the torsional behavior of hRad51 nucleo¯laments are presented. Preliminary results on hRad51-mediated strand exchange are also described. Finally, we develop a single molecule set-up called tomotweezers, combining magnetic tweezers and °uorescence measurements. This set-up allows us to stretch a single DNA molecule, to control its topology and to observe °uorescent proteins near the molecule

Full textIn the first part, we construct a Poincaré duality for pseudo-manifolds with isolated conical singularities. The Poincaré duality is not true in the singular case. In 1980, Goresky and Mac Pherson, introduce the intersection homology for which the rational Poincaré duality remains true for conical singularities. We modifie their cohomology by constructing a non free complex, quasi-isomorphic to the intersection complex but whose cohomology verifies the Poincaré duality. In the second part, we define a geometrical theory of the intersection homology. It results that any intersection cycle can be represented by the cap product of the fundamental class of a manifold with boundary by a class of J-cohomology of this manifold. Full textThe base excision repair enzyme MED1 (also known as MBD4), identified as an interactor of the mismatch repair (MMR) protein MLH1, acts as a thymine and uracil DNA N-glycosylase specific for G:T and G:U mismatches at CpG sites. Methylating agents are a class of antitumor drugs that cause cytotoxicity by inducing O6-methyguanine (O6-meG). O6-meG can direct misincorporation of thymine during replication, generating O6-meG:T mismatches. MED1 was found to exhibit thymine glycosylase activity on O6-meG:T mismatches. We generated mice with targeted inactivation of the Med1 gene and prepared mouse embryonic fibroblasts (MEFs) with different Med1 genotype. Unlike wild type and heterozygous cultures, Med1-/- MEFs failed to undergo G2-M cell cycle arrest and apoptosis upon treatment with the methylating agent Nmethyl-N'-nitro-N-nitrosoguanidine (MNNG). Resistance of Med1-/- MEFs to MNNG was due to a tolerance mechanism, because DNA damage accumulated but did not elicit checkpoint activation. Interestingly, steady state amounts of several MMR proteins are reduced in Med1-/MEFs, in comparison to Med1+/+ and Med1+/+ MEFs. Thus, MED1 has an additional role in DNA damage response to anti-tumor agents and is associated with integrity of the MMR system. Embryonic lethality in mice was caused by homozygosity for the targeted Med1 allele lacking exons 1 to 3, but not for the allele lacking exons 2 to 5. This discrepancy maybe explained by concurrent inactivation of neighboring gene Wdr10. Wdr10 shares a portion of exon 1 with Med1 in the opposite orientation. This suggests that the two genes may represent a functional unit necessary for normal development
