Since ralGDS contained a region highly homologous with the ras p21-interacting domain of RGL, we examined whether ralGDS could interact with ras p21. In the yeast two-hybrid system, ralGDS failed to interact with an effector loop mutant of ras p21. In insect cells, ralGDS made a complex with v-ras p21 but not with a dominant negative mutant of ras p21. ralGDS interacted with the GTP-bound form of ras p21 but not with the GDP-bound form in vitro. ralGDS inhibited both the GTPase-activating activity of the neurofibromatosis gene product (NF1) for ras p21 and the interaction of Raf with ras p21 in vitro. Abstract:
The F1 hybrid sterility in indica/japonica crosses is the major barrier in developing hybrid rice varieties between these two diverse germplasm. The sterility problem in japonica/indica hybrids can be overcome by using wide compatibility genes. The objective of this study was to identify wide compatibility varieties (WCVs) in some tropical japonica rice. Twenty five tropical japonica varieties as male parents were crossed with indica (IR64) and japonica (Akitakomachi) testers as female parents. The crosses were planted following a randomized complete block design with three replications. Varieties having average spikelet fertility of more than 70% with both the indica and japonica testers were rated as WCVs. Result from this study showed that six tropical japonica varieties were classified as WCVs, i.e., Cabacu, Grogol, Kencana Bali, Klemas, Lampung Lawer, and Napa. Hybrid sterility is caused by partial sterility of male and female gametes. The WCVs from the present study can be used in hybrid rice breeding program to solve hybrid sterility in indica/japonica hybrids.

P2P networks are a technology able to deliver real time and video-on-demand services over IP networks. Layered video coding techniques are being introduced due to their ability to deliver streams at different scales (temporal, spatial and SNR) that solve the heterogeneity problem. This eases transmission in the case of limited bandwidth, as the devices can pick and decode the minimum bit rate base layer. Existing work examines layered video in client-server scenarios. In contrast, this paper analyzes scalable coding H.264/SVC over P2P networks based on an SNR-temporal Codec. Due to the interdependency between the different SVC layers, issues of reliability and quality of experience arise unless proper measures are taken to protect the base layer. The authors explore the effectiveness of a combination of P2P strategies, for example, hybrid P2P architecture, P2P locality, and P2P redundancy, to assess the viability and benefits of scalable video coding over P2P. The resulting performance is compared with a state-of-the-art P2P TV platform.

Direct connection and reflective connection are commonly used in traditional Trojans communication model, which have some significant deficiencies in hiding, anti-tracking, and efficiency. To improve the aspects above in traditional Trojan horse communication model, the author proposed a hybrid communication model based on P2P technology. The model uses P2P intranet penetration protocol to build the channel between the controlled station and the control station, and uses P2P node servers for saving the Trojans status and transferring some data between the controlled and the control. The topology of networks for controller will be very flexible. Using the IP traceback technology will be difficult to track the controllers. Using UDP protocol in the hybrid communication model for data transmission will hide the network status. Control sites can work in distributed mode. By using the model, the hiding-ability, anti-traceability and communication efficiency will be greatly improved.

The results showed 25 Hybrid maize tested against Gibberella ear rot there are 7 Hybrid corn hybrids that are resistant to ear rot Kenia-2, NK33, Bima 3, Bima 10 Bima 12 Q, NEI-9008 and X8B649 (check resistance) and moderately resistance there are 9 hybrid that Kenia-3, DK979, NK22, P12, Gumarang, Srikandi putih, Lamuru and Sukmaraga and 9 hybrid to ear rot disease were reacted susceptible and highly susceptible. Whereas from 25 Hybrid maize tested against Diplodia ear rot there are 6 Hybrid corn hybrids resistant to ear rot disease is Kenia - 2, NK33, Bima 3, Bima 12 Q, NEI-9008 and X8B649 (check resistance) while that is moderately resistancet there are 11 hybrid that is Kenia-3, DK979, NK22, P29, Bisi 2, P12, Bima 10, Gumarang, Srikandi putih, Lamuru and Sukmaraga, while 8 hybrid others were reacted susceptible and highly susceptible.

Pure Peer to Peer (P2P) network requires enhancing transportation of chunk video objects to the proxy server in the mesh network. The rapid growth of video on demand user brings congestion at the proxy server and on the overall network. The situation needs efficient content delivery procedure, to the video on demand viewer from the distributed storage. In general scenario, if the proxy server does not possess the required video stream or the chunk of that said video, then the same can be smoothly and rapidly streamed to the viewer. This paper has shown that multitier mesh shaped hybrid architecture composed of P2P and mesh architecture increase the number of requests served by the dynamic environment in comparison with the static environment. Optimized storage finding path search reduces the unnecessary query forward and hence increases the size of content delivery to the desired location.

