Abstract:
The ability to mount an efficient immune response should be an important life-history trait as parasitism can impact upon an individual's fecundity and survival prospects, and hence its fitness. However, immune function is likely to be costly as resources must be divided between many important traits. Whilst many studies have examined host resistance to particular parasite types, fewer have considered general immune responses. Studies that have considered general immune responses tend to do so in vertebrate models. However, the complexity of the vertebrate immune system makes the examination of evolutionary aspects of immune function difficult. Using larvae of the genus Spodoptera (Lepidoptera: Noctuidae) as a model system, this study examines' genetic and phenotypic aspects of innate immunity. The aims were to assess the levels of additive genetic variation maintained in immune traits, to consider possible costs that could maintain this variation, and to assess the role of phenotypic plasticity in ameliorating those costs. A key finding of this study was that high levels of additive genetic variation were maintained in all of the measured Immune traits. Analysis of the genetic correlations between traits revealed potential trade-offs within the immune system and between immune components and body condition. In addition, it was shown that larvae living at high densities invest more in immune function than those living in solitary conditions, suggesting that larvae can minimise the costs of immune function by employing them only when the risk of pathogenesis is high.

Abstract:
Providence virus (PrV) is a member of the Tetraviridae, a family of small, positive sense, single-stranded RNA viruses, which characteristically infect the midgut tissue of heliothine larvae. PrV is the only known tetravirus that replicates in cultured insect cells. The virus comprises a monopartite genome resembling members of the genus Betatetravirus with the capsid precursor protein undergoing autoproteolytic cleavage at its C-terminus consistent with other tetravirus capsid precursor proteins. Analysis of viral cDNA predicted the presence of three potential overlapping gene products (from 5` to 3`): (1) p130, a protein of unrecognized nucleotide or amino acid homology with a 2A-like processing site at its N-terminus; (2) p104, the replicase ORF, which was found to be phylogenetically related to tombus-and umbraviruses replicases. The presence of a read-through stop signal in the p104 ORF was proposed to produce and amino terminal product with a predicted MW of 40 kDa (p40) and (3) the capsid protein precursor (81 kDa) which has two 2A-like processing sites at its N-terminus. Metabolic radiolabelling of viral translation products in persistently infected MG8 cells and in vitro translation of the individual ORFs were performed in order to analyse the expression of PrV gene products. p130 was translated with no evidence of 2A-like processing. Two products of 40 kDa and 104 kDa were translated from the p104 ORF, indicating that the read-through stop signal was likely to be functional. Finally, the capsid protein precursor ORF produced a major translation product of 68 kDa corresponding to the capsid protein precursor as well a peptide of 15 kDa that was attributed to the activity of the second 2A-like site at the N-terminus of the p81 ORF. The subcellular distribution of viral RNA (vRNA) and p40 in MG8 cells was investigated using immunofluorescence and biochemical fractionation. The results showed that p40/p104 and vRNA accumulated in polarized, punctate structures in some but not all MG8 cells and in some cases, co-localization was observed. This thesis concludes that PrV is a novel tetravirus with significant similarities plant carmolike viruses that should be re-classified at the family level.

Abstract:
The success of an invasive alien species is often attributed to the ecological advantage gained from natural enemy release. Numerous factors have been suggested as contributing to the success of Harmonia axyridis as an invasive alien species, including enemy release. This thesis studied the interactions of several parasites with H. axyridis, investigating parasite transmission, growth and virulence as well as host immune responses, thereby shedding light on the potential role of enemy release in the invasion biology of this ladybird. Benefits gained by invasive alien species from enemy release diminish if parasites of native species shift hosts to exploit the novel invader. The fungal ectoparasite Hesperomyces virescens began infecting H. axyridis shortly after it invaded the UK, probably as a result of a host shift from Adalia bipunctata. This study found a rapid increase in H. virescens prevalence over three years in London H. axyridis populations. Laboratory study showed H. virescens transmission and growth to be more efficient on A. bipunctata than the novel host. In addition, reciprocal interspecific transfers of H. virescens strains isolated from A. bipunctata and H. axyridis revealed that the infection characteristics of the fungi from these two hosts differed, suggesting strains may have diverged after the initial shift from A. bipunctata to better exploit the host from which they were derived. Laboulbenialian fungi were previously thought to have negligible impacts on host fitness. A detailed examination of H. virescens infecting H. axyridis found distinct virulence, with infections resulting in a 50% reduction in host lifespan. In addition, chronic H. virescens infection in males caused acceleration in the age-associated decline in body condition while for females, infection triggered fecundity senescence and a faster age-related decline in fertility. While their role in accelerating ageing is debated, the results presented here provide evidence that infectious diseases can drive the ageing process in this insect species. In nature, multiple parasites affecting a single host are common. The effect of co-infection on the virulence caused by two fungal infections was characterised using H. axyridis and A. bipunctata hosts. The ability of two ladybird species to defend against an acute fungal parasite, while infected with the relatively avirulent H. virescens was found to be sex-specific. While for females, the presence of co-infection did not alter the virulence seen in singly infected females, a higher mortality rate existed for co-infected males compared with those infected singly. Previously, H. virescens has been considered to be avirulent, however, this study provides evidence that this chronic fungal parasite may be important when considering the mortality associated with co-infections in the field. The invasive success of H. axyridis has, in part, been attributed to a more vigorous immune ability compared with other competitor species. Previously, field studies have shown that the prevalence of the parasitoid wasp Dinocampus coccinellae in H. axyridis is considerably lower than in the UK primary host of this wasp, Coccinella septempunctata. The extent to which the prevalence asymmetry in the field is driven by differences in host encapsulation response was tested by first comparing the encapsulation ability of C. septempunctata and H. axyridis directed against an artificial implant. Following this, the encapsulation response of D. coccinellae parasitized individuals was assessed and compared between the two host species. While encapsulation ability did not differ between the host species, and D. coccinellae did not affect the immune response of H. axyridis, wasp parasitism did alter the encapsulation ability of C. septempunctata, although it was inconsistent across sexes and populations. Overall, this thesis furthers our understanding of the fungal parasite H. virescens and its association with the notorious invader H. axyridis. The research presented here also demonstrates the use of H. axyridis as a model system in areas other than invasion ecology and furthermore, contributes to understanding the role of infectious disease in the rate of ageing. Finally, sex-specific effects were found across the chapters of this thesis, demonstrating the use of H. axyridis in the study of sex-specific effects of infections.

