Abstract:
The field of wildlife management has been on a collision course with societal values regarding animal use for some time. Although wildlife populations are still managed largely under the “North American model of wildlife conservation” to accommodate consumptive uses, many people, often with different concerns, want to be heard in decisions about wildlife. These human dimensions present a challenge to wildlife management, a field in which policy has been generally driven by experts. This research used several public engagement methods to understand broader attitudes towards wildlife management and how to incorporate them into policy. Participants with varying levels of wildlife experience in British Columbia, Canada, were asked in online and telephone surveys for their attitudes towards, and acceptance of, specific wildlife activities and management practices. Findings indicate a gap between public and expert opinions on invasive (e.g., relocation) and lethal management practices (e.g., problem bears, predator control, protection of endangered species). Problems also emerged with the public view of wildlife feeding. This led to a review of its motivations, types and consequences, and an evaluative framework was proposed to assess when feeding is acceptable. When asked to rate the harm to wildlife caused in various ways (hunting, vehicle collisions, pollution, etc.), experts and the public largely agreed on the relative importance of harms, indicating considerable potential for finding common ground between conservation-oriented and welfare-oriented citizens. However, some current management practices, especially those involving killing animals, lacked broad public support and may be improved upon with public participation in policy development, especially with women, urban residents, those with low wildlife engagement and animal-protectionist values. This could take the form of public polling or increased representation on decision-making committees. Societal expectations for managing human-wildlife interactions in BC documented by this research include ensuring that actions have an appropriate conservation purpose, are controllable, use humane methods, and appear fair to both people and wildlife. Including a broader public, educating both experts and the public on issues of humaneness, and strengthening wildlife and animal protection laws and enforcement, may serve to better align wildlife policy with societal values.

Abstract:
Einführung - Periphere Blutmonozyten spielen eine Rolle in der Pathogenese der Arteriosklerose. Bei spontan hypertensiven Ratten wurden im Vergleich zu normotonen Wistar-Kyoto-Ratten signifikant erhöhte Zahlen aktivierter Monozyten beobachtet [Liu 1996]. Wir untersuchten Monozyten von Patienten mit essentieller Hypertonie und von gesunden Probanden hinsichtlich möglicher Unterschiede in der Aktivierung anhand ihrer Adhäsion an von uns kultivierte (HAEC) und isolierte (HSVEC, HUVEC, HUAEC) humane Endothelzellen. Wir bestimmten die Adhäsionsmoleküle ICAM-1, VCAM-1 sowie E-Selektin und analysierten die Superoxidfreisetzung von humanem Endothel und Monozyten. Methoden - Humane periphere Blutmonozyten wurden mittels Dichtegradienten-zentrifugation und Plastikadhärenz isoliert und mit LPS, Angiotensin II (Ang II), und Ang II nach Vorinkubation mit dem AT1-Antagonisten Eprosartan stimuliert. Die Monozyten wurden auf einschichtigem Endothel ausgesät und die Adhäsion als Prozentsatz der initial gesäten Zellen erfasst. Die durch PMA induzierte Superoxidfreisetzung des Endothels oder der Monozyten wurde mittels Chemilumineszenz bestimmt. Ergebnisse - Monozyten von Patienten mit essentieller Hypertonie hefteten sich im Vergleich zu gesunden Probanden spontan und nach Stimulation mit Ang II signifikant verstärkt an HAEC und HUVEC-Monolayer an. Die Spiegel von ICAM-1 und VCAM-1 waren bei Patienten mit arterieller Hypertonie im Vergleich zu den gesunden Kontrollpersonen signifikant erhöht. Die Chemilumineszenzaktivität postkonfluenter Endothelzellen erhöhte sich nach Stimulation mit Ang II im Vergleich zur Messung ohne vorherige Stimulierung. Nach Stimulation mit PMA oder mit Ang II wurden bei Hypertonikern signifikant höhere Werte für die Chemilumineszenzaktivität der Monozyten gemessen als bei gesunden Kontrollpersonen. Schluss - Mit diesen Versuchen an humanen Monozyten und Endothelzellen wurde ein weiterer Beweis für die Aktivierung der Monozyten von Patienten mit essentieller Hypertonie erbracht. Meine Ergebnisse unterstützen die Sicht einer Monozytenbeteiligung an der Pathogenese atherosklerotischer Läsionen, die mit arterieller Hypertonie in Zusammenhang stehen.
Introduction - Peripheral blood monocytes are involved in the pathogenesis of atherosclerosis. Significantly elevated numbers of activated monocytes were observed in spontaneously hypertensive rats compared to those in normotensive Wistar-Kyoto rats [Liu 1996]. We isolated and cultivated human endothelial cells and examined monocytes from patients with arterial hypertension and healthy volunteers to identify possible differences in their adhesion behavior to human endothelial cells (HAEC, HSVEC, HUVEC, HUAEC). We determined the levels of ICAM-1, VCAM-1 and E-selectin, and we analyzed superoxide release by human endothelium and human monocytes. Methods - Peripheral blood monocytes were isolated by density gradient centrifugation and plastic adherence. Subsets of the samples were stimulated with LPS, Angiotensin II, Angiotensin II following preincubation with the AT1-antagonist eprosartan or left without a stimulant. After incubation, monocytes were seeded onto confluent monolayers of human aortic endothelial cells and the adhesion was determined as the percentage of the initially seeded cells. Oxygen species release induced by PMA was analyzed for endothelium and monocytes in suspension by chemiluminescence. Results - Peripheral blood monocytes of patients with essential hypertension performed a significantly increased spontaneous adhesion and adhesion following stimulation with Angiotensin II to HAEC- and HUVEC-monolayers. Levels of human soluble adhesion molecules ICAM-1 and VCAM-1 were significantly raised in hypertensive patients. Chemiluminescence activity of post confluent endothelial cells was increased after stimulation with Angiotensin II compared to the measurement before stimulation. Following stimulation with PMA or Angiotensin II, significantly higher chem-iluminescence levels were measured in hypertensive patients compared to healthy volunteers. Conclusion - These data indicate that monocytes of patients with essential hypertension may be preactivated. My results support the view of a monocyte involvement in the pathogenesis of atherosclerotic lesions that are related to arterial hypertension.