In this work, we develop a general methodology to assess the level of network awareness and friendliness of P2P-TV applications. The methodology is based on a combination of active and passive measurement techniques and can be applied to any P2P-TV system since it is designed to work considering such systems as a black-boxes. As an interesting case study, we then apply this methodology to PPLive, one of the most popular P2P-TV systems nowadays. Focusing on the video content distribution, we consider several per-path and per-peer metrics, investigating which of them mostly biases PPLive download preferences. Furthermore, in order to refine the picture of PPLive peer selection policy, we not only study the impact of different metrics in isolation, but also assess the joint impact of different metrics at the same time. Our main finding is that PPLive seems mainly bandwidth greedy, but does not show any preference toward peer proximity based on RTT delay; at the same time, our results also suggest that this choice alone may provide a nonnegligible level of geographical clustering among peers as a beneficial side effect.

ABSTRACT p27Kip1 (p27) is often inappropriately downregulated in aggressive human cancers. Although p27 can inhibit cyclin-dependent kinases (CDKs), low p27 does not always correlate with increased CDK activity. Furthermore, cells derived from p27−/− mice respond to antimitogens, maintain restriction point control, and do not deregulate CDKs. Thus, disruption of a p27 function other than CDK inhibition may contribute to the disease state. A yeast two-hybrid screen identified growth factor receptor-bound protein 2 (GRB2) as a p27 binding partner. We now demonstrate that p27 can inhibit GRB2 function by blocking its association with the guanine nucleotide exchange factor SOS. Endogenous p27 is rapidly exported from the nucleus to the cytoplasm in response to mitogen stimulation, where it binds GRB2 concomitant with a decrease in GRB2-associated SOS. As predicted, mitogen-stimulated p27−/− cells maintained their GRB2-SOS complexes for significantly longer. The Ras/mitogen-activated protein kinase pathway does not appear to be deregulated in cells lacking p27 despite excess GRB2-SOS, suggesting that additional control mechanisms are present. A transient-transfection approach was employed to show that p27 can inhibit Ras activation by targeting GRB2 and further revealed that the CDK and GRB2 inhibitory functions of p27 are separable and distinct. Thus, p27 downregulation may compromise control of Ras, one of the most common oncogenic events in human cancer.

A novel urushiol grafted Degussa P25 (urushiol/P25) hybrid nano-material was synthesized, and its structure was characterized. The urushiol/P25 hybrid nanomaterial showed high photoactivity under visible light irradiation (λ  420 nm). Bisphenol A (BPA) can easily approach to P25 surface through the hydrophobic channel between alkyl groups in urushiol. The reaction rate constant of BPA degradation increased by 4.9 times by using 0.5% urushiol/P25 as catalyst than by using bare P25. Radical quenching experiments proved that the superoxide radical anion (O2•−) mediated oxidation pathways were dominant for BPA degradation.

Most of the existing peer-to-peer (P2P) content distribution schemes carry out a random or rarest piece first content dissemination procedure to avoid duplicate transmission of the same pieces of data and rare pieces of data occurring in the network. This problem is solved using P2P content distribution based on network coding scheme. Network coding scheme uses random linear combination of coded pieces. Hence, the above-stated problem is solved easily and simply. Our proposed mechanism uses network coding mechanism in which several contents having the same message are grouped into different groups and coding operation is performed only within the same group. The interested peers are also divided into several groups with each group having the responsibility to spread one set of contents of messages. The coding system is designed to assure the property that any subset of the messages can be utilized to decode the original content as long as the size of the subset is suitably large. To meet this condition, dynamic smart network coding (DSNC) scheme is defined which assures the preferred property, then peers are connected in the same group to send the corresponding message, and connect peers in different groups to disseminate messages for carrying out decoding operation. Moreover, the proposed system is readily expanded to support topology change to get better system performance further in terms of reliability, link stress and throughput. The simulation results prove that the proposed system can attain 20–25% higher throughput than existing systems, good reliability, link failure and robustness to peer churn.