Abstract:
Most, if not all, animals will at some stage in their lives encounter parasites. Some of the most widespread and abundant parasites belong to the Class Trematoda. Trematodes often have a substantial negative impact on individual intermediate host ecology. In this thesis, I investigate ecological and evolutionary consequences of the interaction between the microphallid trematode, Maritrema novaezealandensis, and its intermediate snail and crustacean hosts. Parasites often show a heterogeneous spatial distribution pattern in natural animal host populations. In this study, factors determining the spatial distribution of larval trematodes in Zeacumantus subcarinatus were investigated at two spatial scales (within and among bays). The distribution of shorebird definitive hosts explained a significant amount of the variation in the distribution of trematodes among bays. However, within a bay, other factors override the effect of bird distribution. The influence of larval trematodes on reproduction, survival and population density of Z. subcarinatus was investigated using laboratory and field studies. In the laboratory, it was found that larval trematodes induce castration and mortality of Z. subcarinatus. The field study revealed that the local prevalence of trematodes had a significant negative effect on population density of Z. subcarinatus. Through castration, trematodes act as strong selective agents on snail host life history. The effect of trematodes on life history characteristics (reproductive effort, juvenile growth, size at maturity and susceptibility to trematode infections) were investigated among natural populations of Z subcarinatus. Reproductive effort was not higher for uninfected females from populations where the risk of becoming infected was high. However, offspring from those populations were significantly larger, and laboratory-reared juveniles grew significantly faster than conspecifics from other populations. In addition, size at maturity was negatively correlated with trematode prevalence across snail populations. Z. subcarinatus thus adapts to a high local risk of trematode infection by reaching maturity early, thereby increasing the chance of reproducing. The influence of M. novaezealandensis on the survial of the amphipod host, Paracalliope novizealandiae was examined using experimental infections and field observations. The experimental infections demonstrated that parasite-induced mortality was intensity-dependent. The number of M. novaezealandensis per amphipod was too low to significantly induce host mortality in the field. However, the transmission strategy of this parasite allows it to affect host populations during weather conditions ideal for trematode transmission. Trematode strategies in the second intermediate host are important to the understanding of host-parasite co-evolution and the evolution of parasite life cycles. In this study, potential density-dependent effects at the metacercarial stage on size and fecundity of in vitro adult M. novaezealandensis was examined in both experimentally infected P. novizealandiae and naturally infected Macrophthalmus hirtipes. For this purpose, a method to excyst and cultivate M. novaezealandensis metacercariae to an egg producing stage, was developed. Naturally infected M. hirtipes also harboured larval stages of three other helminths. Crowding effects in the two crustacean hosts were expressed as a decreased volume and smaller egg production of in vitro adult M. novaezealandensis. In addition, interspecific interactions among parasite species were observed in crab hosts. The work in this thesis provides evidence that M. novaezealandensis significantly and negatively affect intermediate host ecology. The heterogeneous distribution of trematodes causes differential effects among host populations with subsequent effects on the life history of snail hosts. In addition, this study demonstrates that parasites interact within their second intermediate host with possible implications for the way parasites exploit their hosts.

