Full textCannabidiol (CBD) has several therapeutic properties such as antiinflammatory, analgesic, anticonvulsivant and anticompulsive. These multiple pharmacological effects occur by different mechanisms. However, CBD exhibits low bioavailability, which may compromise its clinical use. Thus, the development of CBD analogues, more powerful and/or better pharmacokinetic profile would be interest. Here, we evaluated the effects of a fluorinated derivative of CBD, HU-474, comparing its effects with those induced by CBD. Male Swiss mice were submitted to animal models predictive to anti-compulsive (marble burying test) and antinociceptive drugs (hot plate, abdominal writhing and inflammatory hyperalgesia test). To study the mechanisms involved in their effects were used antagonist of the cannabinoid receptor CB1(AM251) and CB2 (AM630). We also evaluated whether HU-474 would induce the classic effects (cannabinoid tetrad) observed after the administration of CB1 receptors agonists. The cannabinoid tetrad is characterized by hypolocomotion, hypothermia, catalepsy, and antinociception. The possible antinociceptive effects and cannabinoid tetrad induced by CBD and HU-474 were compared with those induced by WIN55,212-2, a CB1/2 receptor agonist. CBD (30 and 60mg/kg), and HU-474 (10mg/kg) decreased the number of buried marble, an effect compared with fluoxetine an attenuated by AM251 and AM630. As expected, WIN55,212-2 induced the cannabinoid tetrad, an effect that was not observed after CBD or HU-474 administration. In the hot plate test, HU-474 (30mg/kg) and WIN55,212-2 (5mg/kg) induced antinociceptive effect. Pretreatment with AM251 and AM640 attenuated the effects induced by HU-474 and WIN55,212-2. In the abdominal writhing test induced by acetic acid, CBD (30 and 90mg/kg), HU-474 (30mg/kg) and WIN55,212-2 (3 and 5mg/kg) induced antinociceptive effects characterized by a reduction in the number of writhing. Pretreatment with AM251 only attenuated the effect of WIN55,212-2, but not of the other compounds, while AM630 didn\"t attenuated the effect of any of the tested compounds. In an inflammatory hyperalgesia model induced by carrageenan, CBD (30 and 90mg/kg), HU-474 (3, 10 and 30mg/kg) and WIN55,212-2 (1mg/kg) decreased the intensity of mechanical hyperalgesia measured by electronic von Frey method. The effects of all compounds were attenuated by the pretreatment with AM251 and AM630. These results indicate that HU-474 exhibits anti-compulsive effects similar to CBD, but at lower doses, through a mechanism dependent of the activation of CB1 and CB2 receptors. Furthermore, HU-474 showed antinociceptive properties in all tests at similar or lower doses than CBD. The antinociceptive effects of the three compounds tested were dependent on the activation of CB1 and CB2 receptors, excepted to the writhing test, where the effects of CBD and HU-474 were not attenuated by any of the two antagonists tested. Moreover, unlike WIN55,212-2, CBD and HU- 474 didn\'t induce cannabinoid tetrad. These results showed more significant effects of HU-474, indicating that the addition of fluoride improved the pharmacological profile of CBD. Furthermore, the results with pretreatment with the antagonist AM251 and AM630 allow us to suggest that these effects involve the activation of CB1 and CB2 receptors. Thus, this new compound could be a therapeutical alternative for the treatment of obsessive-compulsive disorder and acute pain in lower doses than CBD