The combination of additive manufacturing and conventional metal forming processes provides the possibility for improvements of forming efficiency and flexibility. Substrate preheating is an implementable technique to improve the interface adhesion properties of the hybrid forming method. The present experiment investigates the adhesion of additive manufactured 316 L steel on P20 and 1045 steel substrates under two substrate temperatures, and the geometrical characterization, interfacial microstructure and mechanical property of the hybrid specimens were compared. As a result, it was found that the ratio of deposition height to the width was reduced and the width was increased under substrate preheating. Tensile results show that the ultimate strength of 1045 and 316 L hybrid specimens was obviously increased, while the properties of hybrid specimens P20 and 316 L were similar, under different substrate temperature conditions. For the hybrid specimens with the metallurgically bonding characteristic, the tensile properties can reach the level of 316 L depositioned specimens fabricated by laser metal deposition (LMD). Furthermore, substrate preheating had little effect on the microstructure of the laser metal deposition zone, and significant influence on the microstructure of the heat affected zone, which was reflected in the difference of the hardness distribution.

The objectives of this study were to determine the nitrogen fixing bacteria compatible with phosphate solubilizing bacteria (PSB), and using those bacteria to increase physiological seed quality and seedling growth of maize female parent. The research consisted of laboratory and field experiments. Laboratory experiment for the isolation and identification of rhizobacteria resulted in 25 Azotobacter and 29 Actinomycetes non-pathogenic isolates capable of fixing nitrogen and PSB selected for compatibility tests were AB3, B28, P12, P14, P24, and P31. The compatibility test showed 25 pairs of BPF with Azotobacter and 16 pairs of BPF with Actinomycetes were mutually compatible. The BPF pair with Azotobacter or Actinomycetes P24-AzL7, P24-AzL9, B28-AcCKB4, P24-AcCKB9, P24-AcCKB20, and P24-AcCKW5 were able to increase the vigor index of hybrid maize female parent seed. Field experiment was arranged in a split plot design with three replications. The main plot was dosage of N-P fertilizer (0%, 25%, 50%, 75%, and 100% of recommendation dosage), and the subplot was 12 rhizobacteria treatments selected from 25 compatible pairs of BPF with Azotobacter and 16 pairs of BPF with Actinomycetes and 1 control. The application of compatible pairs of bacteria had a significant effect on plant height, the number of leaves at 3 and 4 weeks after planting and plant dry weight. However, the best treatment i.e. B28-AcCKB4 was not significantly different with the nutrient broth treatment (as control).<br /><br /><br />

Sclerotinia basal stalk rot (BSR) is a serious fungal disease that reduces yield of global sunflower (Helianthus annuus L.) production. Because limited chemical and biological controls of BSR are available and the present-day hybrids lack sufficient resistance, identification of new sources of resistance is needed to manage the disease in the future. A total of 59 cultivated oilseed sunflower accessions from the Agricultural Research Council, Grain Crops Institute, Potchefstroom, South Africa sunflower collection were evaluated for resistance to BSR in artificially inoculated field trials. Nine accessions from the South African sunflower collection were identified with a disease incidence less than or equal to the moderately resistant sunflower oilseed hybrid. These lines can be used in breeding programs to introgress the genes for resistance to Sclerotinia BSR into other adapted lines, providing a more efficient, durable, and environmentally friendly host plant resistance.

Abstract In this article, I identify and describe multimodal hybrid metaphors—the conceptual representation of two elements represented as merged into a new single ‘gestalt’—represented by the machine and human body domains in “Tetsuo: The Iron Man” (鉄男: Tetsuo), a Japanese avant-garde film. Since “Tetsuo: The Iron Man” portrays the genesis of a man whose body becomes a human-machine hybrid, I explore to what extent this film can act as an example of how hybrid metaphors are conveyed. In line with the ideological function of metaphors, where the use of alternative metaphors may produce different meanings and potentially have different effects on the recipient, I also try to interpret how these hybrid metaphors reveal information about the contemporary Japanese society. Specifically, the ideological analysis considers how the notion of ‘artificial’ and the social phenomena of misogyny, homophobia and social deviance are held to characterise the post-World War II Japanese culture.