Abstract:
Lernaeocera branchialis (Linnaeus, 1767) is a parasitic copepod possessing a complex dual-host lifecycle. The “definitive” gadoid hosts, including Gadus morhua (Atlantic cod), Melanogrammus aeglefinus (haddock) and Merlangius merlangus (whiting), are infected by the fertilised female, which penetrates the host’s ventral aorta or bulbus arteriosus whilst undertaking extensive metamorphosis and a haematophagous lifestyle. The pathogenic effects of this activity upon the host have been well documented and mortality may occur, especially when multiple parasites are present. These negative impacts on cod, particularly juveniles, by L. branchialis have the potential to adversely affect cod aquaculture in the future, and already vulnerable wild cod stocks. This PhD project therefore, investigated the immune response of wild haddock and cultured-cod post-infection by L. branchialis, and the possible mechanisms by which the parasite modulates/evades the host’s immune response. The systemic immune response of both wild haddock and cultured-cod post-infection by L. branchialis depended on the maturation stage of the parasite, and in the former host species, upon the infection intensity. Wild haddock harbouring fully metamorphosed females showed an increase in circulating thrombocytes and a decrease in serum protein levels however; if multiple mature L. branchialis were present the haddock possessed reduced circulating monocytes, and increased circulating thrombocytes and serum anti-trypsin activity. Infection by L. branchialis was also associated with a suppressive effect on haddock serum spontaneous haemolytic activity. These responses were thought to be due to the host trying to counteract the increased damage caused by the massive increase in size and the feeding of the mature parasite, which is more pronounced when multiple parasites are present, resulting in the increase in some parameters and the ‘consumption’ of others. However, the effect of parasite-derived secretions and other pathogens due to observations on wild fish could not be discounted. The laboratory-infection of cultured-cod from two different sources was also performed in order to study the immune response over time. The two groups of cod showed differences in their immune response to L. branchialis. The first group showed suppressed respiratory burst activity of phagocytes, as the parasite reached the early penella sub-stage, whilst no suppression in phagocyte respiratory burst activity was found in the second group. The parasite was found to migrate along the afferent branchial artery of the cod where a thrombus formed and was present throughout its migration into the ventral aorta. At 14 d post-infection, leukocytes expressing Interleukin 8 mRNA were observed within the free-flowing blood at the periphery of the organising thrombus within the lumen of the ventral aorta. This was speculated to aid the recruitment and activation of leukocytes to the site, and the maturation and neovascularisation of granulation tissue. The infection of the second group subsided with the death of the parasite, and none of the parasites metamorphosed past the early penella sub-stage. The live parasites infecting the first group of cod did not possess IgM or complement component C3 binding on their cuticle, however, both IgM and C3 binding occurred on the dead parasites in the second infection trial. This may highlight the importance of these opsonins and the cytotoxic effect of phagocytes in the elimination of L. branchialis by some cod. However, the first infection was terminated as the parasite reached the early penella sub-stage due to a loss of stock cod prior to the study, so the long-term success of the infection can not be concluded. Therefore, the immune response to infection needs to be determined over the entire metamorphosis of L. branchialis to determine whether the infection was successful or not, and preferably in populations with varying susceptibility to L. branchialis. This will not be possible without further studies into the resistance of different stocks of cultured-cod. Many arthropod parasites, such as ticks and salmon lice, have been previously documented to produce pharmacologically active secretions, aiding host invasion and parasite feeding, preventing the host immune response from working effectively against the parasite, all aimed at improving survival of the parasite. Therefore, the effects of the secretory/excretory products (SEPs) produced during the initial infective stage and by the mature, fully metamorphosed female on the immune response of cultured-cod in vitro, and the location of exocrine glands associated with the oral region of the parasite were investigated. The SEPs from the infective stage of the parasite were found not to affect the intracellular hydrogen peroxide (H2O2) production of phagocytes. The practical difficulties in collecting large quantities of the SEPs from the infective stage meant that their effects could not be tested on the other host immune parameters studied. The SEPs from fully metamorphosed female L. branchialis, however, had a number of suppressive effects on the host immune response in vitro including: 1) suppression of the intracellular production of cytotoxic H2O2 during the respiratory burst of phagocytic leukocytes post-PMA stimulation, 2) suppression of the production of macrophage activating factor by leukocytes with a priming effect on naïve phagocyte function, and 3) suppression of the chemo-attraction ‘power’ of zymosan activated cod serum, i.e. anaphylatoxin activity, on head kidney-derived leukocytes. These effects were dose-dependent, and highlight the capacity of L. branchialis to suppress its host’s innate immune response at the local feeding area. Further work is required to establish the mechanisms by which the parasite-derived SEPs suppress these host immune parameters, and to identify which molecules produced by the parasite are responsible. The correlation between these in vitro results, and systemic immune parameters measured from laboratory-infected Atlantic cod and wild infected haddock are discussed. Host immuno-modulation by other arthropod parasites is mediated by pharmacologically active secretions produced by exocrine glands. Therefore, the exocrine glands of the infective and fully metamorphosed female L. branchialis were also investigated in order to identify those that might be responsible for the secretion of host-modifying products. Adult female exocrine glands were mapped using diaminobenzidine (DAB), most commonly known to stain peroxidases and catalases. These compounds are known to be involved in the neutralisation of harmful free radicals which are released during the respiratory burst and tissue damage. Such products may therefore be important protective secretory components at the site of feeding / infection. Exocrine glands were located in the infective stage associated with the oral region, one pair termed the anterior gland complex (AGC), and the other pair extending either side of the oral cone termed the circum-oral glands (CG). These were further investigated using light microscopy and transmission electron microcopy. The AGC and CGs possessed multi-component secretions and they possessed secretory vesicles, abundant and highly active rough endoplasmic reticulum and Golgi apparatus suggesting that protein is an important component of the secretory products. These glands were also observed in the fully metamorphosed females where they had increased in size within the cephalothorax post-metamorphosis. It is hoped that the identification of these glandular structures, which are thought to secrete within the local vicinity of the oral cone, will aid future studies regarding the identification and secretion kinetics of parasite-derived molecules during the infection and feeding process.

An experimental study of laboratory populations of the stored-products moth, Cadra cautella (Lepidoptera: Phycitidae) and its larval parasitoid, Venturia canescens (Hymenoptera: Ichneumonidae) identified and quantified density- and age-dependent demographic characteristics of the host-parasitoid system. Host imago longevity and fecundity depended on larval weight at pupation. Observed effects of C. cautella larval competition for food on larval mortality, stage duration, and weight at pupation were successfully captured in a mathematical model. Host larval age significantly influenced inter-stage cannibalism and susceptibility to mortality resulting from parasitoid oviposition wounds. Both larval parasitoid developmental rates and adult parasitoid attack rates depended on host larval age. Long-term population experiments of host and host-parasitoid populations revealed that host populations fluctuated with a period slightly in excess of host generation time and that parasitoid populations were in synchrony with host populations.

