Abstract:
Les huiles de Jatropha curcas communément appelé pourghère et de neem (Azadirachta indica A. Juss), sont testées comme alternatives aux pesticides chimiques dans l’optique d’y déceler de potentiels effets fongicides tout en préservant l’environnement. Les effets des deux huiles végétales sont comparés à ceux de deux fongicides conventionnels, la griséofulvine 250 mg et la nystatine ainsi que l’alcool 95°, sur cinq champignons phytopathogènes (Aspergillus flavus, Aspergillus niger, Curvularia lunata, Fusarium oxysporum et Rhizopus oryzae). Les observations ont porté sur la mesure du diamètre de la zone d’inhibition exercée par les différentes substances inhibitrices à travers les milieux gélosés. Le dispositif expérimental appliqué a été celui du bloc complètement randomisé de Fisher avec huit répétitions. Les moyennes des diamètres de la zone d’inhibition sont respectivement de 26,51 mm pour la nystatine, 14,53 mm pour la griséofulvine, 4,53 mm pour l’huile de neem, 3,05 mm pour l’huile de jatropha, et 0,00 mm pour l’alcool 95°. L’huile de neem a eu un effet inhibiteur net sur la croissance de l’ensemble des champignons testés. En revanche, l’huile de jatropha a exercé un meilleur contrôle de la croissance des espèces Aspergillus flavus, Aspergillus niger et Rhizopus oryzae. Les deux huiles pourraient être recommandées dans la protection des denrées stockées, notamment les céréales.Mots clés : Extraits végétaux, effet fongicide, zone d’inhibition, lutte alternative.&#x0D; English Title: Effect of Jatropha (Jatropha curcas L.) and neem (Azadirachta indica A. Juss) oils on in vitro devloppement of pathogenic funji Aspergillus flavus, Aspergillus niger, Curvularia lunata, Fusarium oxysporum et Rhizopus oryzae&#x0D; Oils of Jatropha curcas (pourghère) and neem (Azadirachta indica A. Juss) were tested as alternatives to chemical pesticides in a view to finding some potential fungicide effects harmless to environment. The effects of the two natural oils were compared with those of two conventional fungicides, griseofulvin 250 mg and nystatin and also alcohol 95° on five phytopathogenic fungi (Aspergillus flavus, Aspergillus niger, Curvularia lunata, Fusarium oxysporum and Rhizopus oryzae). Observations consisted in measuring the diameter of the zone of inhibition by different substances through the agarose medium. The experimental device applied was the Fisher completely randomized bloc with eight repetitions. Means of inhibition zone diameters were respectively de 26.51 mm for nystatin, 14.53 mm for griseofulvin, 4.53 mm for neem oil, 3.05 mm for jatropha oil, and 0.00 mm for alcohol 95°. The neem oil showed good inhibition effect on the growth of the whole set of tested fungi. In contrast, jatropha oil exhibited a better fungal growth control on Aspergillus flavus, Aspergillus niger and Rhizopus oryzae species. Thus, these two vegetable oils could be recommended in cereal protection in postharvest.Keywords: Vegetal oils, fungicide effects, inhibition zone, alternative control.

Jatropha curcas L. belongs to family Euphorbiaceae, Jatropha curcas is a valuable multi-purpose crop, historically it was used as medicine for wounds and leaves used as drinks against malaria, jatropha plants used to control soil degradation, alleviate erosion, desertification and increase soil fertility, however, in last decades there is more attention to use jatropha oil for produce biodiesel, Jatropha curcas is easily propagated by seeds or stem cutting, it is tolerant for drought for longtime, it is grow well with treated wastewater, also, it can be grown on marginal land. Jatropha curcas seed have about 32-40% valuable oil used to produce biofuel, therefore, it could be the source for biodiesel production particularly in arid and semiarid regions.

