Title: Fluidity and Solidity in Marilynne Robinson’s Housekeeping

Author: Linda Lindqvist

The purpose of this essay is to show that fluidity and solidity constitute a central tension on all levels in Housekeeping, and how this tension leads to a choice of either a fluid or a solid lifestyle and view of the world. I focus on fluidity and solidity in gender roles, in memories, in dreams, in nature, and in different perceptions of reality. By taking a closer look at Ruth’s first-person narration (seeing fluidity as not resisting deformation, while solidity resists deformation), we find that the characters in Housekeeping have fluid and solid traits, but that there is no reality that allows mixed manifestations of these. This results in repression of either fluidity or solidity, which creates tension and feelings of loss. Ruth chooses a fluid lifestyle, thus her memories and dreams become mixed with her present, and this also leads her to become a transient, outside gender roles and traditional small town society. Transience in this novel questions all distinctions conventionally made between dream and reality; male and female. In conclusion, this essay highlights how tension between fluidity and solidity is generated on all levels in Housekeeping, and how this leads to either fluid or solid lifestyles since the characters follow a cultural code that dictates a choice between them.

This thesis is divided into two parts. In the first half, I present the current research results from corporate householding knowledge exploration. They include the categories of corporate householding problems, the related research in data quality and family household, commercial approaches to corporate householding problems, and the various areas where corporate householding applies. In the second half of the thesis, I present the design and implementation of a technical solution to an important type of corporate householding problem - entity aggregation - using a motivational example in account consolidation. The proposed technical approach uses extended COntext INterchange (COIN) technology to manage and process corporate householding knowledge.

En la actualidad el entorno globalizado y constantemente cambiante de los negocios ha llevado a las organizaciones a desarrollar sus servicios rápidamente para poder ser más competitivos en el mercado. Sin embargo, la necesidad de ir probando las diferentes funcionalidades de un servicio en marcha e ir testeando, para acabar ofreciendo una solución final, es considerada una tarea complicada y de muy alto costo. La literatura estudiada muestra que se han generado modelos de diseño de servicios con enfoque ágil, pero orientados a corporaciones ya establecidas o con diseños muy robustos que presuponen altos costos; a la fecha de desarrollo de esta investigación no se han encontrado modelos con una orientación hacia nuevos emprendimientos o startups de servicio. En el presente artículo se propone un modelo ágil para el diseño de servicios, el cual destaca principalmente la agilidad, los bajos costos y la iteración incremental en el desarrollo. El modelo ha sido puesto en marcha en el sector de trabajo del hogar en Lima, Perú, donde se generó una mejora en la satisfacción de los clientes de 2.02 a 3.56.

Myokardinfarkt) mit Hilfe des Ribonuklease Protektion Assay (RPA) und der TaqMan PCR bestimmt. Die linksventrikuläre ANP-mRNA-Expression war in allen untersuchten Modellen unabhängig von der angewendeten Detektionsmethode erhöht. Die mRNA-Expression der Housekeeping Gene mit Hilfe des RPA bestimmt, war in allen untersuchten Modellen im Vergleich zu den Kontrollen unverändert (GAPDH: kleiner Shunt: 105.1+-7.4, großer Shunt: 105.2+-6.8, MI: 88.4+-3.7; 18SrRNA: kleiner Shunt: 110.7+-8.2, großer Shunt: 104.4+-8.9, MI: 107.5+-12.0; Cyclophilin: kleiner Shunt: 96.4+-7.9, großer Shunt: 112.9+-4.9, MI: 95.7+-13.8; PBGD: kleiner Shunt: 81.9+-6.3, großer Shunt: 83.7+-4.7, MI: 79.8+-9.7; % Kontrolle). The purpose of this study was to identify an appropriate left ventricular mRNA as internal standard in gene expression analysis in cardiac hypertrophy and heart failure in the rat. Expression levels of GAPDH, 18SrRNA, Cyclophilin and porphobilinogen desaminase (PBGD) were measured four weeks after induction of either cardiac hypertrophy (small aortocaval shunt) or heart failure (large aortocaval shunt or myocardial infarction) using Ribonuclease protection assay (RPA) and TaqMan PCR. The left ventricular expression of ANP mRNA was increased in all these experimental models independently of the used method. VEZF1 is a DNA-binding transcription factor that is highly conserved in vertebrates. Human VEZF1 has recently been found to interact with transcriptionally active gene promoters and erythroid specific enhancers. It is unclear how the broadly expressed VEZF1 interacts with enhancers in a cell type-specific manner or what functions it plays at these elements. This work begins to address the cell-type specific roles of VEZF1 by creating loss of function models in a human erythroid cell line.

