Full textThis project was designed to examine the effects of oxyhemoglobin on the vascular tone of the middle and basilar cerebral arteries in vitro and to evaluate the validity of using bovine vessels as an animal model of the human cerebral vasculature in which to study cerebral vasospasm. Human vessels were studied in parallel with the bovine arteries to test the validity of the animal model. To further evaluate the model, some experiments were also carried out on canine vessels, because the dog has frequently been used for cerebrovascular studies in the past.
The cow is a better model of the human cerebral vasculature than the dog. Arteries respond to a variety of vasoactive stimuli, in a manner closely resembling human vessels, and can be induced into a prolonged vasospastic state by exposure to oxyhemoglobin. Vasospasm in all three species was independent of endothelial status, functional innervation, or morphological evidence of atherosclerosis. Vasospasm could not be prevented by diltiazem, nicardipine or ascorbic acid, but was partially reversed in some samples by verapamil. Bovine vessels would appear to be exceptionally useful for studies designed to test pharmacological agents for the prevention or therapy of post-hemorrhagic vasospasm and also to determine the pathophysiological mechanisms involved.

Full textMethodology. This investigation, as a component of an international multi-centre study coordinated by the IARC, followed a hospital-based case-control design. Cases consisted of newly diagnosed patients with primary squamous cell carcinoma of the oral cavity, including mouth and oropharynx. Controls were frequency matched to cases by sex, age, and hospital. All subjects were interviewed to elicit detail information on known and putative risk factors.
Oral exfoliated cells were collected from all subjects for detection of HPV DNA using the PGMY09/11 PCR protocol. Antibodies against HPV 16, 18, and 31 capsids were detected in patients' plasma using an immunoassay technique. Logistic regression was used for estimation of odds ratios (ORs) and 95% confidence intervals (CI) of oral cancer for HPV and other candidate risk factors.
Results. A total of 72 cases and 129 controls were recruited. HPV DNA was detected in 19% of cases (14 out of 72), and in 5% of controls (6 out of 129). Analysis for cancers related to Waldeyer's ring (palatine tonsil and base of tongue) showed that the OR of disease for detection of high risk HPV types was 19.32 (95%CI:2.3--159.5), after adjustment for socio-demographic characteristics, tobacco and alcohol consumption. The adjusted OR of disease for HPV 16 seropositivity was 31.51 (95%CI:4.5--219.7). Analysis for non tonsillar oral cancers showed that the OR for detection of high risk HPV DNA in oral cells and for seropositivity were 2.14 (95%CI:0.4--13.0) and 3.16 (95%CI:0.8--13.0), respectively.
Discussion. The results from this study provide evidence supporting a strong association between HPV infection and cancers of the oropharynx, especially those arising from Waldeyer's ring. On the other hand, the association with non tonsillar oral cancers was of much lower magnitude. The biological evidence establishing a firm etiologic link remains to be established for the latter subsites, whereas the association between HPV and Waldeyer's ring carcinomas is consistent with a causal link.

Full textOur laboratory utilizes five human uveal melanoma cell lines (92.1, MKT-BR, OCM-1, SP-6.5, UW-1) that have a previously described metastatic potential from an animal model (MP). We used four methods to characterize the proliferation rate of these five cell lines. We also used a Matrigel invasion assay to assess the invasive ability of the same cell lines in response to the potential chemo-attractants: interleukin-6, Vascular Endothelial Growth Factor, and Fetal Bovine Serum.
From these results we were able to propose a novel classification system for our cell lines: high proliferation and high invasion/high MP, low proliferation and low invasion/low MP, and high proliferation and invasion/no MP.

Full textMethodology. In a prospective cohort of 2075 Brazilian women, cervical specimens were collected for cytology and HPV DNA detection. Information on potential risk factors was obtained by interview at baseline, and at return visits. Follow-up visits were scheduled every 4 months in the first year and every 6 months thereafter.
Results. The prevalence of HPV coinfection was 7.6% among all women, and 21% among HPV-positive women. HPV coinfection was less common among cytologically normal women, and peaked among women with lower grade cytological abnormalities. Both prevalence and incidence of HPV coinfection decreased markedly with increasing age. HPV infection at enrolment was associated with a higher risk of infection with any other HPV type. The adjusted hazard ratios and 95% confidence intervals (95% CI) of acquisition of any HPV type were: 2.2 (95% CI: 1.5--3.2) for oncogenic types excluding HPV 16 and 18 detected at enrolment, and 2.6 (95% CI: 1.7--4.2) for HPV 16 or 18 detected at enrolment. Predictors of HPV coinfection included younger age, higher number of sexual partners, a history of condyloma, and younger age at first sexual intercourse.
Conclusion. HPV coinfections were relatively common, especially among younger women. Prior HPV infections increased the risk of subsequent infections with other types. These results may have implications for vaccine development and public health decisions about vaccination programs.