ABSTRACT&#x0D; The study investigating acute toxicity test of Jatropha curcas L. on Nile tilapia seeds (Oreochromis niloticus L.), had been conducted in May 2017, in Balai Riset Perikanan dan Perairan Umum (BRPPU) Mariana, Banyuasin. This study was aimed to determine the concentration of Jatropha curcas seed powder needed to cause 50% death of Nile tilapia seeds (LT50). This acute toxicity test used biological test with exposure time of 96 hours. The concentrations of Jatropha curcas tested were 0 (control), 240, 288, 346, 415, 498, 597, and 716 ppm which were applied for 96 hours. Data were analyzed using Spearman-Karber method with SPSS. Result showed LC50 of 24 hours was 551,271 ppm, LC50 of 48 hours was 466,513 ppm, and LC50 of 96 hours was 393,892 ppm. Meanwhile, LT50 for 597 ppm of Jatropha curcas was 1285, 166 minutes, and LT50 for 761 ppm was 784,121 minutes. Abstract:
Ce travail porte sur la caractérisation physico-chimique de l'huile de Jatropha Curcas et sa capacité à remplacer l'huile minérale dans les transformateurs de puissance. Ce produit présente plusieurs avantages sur les autres huiles végétales comme l'huile de palme ou l'huile de colza, qui recommandent sa production et son utilisation. En effet, la plante de Jatropha Curcas peut être cultivée sur des sols pauvres à faibles précipitations, évitant ainsi d'utiliser des sols plus fertiles pour sa culture permettant ainsi aux petits exploitants de réserver leurs terres aux cultures de base. Cette plante peut pousser facilement dans des zones où les niveaux de précipitations annuelles sont nettement inférieures à celles requises par d'autres espèces telles que le colza, le tournesol, le soja, le maïs, le palmier à huile et d'autres. Elle peut être cultivée sur tous les types de sol en Indonésie, même sur des terres arides, dans de nombreuses régions de l'Indonésie orientale, inexploitées en raison des difficultés à planter d'autres cultures. En outre, l'huile de Jatropha Curcas est un produit non alimentaire. En faisant subir à l’huile de Jatropha Curcas brute une estérification à base alcaline avec de l'hydroxyde de potassium (KOH), on obtient de l’huile de méthylester de Jatropha Curcas (JMEO) dont la viscosité et l’acidité sont acceptables pour les équipements à haute tension en particulier pour les transformateurs de puissance. Les propriétés physico-chimiques et électriques de JMEO ont été mesurées ainsi que celles de l'huile minérale (MO) pour la comparaison. Pour les propriétés physico-chimiques, il s’agit de la densité relative, la teneur en eau, la viscosité, l'acidité, l'indice d'iode, la corrosivité, le point d'éclair, le point d'écoulement, la couleur, l'examen visuel, et la teneur en ester méthylique. Quant aux propriétés électriques, elles concernent la rigidité diélectrique sous différentes formes de tension (alternative, continu et choc de foudre), les phénomènes de pré-claquage et de claquage sous choc de foudre, les décharges glissantes sur les surfaces de carton comprimé, immergé dans JMEO et MO. Les résultats obtenus montrent que les tensions de claquage moyennes en continu et en choc de foudre des huiles JMEO et MO sont très proches ; la tension de claquage moyenne de JMEO est même plus élevée que celle de l'huile minérale (de type naphténique). La mesure des tensions de claquage des mélanges d'huiles «80% JMEO + 20% MO» et «50% JMEO et 50% MO» montrent que la tension de claquage du mélange «80% JMEO + 20% MO» est toujours supérieure à celle de l'huile minérale sous tensions alternative et continue. Cela indique que le mélange d'huile minérale et de JMEO avec un rapport de 20:80 ne dégrade pas ses performances. Le mélange d'huiles peut se produire lors du remplacement de l'huile minérale par JMEO dans les transformateurs installés et en exploitation. L'analyse des caractéristiques des streamers (la forme, le temps d'arrêt, le courant associé et la charge électrique) se développant dans les huiles JMEO et MO sous tension impulsionnelle de foudre, montre une grande similitude. Aussi, la longueur finale (Lf) et la densité des branches des décharges surfaciques se propageant sur le carton comprimé immergé dans l'huile de Jatropha Curcas de méthylester (JMEO) et de l'huile minérale (MO), sous tensions de choc de foudre positif et négatif (1,2/50 μs), pour deux configurations d'électrodes divergentes (électrode pointe haute tension perpendiculaire et tangente au carton, respectivement), sont fortement influencées par l'épaisseur du carton comprimé. Pour une épaisseur donnée, Lf augmente avec la tension et décroît lorsque l'épaisseur augmente. Lf est plus long lorsque la pointe est positive que lorsque la pointe est négative. Pour une tension et une épaisseur du carton comprimé donnée, les valeurs de Lf dans l’huile minérale et l’huile JMEO sont très proches. [...]<br>This work is aimed at the investigation of the physicochemical characterization of Jatropha Curcas seeds oil and its capacity to be an alternative option to replace mineral oil in power transformers. This product presents several advantages that recommend both its production and usage over those of other vegetable oils as crude palm oil and rapeseeds oil. Indeed, it may be grown on marginal or degraded soils avoiding thus the need to utilize those more fertile soils currently being used by smallholders to grow their staple crops; and it will readily grow in areas where annual rainfall levels are significantly lower than those required by other species such as palm oil, rape-seeds oil, sunflower oil, soybeans oil, corn oil and others. For instance, these plants can grow on all soil types in Indonesia, even on barren soil. The barren soil types can be found in many parts of eastern