In this essay, I examine the role of material culture in Marilynne Robinson's novel Housekeeping (1980) to understand how the prominent presence of material culture introduces complex questions about the relationships among objects, reality, and the self. By recognizing objects' fluidity of meaning, Housekeeping offers its characters a way to see their individuality and conceptions of reality in a similar state of flux. Significantly, it is in the act of recognizing that the socially accepted uses of objects are not necessarily "natural" parts of existence, and, like elements of the natural world, the meanings and uses of these items are susceptible to change and decay that an individual is able to recognize that the self is similarly fluid and moldable, which creates room for both imagination and for the possibility of change.

Abstract:

In this thesis project housekeeping genes in differentiating human embryonic stem cells were investigated. Housekeeping genes are involved in basic functions in the cells and are assumed to be expressed at relatively constant levels across different cell types and experimental conditions. Based on these features, housekeeping genes are frequently used as controls in calibration of gene expression data. Commonly used housekeeping genes in somatic tissues have shown to vary notably in human embryonic stem cells and are therefore inappropriate as reference genes in this unique cell type. In the present work a novel set of gene expression data obtained by profiling of undifferentiated and early differentiating cardiac cells, was analyzed. Stably expressed genes were identified in this data set and were subsequently intersected with a previously proposed set of 292 stable genes in human embryonic stem cells. A resulting set of 73 genes show stability across all investigated cell lines and experimental conditions. These genes are suggested as a more reliable set of reference genes in differentiating human embryonic stem cells than frequently used housekeeping genes in somatic tissue. In addition, a novel set of 20 genes was identified as very stably expressed during the differentiation towards the cardiac lineage. After further validation of stability with RT-PCR, these genes could be useful as controls in studies of human embryonic stem cells that differentiate towards the cardiac lineage.

A caracterização de promotores ubíquos, tecido-específicos ou induzidos sob determinada condição é importante para a produção e liberação comercial de plantas geneticamente modificadas. Este trabalho apresenta a identificação e caracterização de promotores de genes de café (Coffea arabica), uma cultura de grande importância sócio-econômica. Foram selecionados 41 genes candidatos com padrão de expressão constitutivo, fruto-específico ou relacionados com o mecanismo de defesa, através de dados da literatura e da análise in silico em bancos de ESTs disponíveis. A expressão constitutiva ou específica foi confirmada através de PCR quantitativa para somente 10 dos genes analizados. Os experimentos foram realizados em 5 diferentes tecidos/órgãos (raiz, caule, folha, flor e fruto) ou em plantas tratadas com o fungo biotrófico Hemileia vastatrix. Regiões promotoras de 4 genes foram isoladas através de genome walking: CaGAPDH (glyceraldehyde-3-phosphate dehydrogenase como candidato constitutivo), CaAN2 (anthocyanin 2 específico de fruto), CaPR1b e CaNPR1 (basic form of Pathogenesis-Related protein type 1 e Nonexpressor of PR genes, correspondentes a genes induzidos mediante a presença de agentes patogênicos). As regiões cis-reguladoras foram caracterizadas in silico e a funcionalidade dos promotores foi avaliada in planta através da expressão do gene repórter uidA em experimentos de transformação transiente em plantas de tabaco ou tomate. Além do interesse fundamental para a compreensão dos mecanismos de regulação gênica em eucariotos, os resultados desta pesquisa são importantes para o desenvolvimento de plantas transgênicas, apresentando também potencial em programas de melhoramento genético do cafeeiro. The choice of promoter, to confer constitutive, spatial and/or temporal transgene expression, is important in plant biotechnology applications. This study presents the identification and characterization of different gene promoters in coffee (Coffea arabica), a species of major socioeconomic characteristics. Forty one constitutive, fruit-specific or pathogen defense-related genes were identified following literature data or in silico analyses in available EST databases. The expression levels of these genes were verified by quantitative PCR and only 10 genes presented a constitutive or specific expression condition. The expression levels assays were carried out in 5 coffee organs/tissues (root, stem, leaves, flowers and fruits) or in coffee plants challenged with Hemileia vastatrix. The promoter region of 4 genes were isolated and characterized in silico: CaGAPDH (glyceraldehyde- 3-phosphate dehydrogenase as inner control), CaAN2 (anthocyanin 2 fruitspecific), CaPR1b and CaNPR1 (basic form of Pathogenesis-Related protein type 1 and Nonexpressor of PR genes, both pathogen-inducible). The functional analyses of the DNA sequence upstream of these genes were assessed with regard to the uidA reporter gene, via Agrobacterium-mediated transient expression assay in tobacco or tomato plants. The characterization of promoters is not only an approach towards coffee breeding programs, but also provides fundamental data for understanding the mechanisms regulating gene expression in perennial plants