In the southeastern United States, there are limited options for crops that can be grown in rotation with peanut (Arachis hypogaea). Pearl millet (Pennisetum glaucum) has potential as a grain crop, and some hybrids have shown resistance to the peanut root-knot nematode (Meloidogyne arenaria), the primary nematode pest of peanut in this region. The objective of this study was to determine whether pearl millet reduces M. arenaria when planted in rotation with peanut. The experiment was arranged as a randomized, complete-block design with six replications. The rotations were peanut following either 2 years of corn, HGM-100 pearl millet, or TifGrain 102 pearl millet. There were two staggered sequences of each rotation so that a cycle was completed in 2004 and in 2005. Pearl millet did not increase either stem rot or Rhizoctonia limb rot in peanut. In both years, root galling from M. arenaria was lower on peanut following TifGrain 102 (4.6 on a scale of 0 to 10) and corn (4.9) than following HGM-100 (7.5). Peanut yields in 2004 were low and unaffected by the preceding rotation crop; however, in 2005, yields were greater in peanut following 2 years of TifGrain 102 (2320 kg/ha) and corn (2504 kg/ha) than following HGM-100 (1821 kg/ha). The lower yields following HGM-100 were likely due to greater populations of M. arenaria that had developed on the susceptible pearl millet hybrid. Economic analyses showed greater returns above variable costs from the grain crops than from the peanut crops. We conclude that the resistant pearl millet hybrid TifGrain 102 is as effective as corn in limiting population increase of M. arenaria and in enhancing peanut yield compared to the susceptible pearl millet hybrid, HGM-100. Additional research is needed to improve the profitability of pearl millet, and nematode resistance should be an important component of crop improvement programs. ABSTRACT Human immunodeficiency virus type 1 (HIV-1) gp41 plays a critical role in the viral fusion process, and its N- and C-terminal heptad repeat domains serve as important targets for developing anti-HIV-1 drugs, like T-20 (generic name, enfuvirtide; brand name, Fuzeon). Here, we conducted a yeast two-hybrid screening on a human bone marrow cDNA library using the recombinant soluble gp41 ectodomain as the bait and identified a novel gp41 core-binding molecule, designated P20. P20 showed no homology with a current HIV fusion inhibitor, T-20, but had sequence homology to a human protein, troponin I type 3 interacting kinase (TNNI3K)-like protein. While it could bind to the six-helix bundle core structure formed by the N- and C-terminal heptad repeats, P20 did not interrupt the formation of the six-helix bundle. P20 was effective in blocking HIV-1 Env-mediated syncytium formation and inhibiting infection by a broad spectrum of HIV-1 strains with distinct subtypes and coreceptor tropism, while it was ineffective against other enveloped viruses, such as vesicular stomatitis virus and influenza A virus. P20 exhibited no significant cytotoxicity to the CD4+ cells that were used for testing antiviral activity. Among the 11 P20 mutants, four analogous peptides with a common motif (WGRLEGRRT) exhibited significantly reduced anti-HIV-1 activity, suggesting that this region is the critical active site of P20. Therefore, this peptide can be used as a lead for developing novel HIV fusion inhibitors and as a probe for studying the membrane-fusogenic mechanism of HIV.

Abstract The involvement of SMALL AUXIN-UP RNA (SAUR) proteins in leaf senescence has been more and more acknowledged, but the detailed mechanisms remain unclear. In the present study, we performed yeast two-hybrid assays and identified SAUR49 as an interactor of SENESCENCE SUPPRESSED PROTEIN PHOSPHATASE (SSPP), which is a PP2C protein phosphatase that negatively regulates Arabidopsis leaf senescence by suppressing the leucine-rich repeat receptor-like protein kinase SENESCENCE-ASSOCIATED RECEPTOR-LIKE KINASE (SARK), as reported previously by our group. The interaction between SAUR49 and SSPP was further confirmed in planta. Functional characterization revealed that SAUR49 is a positive regulator of leaf senescence. The accumulation level of SAUR49 protein increased during natural leaf senescence in Arabidopsis. The transcript level of SAUR49 was upregulated during SARK-induced premature leaf senescence but downregulated during SSPP-mediated delayed leaf senescence. Overexpression of SAUR49 significantly accelerated both natural and dark-induced leaf senescence in Arabidopsis. More importantly, SAUR49 overexpression completely reversed SSPP-induced delayed leaf senescence. In addition, overexpression of SAUR49 reversed the decreased plasma membrane H+-ATPase activity mediated by SSPP. Taken together, the results showed that SAUR49 functions in accelerating the leaf senescence process via the activation of SARK-mediated leaf senescence signaling by suppressing SSPP. We further identified four other SSPP-interacting SAURs, SAUR30, SAUR39, SAUR41 and SAUR72, that may act redundantly with SAUR49 in regulating leaf senescence. All these observations indicated that certain members of the SAUR family may serve as an important hub that integrates various hormonal and environmental signals with senescence signals in Arabidopsis.

Power to gas facilities (P2G) could absorb excess renewable energy that would otherwise be curtailed due to electricity network constraints by converting it to methane (synthetic natural gas). The produced synthetic natural gas can power gas turbines and realize bidirectional energy flow between power and natural-gas systems. P2G, therefore, has significant potential for unlocking inherent flexibility in the integrated system, but also poses new challenges of increased system complexity. A coordinated operation strategy that manages power and natural-gas network constraints together is essential to address such challenges. In this paper, a novel low-carbon economic environmental dispatch strategy is presented considering all the constraints in both systems. The multi-objective black-hole particle swarm optimization algorithm (MOBHPSO) is adopted. In addition to P2G, a gas demand management strategy is proposed to support gas flow balance. A new solving approach that combines the effective redundancy method, trust region method, and Levenberg-Marquardt method is proposed to address the complex coupled constraints. Case studies that use an integrated IEEE 39-bus power and Belgian high-calorific 20-node gas system demonstrate the effectiveness and scalability of the proposed model and optimization method. The analysis of dispatch results illustrates the benefit of P2G for the wind power accommodation, and low-carbon, economic, and environmental improvement of integrated system operation.