Indonesia that remain untapped because of the difficulty planted with other crops. Moreover, jatropha curcas oil is nonfood crops. Jatropha Curcas oil was processed by alkali base catalyzed esterification process using potassium hydroxide (KOH) to produce Jatropha Curcas methyl ester oil (JMEO) has a viscosity and a acidity that are acceptable for high voltage equipment especially in power transformer. The physicochemical and electrical properties of JMEO were measured as well as those of mineral oil (MO) for comparison. The physicochemical properties cover relative density, water content, viscosity, acidity, iodine number, corrosivity, flash point, pour point, color, visual examination, and methyl ester content. Meanwhile the electrical properties cover dielectric strength under AC, DC and lightning impulse voltages, pre-breakdown / streamers under lightning impulse voltage, creeping discharge over pressboard immersed in JMEO and MO. The obtained results show that the average DC and lightning impulse breakdown voltages of JMEO and MO are too close, even the average AC breakdown voltage of JMEO are higher than that of mineral oil (napthenic type). The measurement of breakdown voltages of two oil mixtures namely “80% JMEO + 20% MO” and “50% JMEO and 50% MO” shows that the breakdown voltage of the first mixture (i.e., “80%JMEO+20%MO”) is always higher than that of mineral oil under both AC and DC voltages. This indicates that mixing 20:80 mineral oil to JMEO ratio does not degrade its performance. The mixing of oils can occur when replacing mineral oil by JMEO in installed transformers. The analysis of the streamers characteristics (namely; shape, stopping length, associated current and electrical charge) developing in JMEO and MO under lightning impulse voltages, shows that these are too close (similar). It is also shown that the stopping (final) length Lf and the density of branches of creeping discharges propagating over pressboard immersed in Jatropha Curcas methyl ester oil (JMEO) and mineral oil (MO), under positive and negative lightning impulse voltages (1.2/50 μs), using two divergent electrode configurations (electrode point perpendicular and tangential to pressboard), are significantly influenced by the thickness of pressboard. For a given thickness, Lf increases with the voltage and decreases when the thickness increases. Lf is longer when the point is positive than with a negative point. For a given voltage and thickness of pressboard, the values of Lf in mineral oil and JMEO are very close. It appears from this work that JMEO could constitute a potential substitute for mineral oil for electrical insulation and especially in high voltage power transformers

Abstract:
Parmi les technologies de solaire thermodynamique, la technologie du linéaire de Fresnel semble la plus adaptée aux régions d’Afrique Sub saharienne. Cela en raison de la simplicité de la technologie. C’est dans cette optique qu’un collecteur de type linéaire de Fresnel d’une superficie de 7,5 m² de miroirs a été construit au laboratoire énergies renouvelables et efficacité énergétique (LabEREE). La construction du collecteur s’est faite en utilisant en priorité les matériaux disponibles localement afinde rendre la technologie plus accessible aux populations locales et de réduire les coûts de fabrication. Des tests sont effectués sur le collecteur afin de déterminer ses rendements optiques, thermiques et globaux. Dans un premier temps, une revue bibliographie des différents collecteurs de type linéaire de Fresnel nous a permis d’identifier les variantes, de cette technologie, les plus adaptées au contexte dela région . Un modèle thermique et un modèle optique ont été mis en place comme outils de dimensionnement et d’optimisation du collecteur. Les résultats expérimentaux obtenus ont été utilisés pour valider les différents modèles mis en place. Le rendement global du collecteur obtenu expérimentalement est de 21% et il a un facteur de concentration local de 6<br>Among the different technologies of concentrated solar power plant, the linear Fresnel, thanks to its simplicity, appears the most adapted to rural area of Sub Sahara region. A linear Fresnel collector of 7.5 m² has been built in “laboratoire énergies renouvelables et efficacité énergétique (LabEREE)”. The collector have been designed using material available locally by local man power. This reduces the total cost of the technology and makes it affordable for local population. The collector has been characterized in order to find optical, thermal and global efficiencies. In first time, a review on different linear Fresnel collector allows finding the technology that is most adapted to the Sub-Saharan region. An optical and thermal model of the collector has been done as a tool for designing and optimisation. The experimental results enable to validate the different models done. The collector has an effective concentration factor of 6 and a global efficiency of 21%

Abstract:
Convaincu de l’intérêt et du potentiel des matériaux naturels et des déchets industriels, cette thèse a contribué à la mise au point de matériaux de stockage de la chaleur (TESM) pour les CSP en Afrique de l’Ouest. Plus spécifiquement, ce travail de recherche a porté sur la valorisation de la latérite du Burkina Faso, des cendres de foyer des centrales à charbon de la société SONICHAR au Niger, des résidus en carbonate de calcium (chaux) de l’industrie de production de l’acétylène au Burkina Faso et l’huile végétale de Jatropha curcas de la société Belwet au Burkina Faso. Les résultats de cette étude ont permis de montrer que l’huile de Jatropha curcas peut être considérée comme une alternative viable aux fluides de transfert et aux TESM conventionnels pour les CSP fonctionnant à 210 °C. Les matériaux élaborés à partir des cendres de foyer et de la latérite présentent un caractère réfractaire en raison de la présence de mullite et de spinelle. L’ajout de chaux permet de réduire le point de fusion tout en préservant le caractère réfractaire et conducteur des phases obtenues. En raison de leurs stabilités, et l’absence de conflit d'utilisation, les matériaux obtenus peuvent être utilisés comme TESM dans CSP à des températures allant jusqu’à 900 °C<br>Convinced of the interest and potential of natural materials and industrial waste, this thesis has contributed to the development of heat storage materials (TESM) for CSPs in West Africa. More specifically, this research focused on the valorization of laterite from Burkina Faso, the bottom ashes from the coal-fired power plants of SONICHAR in Niger, residues of calcium carbonate (lime) from the acetylene in Burkina Faso and the vegetable oil of Jatropha curcas from the company Belwet in Burkina Faso. The results of this study showed that Jatropha curcas oil can be considered as a viable alternative to conventional HTF and TESM for CSP operating at 210 °C. The materials elaborated from bottom ashes and laterites present a refractory character due to the presence of mullite and spinel. The addition of lime makes it possible to reduce the melting temperature while preserving the refractory and conductive character of the obtained phases. Due to their stabilities, and the absence of conflict of use, the obtained materials can be used as TESM in CSP at temperatures up to 900 °C

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior<br>O pinhÃo manso (Jatropha curcas L.) à considerado uma fonte potencial para a produÃÃo de biocombustÃveis, tendo por base a alta produtividade e qualidade do Ãleo extraÃdo de suas sementes. Entretanto, seu uso à inviabilizado para alimentaÃÃo humana e animal devido à presenÃa de fatores antinuticionais como a curcina e, em especial, os Ãsteres de forbol. A utilizaÃÃo de ferramentas biotecnolÃgicas, como a cultura de tecidos, pode vir a minimizar ou solucionar essas limitaÃÃes. Nesse contexto, o objetivo desse trabalho foi propagar in vitro plantas inteiras via cultivo de Ãpices caulinares obtidos a partir de sementes de pinhÃo manso germinadas in vivo e in vitro; determinar a reprodutibilidade de um protocolo de regeneraÃÃo disponÃvel na literatura e definir, atravÃs de estudos histolÃgicos, a origem da planta regenerada in vitro. Para a propagaÃÃo in vitro utilizou-se Ãpices caulinares, como explantes, que foram colocados em meio MS suplementado com as citocininas BAP (6-benzilaminoprina), KIN (cinetina) e 2-iP (2-isopenteniladenina) na concentraÃÃo de 2,0 mg.L-1. O BAP apresentou os melhores resultados no desenvolvimento dos Ãpices caulinares. Este e a giberelina foram utilizados juntos no mesmo meio ou separados, para a induÃÃo do alongamento dos mesmos. O Ãcido indol-3-butirÃco (AIB) foi usado para o enraizamento. A suplementaÃÃo do meio de cultura com BAP foi eficiente no desenvolvimento dos Ãpices caulinares e o alongamento apresentou Ãxito com o uso do GA3 individualmente. O enraizamento com o AIB foi conseguido. A fim de determinar a reprodutibilidade do protocolo de organogÃnese, cotilÃdones de sementes de pinhÃo manso germinadas in vitro foram colocados em meio MS suplementado com BAP e AIB para a induÃÃo da formaÃÃo de partes aÃreas, estas foram alongadas com BAP e enraizadas com AIB. AlÃm disso, os explantes cotiledonares foram colocados com a face abaxial e adaxial em contato com o meio de cultivo a fim de estabelecer a melhor posiÃÃo do mesmo na regeneraÃÃo in vitro. Plantas inteiras de J. curcas L. foram obtidas a partir da utilizaÃÃo do protocolo testado. A melhor posiÃÃo do segmento cotiledonar foi a face adaxial em contato com o meio, onde 56% dos explantes formaram partes aÃreas. A anÃlise histolÃgica foi feita com a coleta de explantes cotiledonares em intervalos sequenciais de 0, 5, 10, 15 e 25 dias de permanÃncia do meio de regeneraÃÃo. O estudo anatÃmico possibilitou o acompanhamento da organogÃnese do pinhÃo manso, no qual cÃlulas parenquimÃticas prÃximas à regiÃo adaxial do explante cotiledonar se dividiram e formaram as partes aÃreas. However, its use is unfeasible for human and animal food due to antinutritional factors like the curcin protein and specially the secondary metabolites â phorbol esters. The application of biotechnological tools, like tissue culture, can be used to overcome these limitations. In this context, the aim of this work was to propagate whole plants in vitro using shoot cultures obtained from physic nut seeds germinated in vitro and in vivo; to determine the reproducibility of a regeneration protocol available in the literature, and to define through histological studies, the origin of the regenerated plant in vitro. For the in vitro propagation, shoots were used as explants sources, which were placed on MS medium supplemented with the cytokinins BAP (6- benzylaminopurine), KIN (kinetin) e 2-iP (2- isopentenyl adenine), 2,0 mg.L-1. BAP showed the best results in the development of the shoots. This regulator was used alone or in association with gyberelins for the elongation of the shoots. The indole-3-butyric-acid was used for the rooting. The supplementation of the medium culture with BAP was efficient in the development of the shoots and the elongation was superior with the use of GA3 alone. The rooting was achieved with AIB. To determine the reproducibility of an organogenesis protocol, cotyledons from physic nut seeds germinated in vitro were placed on MS medium supplemented with BAP and AIB for the promotion of