Abstract:
This thesis project analyzes the influence of the built environment on subjectivity in late twentieth-century American fiction. Marilynne Robinson's Housekeeping, I contend, uses domestic architecture to inform the concept of female-as-subject, specifically through additive and subtractive construction techniques. These techniques influence public and private definitions of female, as they are present at the level of the individual house as well as the level of the town of Fingerbone as a cohesive unit. Jeffrey Eugenides's The Virgin Suicides interrogates ethnic subjectivity in a suburban setting. A strict grid orders the suburban landscape which, although visually unifying a group of ethnically diverse individuals, ultimately isolates the subjects--down to the scale of the single-family home--as it encourages collective memory, ethnic repression, and the standardization of perspective. In these novels, architecture is an agent which negotiates subjectivity and the vehicle that communicates ideology through and with the material reality of these homes. Reading architecture helps us understand who these characters are and how the built environment shapes the way they have come to define themselves and others as individuals, which ultimately allows for the subversion of such definitions.

Abstract: The choice of promoter, to confer constitutive, spatial and/or temporal transgene expression, is important in plant biotechnology applications. This study presents the identification and characterization of different gene promoters in coffee (Coffea arabica), a species of major socioeconomic characteristics. Forty one constitutive, fruit-specific or pathogen defense-related genes were identified following literature data or in silico analyses in available EST databases. The expression levels of these genes were verified by quantitative PCR and only 10 genes presented a constitutive or specific expression condition. The expression levels assays were carried out in 5 coffee organs/tissues (root, stem, leaves, flowers and fruits) or in coffee plants challenged with Hemileia vastatrix. The promoter region of 4 genes were isolated and characterized in silico: CaGAPDH (glyceraldehyde- 3-phosphate dehydrogenase as inner control), CaAN2 (anthocyanin 2 fruitspecific), CaPR1b and CaNPR1 (basic form of Pathogenesis-Related protein type 1 and Nonexpressor of PR genes, both pathogen-inducible). The functional analyses of the DNA sequence upstream of these genes were assessed with regard to the uidA reporter gene, via Agrobacterium-mediated transient expression assay in tobacco or tomato plants. The characterization of promoters is not only an approach towards coffee breeding programs, but also provides fundamental data for understanding the mechanisms regulating gene expression in perennial plants

Abstract:
The genetic information is identical within the organism but the mechanisms by which different cell types achieve specialized functions interpreting the same set of instructions is not completely understood. It is now increasingly accepted that the combination of different genomic elements, both promoters and enhancers, favors the recruitment of different TFs, which in turn promotes the assembly of different pre-initiation complexes, guaranteeing heterogeneity in transcriptional outputs across different tissues. Nevertheless, the cis-regulatory elements and the transcriptional rules that control and maintain the expression of constitutively active genes are still poorly characterized. Specifically, whether the constitutive activity of promoters and enhancers relies on entirely distinct or instead shared regulators is unknown. By dissecting the cis-regulatory repertoire of macrophages, we found that the ELF subfamily of ETS proteins selectively bound within 60 bp from the transcription start sites of highly active housekeeping genes. ELFs also bound constitutively active, but not poised macrophage-specific enhancers and promoters. The role of ELFs in promoting constitutive transcription is suggested by multiple evidences: ELF sites enabled transcriptional activation by endogenous and minimal synthetic promoters; ELF recruitment was stabilized by the transcriptional machinery, and ELF proteins mediated recruitment of transcriptional and chromatin regulators to core promoters. These data indicate that a distinct subfamily of ETS proteins imparts high transcriptional activity to a broad range of housekeeping and tissue-specific cis-regulatory elements, which is consistent with the role of an ETS family ancestor in core promoter regulation in a lower eukaryote.