<p>Maize (Zea mays L.) is the second most important cereal crop of Indonesia after rice. The productivity of corn in Central Java province in 2015 was increased that considered as relatively low. Maize productivitiy needs to be improved to supply the requirement of people through of plant spacing and use the hybrid maize varieties. The aims of study was to determine the effectiveness level of plant spacing, to test the best hybrid maize varieties, and test the combination of two treatments to optimize the yield potential hybrid maize. This study has been conducted from June until October 2017, at Desa Srimartani, Kecamatan Piyungan, Kabupaten Bantul, Daerah Istimewa Yogyakarta. This study was arranged in Split Plot Design consisting of two factors that have been researched, varieties (ABCD, EFGH, NK6172, P27 and DK77) and plant spacing (70 x 20 cm, 75 x 20 cm, and 75 x 25 cm). The results show that treatment of varieties mostly can increase all of variable analysis but there is no interaction on both treatment except plant height, fresh and dry biomass weight. Variable of plant height, tasseling and silking, cob weight with husk, cob weight without husk, kernel seeds weight per plot, 100 seeds weight, yields per hectare show that the highest average is variety of P27. While treatment of plant spacing has no effect because the interval between treatments plant spacing is too close therefore it shows insignificantly different result. The highest dry kernel product on variety treatment P27 (8,82 ton ha-1), followed by DK77 (8,53 ton ha-1), ABCD (8,50 ton ha1), NK6172 (8,34 ton ha-1), and EFGH (7,13 ton ha-1).</p>

