Abstract:
Artykuł dotyka kwestii redukcji hałasu toczenia emitowanego przez wagon towarowy poprzez zastosowanie kompozytowej wstawki hamulcowej. Przedstawiono możliwości zastosowania kompozytowej wstawki hamulcowej typu LL, nie posiadającej homologacji UIC, w wagonach towarowych poruszających się w transeuropejskim systemie kolei. Publikacja zawiera analizę obecnie obowiązujących w Unii Europejskiej przepisów, regulujących interoperacyjność kolei w zakresie układu hamulcowego oraz przedstawia sposób postępowania umożliwiający zastąpienie wstawek żeliwnych kompozytowymi w istniejącym taborze. W artykule zaprezentowano ponadto wyniki prób stanowiskowych i eksploatacyjnych kompozytowej wstawki hamulcowej FR510 przeprowadzone w Polsce i w Maroku oraz porównanie właściwości tego materiału z wynikami wstawek typu LL, które uzyskały homologację UIC w projekcie EuropeTrain.

Abstract:
Induced pluripotent stem cells (iPSC) technology, which allows direct reprogramming of somatic cells to a pluripotent state, is a promising tool for gene-function studies disease modelling, drug screening, toxicology tests and to generate knockout animal models. The goal of the current work was to close the gap in knowledge with regard to the molecular biological background for rabbit iPS work by isolating the putative pluripotency genes from the rabbit, based on the sequences published for other species. The sequence of known pluripotency genes (Oct4, Sox2, Klf4, c-Myc, Nanog) were analysed and primers designed based on the similarity of sequences. Sequences of each individual rabbit pluripotency gene was compared to other vertebrates (e.g. human, mouse, bovine) phylogenetically. Rabbit ESCs and blastocyst stage embryos were collected from superovulated rabbits to isolate total RNA. Genes of interest were amplified using RT-PCR and electrophoretically separated for cDNA fragment isolation. Isolated and subcloned cDNA fragments were sequenced and analysed. Our results showed that after restriction digestion the size of amplified and cloned rabbit Oct4, Sox2, Klf4, c-Myc and Nanog gene fragments correspond to the expected amplicon size. Furthermore, sequence confirmation by DNA sequencing has been completed in the case of Oct4, c-Myc, Klf4 and Nanog. The homology of these genes to that of their mouse and human orthologs were as follows: Oct4: at Na level 79% homologue to mouse, 85% homologue to human, at Aa level 81% homologue to mouse, 90% homologue to human; Klf4: at Na level 98% homologue to mouse, 85% homologue to human, at Aa level 95% homologue to mouse, 84% homologue to human; c-myc: at Na level 88% homologue to mouse, 92% homologue to human, at Aa level 91% homologue to mouse and 94% homologue to human; Nanog: at Na level 71% homologue to mouse, 78% homologue to human, at Aa level 55% homologue to mouse, 66% homologue to human. In conclusion, we have revealed differences at both Na and Aa level in all four major rabbit pluripotency gene sequences in comparison to their mammalian orthologs which might partially explain difficulties in generation of rabbit iPSC capable of germline transmission. Our further goal is to apply rabbit specific pluripotency genes to reprogram somatic cells and generate iPSC more efficiently than by using mouse or human genes. A fast neutron mutant in Arabidopsis (Columbia) was identified that exhibits enhanced downy mildew (edm1) susceptibility to several Peronospora parasitica isolates, including the RPP7-diagnostic isolate Hiks1. The mutation was mapped to chr.4 and physically characterised as a 35kb deletion spanning seven genes. One of these genes restored wild-type resistance to all of the P. parasitica isolates. This gene (AtSGT1b) encodes a predicted protein that is orthologous to yeast SGT1, originally described as a key regulatory protein in centromere function and ubiquitin-mediated proteolysis. AtSGT1b contains three tatratrico-peptide repeats at the N terminus followed by a bipartite “CS” (CHORD containing Sgt) domain and an SGT specific (SGTS) domain at the carboxyl terminus. Altered expression of this gene is being investigated in Arabidopsis and Brassica olarecea to determine its potential use for crop improvement.

Skandal Dieselgate obnażył wszystkie niedoskonałości systemu homologacji pojazdów obowiązującego w Unii Europejskiej. Oprócz niedokładnych testów po stronie organów państw członkowskich i oszustw producentów problem tkwił w procedurach homologacji typu, które były nieprzejrzyste i stwarzały wiele możliwości, by nie egzekwować przepisów w interesie producentów pochodzących z państwa członkowskiego, którego organ badał zgodność pojazdów z przepisami. Nowe rozporządzenie ramowe w sprawie homologacji typu ma na celu rozwiązanie tych problemów poprzez wprowadzenie nadzoru Komisji, dodanie nadzoru rynkowego, przyznanie organom udzielającym homologacji typu większych kompetencji i koordynację działań państw członkowskich. Obszarem szczególnie rozbudowanym, a wcześniej niemal całkowicie pomijanym, są kontrole rzeczywistych pojazdów, których model został już homologowany, w celu sprawdzenia, czy rzeczywiste samochody wprowadzane na rynek rzeczywiście są zgodne z przepisami homologacyjnymi. Autor argumentuje, że rezygnacja z utworzenia unijnej agencji ds. homologacji typu pojazdów byłaby lepszym wyborem niż próba koordynacji i nadzoru tak wielu organów krajowych, a także dużo bardziej skutecznym w osiągnięciu jednolitego stosowania TAFR. Z tego powodu, pomimo usunięcia wszystkich głównych wad obecnego rozporządzenia, można je uznać za straconą szansę.