Abstract:
Lernaea spp. was identified as a widespread problem in West Malaysia and various aspects of the parasite were studied. A survey of the distribution of Lernaea spp. in West Malaysia revealed its presence in all the 8 government owned fish breeding stations, 104 privately owned farms and 4 out of 5 consignments of imported fingerlings. The Lernaea spp. present in these ponds were identified as L. piscinae and L. cyprinacea "Asian" form (=b.elegans) and its morpha forms, L. ctenopharyngodonis and L.guadrinucifera. The life cycles of L. cyprinacea and L. piscinae were determined under laboratory conditions. Infection of ~. auratus with the offspring of L. cyprinacea "Asian" form produced adult females similar to the maternal form and another form identified as b. ctenopharyngodonis. A. nobilis, infected with L. cyprinacea "Asian" form and L. piscinae produced only species identical to the maternal form. A high degree of polymorphism was revealed and was further investigated. Morphometric studies on the larval stages and adult female parasites were investigated and reliable characteristics for the identification of Lernaea is discussed. The distribution frequency of b. cyprinacea on 3 host species and L. piscinae on A. nobilis in aquarium tanks was goodness of tested for/fit with the theoretical negative binomial distribution. The bases of fins were found to be the preferred site of infection for both species. studies on growth performance of A. nobilis infected with L. piscinae in ponds, revealed a significant reduction in Specific Growth Rates. Infected fish had a higher mortality than uninfected fish. A decrease in parasite infection on the body surface after 3 months was associated with its appearance in the eye. A challenge infection did not establish and the fish were suspected of being immune. Histopathological studies showed a typical inflammatory response with the formation of a granuloma. Eosinophilic granular cells, lymphocytes, and club cells which were identified in the hosts immune to the infection, were believed to play an important role in the rejection of parasites.

From simple beginnings, when only one host was required, numerous parasitic organisms have evolved complex life-cycles involving two or more host species. For example, trematode parasites reproduce in vertebrates, their definitive host, but their current life cycle also typically involves two intermediate hosts that were added during the course of evolution. Vertebrates are often considered to be the ancestral hosts of trematodes although other scenarios exist. While multi-host life cycles are observed in distantly related groups of parasites, their evolution remains largely unexplored. In trematodes, while recent phylogenetic studies have shed light on the sequence along which the different hosts were incorporated in the cycle, conditions that favoured the evolution of such complex life cycles can only be hypothesized. However, one opportunity to understand the force shaping the evolution of complex life cycles is provided by the few trematode species in which the classical three-host cycle facultatively reverts to a shorter cycle (i.e. life cycle abbreviation). In this study, the effects of different environmental factors on the life history strategy of the trematode Coitocaecum parvum were investigated using laboratory and field studies. C. parvum is able to abbreviate its life cycle from three to two hosts by maturing early (i.e. progenesis) and producing eggs inside the second intermediate host; both life history strategies occur simultaneously in C. parvum populations. Environmental factors such as predator densities should strongly influence parasite life history strategies. In fact, this study shows that laboratory reared Coitocaecum parvum adopt preferentially the normal three-host cycle when chemical cues from the definitive host are added to their environment, while the shorter cycle is favoured when these cues are absent. However, in nature, multiple environmental factors are likely to be perceived by parasites. Consequently, C. parvum�s ability to adapt its developmental strategy to definitive host densities may be confounded by the complex combination of various environmental parameters. Within-host competition between parasites sharing a common host is also likely to influence individual life history strategies. Parasites could then use alternative life strategies to adaptively respond to intraspecific and interspecific competition. Indeed, this study found that C. parvum preferentially adopts the abbreviated cycle in the presence of competitors. However, in interspecific competition, C. parvum�s strategy also depends upon the competitor species, possibly influenced by the other species� transmission route. Furthermore, intensity of intraspecific competition proved to constrain C. parvum�s ability to use the abbreviated life cycle. Finally, genetic relatedness between co-infecting C. parvum individuals seems to affect parasite life strategy through kin selection: closely related individuals are more likely to adopt the same developmental strategy, when they share a host, than unrelated ones. C. parvum individuals adopting the abbreviated cycle are enclosed within a cyst in their intermediate host and must produce eggs by self-fertilization, the most severe case of inbreeding. It was hypothesized that their offspring would have reduced fitness due to inbreeding depression, therefore selecting against the shorter cycle. However, this study found no difference in the survival and infection success of offspring produced through the abbreviated and normal cycles. Furthermore, no evidence for a genetic basis of life cycle abbreviation was detected: the same proportion of offspring from both reproductive strategies adopted the shorter life cycle. The work in this thesis provides evidence that although life cycle abbreviation provides Coitocaecum parvum with a viable alternative life strategy, numerous factors promote or restrict the adoption of this strategy. While this life history strategy has no detectable effect on parasite fitness, both environmental parameters and within-host competition affect C. parvum life-history strategies, alternatively selecting for either the shorter or normal life cycle. Overall, the complexity of the parasite environment could maintain both developmental strategies in C. parvum populations and, on a broader scale, could have influenced the evolution of complex life cycles in parasites.

Abstract:
This thesis investigates aspects of the community ecology of rabbit parasites with particular emphasis upon the gut helminths, utilising a 23 (later extended to 26) year time series of rabbits and their parasites. A clearer understanding of parasite communities can lead to more effective biological control strategies. Rabbits are regarded as a serious pest species throughout Europe and the Antipodes and the use of the myxomatosis virus, as a biological control agent, has already been tried and failed. However, a clearer picture of the parasite community may offer future possibilities for control. Additionally, the rabbit is a good model for other grazing species, as it carries a similar gut helminth community. Drug resistance is an increasing problem in a wide range of parasites. A clearer appreciation of parasite communities could also aid in the search for effective and environmentally sound pathogen control strategies (e.g. via cross immunity or competition with benign species). Theoretical models have revealed the importance of aggregation to the stability of the host parasite relationship, to parasite evolution and to interspecific parasite interactions. A number of models have considered the effect of varying aggregation upon these dynamics with differing outcomes to those where aggregation was a fixed parameter. Here the stability of the distribution for each of the rabbit helminths was examined using Taylor's power law. The analyses revealed that aggregation was not a stable parameter but varied with month, year, host sex, host age, and host myxomatosis status. Evidence for the existence of interspecific parasite interactions in natural systems has been equivocal. Factors influencing parasite intensity were evaluated for the gut helminth. A network of potential interactions between the parasites was revealed. Only month was shown to be of greater influence on the community. Following, from the above analyses, a community model was constructed which incorporated both seasonal forcing and interspecific parasite interactions, with interaction mediated via host immunity. One unexpected emergent property was an interaction between the seasonality and the immune decay rate with slower immune decay resulting in a shift of the immune response out of phase with the species against which it was produced. The model was also used to assess the potential effects of two control strategies, an anticestodal and a single species vaccine. The vaccine had greater effects on the whole community than the anticestodal because of the immune- mediated interactions. The host is also an integral part of the community as the parasite dynamics are linked with that of their host. Therefore an assessment of the parasites' impact upon host condition and fecundity was also undertaken. This revealed a variety of positive and negative associations between the parasites and their host, with potential implications for future host control strategies. This study has shown that ignoring parasite-parasite or parasite-host interactions and interactions of both the host and the parasite with the external environment, could result in a poor description of the community dynamics. Such complexities need to be considered and incorporated into theory if future control strategies for either host or parasites are to be effective.