Pig breeding is an industry the development of which makes it possible to provide an increase in accelerated meat production due to the precocity of animals, feed recoupment by weight gain, maximum adaptability to the conditions of industrial production at complexes. This makes it possible to provide an uninterrupted supply of the population with products in the shortest possible time. Final assessment of meat productivity is set after slaughter of animal on the basis of quantitative and qualitative carcass parameters, which are subdivided into slaughter and meat traits. Performance of pigs is determined by quantity of the products obtained from them and suitable to be used in food products. The life determination of meat traits makes it possible to carry out their preliminary assessment. The aim of the research – to determine the effect of hybrid boars (Duroc × Pietrain) on the progeny’s meat performance. A graded cutting and boning of five left half-carcasses of pigs of each genotype was conducted to determine the morphological composition and meat content in carcasses. Analysing the data, it should be noted that animals of genotype (BLW×Y)×(D×P) by slaughter traits surpassed their coevals in the control group for pre-slaughter live weight by 2.0 kg (2%), weight of pair carcass – by 3.8 kg (5.4%), slaughter output – by 6.3 p.p., respectively. Young animals of genotype (BLW×Y)×(D×P) surpassed coevals of other experimental groups on the pre-slaughter live weight by 0.6-1.6 kg, or 0.6-1.6%, by weight of pair carcass – by 0.4-1.1 kg, or 0.6-1.6%, by slaughter output – by 0.9-2.4 p.p. respectively. The variation ratio for slaughter yield ranged 1.56-3.86%, indicating the uniformity of index in all the experimental groups. The degree of variability in pair carcass weight was within 0.57-4.00%. A high variability of this trait was noted in the control group animals of genotype (BLW×BM)×D, and was 4.00%. It was determined that the highest percentage in composition of chilled carcasses is taken by shoulder cut. So, for the young animals of genotype (BLW×Y)×(D×P), it takes 34.17%, that is 0.07 p.p. higher than in the control group and 0.21-0.27 p.p. than in other experimental groups. The yield of spinal rib cut was higher with combination of the I experimental group and was 31.90%, that is 0.13 p.p. higher than with the control group hybrids and 0.3-0.9 p.p. compared with the other experimental genotypes. As for specific weight of the rear third part of carcass, the highest figure was observed in the animals of combination (BLW×Y)×(D×P) – 34.83%, that is 0.7 p.p. higher compared with animals in the control group. According to this indicator, young animals of genotype (BLW×Y)×(D×P) surpassed their coevals of the other experimental groups by 0.39-0.63 p.p. respectively. The most valuable part of pork is the meat, consisting mainly of muscle tissue, concentrated in the skeletal muscle area. The definition of “meat” includes muscle, fat and connective tissue. The most important and useful part of the muscle tissue is proteins. Adipose (fat) tissue is a type of loose connective tissue, the cells of which are filled with fatty inclusions. The fat in pigs is deposited under the skin almost uniformly (depending on the genotype). Uniform deposition of adipose tissue between the muscle fiber bundles gives “marbling” to meat, thus enhancing its taste, nutritional and culinary values. When analysing morphological content of chilled half-carcass of pigs of different genotypes, it was determined that the young animals of genotype (L×Y)×(D×P) by meat yield significantly surpassed the control group coevals by 1.8 p.p. (R≤0.001) and coevals of other experimental groups – by 0.7-1.1 p.p., respectively. Carcasses of pigs of genotypes (BLW×BM)×(D×P) and (BLW×Y)×(D×P) also surpassed the control group of animals of genotype (BLW×BM)×D on the meat yield by 1.1-0.7 p.p. (P≤0.05), respectively. Carcasses of pigs of genotype (L×Y)×(D×P) had less lard in the body: so, the lard content in carcass was by 1.5 p.p. lower compared with the control group animals of (BLW×BM)×D (P≤0.01). Content of bones in carcasses of pigs of all the groups was within 11.5-11.7% with a trend towards reduction of this indicator in animals of experimental groups by 0.1-0.2 p.p. (P≤0.05). For the comparative assessment of carcasses an indicative criterion is the ratio of tissues: meat/bone – “meat content index” and meat/fat – “lean meat index”. Having analysed the data we can say that the highest index of “meat content” was obtained in animals of genotype (L×Y)×(D×P) – 5.62. When defining index of “lean meat”, animals of genotype (L×Y)×(D×P) surpassed all the groups of animals with 3.81 index, which exceeded the index for the animals in the control and experimental groups. Conclusions. It was determined that hybrid young animals of genotype (BLW×Y)×(D×P) surpassed their coevals of other experimental groups on the pre-slaughter live weight by 0.6-1.6 kg, or 0.6-1.6% by weight of pair carcass – by 0.4-1.1 kg or 0.6-1.6%, on slaughter output – by 0.9-2.4 percentage points, respectively. On the specific weight of rear third part of carcass the highest figure was observed in the animals of combination (BLW×Y)×(D×P) – 34.83%, that is 0.7 p.p. higher compared with animals in the control group. On this trait young animals of genotype (BLW×Y)×(D×P) surpassed their coevals of other experimental groups by 0.39-0.63 percentage points, respectively. Progression through the G1/S transition commits cells to synthesize DNA. Cyclin dependent kinase 2 (CDK2) is the major kinase that allows progression through G1/S phase and subsequent replication events. p27 is a CDK inhibitor (CKI) that binds to CDK2 to prevent premature activation of this kinase. Speedy (Spy1), a novel cell cycle regulatory protein, has been found to prematurely activate CDK2 when microinjected into Xenopus oocytes and when expressed in mammalian cells. To determine the mechanism underlying Spy1-induced proliferation in mammalian cell cycle regulation, we used human Spy1 as bait in a yeast two-hybrid screen to identify interacting proteins. One of the proteins isolated was p27; this novel interaction was confirmed both in vitro, using bacterially expressed and in vitro translated proteins, and in vivo, through the examination of endogenous and transfected proteins in mammalian cells. We demonstrate that Spy1 expression can overcome a p27-induced cell cycle arrest to allow for DNA synthesis and CDK2 histone H1 kinase activity. In addition, we utilized p27-null cells to demonstrate that the proliferative effect of Spy1 depends on the presence of endogenous p27. Our data suggest that Spy1 associates with p27 to promote cell cycle progression through the G1/S transition.

Abstract:
Oligonucleotide-directed mutagenesis was used to construct chimeric cDNAs that encode the extracellular and transmembrane domains of the vesicular stomatitis virus glycoprotein (G) linked to the cytoplasmic domain of either the immunoglobulin mu membrane heavy chain, the hemagglutinin glycoprotein of influenza virus, or the small glycoprotein (p23) of infectious bronchitis virus. Biochemical analyses and immunofluorescence microscopy demonstrated that these hybrid genes were correctly expressed in eukaryotic cells and that the hybrid proteins were transported to the plasma membrane. The rate of transport to the Golgi complex of G protein with an immunoglobulin mu membrane cytoplasmic domain was approximately sixfold slower than G protein with its normal cytoplasmic domain. However, this rate was virtually identical to the rate of transport of micron heavy chain molecules measured in the B cell line WEHI 231. The rate of transport of G protein with a hemagglutinin cytoplasmic domain was threefold slower than wild type G protein and G protein with a p23 cytoplasmic domain, which were transported at similar rates. The combined results underscore the importance of the amino acid sequence in the cytoplasmic domain for efficient transport of G protein to the cell surface. Also, normal cytoplasmic domains from other transmembrane glycoproteins can substitute for the G protein cytoplasmic domain in transport of G protein to the plasma membrane. The method of constructing precise hybrid proteins described here will be useful in defining functions of specific domains of viral and cellular integral membrane proteins.