Danos causados por doenças virais estão entre os principais fatores limítrofes da produtividade do feijão-caupi. Em condições de estresse os fatores de transcrição (TFs) participam ativamente das etapas iniciais do processo de detecção e sinalização, regulando a expressão de vários grupos gênicos. Neste sentido, objetivou-se caracterizar TFs da família MYB e avaliar sua expressão diferencial frente à infecção viral, bem como determinar genes de referência (RGs) para normalização dos dados em RT-qPCR sob diferentes condições de estresse e controles. Por meio de análises in silico no banco NordEST, identificamos no transcriptoma do feijão-caupi um total de 86 candidatos a TF MYB, classificados em três subfamílias. A análise dos componentes estruturais do domínio R2R3-MYB permitiu observar a conservação dos aminoácidos característicos desta classe protéica em feijão-caupi. Por sua vez, o padrão de distribuição em pseudocromossomos de Phaseolus vulgaris indicou que genes MYB sofreram duplicações em tandem e intercromossomais, contribuindo para sua expansão no feijão-caupi. A análise filogenética formou 18 subclados, apoiados pela estrutura dos motivos funcionais da região C-terminal das proteínas. Das tags SuperSAGE diferencialmente expressas sob infecção viral, três foram reguladas positivamente, indicando a participação de candidatos MYB na resposta ao estresse viral. Dos sete RGs avaliados em três conjuntos experimentais, β-tubulina, Skip16 e Act2/7 + Skip16 foram as melhores combinações para seca, salinidade e vírus, respectivamente, podendo ser recomendados como normalizadores para estudos de expressão diferencial em feijão-caupi. Neste estudo identificamos a maior família de TFs em plantas observando sua participação ativa na resposta de defesa contra estresses em feijão-caupi.
Damages caused by viral diseases are among the main factors affecting the cowpea productivity. Under stress conditions, transcription factors (TFs) actively participate in the initial stages of the detection and signaling process by regulating the expression of various gene groups. In this sense, the objective of the present work was to characterize members of the MYB TF-family and evaluate their differential expression under viral infection, also determining reference genes (RGs) for data normalization in RT-qPCR under different stress and control conditions. Using in silico approaches to analyze the NordEST databank, a total of 86 MYB TF-candidates could be identified, being classified into three subfamilies. An analysis of the structural components of the R2R3-MYB domain allowed the identification of conserved amino acid residues of this protein class in cowpea. In turn, the MYB distribution pattern in the pseudochromosomes of Phaseolus vulgaris indicated that MYB members suffered in tandem and interchromosomal duplications, contributing to their expansion in cowpea. Phylogenetic analysis formed 18 subclades, supported by structural features of motifs in the C-terminal region of the protein. Of differentially expressed SuperSAGE tags under viral infection, three were upregulated, indicating the involvement of MYB candidates in response to viral stress. Considering the seven tested RGs under three experimental conditions, β-tubulin, Skip16 and Act2/7 + Skip16 were the best combinations for drought, salinity and viruses, respectively, recommended as normalizer genes in studies of differential expression in cowpea. In the present work we identified members of the largest family of TFs in plants observing their active participation in defense against stress response in cowpea.