Where is technology heading? And how will our behaviors towards these new innovations look? This project questions the direction of ”beyond smart” products, through scenarios within our everyday life. The work is both critical and speculative. Speculative in the sense that it is speculating in how a future scenario with ”beyond smart” products would look like. And by beyond smart I refer to the fact that the products would posses extreme features. Such as complexity and irationalism. But also to be spontaneous and self aware. At the same time the work is questioning, and perhaps criticising, if this sort of development really is beneficial for us or not. If we would turn to technology and smart products for a solution to our desires and problems, how would our human relationships towards each other look? And also how would we interact with these future products and what would the consequences of that be?

Abstract:
I have in this thesis composed music for a concert using a string quartet and a jazz quartet combined, which for me is an entirely new group of instruments put together. This thesis follows my process writing the compositions from the beginning until the end. The purpose of the thesis has been to explore how to integrate a string quartet and a jazz quartet combined in my compositions.  I have, while composing, examined how the double bass relates to the instrumentation and how the bass could be used in melodies, voicings and bass lines. The goal has also been to perform these compositions during my graduation concert.   In the thesis I describe the different methods I have used to fulfil the purpose; “learning-by-doing”, inspirations and interviews, and I tell the reader about the experiences made and the knowledge I have acquired by using these. Furthermore, I give an account of the concert-material made and explain how I merged the instruments together in the compositions. Towards the end of my thesis, I share the insights and understandings that has arisen along the composing process. I ascertain that I by doing the project have developed as a composer as well as a double bass player.  This thesis could be interesting for anyone who is involved or interested in music, composing, double bass or the creative process.

Konserten tog plats den 2 mars på Kungliga Musikhögskolan i Stockholm. Vi var en ensemble på åtta personer och musiken jag hade skrivit till den här konserten fokuserade på att interagera jazzkvartett och stråkkvartett. Temat på konserten var "Humane Strings" och ordet Humane betyder några av de fina egenskaper vi människor har. Så som; att vara medkännande, vänlig, godhjärtad, generös och att vara öppenhjärtad. Så ofta glömmer vi bort våra vackra egenskaper och vi tänker för mycket på de negativa karaktärsdragen vi har. Min idé var att om vi kunde förtstärka våra goda egenskaper så kanske världen skulle vara en bättre plats. Detta tema försökte jag gestalta i musiken skriven för konserten.