Most organisms are rarely infected with just a single species of parasite and are usually simultaneously infected with a range of species. Thus, the parasite fauna of a host represents an entire community composed of multiple individuals from many different species. In nature, it is within the host that parasites can encounter conspecifics and individuals from other species. As in any ecosystem, while such interactions between parasites can be antagonistic due to competition or conflicting interests, association between different species can also be beneficial. In this thesis, I investigated patterns of associations between parasites in the New Zealand cockle Austrovenus stutchburyi through a combination of descriptive and experimental studies employing both standard ecological field techniques and molecular biology methods. It was found that the presence and infection intensity of various parasites species are not independent of each other. Among cockles, an association was found between two trematode taxa, i.e. between the infection intensity by foot-encysting echinostomes and the metacercariae of Gymnophallus sp. It was also found that the presence of the parasitic copepod Pseudomyicola spinosus was associated with greater infection intensity by the echinostomes but not Gymnophallus sp. While it was postulated that the positive association between the echinostomes and Gymnophallus sp. was due to the latter�s preference to infect cockles that are stranded on the sediment surface as a result of heavy echinostome metacercariae burden in their foot, a field experiment found that Gymnophallus cercariae did not preferentially infect cockles that have been forced to remain above the sediment surface as opposed to those that were forced to remain buried. Meanwhile, the two species of echinostomes known to encyst in the cockle�s foot, Acanthoparyphium sp. and Curtuteria australis, were found to represent cryptic species complexes. The presence of such cryptic species means that it is possible that some potential interspecific interactions are overlooked. A study of the population structure of Gymnophallus sp. found that each cockle contains multiple genetically distinct individuals and that clonal individuals rarely co-occur in the same cockle. This adds to the growing body of evidence suggesting that in addition to acting as a means of reaching the definitive host, the second intermediate host also acts to promote genetic diversity by accumulating cercariae shed by multiple first intermediate hosts in the environment. An experimental infection study conducted with Curtuteria australis cercariae deriving from singly-infected first intermediate hosts revealed that different clonal lineages varied with respect to their contribution to host manipulation. It was found that while certain lineages have a preference for host manipulation, others tend to adopt a "hitch-hiker"-type life history strategy. However, this genetic predisposition was also found to be a phenotypically plastic trait, as the presence of a higher number of manipulators seems to encourage newly-arrived cercariae to become manipulators, regardless of clonal lineage. This thesis provides evidence that patterns of interactions can affect various aspects of parasite life history. Apart from host condition, parasites can also be affected by other parasites. Studying the dynamics of mixed infections can provide informative insights for evolutionary and ecological research.

The ecology of Protocalliphora parasitism was studied for a two-year period in two species of cavity nesting passerine birds breeding in southwestern Quebec, the eastern bluebird (Sialia sialis Linnaeus) and the tree swallow (Tachycineta bicolor Vieillot). Both species occupied next boxes in open field habitats. Nests were parasitized primarily by Protocalliphora sialia Shannon and Dobroscky.
The relationship between the number of Protocalliphora larvae per nest and selected nesting variables was examine for the two species. The variables included hatching date of the host young, nest material volume, number of fledglings, and percent canopy cover above the nest box. In bluebirds, the number of larvae per nest was significantly correlated with nest material volume in 1989 and number of fledglings in 1990. In tree swallows, the number of larvae per nest was significantly correlated with hatching date and number of fledglings in 1989.
Information on reproduction in Protocalliphora was obtained. Gravid flies were captured in active nests up to the last end of the second week of the nestling period. P. sialia was determined to be at last bivoltine through mark, release, and recapture studies. It was determine that nulliparous P. sialia can complete egg development within 9 or 10 days after eclosion. The practicality of a new Protocalliphora trapping technique and a mark, release, and recapture technique was demonstrated. (Abstract shortened by UMI.)