Os bastonetes Gram positivos irregulares (BGPIs) compõem um grupo de espécies bacterianas com ampla diversidade fenotípica e que podem estar presente no meio ambiente, na microbiota humana e de animais. A identificação acurada de BGPIs em nível de gênero e espécie empregando métodos bioquímicos convencionais é bastante limitada, sendo recomendado, portanto, o uso de técnicas moleculares. No presente estudo, foram identificadas amostras de BGPIs oriundas de espécimes clínicos de humanos, de produtos farmacêuticos e de áreas limpas através da análise de sequencias do gene 16S rRNA e de outros genes conservados (housekeeping genes). Os resultados obtidos pelo sequenciamento dos genes 16S rRNA e rpoB demonstraram C. striatum multi-resistente (MDR) como responsável por surto epidêmico em ambiente hospitalar da cidade do Rio de Janeiro. Quinze cepas de C. striatum foram isoladas em cultura pura a partir de secreção traqueal de pacientes adultos submetidos a procedimentos de entubação endotraqueal. A análise por eletroforese em gel de campo pulsado (PFGE) indicou a presença de quatro perfis moleculares, incluindo dois clones relacionados com cepas MDR (PFGE I e II). Os dados demonstram a predominância de PFGE I entre cepas MDR isoladas de unidades de terapia intensiva e enfermarias cirúrgicas. Uma potencial ligação causal entre a morte e a infecção por C. striatum MDR (PFGE tipos I e II) foi observada em cinco casos. Adicionalmente, acreditamos que este seja o primeiro estudo de identificação de espécies de Nocardia relacionadas com infecções humanas pela análise da sequencia multilocus (MLSA) no Brasil. Diferente dos dados observados na literatura (1970 a 2013) e obtidos pelos testes fenotípicos convencionais, a caracterização molecular de quatro lócus (gyrB-16S-secA1-hsp65) permitiu a identificação das espécies N. nova, N. cyriacigeorgica, N. asiatica e N. exalbida/gamkensis relacionadas com quadros de nocardiose em humanos. Cepas de N. nova isoladas de diferentes materiais clínicos de um único paciente apresentaram padrões de susceptibilidade antimicrobianos idênticos e dois perfis PFGE, indicando a possibilidade de quadros de co-infecção por N. nova em humanos. Em outra etapa da investigação, amostras de BGPIs obtidos de ambientes de salas limpas que não puderam ser identificadas por critérios convencionais foram submetidas a análise da sequência do gene 16S rRNA e caracterizadas 95,83% em nível de gênero e 35,42% em espécies. Para gêneros mais encontrados no estudo, foram analisados os genes rpoB e recA de dezessete cepas de Microbacterium e utilizado o MLSA para a identificação de sete cepas identificadas como Streptomyces. Os ensaios permitiram a identificação de três cepas de Microbacterium e de uma única amostra de Streptomyces ao nível de espécie. A análise da sequencia do gene rpoB também se mostrou eficaz na identificação de espécies de cepas de Corynebacterium. Finalmente, para as cepas ambientais pertencentes à classe Actinobacteria os dados morfológicos, bioquímicos e genotípicos permitiram documentar a cepa 3117BRRJ como representante de uma nova espécie do gênero Nocardioides, para o qual o nome Nocardioides brasiliensis sp. nov. e as cepas 3712BRRJ e 3371BRRJ como representante de um novo gênero e espécie para o qual o nome Guaraldella brasiliensis nov. foi proposto.
Irregular Gram positive rods (coryneform bacteria; IGPRs) comprise a group of species that has a wide phenotypic diversity and makes the conventional identification limited and that may be present in the environment, humans and animals hosts. In order to provide further accurate identification of these microorganisms in a genus and species terms it is recommended the use of molecular methods. In this study, we analyzed the 16S rRNA gene sequences and other conserved genes (housekeeping) in order to elucidate the identification of clinical isolates, pharmaceutical and clean room areas. We have documented a nosocomial outbreak caused by multidrug-resistant (MDR) C. striatum in Rio de Janeiro. C. striatum identification was confirmed by 16S rRNA and rpoB gene sequencing. C. striatum was mostly isolated in pure culture from tracheal aspirates of adults undergoing endotracheal intubation procedures. The analysis by pulsed-field gel electrophoresis (PFGE) indicated the presence of four PFGE profiles, including two related clones of MDR strains (PFGE I and II). The data demonstrated the predominance of PFGE I, mostly isolated from patients of intensive care units and surgical wards. A potential causal link between death and MDR C. striatum (PFGE I and II) infection was observed in five cases. We also performed the identification of Nocardia species of human infections by multilocus sequence analysis (MLSA) and characterized their antimicrobial and phenotypic profiles. An overview from 1970 to 2013 of the case reports on Nocardia species related to human infections, except mycetomas, in Brazil, was also accomplished. Molecular characterization by four-locus (gyrB-16S-secA1-hsp65) has provided the species identification N. nova, N. cyriacigeorgica, N. asiatica and N. exalbida/gamkensis. N. nova strains isolated from different clinical specimens of one patient showed identical antimicrobial susceptibility patterns. PFGE analysis performed to determine the genetic relatedness of these N. nova strains two distinct profiles, which were designated A and B. This is the first report on identification of Nocardia species by MLSA in Brazil. The IGPRs obtained in clean room environments that could not be identified by conventional criteria were studied by 16S rRNA gene sequence analysis that allowed the identification of 95.83% at genus level and of 35.42% at the species level. The most common genus found in the clean room environments were Microbacterium and Streptomyces. The analysis of rpoB and recA genes sequence of seventeen Microbacterium strains contributed to the identification of three strains at species level. MLSA of seven Streptomyces strains identified a single sample at the species level. Moreover, the rpoB gene sequence analysis was effective in identifying Corynebacterium strains at species level. A new genus and species were found among clean room environmental strains belonging to the Actinobacteria class. Based on the morphological, genotypic and biochemical data presented in this study the 3117BRRJ strain represent a novel specie of the genus Nocardioides, for which the name Nocardioides brasiliensis sp. nov. was proposed and the 3712BRRJ and 3371BRRJ represent a new genus and species for which the name Guaraldella brasiliensis nov. were proposed.