Die Verpackung und Spaltung konkatemerer DNA ist ein essentieller Prozeß bei der Reifung von Virionen. Die an diesem Prozess maßgeblich beteiligten Proteine werden als Terminase bezeichnet. Die Inhibition der HCMV Terminase bietet einen attraktiven alternativen Ansatzpunkt für die antivirale Therapie. Zur Inhibition der Terminase Aktivität wurden die neuen Benzimidazol D-Ribonukleosid Derivate BTCRB und Cl4RB auf ihre antivirale Wirkung analysiert. Die neuen Benzimidazol D-Ribonukleosid Derivate BTCRB und Cl4RB zeigten sowohl gegen den HCMV Laborstamm AD169 als auch gegen klinische HCMV-Isolaten eine Wirkung. Weiterhin wurde die Wirksamkeit der Substanzen auf anderen Herpesviren getestet. Interessanterweise zeigten BTCRB und Cl4RB sowohl einen Effekt gegen Varicella-Zoster-Virus (VZV) als auch Ratten-Cytomegalovirus (RCMV), wohingegen der Effekt gegen Herpes-simplex-Virus Typ-1 (HSV-1) und Maus-Cytomegalovirus (MCMV) gering war. Infolgedessen eignen sich beide Substanzen BTCRB und Cl4RB als attraktive alternative Inhibitoren für die weitere Entwicklung einer antiviralen Therapie.
DNA packaging is the key step in viral maturation and involves binding and cleavage of viral DNA containing specific DNA-packaging motifs. This process is mediated by a group of specific enzymes called terminase. The development for an inhibitor of HCMV terminase would be of great value, because it would act subsequent to DNA synthesis and block the first steps in viral maturation. Therefore we characterized two inhibitors targeting the HCMV terminase, 2-bromo-4,5,6-trichloro-1-(2,3,5-tri-O-acetly-ß-D-ribofuranosyl) benzimidazole (BTCRB) and 2,4,5,6-tetrachloro-1-(2,3,5-tri-O-acetly-ß-D-ribofuranosyl) benzimidazole (Cl4RB). By using viral plaque formation, viral yield, viral growth kinetics and electron microscopy, we demonstrated that two compounds BTCRB und Cl4RB are highly active against HCMV AD 169 and HCMV clinical isolates. In addition, the antiviral effect on other herpesviruses was determined. Interestingly BTCRB was active all tested herpesviruses. The best effects were observed on VZV-and RCMV-infected cells. Therefore new compounds might be promising attractive compounds for antiviral therapy in the future.

Abstract:
Worldwide, an estimated 9.1 billion birds may need to be killed on farm each year. As of January 2013 the use of manual cervical dislocation (MCD) as a killing method for poultry on-farm has been heavily restricted through new EU legislation (EC 1099/2009) on the Welfare of Animals at the Time of Killing, following reported welfare concerns. The method by which birds are killed on farm is crucial to poultry welfare on a large scale. The overall aim of this project was to design a mechanical device conforming to the new legislation to kill poultry humanely on-farm and provide a competitive replacement for MCD. Following a survey and a literature review, four mechanical devices were designed and prototyped: Modified Armadillo (MARM); Modified Pliers (MPLI); Modified Rabbit Zinger (MZIN) and a Novel mechanical cervical dislocation glove (NMCD). The devices were tested for killing efficacy in three laboratory experiments, assessing their performance in poultry cadavers (Study 1), anaesthetised birds (Study 2) and live conscious birds (Study 3). The reliability and welfare impact of the devices, along with comparisons with a control method (MCD) were evaluated via post-mortem analysis, reflex and behaviour durations, and characteristics of electroencephalography (EEG) analysis. Due to consistently high kill success rates and rapid loss of reflexes, as well as short durations of EEG activity indicating consciousness across three laboratory experiments, the NMCD device was shown to have the most promise as a mechanical device to be used as an alternative to MCD for poultry stock-workers and keepers. The final experiment explored the user-reliability and practicality of the NMCD device in two relevant commercial environments (a layer hen farm and a broiler farm). When applied by multiple users, the NMCD device did not match the killing performance of MCD, however it did show promise and the study highlighted the need for further refinement in the training protocol in order to encompass the wide variation in MCD techniques and experience. The result of this project is a novel on-farm mechanical killing device, which shows great potential in laboratory experiments and competed with the traditional MCD method in commercial environments. Further training refinements are required in order to develop the device into a marketable product which any individual could purchase and use as a humane method for killing poultry.