The intracellular parasite Trichinella is a genus in phylum Nematoda that contains six named species including Trichinella spiralis and Trichinella pseudospiralis. These parasites infect a large variety of wild and domestic animals, human beings and a few species of birds. The parasitic strategies and the pathological effects on the host between trichinella spiralis and Trichinella pseudospiralis are quite different. The purpose of this study is to gain an understanding of the physiological, immunological and pathological differences between these two species of Trichinella using infections in the mouse as a model. In the course of this research I have attempted to answer the following questions: A) Is cortisol a factor in the differences of the host response to Trichinella spiralis or Trichinella pseudospiralis? B) Are there differences in leukocyte response in the peripheral blood of Trichinella infected mice? C) Are there differences in the up-regulation or down-regulation of cell surface molecules on leukocytes in the spleens of Trichinella infected mice? D) Is there a difference in the degree of muscle damage (as measured by creatine phosphokinase) when infections by the two species of Trichinella are compared? E) Are there differences in angiogenesis and collagen deposition in Trichinella infected mice and are these differences related to cortisol? F) Is nitric oxide a component in parasite killing and are there differences in nitric oxide production in host mice when the two species of Trichinella are compared? My research has shown that there are significant differences in the parasitic strategies and pathological consequences in mice infected with one or the other of the two species of Trichinella. The two species appear to generate different immune and inflammatory responses from the host. Trichinella pseudospiralis is much less damaging to the host, generates a very different peripheral blood response, stimulates the production of substantially greater levels of serum cortisol, generates a significantly different profile in cytokine production presents a very different cell surface antigen profile and does not produce a collagen nurse cell or generate an angiogenic response when compared to T spiralis. In addition, I have shown a role for nitric oxide in parasite killing and a role for serum cortisol in larval survival. I have also shown that cortisol has no role in either collagen deposition or the angiogenic response in Balb/c mice under the experimental conditions detailed here.

Natural selection should favour host defenses that reduce a host’s exposure to parasites or reduce their negative effects. One strategy that resolves the substantial costs of host immunity and/or tolerance is to avoid infective stages altogether. For fish, behavioural avoidance is well-known for defense against aquatic predators, but it is poorly known for defense against parasites. I used a model system that is amenable to experimental manipulation to test the behavioural avoidance hypothesis for fathead minnows exposed to the larvae of two of their common flatworm parasites. First, I showed that minnows exposed to a liver encysting trematode, Ornithodiplostomum sp. showed an increase in lipid peroxidation, an indicator of oxidative stress, persisting through worm development. Three lines of evidence provided support for the behavioural avoidance hypothesis. First, shoal area decreased in groups of minnows exposed to O. ptychocheilus cercariae compared to those exposed to cues from other aquatic threats. Second, average worm numbers were 50 % lower in fish confined to artificial shoals compared to non-shoaling minnows, indicating that shoaling reduces risk of exposure. The third experiment showed that minnows within the centre of shoal reduced their risk of infection by 67%. Taken together, these results demonstrate a cost of trematode infection on minnows, that minnows can detect infective larvae within the water column, and that social living reduces a hosts’ risk of exposure.

The rate of recombination and parasite load of species of anuran from Peruvian Amazonia were examined to test the idea that recombination functions to diversify progeny in order to resist the continual counter-adaptation of parasites. The anurans were found to be hosts to over 32 species of macroparasite, as well as a wide variety of protistan and moneran parasites. It was found that a combination of three variables (diet, habitat, abundance), together with host body size accounted for a substantial proportion of the variation in mean parasite richness and parasite species distribution among host species. The relationship between parasites and ecology was found to be independent of host phylogeny. The demonstration of substantial environmental heterogeneity in parasitization predicates that a positive relationship between parasite richness and recombination should be found in this case. This prediction was supported by the data: highly parasitized species of anuran had higher rates of recombination. This is the first study to demonstrate a direct positive relationship between recombination (rather than sex) and parasites.

A comunidade parasitária do lagarto Tropidurus torquatus em uma área de campo rupestre localizada no estado de Minas Gerais, sudeste do Brasil, foi estudada. Dos 110 hospedeiros analisados, 92 (83,6%) estavam infectados. Foram encontradas cinco espécies de helmintos, três de nematóide: Physaloptera lutzi, Parapharyngodon sp. e Oswaldofilaria sp., uma espécie de cestóide e um cistacanto não identificados. Apenas as espécies de nematóides apresentaram dados suficientes para analises ecológicas, sendo P. lutzi a espécie mais prevalente (67,3%) e com maior intensidade parasitária (4,86±4,85). Somente para Oswaldofilaria sp. foram encontradas diferenças significativas na prevalência e intensidade parasitária entre hospedeiros fêmeas e machos, esses últimos apresentando os maiores valores. A intensidade parasitária total e de P. lutzi também diferiram entre os sexos de hospedeiros com machos apresentando os maiores valores. O tamanho corporal do hospedeiro correlacionou-se de forma positiva com a intensidade de infecção de todas as espécies de nematóides. A sazonalidade local não influenciou de forma significativa a estrutura da comunidade componente de parasitos estudada. Aspectos do hospedeiro como dieta, dimorfismo sexual e comportamento (territorialiasmo e forrageamento) representaram importantes fatores na determinação da estrutura dessa comunidade de helmintos. De maneira geral, a comunidade parasitária apresentou baixa riqueza de espécies e um caráter não interativo, características comuns em comunidades parasitárias de répteis como hospedeiros.
The helminth community of the lizard Tropidurus torquatus from a rocky outcrop located in Minas Gerais state, southeast Brazil, was studied. Ninety two of the one hundred ten individuals examined (83,6%) harbored helminths. A total of five helminth species were found, tree nematodes: Physaloptera lutzi, Parapharyngodon sp. and Oswaldofilaria sp., one cestode species and one acanthocaphalan cystacanth not identified. Only nematode species had sufficient data to perform ecological analysis with Physaloptera lutzi showing the highest prevalence (67,3%) and mean intensity (4,86±4,85). Prevalence and intensity of infection between males and females hosts differed only for Oswaldofilaria sp., with males showing the highest values. Total and P. lutzi intensity of infection were different among males and females hosts, with males showing the highest values. The host body size was positively related to intensity of infection for all nematode species. Local seasonality had no influence in the helminth community structure. Host diet, sexual dimorphism and behavior (territorialism, forage strategy) represented important factors in the structuring of this parasite community. In general lines, the helminth community was species poor and non interactive, representing a typical community structure found in herps hosts.