Gas-fired power units (GFUs) are the best technology in recent years due to lower natural gas prices, higher energy transformation performance, and lower CO2 emission, as compared to the conventional power units (CPUs). A permanent storage facility called power-to-gas (P2G) technology can provide adaptation of ever-increasing renewable energy sources (RESs) fluctuations in power system operations, as well as reduce dependency to buy natural gas from the gas network. High investment and utilization expenditures of state-of-the-art P2G technology do not lead to economically effective operation individually. Therefore, in the present paper, an integrated GFUs-P2G-wind power unit (WPU) system is proposed to determine its optimal bidding strategy in the day-ahead energy market. A robust optimization approach is also taken into account to accommodate the proposed bidding strategy within the electricity price uncertainty environment. This problem was studied by using a case study that included a P2G facility, GFU, and WPUs to investigate the effectiveness and capability of the proposed robust bidding strategy in the day-ahead energy market. Simulation results indicate that the obtained profit increase by introducing the integrated energy system, and the P2G facility has a significant effect on participating GFUs, which have gas-consumption limitations in order to achieve maximum profit. Moreover, as it can be said, the amount of purchased natural gas is decreased in the situations, which do not have any gas-consumption limitations. Furthermore, the proposed system’s operation in the robust environment provides more robustness against electricity price deviations, although it leads to lower profit. In addition, deploying P2G technology causes about 1% incrementation in the introduced system profit.

We report a facile easy method to deposit anatase titania (TiO[Formula: see text] on cellulose paper. The anatase TiO2/cellulose paper (ATCP) was characterized by scanning electron microscopy, transmission electron microscope, energy dispersive X-ray spectrometer, X-ray diffraction, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, and thermogravimetric analysis. This hybrid paper with the anatase TiO2 content of around 13.86[Formula: see text]wt.% can serve as an eco-friendly flexible photocatalyst, which can rapidly degrade blue indigo carmine dye into a colorless solution within 30[Formula: see text]min under UV radiation. Moreover, compared to commercially available TiO2 P25 and anatase TiO2 powder, a faster decomposition rate of indigo carmine dye was acquired when using ATCP. These results suggest that this hybrid paper might be useful in the treatment of organic dye wastewater.

ABSTRACT Antitermination in bacteriophage P22, a lambdoid phage, uses the arginine-rich domain of the N protein to recognize boxB RNAs in the nut site of two regulated transcripts. Using an antitermination reporter system, we screened libraries in which each nonconserved residue in the RNA-binding domain of P22 N was randomized. Mutants were assayed for the ability to complement N-deficient virus and for antitermination with P22 boxBleft and boxBright reporters. Single amino acid substitutions complementing P22 N− virus were found at 12 of the 13 positions examined. We found evidence for defined structural roles for seven nonconserved residues, which was generally compatible with the nuclear magnetic resonance model. Interestingly, a histidine can be replaced by any other aromatic residue, although no planar partner is obvious. Few single substitutions showed bias between boxBleft and boxBright, suggesting that the two RNAs impose similar constraints on genetic drift. A separate library comprising only hybrids of the RNA-binding domains of P22, λ, and φ21 N proteins produced mutants that displayed bias. P22 N− plaque size plotted against boxBleft and boxBright reporter activities suggests that lytic viral fitness depends on balanced antitermination. A few N proteins were able to complement both λ N- and P22 N-deficient viruses, but no proteins were found to complement both P22 N- and φ21 N-deficient viruses. A single tryptophan substitution allowed P22 N to complement both P22 and λ N−. The existence of relaxed-specificity mutants suggests that conformational plasticity provides evolutionary transitions between distinct modes of RNA-protein recognition.