Full textA model of mammary carcinogenesis in mice demonstrated no significant biochemical change relating to potential drug resistance among normal, preneoplastic and neoplastic mammary tissues. This is of particular interest since human breast cancer is usually quite drug sensitive at the start of the treatment.
Human colon cancer is probably induced by carcinogenic compounds present in the environmental dietary elements and also exhibits de novo resistance to most antineoplastic drugs. Most of the biochemical elements examined were found to be present in these colon tissues. Additionally, the detoxification pathways including glutathione and its related enzymes, were found to be significantly elevated in colon tumor versus normal adjacent mucosa. The molecular characterization of glutathione S-transferase (GST) isoenzymes using antibodies and cloned molecular probes to specific enzyme forms showed that the anionic form of GST (GST-$ pi)$ is greater in tumors than in the respective normal mucosa. The neutral form (GST-$ mu)$ is conversely decreased in tumor relative to normal. The cationic (GST-$ alpha)$ is present only in 30% of the samples examined, with no difference between tumor and adjacent normal mucosa. The expression of cytochrome P-450 isoenzymes was also examined in a similar fashion; a phenobarbital-inducible form was expressed in most colon tissues examined, and expression of the polycyclic aromatic hydrocarbons-inducible P$ sb3$-450 was present in some colon tissues while mRNA for P$ sb1$-450 was not detected in any. The biochemical alterations found in human colon could be the targets of therapeutic manoeuvers to enhance the efficacy of antineoplastic treatment of human colon cancer.

Full textObjectives. (1) To measure the risk of incident neoplastic cervical lesions over time related to prior cumulative and persistent HPV infections. (2) To evaluate the influence of HPV viral burden on lesion risk longitudinally. (3) To estimate the progression rates and sojourn time for precursor squamous intraepithelial lesions (SILs) and how they relate to HPV infection status.
Design and methods. In 1993, the Ludwig-McGill study team began a large longitudinal study of the natural history of HPV infection and cervical neoplasia in the city of Sao Paulo, Brazil. Follow-up involved repeated measurements on individual subjects over time. 2462 women were enrolled into the study and were seen every 4 months in the first year (0, 4, 8 and 12 months), and twice yearly thereafter for a period of up to eight years. In addition to obtaining risk factor information via questionnaire, cervical specimens were taken for Pap cytology and HPV testing at every visit. Statistical analyses entailed: (1) using different modalities for defining HPV persistence by type and intensity; (2) using modeling approaches that take into account the repeated measurements of HPV and SIL over time within individuals; (3) analyzing changes in transition states between different cervical lesion grades and the rate of progression from one state to the next.
Rationale. A longitudinal, repeated measurement cohort investigation, such as this one, permits an accurate and unbiased assessment of the relationship between cumulative HPV exposure and lesion incidence. An elevated relationship between persistent HPV infections and SIL incidence supports the proposal for the application of type-specific molecular HPV DNA testing as a screening tool for the detection of cervical neoplasia. Better understanding of the natural history of disease can help in developing effective and efficient public health programs in prevention for cervical cancer.

Full textACTH was purified from extracts of guinea-pig anterior pituitaries and characterized in terms of its amino acid composition and molecular weight using IS-MS and HPLC. Guinea-pig ACTH was found to have a similar activity to that of human ACTH with respect to the maximal steroid output of corticosterone and aldosterone. However, it proved to be slightly more potent in terms of the concentration which elicited half-maximal steroid secretion. Under the assay conditions used, guinea-pig ACTH does not seem to a superagonist as suggested by a previous study. Combining amino acid compositions, mass spectrometric data, and the recent determination of the cDNA sequence for guinea-pig ACTH, the identification of various purified biosynthetic derivatives of guinea-pig POMC was facilitated.
Joining peptide, a major product of POMC processing, was found in extracts of both anterior and neurointermediate lobes. The purified peptide corresponded exactly in amino acid composition and mass to the predicted structure established by the CDNA sequence. Also, by using ion-spray mass spectrometry, post-translational modifications of other products of intermediate lobe processing were observed. N- and O-acetylation of $ alpha$-melanotrophin, partial O-phosphorylation of corticotrophin-like intermediate lobe peptide and carboxyl-terminal amidation of $ beta$-melanotrophin were identified. Full textNonalcoholic fatty liver disease (NAFLD) has become a worldwide, chronic liver disease of increasing clinical significance. It is closely associated with the rising epidemics of obesity and insulin resistance. Up to 17% of the United States population may progress from the disease stage characterized as simple, benign steatosis to the more severe, inflammatory stage of nonalcoholic steatohepatitis (NASH). This progression occurs through 2nd 'hits' of increased oxidative stress and inflammation to a liver that has been sensitized by lipotoxic stress. NASH is also characterized by increased collagen deposition resulting in fibrosis and architectural rearrangement of the liver. Progressive NAFLD is currently recognized as an important contributor to the development of cryptogenic cirrhosis and subsequent liver-related mortalities (estimated at 30-40% in these patients).
The pathological progression of NAFLD, as described by the 'two hit' hypothesis, characterizes the different stages of liver injury. However, the mechanism(s) responsible for the progression to NASH are unknown. Profiling global gene expression and metabolite patterns in human liver samples representing the full spectrum of progressive human NAFLD may reveal potential mechanisms of progressive disease. Human liver samples representing each stage of NAFLD progression were analyzed by methodologies such as high-throughput microarrays, high resolution mass spectrometry, and protein immunoblot techniques. Bioinformatics tools and gene expression/regulation database software were utilized in several studies to characterize the altered hepatic profiles of these patients.
Hepatic transcriptomic profiles of ADME (absorption, distribution, metabolism and elimination) and ER (endoplasmic reticulum) stress response genes exhibited initiated hepatoprotective responses in patients with NASH. The endogenous pathways of BA (bile acid) synthesis and BCAA (branched chain amino acid) metabolism also showed evidence of coordinately regulated alterations in response to disease-induced stress in NASH. The transcriptional regulation of the investigated pathways was confirmed by transcription factor binding sites enrichment analysis. The collective response to hepatic stress in human NAFLD, demonstrates a coordinated, hepatoprotective intent that may be utilized for future therapeutics in the battle against progressive liver disease.