Full textPost-transcriptional regulation plays pivotal roles from assembly to function of the nervous system. In metazoans, the ELAV/Hu genes constitute a conserved multigene family of pan-neuronal RNA-binding protein involved in post-transcriptional regulation. The elav (embryonic lethal abnormal visual system) gene is the first described family member, it encodes a vital protein and is restricted to dipterans. Elucidating the functions of the more ancient Drosophila paralogs, fne (found in neurons) and rbp9 (RNA-binding-protein-9), has been the focus of this work, because of (1) the close relationship of these genes to the vertebrate orthologs, (2) the availability of viable null mutants.In collaboration with the laboratory of Brian Oliver, NIH, USA, we used RNA-Seq to analyze the head transcriptomes of fne- mutants and a wild type reference strain (WT). The RNA-Seq data was searched for differences in transcript levels using the programs Cuffdiff and DESeq. Alternative splicing was examined using a suite of programs called Spanki (SPlicing ANalysis Kit), whose development we participated in. Spanki utilizes only sequence reads spanning exon-junctions to compare pairs of mutually exclusive alternative splicing events.Initial analyses included sex-specific comparisons in WT transcriptomes. Somatic sexual determination and behavior in Drosophila are under the control of a well characterized genetic cascade initiated by Sxl (Sex lethal), but the targets downstream of this canonical pathway remain largely unknown. As expected, the genes Sxl, tra (transformer), msl2 (male-specific lethal gene), dsx (doublesex) and fru (fruitless), which belong to the canonical sex-determination/dosage compensation pathways were identified by our analyses. In addition, new genes whose transcript expression levels differ between the sexes were identified and validated by qPCR. Further, I obtained evidence for previously undocumented roles of tra and tra2 (transformer 2) in the male soma. Finally, a sex-biased alternative splicing event was identified in fne, whose regulation is independent of tra or tra2, but dependent upon Sxl. This makes of fne the first Sxl-dependent, tra/tra2-independent target identified in a sex determination/differentiation pathway, previously been predicted to exist based upon other studies.The data was analyzed to identify functional networks controlled by fne. I found that it affects the expression of several genes at the level of transcript expression and alternative splicing involved in synaptic functions. They include of unc-13, n-syb (neuronal Synaptobrevin), tomosyn, brp (bruchpilot) and Clc (Clathrin light chain). Further, genetic interactions between fne and shi (shibire) or nrg (neuroglian), which also possess synaptic functions, reveal synthetic lethality in the double mutants. In addition, fne mutant males display a fruitless-like chaining behavior, also consistent with a function at the synapse. Thus the fne gene links post-transcriptional regulation to synaptic function and behavior.Finally, I used an evolutionary approach to distinguish newly evolved functions, i.e. specific to fne or rbp9, from those that are shared and thus more likely to be ancestral. Molecular, anatomical and behavioral approaches in parallel analyses of fne and rbp9, show that they possess both gene-specific and overlapping functions. The latter is evident from synthetic lethality in early adulthood of double mutants. My results distinguish different gene categories with respect to their regulation: genes whose expression is affected (1) only by fne (2) only by rbp9 (3) both by fne and rbp9 in redundant or synergistic ways. Finally, I showed that male-male interactions dramatically differ between fne versus rbp9 mutants, revealing the emergence of a new (or loss of an ancestral) function in behavior

This thesis addresses the topic of geopolitical power calculations from the Chinese perspective. A unique aspect of China's assessment of this subject is the use of an analytical tool called "Comprehensive National Power" (CNP) which aims to measure the power of countries and to rank them according to the results found. This Chinese method evaluation, in most of its variations, emphasizes political power as the basis of state power, as well as considering that these units are responsible for the configuration and definition of the International System, not the opposite the other way around. By doing so, it visualizes international relations from a different parameter of Western mainstream approaches, which are mostly based on material power and the power of structure constraint. This dissertation deals with this matter by contextualizing the reader from the initial theoretical foundations on state power and power in the Chinese context, in the presentation of the various CNP versions in China, and, finally, using the analysis method of Yan Xuetong (2008;2011) to evaluate the evolution of China's CNP in the Hu Jintao period (2003-2013). In this way, the thesis presents the relations between China's action - in the established time interval - and the CNP, aiming to identify how the country uses this in the assessment of the International System.
Esta dissertação aborda o tema de cálculos de poder geopolítico a partir da perspectiva chinesa. Um aspecto singular da avaliação da China a respeito deste tema é a utilização de uma ferramenta analítica chamada “Comprehensive National Power” (CNP) que objetiva mensurar o poder dos países e hierarquizá-los a partir dos resultados encontrados. Este método de avaliação chinês, na maioria de suas variações, enfatiza o poder político como a base do poder de um Estado, além de considerar os que estas unidades são responsáveis pela configuração e definição do Sistema Internacional, não o movimento contrário. Desta maneira visualiza as relações internacionais a partir de um parâmetro diferente das abordagens ocidentais de mainstream, baseadas no poder material e no poder do constrangimento da estrutura. A presente dissertação aborda esta matéria contextualizando o leitor desde os fundamentos teóricos iniciais sobre poder e poder estatal no contexto chinês, na apresentação das diferentes versões do CNP da China, e, por fim, utilizando o método de análise de Yan Xuetong (2008; 2011) para avaliar a evolução do CNP da China no período Hu Jintao (2003-2013). Desse modo, o trabalho apresenta as relações entre a ação da China no interregno temporal estabelecido e o CNP, objetivando identificar de que maneira o país utiliza desta ferramenta em sua avaliação do Sistema Internacional.