La manipulation du comportement de l'hôte est une stratégie couramment utilisée chez les organismes parasites, et ce afin de favoriser leur transmission et/ou leur survie. La compréhension d'une telle stratégie est au cœur de plusieurs disciplines appliquées de la parasitologie, telles que l'épidémiologie et la médecine, mais est aussi d'une grande importance d'un point de vue écologique et évolutif. Dans le cadre de cette thèse, notre objectif était d'améliorer l'état des connaissances sur un type de manipulation encore très peu connu : la manipulation garde du corps, une stratégie initialement décrite chez les insectes parasitoïdes. À la suite de leur développement larvaire, certaines guêpes parasitoïdes (Braconidae) ont la capacité d'usurper le comportement de leur hôte et l'obligent ainsi à se comporter comme un véritable garde du corps pour protéger leur cocon des ennemis naturels. Basé sur le système associant la guêpe parasitoïde Dinocampus coccinellae et un de ses hôtes la coccinelle maculée Coleomegilla maculata, le présent travail aborde cinq aspects particuliers de la manipulation parasitaire : i) Efficacité de la protection par un garde du corps et coûts liés à une telle stratégie, ii) Contraintes énergétiques pour les parasites manipulateurs, iii) Diversité et l'évolution de la manipulation garde du corps, iv) Survie des hôtes à la manipulation par un parasitoïde et v) Effets de la nutrition de l'hôte sur une association hôte–parasitoïde manipulateur. Nos résultats apportent plusieurs éléments de réponse quant à l'écologie et l'évolution de la manipulation garde du corps, et notamment en ce qui a trait aux coûts énergétiques associés à une telle stratégie ainsi qu'au rôle de la ressource dans cette étroite association. Host behavioral manipulation, a widespread strategy among parasitic organisms, has evolved to favor the transmission and/or survival of parasites. Understanding such a strategy is at the heart of applied aspects of parasitology, such as epidemiology and medicine, but is also important for evolutionary and ecological reasons. The aim of this study was to improve our knowledge on a category of manipulation that remains poorly documented: the bodyguard manipulation, initially described on parasitoid insects. Following their larval development, certain parasitic wasps (Braconidae) are able to usurp the behavior of their host and force them to serve as bodyguards to protect parasitoid cocoon against natural enemies. Studying the association between the parasitic wasp Dinocampus coccinellae and one of its host the spotted lady beetle Coleomegilla maculata, this work is structured in five axes: i) Efficiency of the bodyguard protection and associated costs, ii) Energetic constraints for manipulative parasites, iii) Diversity and evolution of bodyguard manipulation, iv) Host survival from parasitism and v) Host nutrition effects on a host–manipulative parasitoid association. Our results provide original findings about the ecology and evolution of the bodyguard manipulation, for instance the energetic costs associated with host manipulation or the role of host nutrition in this type of association. New perspectives are identified in order to stimulate future research in this area

Some parasites may modify the behavior of their hosts. Altered behaviors may: 1) benefit the host in that they defend against the pathogen, 2) benefit the pathogen and represent manipulations of the host response, and 3) benefit neither the host or the pathogen and simply be a product of the host response to infection. In this thesis I examine four host/parasite systems. For each system, I explore host/parasite behavioral interactions, and examine them with regard to selective pressures that may be acting on both the host and the parasite. I test the Hamilton and Zuk hypothese in 26 species of lizards. I find an inverse relationship between a lizard species' brightness and parasite prevalence. My result lend credence to criticisms of the Hamilton and Zuk Hypothesis. If infection does occur, animals may alter their behavior to impair the growth and reproduction of the parasite. To test this prediction, I examine behavioral thermoregulation in two strains of the snail Biomphalaria glabrata, one resistant to, and one susceptible to, the parasite Schistosoma mansoni. The preferred temperature of infected snails drops five weeks after exposure to the parasite. I propose the hypothesis that pathogen-induced host defense responses result in altered host behaviors and enhanced predation. In particular, I examine the effects of the acute phase response (a physiological response whose symptoms include fever, reduced activity and malaise) on antipredatory behavior in bullfrog (Rana catesbeiana) tadpoles. This host response is associated with the preliminary stages of infection with many pathogens yet its behavioral effects have received little attention. I find that the stereotypical effects of the acute phase response can lead to increased predation. I suggest that altered behaviors may afford some parasites a potential pathway to their next host. I examine the behavioral effects of a yeast, Candida spp., a single-host parasite species in its natural host, the red-legged frog (Rana aurora). Infected tadpoles exhibit the same behavioral modifications that are noted in bacteria injected bullfrog tadpoles. These results suggest that some altered behaviors may occur due to a host response to infection and not due to parasitic manipulation.