aerial which were elongated with BAP and rooting with AIB. The cotyledonary explants were placed with the abaxial and adaxial face in contact with the medium to establish the best position of it in the in vitro regeneration. Whole plants of J. curcas L. were obtained using the protocol tested. The best position of the cotyledonar segment was the adaxial face in contact with the medium, where 56% of the explants formed shoots. Entretanto, seu uso é inviabilizado para alimentação humana e animal devido à presença de fatores antinuticionais como a curcina e, em especial, os ésteres de forbol. A utilização de ferramentas biotecnológicas, como a cultura de tecidos, pode vir a minimizar ou solucionar essas limitações. Nesse contexto, o objetivo desse trabalho foi propagar in vitro plantas inteiras via cultivo de ápices caulinares obtidos a partir de sementes de pinhão manso germinadas in vivo e in vitro; determinar a reprodutibilidade de um protocolo de regeneração disponível na literatura e definir, através de estudos histológicos, a origem da planta regenerada in vitro. Para a propagação in vitro utilizou-se ápices caulinares, como explantes, que foram colocados em meio MS suplementado com as citocininas BAP (6-benzilaminoprina), KIN (cinetina) e 2-iP (2-isopenteniladenina) na concentração de 2,0 mg.L-1. O BAP apresentou os melhores resultados no desenvolvimento dos ápices caulinares. Este e a giberelina foram utilizados juntos no mesmo meio ou separados, para a indução do alongamento dos mesmos. O ácido indol-3-butiríco (AIB) foi usado para o enraizamento. A suplementação do meio de cultura com BAP foi eficiente no desenvolvimento dos ápices caulinares e o alongamento apresentou êxito com o uso do GA3 individualmente. O enraizamento com o AIB foi conseguido. A fim de determinar a reprodutibilidade do protocolo de organogênese, cotilédones de sementes de pinhão manso germinadas in vitro foram colocados em meio MS suplementado com BAP e AIB para a indução da formação de partes aéreas, estas foram alongadas com BAP e enraizadas com AIB. Além disso, os explantes cotiledonares foram colocados com a face abaxial e adaxial em contato com o meio de cultivo a fim de estabelecer a melhor posição do mesmo na regeneração in vitro. Plantas inteiras de J. curcas L. foram obtidas a partir da utilização do protocolo testado. A melhor posição do segmento cotiledonar foi a face adaxial em contato com o meio, onde 56% dos explantes formaram partes aéreas. A análise histológica foi feita com a coleta de explantes cotiledonares em intervalos sequenciais de 0, 5, 10, 15 e 25 dias de permanência do meio de regeneração. Se analiza la variabilidad morfológica de Jatropha curcas en 20 poblaciones cultivadas en las regiones de Lambayeque, Piura, Tumbes, San Martín y Ucayali. Se muestrearon 123 individuos de 2-4 cm de dap; se registraron ocho parámetros morfométricos como: altura total de la planta, número de ramas de la planta, número de lóbulos del limbo, longitud del limbo, del pecíolo, del fruto, largo y ancho de la semilla. Los parámetros fueron analizados mediante la prueba de comparaciones de medias ANOVA (p menor 0.05); a los que mostraron diferencias significativas se realiza el análisis de componentes principales. Existen diferencias significativas de seis parámetros entre las poblaciones; respecto a la altura total de la planta, el mayor valor se registra para la población de La Libertad, y el menor valor para la población de San Isidro; respecto a la cantidad de ramas, el mayor valor se registra para Olmos, y el menor valor para Chazuta; respecto a la longitud del limbo y el pecíolo, el mayor valor se registra para Tambo Grande, y el menor valor para Olmos; respecto a la cantidad de lóbulos, el mayor valor se registra para Tambo Grande, y el menor valor para Curimaná; respecto a la longitud del fruto, el mayor valor se registra para la población de Oidor y el menor valor en la población Yarinacocha. Los resultados del Análisis de Componentes Principales mostraron que el 66.9% de la varianza correspondió a los 2 primeros componentes. No se observaron grupos claramente definidos; sin embargo, las poblaciones de la región costera formaron grupos distintos con respecto a las poblaciones de la región amazónica. A total of 123 individuals with DBH 2-4 cm were sampled, eight morphometric parameters were evaluated such as total plant height, number of branches of the plant, number of lobes of the lamina, length of blade, petiole, fruit and seed. The parameters were analyzed by Paired t test ANOVA (p less than 0.05), those with significant differences were performed principal components analysis. There are significant differences among the populations in seven parameters; regarding the total plant height, the highest value recorded was in La Libertad, and the lowest value for the population of San Isidro; regarding the number of branches, the highest value recorded was in Olmos, and the lowest value for Chazuta; regarding the length of the blade and petiole, the highest value recorded was found in Tambo Grande, and the lowest value in Olmos, regarding the number of lobes, the highest value recorded was in Tambo Grande, and the lowest value in Curimaná; regarding the length of the fruit, the highest value recorded was found in Oidor and the lowest in the population Yarinacocha. The Principal Component Analysis showed that 70.9% of the variance corresponds to the two first components. Groups clearly defined were not observed; however, the populations of the coastal region formed groups distinct from those of the jungle region. CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior<br>Jatropha curcas L. is a plant native to America and belongs to the