A novel chalconyl homologous series of liquid crystalline derivatives; RO - C6H4- CH : CH - COO - C6H4-CO - CH : CH - C6H4 - OC8H17 (n) (para) have been synthesized and studied with a view to correlate the thermotropic liquid crystal (LC) behavioural properties and the molecular structure of a substance. Present novel series consisted of eleven homologue members (C1 to C16) whose, only nematogenic mesomorphism commences from C3 homologue and continued upto C16 homologue in enantiotropic manner with absence of smectogenic character. Transition temperatures and textures of nematic phase were determined using an optical polarizing microscopy equipped with a heating stage (POM). Transition curve Cr-N/I behaved in normal manner. N-I transition curve exhibited odd-even effect and deviated from its normal descending tendency from and beyond C8 homologue and adopted serpantile shape from C8 to C16 homologue in deviating manner. Textures of the nematic phase are threaded or schlieren. Analytical and spectral data supported molecular structures of homologues. Thermal stability for nematic is 112.66 and the degree of mesomorphism vary from 5.00C to 44.00C at the C12 andC4 homologue respectively. It is a middle ordered melting type series, whose relative group efficiency order for nematic derived.

ABSTRACT Surface proteins are important factors in the interaction of probiotic and pathogenic bacteria with their environment or host. We performed a comparative bioinformatic analysis of four publicly available Lactobacillus genomes and the genome of Lactobacillus salivarius subsp. salivarius strain UCC118 to identify secreted proteins and those linked to the cell wall. Proteins were identified which were predicted to be anchored by WXL-binding domains, N- or C-terminal anchors, GW repeats, lipoprotein anchors, or LysM-binding domains. We identified 10 sortase-dependent surface proteins in L. salivarius UCC118, including three which are homologous to mucus-binding proteins (LSL_0152, LSL_0311, and LSL_1335), a collagen-binding protein homologue (LSL_2020b), two hypothetical proteins (LSL_1838 and LSL_1902b), an enterococcal surface protein homologue (LSL_1085), a salivary agglutinin-binding homologue (LSL_1832b), an epithelial binding protein homologue (LSL_1319), and a proteinase homologue (LSL_1774b). However, two of the genes are gene fragments and four are pseudogenes, suggesting a lack of selection for their function. Two of the 10 genes were not transcribed in vitro, and 1 gene showed a 10-fold increase in transcript level in stationary phase compared to logarithmic phase. The sortase gene was deleted, and three genes encoding sortase-dependent proteins were disrupted. The sortase mutant and one sortase-dependent protein (mucus-binding homologue) mutant showed a significant reduction in adherence to human epithelial cell lines. The genome-wide investigation of surface proteins can thus help our understanding of their roles in host interaction.

Abstract:
ABSTRACT The β-lactam resistance gene mecA was acquired by Staphylococcus aureus from an extraspecies source. The search for the possible origin of this gene has led to the identification of a close structural homologue of mecA as a native gene in the animal species Staphylococcus sciuri. Surprisingly, the overwhelming majority of S. sciuri isolates were fully susceptible to β-lactam antibiotics in spite of the ubiquitous presence of the mecA homologue in the bacteria. We now describe two unusual S. sciuri strains isolated from humans—SS-37 and SS-41—that showed resistance to methicillin associated with high rates of transcription of the mecA homologue and production of a protein resembling penicillin binding protein 2a, the gene product of S. aureus mecA. In strain SS-37 increased transcription of the mecA homologue was related to insertion of an IS256 element upstream of the structural gene, and strain SS-41 had single nucleotide alterations in the promoter region of the mecA homologue which appear to be related to up-regulation of the rate of transcription. A third methicillin-resistant human isolate of S. sciuri that carries both the native mecA homologue and a methicillin-resistant S. aureus (MRSA) type mecA, strain K3, was now shown to be unstable in the absence of drug selection, causing the segregation of antibiotic-susceptible cells accompanied by the loss of the MRSA type mecA. These observations illustrate the remarkable variety of strategies available to bacteria for acquiring mechanisms of drug resistance in the in vivo environment.

The purpose of this article is to define the Hochschild and cyclic homologies of an algebraic space $X$. We consider two definitions of Hochschild homology of $X$ and show that these definitions coincide in the case of a separated algebraic space. Moreover, we construct a shuffle product on the Hochschild homologies of $X$. Finally, we show that one has a pairing between the Hochschild homologies and cohomologies of $X$. Résumé Le but de cet article est de définir les homologies de Hochschild et les homologies cycliques d'un espace algébrique $X$. Nous considérons deux définitions de l'homologie de Hochschild et nous montrons que ces définitions sont les mêmes en cas d'un espace algébrique séparé. De plus, nous construisons un produit shuffle sur les homologies de Hochschild de $X$. Enfin, nous montrons qu'on a un appariement entre les homologies et les cohomologies de Hochschild de $X$.