The current study investigated the influence of intensified daily interactions with living animals in the classroom on the development of empathy among Japanese children. Specifically, the study examined (a) the effect of introducing animals into the classroom on children's empathic behaviors and attitudes, and (b) the generalization of this animal-directed empathy to humans.

Abstract:
Die Magnetresonanztomografie (MRT) ist ein wichtiges diagnostisches Mittel um kartilaginäre Läsionen frühzeitig erkennen und effektiv behandeln zu können. MRT Untersuchungen erlauben die Diagnose von kartilaginären Schäden ohne den Einsatz von radioaktiver Strahlung, wie sie im Röntgen oder CT Verwendung findet. Obwohl MRT Untersuchungen als generell sicher gelten, gibt es bisher keine eindeutigen Untersuchungen über die Auswirkung von hochfrequenten, starken Magnetfeldern auf humane Zellen. Die Wirkung von Magnetfeldern, wie sie in der MRT Diagnostik verwendet werden, auf Chondrozyten und den unter anderem von Chondrozyten gebildeten Knorpel ist bis jetzt nur unzureichend untersucht. Ziel der vorliegenden Arbeit war es daher, die Auswirkungen eines 3 Tesla Magnetfeldes auf die Proliferationsrate und Genexpression humaner Chondrozyten zu evaluieren. Dafür wurden humane Chondrozyten einer 3 T MRT Sequenz ausgesetzt, wie sie zur Untersuchung am Knie verwendet wird. Die Proliferationsrate 1, 5 und 10 Tage nach Exposition wurde mit Hilfe eines WST-1 Proliferations Assay bestimmt und mit jener einer Kontrollgruppe verglichen. Gleichzeitig wurde die RNA Expression von Proteinen bestimmt, die entweder Chondrozyten- oder Apoptosespezifisch sind. Beim Betrachten der Ergebnisse zeigte sich, dass Chondrozyten, die einem 3 T Magnetfeld ausgesetzt waren, 10 Tage nach Exposition eine im Vergleich zur Kontrollgruppe signifikant gesteigerte Proliferationsrate aufwiesen. Apoptose spezifische RNA wurde an den Tagen 1 und 5 nach Exposition im Vergleich zur Kontrollgruppe geringer exprimiert
Introduction: Magnetic resonance imaging is an important diagnostic tool used to evaluate the size of cartilage damage in chronic osteoarthritis or after traumatic injuries. While multiple studies are involved in evaluating effects of weak EMFs on human cells, there is very little evidence concerning the effects of varying magnetic fields as they are used in MRI. Aim of this study was therefore to expose human chondrocytes to a 3 T MRI used for musculoskeletal examinations and determine the effects on proliferation and gene expression levels after 1, 5 and 10 days. Methods: Human chondrocytes from healthy donors were exposed to a 3 T varying magnetic field used for knee MRI examination. Proliferation rate was evaluated using a WST-1 proliferation assay. RNA expression of chondrocyte specific as well as apoptosis related proteins were examined 1, 5 and 10 days after exposure to MRI. Results: Human chondrocytes exposed to a musculoskeletal MRI showed an increased proliferation rate 10 days after exposure compared to non-exposed cells. Initial proliferation rate was not significantly decreased 1 day after exposure. PARP was down-regulated 1 and 5 days after MRI. Conclusion: Using a 3 T MRI following a protocol of musculoskeletal examinations, an increased proliferation rate of human chondrocytes was observed 10 days following exposure. This might be due to a down-regulation of apoptosis related proteins which occured earlier after MRI stimulation. Further studies are needed to examine long-term effects of musculoskeletal MRI on cartilage.
