A restriction map of the housefly mitochondrial DNA was constructed. The putative arrangement of the major coding regions was determined by two methods. In the first, Drosophila clones of known gene content were hybridized to housefly restriction fragments on Southern blots. In the second, rare restriction sites and clusters of restriction sites were used to align the housefly restriction map with that of Drosophila yakuba.