Euphorbiaceae family. Currently it is gaining economical interest mainly because it is an oilseed crop with potential to produce biodiesel. However, presence of phorbol esters (a class of diterpenes) that are the major toxic constituents of the seeds, limits a better usage of the plant, by making the use of the residue, obtained after the oil extraction from the seeds, unfeasible for animal feed, due to its pro-carcinogenic activity and inflammatory action. Proteomic analysis of the plastids isolated from developing seeds of Jatropha is important because the synthesis of fatty acid as well as phorbol esters, the two most attractive compounds in the study of Jatropha curcas, occur in plastids. Proteomic analysis of this organelle is crucial to better understand and explore not only the biosynthetic pathway of these two compounds but other metabolic pathways , and addtionaly providing foundation for researchs that aimed to develope genotypes with more suitable characteristics for industrial applications. In this study, we performed a proteomic analysis of plastids isolated from the endosperm of developing Jatropha curcas seeds that were in the initial stage of deposition of protein and lipid reserves. Proteins extracted from the plastids were digested with trypsin, and the peptides were applied to an EASY-nano LC system coupled online to an ESI-LTQ-Orbitrap Velos mass spectrometer, and this led to the identification of 1103 proteins representing 804 protein groups, of which 923 were considered as true identifications, and this considerably expands the repertoire of J. curcas proteins identified so far. Of the identified proteins, only five are encoded in the plastid genome, and none of them are involved in photosynthesis, evidentiating the nonphotosynthetic nature of the isolated plastids. Homologues for 824 out of 923 identified proteins were present in three different plastids proteins databases i.e. PPDB, SUBA and PlProt, while homologues for 13 proteins were not found in any of these three databases but were marked as plastidial by at least one of the three prediction programs used (TargetP, ChloroP and PlantMPloc). Functional classification showed that proteins belonging to amino acids metabolism comprise the main functional class, followed by carbohydrate, energy, and lipid metabolisms. The small and large subunits of Rubisco were identified, and their presence in plastids is considered to be an adaptive feature counterbalancing for the loss of one-third of the carbon as CO2 as a result of the conversion of carbohydrate to oil through glycolysis. While several enzymes involved in the biosynthesis of several precursors of diterpenoids were identified, we were unable to identify any terpene synthase/cyclase, which suggests that the plastids isolated from the endosperm of developing seeds do not synthesize phorbol esters. In conclusion, this study provides insights into the major biosynthetic pathways and certain unique features of the plastids from the endosperm of developing seeds at the whole proteome level.<br>O pinhÃo manso (Jatropha curcas L.) à uma planta nativa da AmÃrica, pertencente à famÃlia Euphorbiaceae. Entretanto, a presenÃa de Ãsteres de forbol (uma classe de diterpeno), que sÃo os principais constituintes tÃxicos das sementes, limita uma melhor utilizaÃÃo dessa planta, por inviabilizar o uso do resÃduo de extraÃÃo do Ãleo das sementes na alimentaÃÃo animal, bem como, por apresentar atividade prÃ-carcinogÃnica e aÃÃo inflamatÃria. A anÃlise proteÃmica de plastÃdeos, isolados de sementes em desenvolvimento de pinhÃo manso, à uma importante vertente de estudo, pois tanto a sÃntese de Ãcidos graxos como dos Ãsteres de forbol, os dois compostos mais atrativos no estudo de Jatropha curcas, ocorrem nos plastÃdeos. O estudo proteÃmico dessa organela torna-se crucial para melhor compreender e explorar nÃo somente as vias biossintÃticas desses dois compostos, como de outras vias metabÃlicas, alÃm de proporcionar um conjunto de dados que pode ser utilizado em pesquisas voltadas para o desenvolvimento de genÃtipos com caracterÃsticas mais adequadas para aplicaÃÃes industriais. No presente trabalho, realizou-se uma anÃlise proteÃmica de plastÃdeos isolados do endosperma de sementes em desenvolvimento do pinhÃo manso, que estavam nos estÃgios iniciais de deposiÃÃo de lipÃdios e proteÃnas de reserva (25-30DAA), confirmados por meio de anÃlises histolÃgica e histoquÃmica. As proteÃnas extraÃdas dos plastÃdeos foram digeridas com tripsina e os peptÃdeos foram aplicados no sistema de nano-LC EASYII acoplado online ao espectrÃmetro de massa nano ESI LTQ-Orbitrap velos, o que resultou na identificaÃÃo 1103 proteÃnas, representando 804 grupos de proteÃnas, dos quais 923 foram consideradas identificaÃÃes verdadeiras. Isso expandiu consideravelmente o repertÃrio de proteÃnas do pinhÃo manso atà agora identificas. Dentre as proteÃnas identificadas, apenas 5 sÃo codificadas pelo genoma plastidial, e nenhuma delas està envolvida na fotossÃntese, o que evidencia a natureza nÃo fotossintÃtica dos plastÃdeos isolados. HomÃlogos de 824, dentre as 923 proteÃnas identificadas, estavam presentes nos bancos de dados PPDB, SUBA e PlProt, enquanto homÃlogos para 13 proteÃnas nÃo foram encontrados em nenhum dos trÃs bancos de dados plastidiais, mas foram detectados como plastidiais por pelo menos um dos trÃs programas de prediÃÃo de localizaÃÃo subcelular utilizados (TargetP, ChloroP, PlantMPloc). A classificaÃÃo funcional mostrou que a maioria das proteÃnas identificadas pertencia ao metabolismo dos aminoÃcidos, seguido dos metabolismos dos carboidratos, energÃtico e dos lipÃdios. As subunidades maiores e menores da Rubisco foram identificadas, e sua presenÃa nos plastÃdeos foi considerada uma caracterÃstica adaptativa para contrabalancear a perda de um terÃo do carbono na forma de CO2 como um resultado da conversÃo de carboidratos em Ãleo atravÃs da glicÃlise. Apesar de enzimas envolvidas na biossÃntese de diversos precursores dos diterpenÃides terem sido identificadas, nÃo foi detectado nenhuma terpeno sintase/ciclase, o que sugere que os plastÃdeos isolados do endosperma de sementes em desenvolvimento nÃo sintetizam Ãsteres de forbol, apesar de uma grande quantidade desse composto ser encontrada neste tecido. Como conclusÃo, o presente trabalho proporciona insights sobre as principais vias biossÃntÃticas, e sobre caracterÃsticas peculiares dos plastideos isolados do endosperma de sementes em desenvolvimento.