This thesis explores and analyses the practices, role and significance of housekeeping in the royal bedchambers at Hampton Court in the period between 1689 and 1737. Specifically, it seeks to chart contemporary practices of care and maintenance and to situate this work within the context of the rooms that lay at the heart of the late-Stuart and early-Hanoverian courts. A broad range of primary sources have been used to inform the study, in particular the records of the Lord Chamberlain and the Great Wardrobe. These archives are considered in relation to the material culture of the interiors at Hampton Court and are placed within the broader framework of social and political histories of the period. The thesis is divided into two main parts, the first of which explores the context of the royal bedchambers at Hampton Court. Starting with the premise that the practices of housekeeping were shaped by the specific environment in which they operated, it provides an exposition of the dual significance of this area of the palace as a space for magnificent court ceremony and as a retreat for the rituals of royal private life. Developing these findings, the second part of the study discusses housekeeping practices and the servants who undertook this work. In particular, it focuses on the identity, role and status of the lower ranking female servants of the bedchamber department, the Keeper of the Standing Wardrobe and the Privy Lodgings and the craftsmen of the Great Wardrobe. Throughout the discussion of these individuals, and the practices of care and maintenance, is framed by an analysis of the motivations for good housekeeping at court, and the meanings that were ascribed to this work. The research contained within this thesis contributes to our knowledge of the royal bedchambers at Hampton Court, in a key phase of the palace's history. by offering a more complete picture of how these rooms were inhabited by domestic servants as well as the monarch. New light is also shed on the many long periods when Hampton Court lay empty and the spheres of activity that took place in the absence of the court. The findings of this thesis demonstrate the significant role played by housekeeping as an essential underpinning for the use of Hampton Court as a royal home and as a splendid place of court.