Abstract:
Trygonorrhina fasciata (Rhinobatidae) specimens naturally infected by three monogenean species were captured and maintained in marine aquaria to promote a continuous parasite load. Monogenean eggs recovered from aquaria provided larvae for descriptions and life history experiments. I describe the adult, larva and post-larval development of a new species of hexabothriid, Branchotenthes octohamatus, from the gills. This is the first monogenean larva described with only eight hooklets. This character may be useful to help resolve problematic relationships within the Hexabothriidae and offers insight into more general hypotheses about relationships within the Monogenea. I also redescribe the adult of Calicotyle australis (Monocotylidae) from the cloaca and describe the larva. The number and arrangement of larval ciliated epidermal cells and sensilla was revealed using silver nitrate. I redescribe Pseudoleptobothrium aptychotremae (Microbothriidae) adults from the skin of T. fasciata, representing a new host and locality record. Larval anatomy and post-larval development are also documented. The presence of six needle-like spicules in the larval haptor is confirmed, supporting an earlier theory that spicules are ancestral vestiges. My studies revealed three different egg hatching, host finding strategies and larval ‘types’. Branchotenthes octohamatus has a ‘sit-and-wait’ strategy, entirely dependent on mechanical disturbance to stimulate eggs to hatch. Larvae are unciliated, cannot swim, lack pigmented eyespots and show no photo-response but may survive for more than two days after hatching at 22ºC. In contrast, eggs of C. australis hatch spontaneously with a strong diurnal rhythm in the first few hours of daylight when exposed to a LD12:12 illumination regime. Larvae are ciliated and can swim, have pigmented eyespots, are photo-positive and can remain active and survive for up to 24 h after hatching at 22ºC. Eggs of P. aptychotremae may have a ‘bet-hedging’ strategy. Some eggs hatch spontaneously and rhythmically in an LD12:12 regime during the last few hours of daylight but their low hatching success rate suggests that other eggs may require a different cue provided by the host. Larvae are ciliated, can swim, lack pigmented eyespots, show no photo-response and remain active for only a few hours at 22ºC. Experiments using the fluorescent dye, 5(6)-carboxyfluorescein diacetate N-succinimidyl ester (CFSE) revealed B. octohamatus on gills of T. fasciata within 30 min of exposure to the host. This provides strong evidence that larvae invade the gills directly via the host’s inhalant respiratory current and do not migrate after initial attachment elsewhere. Five rhinobatid species (Aptychotrema vincentiana, T. fasciata, Trygonorrhina sp. A, A. rostrata and Rhinobatos typus), with overlapping distributions spanning west, south and east Australian coastal waters were surveyed for monogeneans at four locations between Fremantle, Western Australia and Stradbroke Island, Queensland. Genetic homogeneity, using the mitochrondrial gene Cytochrome b (cytb) and the nuclear marker, Elongation factor-1 alpha (EF1a), was observed for all Branchotenthes and Calicotyle specimens irrespective of collection locality or rhinobatid species. Genetic homogeneity was observed for Pseudoleptobothrium specimens collected in western and southern Australia. However, local genetic heterogeneity was apparent among Pseudoleptobothrium specimens collected from two sympatric host species in New South Wales. Analyses revealed a highly divergent clade, indicating a morphologically cryptic, ancestral species.

Abstract:
Ceratomyxa shasta infects salmonids in the Pacific Northwest (PNW) of North America, occasionally causing losses in wild and captive populations. Host-specific parasite genotypes (O, I, II, III) were previously characterized molecularly using markers in the ribosomal DNA and phenotypically by type host in the Klamath River, CA/OR. This thesis sough to re-examine the composition of the parasite population elsewhere in the PNW and to further evaluate the host specificity of each genotype. I surveyed salmonids native to the PNW primarily from the Fraser, the Columbia and the Sacramento River basins. I also conducted sentinel studies on the Willamette and Deschutes Rivers that exposed native and non-native salmonids to parasite populations above and below migration barriers. These studies expanded upon the known host range of each genotype: O was specific to rainbow/steelhead (Oncorhynchus mykiss) and coastal cutthroat trout (O. clarkii); I was specific with Chinook salmon (O. tshawytscha); II was non-specific, being detected in six species, but predominating in coho (O. kitsuch), chum (O. keta), and pink (O. gorbuscha) salmon; and III was also non-specific infecting many native and non-native salmonids. Hosts infected with genotype III were considered “adequate” if myxospore development occurred, and included brook char (Salvelinus fontinalis), brown (Salmo trutta), rainbow, cutthroat, and steelhead trout; but Chinook, kokanee/sockeye (O. nerka) and Atlantic salmon (Salmon salar) were less suitable. The distribution, life history, and phylogenetic traits of each salmonid host are factors that potentially explain the host specificity and the spatial and temporal patterns of each genotype. Data collected in this thesis provide evidence that host-specific C. shasta coevolved with Pacific salmonids, adapting unique host-parasite relationships over time.

Abstract:
The freshwater polychaete Manayunkia speciosa was identified as an obligate host of the salmonid parasite Ceratomyxa shasta in 1997, prompting increased research on the small benthic invertebrate. Ceratomyxa shasta infection in fish can cause mortality, and presents a disease risk for both hatchery and wild salmon and trout. Ceratomyxa shasta is endemic to rivers of the Pacific Northwest, and its effects have been particularly well documented in the Klamath River, Oregon and California. One option for managing C. shasta impacts is by decreasing densities of M. speciosa through habitat manipulation, thus decreasing amplification of the parasite. The Klamath River is regulated by irrigation and hydropower dams, thus manipulating the hydrograph to destabilize habitat is a possibility. Decreasing habitat through flow manipulation requires a thorough understanding of the hydraulic environment of polychaete habitat, and how that environment changes with discharge. This thesis proposes an influence diagram of physical variables driving M. speciosa density, and investigates several of them. Samples were collected for enumerating M. speciosa density from nine sites in the Klamath River over 15 months, and seasonal density changes were examined, as were the relationships between density and hydraulic variables (depth, average velocity, substrate size, Reynolds number, Froude number). Density increased directly with depth and inversely with velocity, and was greater on small (silt, sand) and large (boulder, bedrock) substrate relative to medium substrate (gravel, cobble). Density was highest in the summer (July, August, September), and there was evidence that summer densities were influenced by spring discharges through the mechanism of substrate mobilization. Differences in infection prevalence among seasons and habitats were also investigated; however, very low overall incidence of infection limited any conclusions. Based on these results, it is recommended that habitat modeling for management of M. speciosa populations include a habitat stability component that incorporates s whether peak discharge the previous year surpassed a stability threshold.