Abstract This research aims to examine Tsung-Hsien Hu’s pirate-resisting actions in Ming Dynasty. From his behavior style we can analyze how he established human resource and handled sophisticated relationships in officialdom via adept political skills. Through Tsung-Hsien Hu’s participating in the pirate-resisting war, his operations and contributions in this war can be revealed. The pirates were roughly annihilated because of a series of his key strategies; therefore, Tsung-Hsien Hu is an important figure of the pirate-resisting war development in Ming Dynasty. Facing pirates’ huge domination and Ming Dynasty’s weak military strength after taking over as the governor of Jei-Chian, Tsung-Hsien Hu employed strategies and tactics to gradually destroy two prevailing branches of pirates reined in by Shi-Hai and Wang-Chi, which was an impressive contribution. To eliminate pirates as his goal, Tsung-Hsien Hu behaved without precise ethics, gaining criticism for his risk-taking characteristics. With the then government, however, controlled by Yan-Song and the emperor Ming Shizone emotionally unpredictable and unfathomable, Tsung-Hsien Hu was quite fitting into this post—He played well his role at right time and right place with good relationship with the emperor to facilitate war progress; besides, he could judge people’s worth and know how to best employ them, making his subordinates willing to render loyal service to him. That Tsung-Hsein Hu’s life ended in a tragedy, wherein he exhibited great capability in undergoing a variety of extreme experiences, arose form his not minding trifle formalities, his handling matters at his discretion and rotten life. Besides his personal characteristics, political environment had also a huge impact on his behavior patterns. Though dealing with Ming Shizone and chaotic dynasty, Tsung-Hsien Hu proceeded with military victory. He deserves a reasonable historic evaluation from people without considering his contentious characters.

The invention of paper, with it’s wide range of uses, is a significant contribution to human beings. In aesthetics, paper begins to show it’s magnificent beauty gradually, turning into countless amazing works of art. In particular, various paper-carving techniques display different wonderful colors. In the past, we mostly appreciated the fine arts in a static way, presented in writing, flat, or only using a single material. As paper carving evolved, it became more plastic and diverse. Now, it is presented as games and animations. It is both static and dynamic ways. Therefore, with the creator’s efforts, paper carving has great potential. In recent years, with the development of science and technology, the invention of tablet computers prompted the transformation from traditional painting to electronic painting. In the traditional story-painting form, we only used paper carving to create illustrations. In electronic painting, however, we are using paper carving less to tell a story. Therefore, I initiated an idea: to research how to create a paper carving art with electronic painting, making it more flexible, vivid, and more meta-faceted. This research studies the characteristics and methods of the traditional paper carving art from both nationwide and global; it also analyzes the different modes of interactive operation in electronic drawing. The result of this research is not only regarding the painting of illustrations and paper carving characteristics using an innovative combination of different media, but, more importantly it can push the art field to develop into new areas and trends. Further promotion of this thought enables more talents to contribute to the paper carving art, giving paper carving an unlimited vision and a better future! In addition, the research also explores current environmental issues presented in a story, “Ecological Conservation.” The story covers the continuous progress of time, how to preserve original species, and how to achieve peaceful coexistence, which is worthy of human thinking and reflection. The hope is for electronic painting to facilitate the interactions between viewers, especially to enhance the friendship between parents and their children. Science and technology products are no longer just a substitute for parent-child relations. Instead, they become a key factor in how people interact – enabling love and mutual exchange between us!