Biodiesel is primarily produced by transesterification of edible oils. Increasing concern about using food supplies for fuel has generated interest in alternative raw materials. Furthermore, there are numerous steps between harvesting of oilseeds and final production of biodiesel that can be integrated, thereby simplifying the process and making it more suitable for distributed production. Hence, in this study, the production of biodiesel via in situ transesterification of non-edible Jatropha curcas seed has been investigated. The main aim was to investigate the parameters of the process, with a view to reducing the substantial excess of methanol required. A significant secondary aim was to investigate the possibility of utilising other compounds that come out from the process. “Design of experiments” was employed to study the parameters at lab-scale, with the matrix boundary being determined beforehand using one-at-a-time experiments. The reduction of methanol excess was attempted by use of two co-solvents, hexane and diethylmethane (DEM), and by replacing methanol with methyl acetate. It was found that in situ transesterification run using particle sizes below 0.71 mm, a 400:1 molar ratio of methanol to oil, 60 minutes, and a minimum of 300 rpm mixing intensity yielded the highest biodiesel yield of 83 wt %. NaOH concentration and reaction temperature were not found to be significant variables, and were set at 1.0 N and 30oC respectively. DEM was a more effective co-solvent than hexane. The addition of DEM to the process at 400:1 molar ratio experiment increased the yield from 83 to 92 wt %. When methyl acetate was used to replace methanol, the requirement of molar ratio of solvent:oil reduced significantly to 175:1 to achieved 86.8 wt% of biodiesel. The solid meal was shown to contain substantial amounts of protein, making it a valuable co-product stream. Previously J. curcas meal had had little value as animal feed due to its toxicity, but this may be reduced or removed by this process.

Interest and investment in biofuel production is growing in response to political and economic drivers associated with concerns over fossil fuel depletion; the need for energy security; greenhouse gas emission reduction demands; and the perceived potential for biofuel production to contribute to rural development and poverty alleviation. Despite this, empirical data on the impacts of biofuel crops are limited and their potential role in enabling livelihood diversification activities in sub-Saharan Africa is under-explored. Addressing this gap, this research takes an integrated case study approach to assess the impacts of different Jatropha curcas cultivation models on rural livelihoods across a range of study sites in Malawi. The research develops and adopts an integrated research methodology that combines the Sustainable Livelihoods Framework (SLF) with the diffusion of innovation model, using participatory and conventional social science research methods to achieve a multi-scale analysis spanning from local to national levels. Findings show that the overall approach to cultivation (from planting method, through production and processing to marketing) is more important in providing a livelihood contribution than the model of cultivation used. When Jatropha is promoted as a boundary fence, the crop and its by-products (such as seedcake fertiliser) can contribute to all 5 livelihood asset groups (natural, physical, financial, human and social capitals): directly or indirectly through financial capital substitution. However, a complex set of factors are involved in farmers' adoption decisions, .which intimately link the livelihood context, communication channels and the characteristics of the innovation itself. This results in varying numbers of households cultivating the crop in the case study villages. In addition, constraints identified at the local to national level mean livelihood benefits are yet to be fully realised. Communication amongst the diverse range of stakeholders involved in Jatropha cultivation in Malawi is characterised by isolated networks, individuals and groups at the local to sub- national level. At the national level, the private sector dominates policy dialogue. Moreover, relevant legislation and policy is absent, hindering the development of the sector. Policy recommendations and action points for actors are presented in order to address these constraints. At the national level, it is recommended that the government of Malawi should prioritise rural development as a policy goal for the future of national biofuel production and take steps to develop a coherent biofuels policy which is fully integrated into national development policies. At local to sub-national levels, stakeholder communications must be improved in order to facilitate knowledge management and governance of the sector, and investments must be made in the extension services network.