One purpose of our study was to investigate if equality in work time and parental leave in a relationship affect the time that parents spend with their children. Our first hypothesis was that increased equality in work time and parental leave leads to a reduction of the parents’ total time with their children, ie the children spend more hours in pre-school and begin pre-school earlier. In order to test our hypothesis, we investigated the relationship between the woman’s and the man’s work time and parental leave and the time that the children spend in pre-school as well as at what age they began pre-school by using two surveys. Our study showed a significant coherence between both working women’s work time and the children’s time in pre-school as well as women’s parental leave and the children’s age at start in pre-school. We did not find any corresponding coherence for men. This indicates that it is the women’s work situation which, to a large degree, determines the children’s time in pre-school and at what age they start pre-school. Hence, increased equality in work life, meaning that women work more, probably has the consequence that the children spend more time in pre-school and also start pre-school earlier. The second purpose of our study was to investigate parents’ own experience of the equality in their relationship, compared to their actual equality regarding work time, parental leave, housekeeping, time with their children and own time. Our second hypothesis was that couples feel much more equal than their actual division of work time, parental leave, housekeeping, time with the children and own time indicates. We found some coherence between experienced equality and especially division of work time and housekeeping. Our study showed, however, that most couples feel that they have equality, despite an unequal division of work time and parental leave.
Ett syfte med vår studie var att undersöka huruvida jämställdhet i arbetstid och föräldraledighet i ett parförhållande påverkar den tid föräldrar tillbringar med sina barn. Vår första hypotes var att ökad jämställdhet i arbetstid och föräldraledighet leder till att föräldrars totala tid med barnen minskar, dvs. att barnen tillbringar fler timmar i förskolan och inskolas tidigare. För att testa denna hypotes undersökte vi, med hjälp av två enkätstudier, sambanden mellan kvinnans och mannens arbetstid och föräldraledighet, och den tid som barnen tillbringar i förskolan samt vid vilken ålder de skolades in i förskolan. Våra studier visade ett signifikant samband mellan såväl yrkesarbetande kvinnors arbetstid och barnens antal timmar i förskolan som mellan kvinnors föräldraledighet och barnens ålder vid förskolestart. Motsvarande samband för männen saknades. Detta indikerar således att det är kvinnornas arbetssituation som till betydande del avgör barnens närvarotid i och ålder vid start i förskolan. Ökad jämställdhet i arbetslivet, innebärande att kvinnor arbetar mer, leder sannolikt till att barnen tillbringar mer tid i förskolan och inskolas tidigare. Ett andra syfte med vår studie var att undersöka föräldrars egen upplevelse av jämställdhet i sin parrelation, i förhållande till deras jämställdhet i arbetstid, föräldraledighet, fördelning av hemarbete, tid med barnen och egen tid. Vår andra hypotes var att par upplever sig som betydligt mer jämställda än vad fördelning av arbete, föräldraledighet, hemarbete, tid med barnen och egen tid indikerar. Vi fann i vår studie att det finns ett visst samband mellan upplevd jämställdhet och framförallt fördelning av arbetstid och hemarbete. De flesta par upplever sig dock som jämställda, trots en relativt ojämlik fördelning av arbetstid och föräldraledighet.

Abstract:
Bacteria have the ability to sense different environmental signals. When an environmental stress is detected, bacteria rapidly adjust their gene expression profile to be able to survive and thrive. The transduction of such environmental signals often requires the coordinated involvement of several factors that constitute complex regulatory networks. Hence, depending on the combination of signals, a unique gene expression profile required to adapt to the specific stress conditions is generated. Proteins are the best-known regulatory factors. However, non-proteinaceous molecules are also important in signal-responsive control of bacterial gene expression. Alarmones are low molecular weight non-proteinaceous regulatory factors which can characteristically be rapidly turned-over to mediate instant changes in gene expression. One such alarmone is the modified nucleotide ppGpp, which directly binds to RNA polymerase to alter its activity. The levels of this alarmone are expected to rapidly increase in response to any environmental stress that result in slow proliferation. DksA, a putative ppGpp co-regulator that likewise directly targets RNA polymerase, has been suggested to be required for both the positive and negative regulation mediated by ppGpp in Escherichia coli. This thesis describes dissection of the role of ppGpp and DksA on transcriptional regulation, primarily using the fim genetic determinant that encodes for the type 1 fimbriae. Type 1 fimbriae are involved in adhesion to abiotic surface and initial adhesion/invasion of bladder cells, as well as in biofilm formation. We found that ppGpp regulates phase variation by increasing the sub-population of cells that express the fimbriae. The effect of ppGpp was ultimately traced to its role in transcription of the fimB gene that encodes a recombinase involved in the phase variation process (paper 1). In contrast, we unexpectedly found that lack of DksA causes an increase, rather than a decrease, in transcription from the fimB P2 promoter in vivo. However, in vitro transcription studies demonstrated that ppGpp and DksA, both independently and co-dependently, stimulate transcription from the fimB P2 promoter. These seemingly contradictory results from the in vivo and in vitro transcriptional studies were shown to be, at least in part, a consequence of the increased association of Gre-factors with RNA polymerase that can occur in the absence of DksA in vivo (paper 2). The results outlined above have implications for the role of ppGpp and/or DksA in global gene expression. Using gene expression profile (microarray analysis) during the transition from logarithmic to stationary phase of E. coli, we found that while most of the genes regulated by ppGpp and DksA are regulated in the same direction by the two factors, many were not. In addition to the fim genes, genes involved in flagella functioning, taxis responses, and a few genes encoding different transport systems are also differentially regulated in ppGpp- and DksA-deficient strains in vivo. Our results clearly indicate that the effect of deficiencies in ppGpp and DksA is far more complex than phenotypic similarity of the corresponding mutants anticipated by the proposed concerted action of ppGpp and DksA on gene expression (paper 2 & 3).