In the ancient times, there used to be caravans made up of the Hu shang (businessmen of minorities in the north and west) conducting a long-distance transportation for sale, i.e. a kind of transshipment trade. They set up commercial beach heads along the Silk Road, and spread their cultures over transactions. This paper, drawing on the archeological documents and literature, analyzes such issues as the types, the scale and journey of ethnic groups constituting the Silk Road Caravan, togethet with the monks and merchants traveling along. The aim of this paper attempts to disclose some characteristics 0f those trade activites engaged in by the Hu shang on the ancients Silk Road. Except for mutual business trade that was the main trade method between Tang dynasty and frontier nationalities , during some series of developing frontier , they may adopted force , political marries , giving some reputation and giving some gift , altogether giving trade that kept a friendly political relationship between Tang dynasty and frontier was a kind of important trade method On the other hand, Hu Shang is a special community, and their appearance and acts different from common run arouse the noble` interest, thus they prefer to mention every aspect of this special community. In this paper they are divided into the jewelry merchant、fragrance merchant、finance merchant、dining merchant ; the ordinary merchant. And according to records of literary sketches the social environment of their period and influences of their business activities reflected by this environment are discussed in the paper. About the narration of jewelry merchants, the reason why they frequently appears in literary sketches and the cause of magic is mainly discussed. The discussion of Hu Sang intends to prove the universality and importance of them during that time in China

Huyuan wrote General Rules for Rhyme at All Times and in All over the World(《古今中外音韻通例》) in the 1886A.D, and it is the assembly of Huyuan’s phonological theory. He employed phonology to make a detailed analysis on old Chinese sound, modern one and dialectal one. Besides, he also recorded dialectal accent characteristics in Yangze River and Huai River Districts, especially Nanking and Yangzhou, in late 19th century. It is a masterpiece outstanding in application of phonology and dialect investigation. The previous scholars mainly focused on analyzing the phonetic system of the Fifteen Phonology Map(十五總圖) and reached a general conclusion that it took the Jianghuai mandarin Hongchaopian as the basic dialectal accent. However, the researches rarely mentioned Huyuan’s achievements in dengyunxue(等韻學), archaic Chinese phonology and modern phonology. The seven chapters of this thesis are divided into four topics, which include Huyuan’s Voice and Rhyme Theory, Archaic Chinese Phonology, Dialectology and Translatology. The most important characteristics of it is taking the phonology map as the basis of all voices and extending it to the research on ancient voice, dialectal voice and translation. Generally speaking, Huyuan had been extremely elaborate in the analysis of voice, and he was to some extent capable in dialectal accent investigation. However, he was limited by the lack of recording medium, correct cognition of diachronic and synchronic variation and he was influenced by Neo-Confucianism. All these factors lead him being unable to break away from the research scope of the phonology scholars in Qing dynasty, thus the research was incapable in competing with the western phonology theory later.

To realize the relationship between the CCP and the intellectuals, we should review the historical development. Early in the 1940s, the CCP has already progressed the method of “using” the intellectuals. In 1949, the new regime was built by the CCP. Furthermore, they spread this strategy to whole mainland. Obviously, it was a big change and challenge to the contemporary Chinese intellectuals. Hu Feng, as an intellectual and a “comrade”, whose political position had been closed to the CCP. However, Hu Feng still had some opinions which differ from the CCP. In 1954, Hu Feng submitted his proposal which criticizing the CCP’s literature and art policy. In 1955, Mao ZeDong judged this case as “Hu Feng counterrevolutionary group”. After that, Hu Feng and his companions were arrested and investigated by the government. The Hu Feng Incident seemed to be such a literary persecution and injustice case. This research mainly focuses on the Hu Feng Incident. Firstly, Chapter 1 is the “Introduction”, which introduces the reasons of this research and the changing thought of modern Chinese intellectuals. Secondly, the focus of Chapter 2 is the life course and career pattern of Hu Feng. Chapter3 is the narration of the big change to the intellectuals on 1949. Chapter 4, which includes the main idea of this research, not only analyzes the causes of the Hu Feng Incident, but also seeks the crucial role of Mao. In Conclusion, Chapter 5 summarized the reflection and the vision upon this research. The writer reviews Mao’s personal authority and the political culture of the CCP. Finally, the special relationship between the CCP and the Chinese intellectuals will still be a major issue, that will need further study on it.