Conselho Nacional de Desenvolvimento Científico e Tecnológico<br>In this work was determined, for the first time, the volatile composition from fresh and dried leaves of six accessions (named PM-2, PM-7, PM-10, PM-11, PM-12 e PM-14) and a cultivar (EPAMIG, named here EMB) of Jatropha curcas L. The volatile compounds was extracted by hydrodistillation and then analyzed by GCMS. Thus, it was possible to observe that the (Z)-3-hexen-1-ol was the major compound in the fresh leaves (>70% in all accessions). Alcohols also predominated in the dried leaves, mainly (Z)-2-penten-1-ol and (Z)-3-hexen-1-ol, but others constituents were identified in significant proportions included benzenoid compounds such as benzaldehyde. The influence of drying on volatile composition from leaves of J. curcas was evaluated on the PM-2 accession. Besides, this work also describes the development of a High Performance Liquid Chromatograpy (HPLC) method for differentiation of six accessions and a cultivar (EPAMIG) from fresh and dried leaves of J. curcas. The methanol extracts of the leaves were analyzed by HPLC-DAD and after stages of optimizations, we obtained the fingerprint chromatograms. For a comparative analysis of the accessions and a cultivar, were applied chemometric tools of Principal Component Analysis (PCA) and Hierarchical Cluster Analysis (HCA), which it was possible to discriminate the samples from dried leaves according to their origins. The methanol extracts of the leaves were submitted to cytotoxicity assay against four tumor cell lines (HCT-8, HL-60, SF-295 and MDA-MB435), presenting IC50 values of 25.13 μg/ml, against HL-60 cell line for EMB sample considered promising by National Institute of Cancer, USA (NCI). The biological activity of this extracts was evaluated on the development of Spodoptera frugiperda, considered as a primary pest in corn crop. It was observed 60% larval mortality.<br>No presente trabalho foi determinado, pela primeira vez, os compostos voláteis das folhas frescas e secas de Jatropha curcas L. pela técnica de hidrodestilação, para seis diferentes acessos (denominados de PM-2, PM-7, PM-10, PM-11, PM- 12 e PM-14) e uma cultivar (EPAMIG, denominada de EMB). As análises foram realizadas através de CG/EM, obtendo-se o (Z)-3-hexen-1-ol (75,7±18,2%) como composto majoritário nas folhas frescas, com percentuais superiores a 70% entre os acessos. Nas folhas secas obteve-se como compostos majoritários o (Z)-2- penten-1-ol (14,9±11,3%), (Z)-3-hexen-1-ol (9,6±9,1%) e o benzaldeído (3,9±2,7%). Um experimento de secagem foi realizado para o acesso PM-2 com intuito de verificar o efeito da secagem na composição dos voláteis das folhas de J. curcas. Neste trabalho também foi desenvolvido um método por Cromatografia Líquida de Alta Eficiência (CLAE) para diferenciação dos seis acessos e da cultivar (EPAMIG) para folhas frescas e secas de J. curcas. Os extratos metanólicos das folhas foram analisados por CLAE-DAD e após etapas de otimizações, foram obtidos os cromatogramas fingerprints. Para uma análise comparativa entre os acessos e a cultivar, aplicaram-se ferramentas quimiométricas de Análise dos Componentes Principais (PCA) e Análise de Agrupamentos Hierárquicos (HCA), pelas quais foi possível diferenciar as amostras das folhas secas de acordo com suas procedências. Os extratos metanólicos também foram submetidos a ensaios de citotoxicidade in vitro para 04 linhagens de células tumorais humanas (HCT-8, HL-60, SF-295 e MDA-MB- 435), obtendo-se CI50 25,13 μg/mL, na linhagem HL-60 para a amostra da cultivar (EPAMIG). Este resultado está dentro do intervalo proposto pelo National Institute of Cancer, USA (NCI) para considerar extratos como promissores. A atividade biológica destes extratos também foi avaliada sobre o desenvolvimento de Spodoptera frugiperda, principal praga do milho, apresentando 60% de mortalidade larval.