ABSTRACT Pak1 protein kinase of Schizosaccharomyces pombe, a member of the p21-GTPase-activated protein kinase (PAK) family, participates in signaling pathways including sexual differentiation and morphogenesis. The regulatory domain of PAK proteins is thought to inhibit the kinase catalytic domain, as truncation of this region renders kinases more active. Here we report the detection in the two-hybrid system of the interaction between Pak1 regulatory domain and the kinase catalytic domain. Pak1 catalytic domain binds to the same highly conserved region on the regulatory domain that binds Cdc42, a GTPase protein capable of activating Pak1. Two-hybrid, mutant, and genetic analyses indicated that this intramolecular interaction rendered the kinase in a closed and inactive configuration. We show that Cdc42 can induce an open configuration of Pak1. We propose that Cdc42 interaction disrupts the intramolecular interactions of Pak1, thereby releasing the kinase from autoinhibition.

Corn is an important food crop in the world. Downy mildew as a corn main disease causes a decrease of crop yields. Downy mildew can be prevented by seed treatment. The study aims to determine the effect of washing and adding fungicides on seeds to downy mildew attack, growth, and hybrid corn yields. The research conducted in Wedomartani Experimental Field, Ngemplak, Sleman with experiment in June – October 2018. Field experiment with Randomized Complete Block Design (RCBD) was set up, using single factor, which is seeds of P27 variety washed and added fungicides. There are 10 treatments: control, washed seeds, washed seeds + fungicide metalaxyl 3g/kg + dimetomorph 2g/kg, washed seeds + fungicide metalaxyl 3g/kg + dimetomorph 3g/kg, washed seeds + fungicide metalaxyl 3g/kg + dimetomorph 4g/kg, washed seeds + fungicide metalaxyl 3g/kg + dimetomorph 5g/kg, washed seeds + fungicide metalaxyl 5g/kg + dimetomorph 2g/kg, washed seeds + fungicide metalaxyl 5g/kg + dimetomorph 3g/kg, washed seeds + fungicide metalaxyl 5g/kg + dimetomorph 4g/kg, washed seeds + fungicide metalaxyl 5g/kg + dimetomorph 5g/kg. The results showed that control effective to prevent downy mildew attack, increasing growth and hybrid corn yields. Washed seeds cause high level of downy mildew attack, slow growth, and low crop yields. Seed treatments with fungicides is not effective, because give same affect with control.

Abstract Iron (Fe) deficiency limits the yield of fruit trees. When subjected to Fe deficiency, H+ secretion increases in the rhizosphere of dicotyledonous plants and pH decreases. This leads to the acidification of the soil and promotes Fe3+ to Fe2+ conversion, which plants can better uptake. This study investigated the relationship between two inhibitory transcription factors (ethylene response factors MbERF4 and MbERF72) and the H+-ATPase gene MbHA2. Two species of apple woody plants were studied: the Fe-inefficient Malus baccata and the Fe-efficient Malus xiaojinensis. Yeast one-hybrid and electrophoretic mobility shift assays showed that both MbERF4 and MbERF72 bind to the GCC cassette (AGCCGCC) of the MbHA2 promoter. Moreover, yeast two-hybrid and bimolecular fluorescence complementation assays showed that MbERF4 interacts with MbERF72. Furthermore, β-glucuronidase and luciferase reporter assays showed that the MbERF4- and MbERF72-induced repression of MbHA2 expression is synergistic. Virus-induced gene silencing of MbERF4 or MbERF72 increased MbHA2 expression, and thus lowered the rhizosphere pH in M. baccata. Consequently, the high expressions of MbERF4 and MbERF72 induced by Fe deficiency contributed to the Fe sensitivity of M. baccata. Moreover, the low expressions of MxERF4 and MxERF72 contributed to the Fe-deficiency tolerance of M. xiaojinensis via different binding conditions to the HA2 promoter. In summary, this study identified the relationship of two inhibitory transcription factors with the H+-ATPase gene and proposed a model in which ERF4 and ERF72 affect the rhizosphere pH in response to Fe deficiency.

Recently, BiPO4has been proved to be an efficient photocatalyst which exhibits more attractive activity than that of TiO2-P25 under UV irradiation. However, photocatalytic oxidation-reduction ability of BiPO4under visible light still needs to be improved. Conjugated polymer just like polyaniline was demonstrated to be a new and efficient visible co-photocatalyst in the field of pollutant governance. As far as we know, the attempt to enhance the photochemical ability of BiPO4by employing polyaniline as a co-photocatalyst is still very limited. Hence, in order to testify the potential enhanced performance of such heterostructure, BiPO4-polyaniline heterostructure was prepared via hydrothermal method and hybridization in this paper. The experimental results indicated that several typical pollutants such as organic dyes and heavy metal ions can be effectively removed by BiPO4-polyaniline under visible light. The enhanced removal mechanism was attributed to the high separation efficiency of photo-induced electron–hole pairs and more efficient absorption of visible light after the loading of polyaniline on the surface of BiPO4as an out-layer compared with single BiPO4.