The Tang Dynasty is the peak of China''s feudal society. The women in this dynasty, the costumes blend of Chinese and Western culture expertise and the popularity of Hu apparel, is unprecedented. However, the reasons for creating the prevalence of this Hu apparel include political, social, and economic. In this paper, we first explore the background of the development of Hu apparel, respectively is political openness, the integration of Eastern and Western cultures, as well as the high development of economy. Next, through the above background, we can see the Tang Dynasty clothing apparel subject to external influence and the popular clothing of Hu apparel. The significant feature for Tang Dynasty advocating Hu apparel is that many women wear Hu apparel. The popular of Hu dance also has great influence on changes in women''s clothing, From advocating the Hu dance, developed to imitate Hu apparel. For example, Hu hat and Hu boots which has heavy correlated with Hu dance and the narrow sleeves, lapels, small pants, leather belt is characteristic for women at the time. In addition, other nations have also affected the women wear in Tang Dynasty, such as "Uighur equipment", etc. The Hu apparel style that popular in the Tang Dynasty can be confirmed by the Tang dynasty painting works of art, Tombs character murals, fresco, and around the excavation of cultural relics, such as pottery, Tang Dynasty, etc. Tang has a strong cultural compatibility, can integration of culture in other countries. Therefore, Tang women dressed Hu apparel have become fashionable at the time. The morale of dressing Hu apparel also influence Tang Women''s Clothing, such as diversification of wearing clothing and rich pattern of clothing. Tang’s apparel is colorful, in this paper, we take a depth study on the pattern of Tang’s clothing and influence, and can see that it contains the characteristics of age, ethnic, geographical, and cultural.

Full textWe have investigated the use of the E. coli lac operator-repressor system to regulate the expression of genes introduced into mammalian cells by gene transfer. We find that the bacterial lac repressor protein encoded in a suitable expression vector is synthesized in mammalian cells in culture, assembles into a tetramer, enters the nucleus to some extent, and represses expression of another gene that has one or several lac operator sequences inserted into any one of several sites in the promoter region of the gene. Derepression can be achieved by exposure of the cells to IPTG. From a practical point of view of an inducible genetic switch, this system confers an induction level of somewhere between 10- and 20-fold in most of the cases we have tested. That is not better than those that have been achieved with heat shock, mouse mammary tumor virus, and metallothionein promoters. There may, however, be some situations and some promoters for which the use of the lac operator system is advantageous. We have also shown that this lac control system can be used to regulate the expression of genes introduced into Xenopus oocytes by micro-injection.
At present, we have been trying to revise this system by using a newly developed promoter containing a symmetric lac operator sequence inserted at the various strategic points within the human metallothionein IIA promoter and enhancer regions, which consist of several positive control elements, in order to achieve induction ratios of a factor of 100 or more. We have also generated a new lacI gene which encodes a repressor containing at its carboxyl terminus the nuclear localization signal of the SV40 large-T antigen. Overall, by combining the newly developed lac control promoters with the new repressor producing cell lines, we hope to generate an inducible expression system with large induction ratios that can be used as a general genetic switch in the future.

ABSTRACT A simple identification of Shan-Shu is books encouraging people to be philanthropic. This kind of books help populations develop their good deed and are popular in the street. Although Shan-Shu is not weighted as much as classics, it has contributed incredibly to civilizer at the Chinese society. Therefore, to completely explore Shan-Shu, this presenting dissertation focus on the Song Dynasty in which Shan-Shu became as a category. Since this dissertation looks at the period of time that Shan-Shu developed as a subject, realizing the origin of Shan-Shu is an important task. Moreover, by discussing and analyzing those books finished at the Song Dynasty from different points of view, a more thoughtful opinion can be generated. The presenting dissertation is composed by eight chapters. The first three chapters are Introduction, The Structure and Division of Shan-Shu at the Song Dynasty, and The Historic Background of Shan-Shu Development at the Song Dynasty, respectively. These three chapters concentrate on background information of Shan-Shu and investigate questions such as why Shan-Shu is formed in the Song Dynasty and what the origin is. Conceptual problems of Shan-Shu are emphasized in the following three chapters, Chapter Four: The Development of Retribution in Shan-Shu during the Song Dynasty, Chapter Five: The Belief in Si-Ming Shen of Shan-Shu, and Chapter Six: The Subjects Illustrating in Shan-Shu during the Song Dynasty. These three chapters reveal the conceptual problems of Shan-Shu thought profoundly discussing issues about the blend of retribution from three religions, the belief in Si-Ming Shen and the topics of Shan-Shu. The last two chapter, Chapter Seven: The Value, Influence and Development of Shen-Shu at the Song Dynasty, and Chapter Eight: Conclusion summarize the research results, evaluate Shen-Shu at the Song Dynasty and its shaping afterward, all of which can provide useful information for future studies.
