Full textThe gonadotrophic hormones are used for reproduction control in farming animals. Up to now, these hormones were extracted from animal fluids or tissues. The company CEVA Santé Animal has recently decided to produce recombinant versions of these hormones. The objective of this study was to obtain a pure and biologically active recombinant equine chorionic gonadotropin (reCG) after expression in CHO mammalian cells. The production, purification and characterization steps have been developed. CHO cells were grown in Erlenmeyer flasks with different culture media. Two media were selected based on their cell growth potency and of the amount of reCG produced. By using a bioreactor to control key parameters (temperature, pH), the production process was then optimized. The recombinant proteins that accumulated in the supernatant of the two conditions were called reCG 1 and reCG 2. A 3-steps purification process was then developed using reCG 1. Several resins and chromatographic conditions were screened in microplates. Multimodal resins were used to eliminate the main contaminants thanks to their selectivity. reCG aggregates were efficiently eliminated by a chromatographic step with an anionic resin. The overall purification process was finally validated for reCG 1 and reCG 2. Purity and yield were respectively, 98 % and 80 % for the two reCG. We verified that the in vitro and in vivo activities of reCG 1 and reCG 2 were comparable to those of the CG extracted from natural sources. The in vivo assays also confirmed previous studies showing that the degree of glycosylation of an hormone, and most notably their level of sialytation, is important for their biological activity

Full textThe equine luteinizing hormone (eLH) and the equine chorionic gonadotrophin (eCG) activate cell signaling from two distinct GPCRs in non-equine species: the LH receptor (LHR) and the FSH (Follicle stimulating Hormone) receptor (FSHR). Structural modifications of glycanic chains of eLH or conformational modification of eCG consecutive to its interaction with an antibody (Ab) both led to a modulation of the native hormone’s biological efficacy. In order to understand the molecular mechanisms induced by the 2 type of hormone modifications, we have studied their impact on 2 of the signaling pathways triggered by FSHR: the G protein-and the b-arrestin-dependent pathway. Our results demonstrate that modifications induce signaling bias. We conclude that our finding might lead the way to a new generation of gonadotrophins’ modulating drugs with promising applications in animal breeding and in Human medicine

Full textBackground. Hypospadias is the most common malformation affecting the male genitalia. Although the causes remain often unknown, endocrine, vascular and environmental factors have been implicated. However, the genetic basis is probably underestimated. Objectives. To study the genetic factors implicated in sexual differentiation. Methods. A cohort of 284 children born with hypospadias has been established since 2011. DNA was extracted from blood lymphocytes and fibroblasts obtained after a foreskin biopsy. Sanger sequencing of AR, MAMLD1 and NR5A1, SNP-array analysis, and Whole Exome Sequencing (WES) were performed. A second cohort of 1,311 pregnant women was established to evaluate an association between maternal fhCGb and hypospadias. Results. No mutation in AR and MAMLD1 was found. Non-described variations of AR, MAMLD1 and NR5A1 were identified without any splicing activity. Some polymorphisms in AR gene had a MAF significantly higher (p<0.01). A heterozygous intragenic duplication of NR5A1 was found. Pangenomic analysis included 35 anomalies (15.7%) that could be a potential cause of isolated, distal, non-hereditary hypospadias encompassing 25 candidate genes. 22 variants was classified as pathogenic on WES. Finally, a significant difference of fhCGb for severe types was identified even after adjustment for placenta dysfunction (p<0.03). Conclusion. Our hypothesis about “somatic” mutations in 3 candidate genes was not ascertained. However, pangenomic analysis found 15.7% anomalies which could be likely linked to hypospadias. The use of WES could be an attractive method for exploring further these results as we identified 22 variants in 30 cases of familial hypospadias
