Abstract:
The aim of the present study was to assess the effect of IVM treatment on the developmental competence of oocytes recovered from repeated laparoscopic ovum pickukp (LOPU) in goats. A total of 94 LOPU sessions were performed on 33 adult goats of the Saanen and Alpine breeds. Females were synchronized (Day 0) during the nonbreeding season by inserting vaginal sponges (45 mg of fluorogestone acetate, Intervet, Boxmeer, The Netherlands). At Day 8, an i.m. injection of 50 μg of cloprostenol (Estrumate; Schering-Plough Animal Health, Pointe-Claire, Quebec, Canada) was administered. Porcine FSH (Stimufol, Merial, Brussels, Belgium, 160 mg/goat) was administered in 5 injections at 12-h intervals, starting on Day 8. The LOPU took place under general anesthesia on Day 11, and follicles ≥2 mm were aspirated with an 18-gauge needle connected to a controlled vacuum system. Vaginal sponges were removed at the time of LOPU. Treatments were repeated 2 times in a 2-week interval scheme (2 goats and 1 goat were excluded from the experiment during the second and third LOPU sessions, respectively). Cumulus–oocyte complexes were washed and evaluated for quality (graded from 1 to 3). Oocytes recovered from unstimulated slaughterhouse-derived ovaries served as a control. Cumulus–oocytes complexes from Grades 1 and 2 were submitted to IVM in TCM-199, supplemented with 100 μm of cysteamine and either 10 ng mL–1 of epidermal growth factor (EGF) or 10% follicular fluid and 100 ng mL–1 of ovine FSH (FF-FSH). Matured oocytes were then submitted to IVF and in vitro development as described by Cognié et al. (2004 Reprod. Fertil. Dev. 16, 437–445). Over the 94 LOPU sessions, 20.4 ± 0.9 follicles were aspirated (mean ± SEM), allowing the recovery of 12.3 ± 0.7 COC per goat and per session, of which 80.1% were suitable for IVM (Grades 1 and 2). Results of in vitro production are detailed in the table. The IVM treatment did not significantly affect cleavage or blastocyst development rates in oocytes derived from slaughterhouse ovaries. Cleavage rates were significantly decreased in LOPU-derived oocytes when compared with control oocytes. For LOPU-derived oocytes, cleavage and final blastocyst development rates were increased significantly and kinetics of embryo development were accelerated when FF-FSH was used during IVM as compared with EGF. The IVM with FF-FSH allowed us to produce 4.1 blatocysts per goat per LOPU session. These results demonstrate the interest in LOPU for goat embryo production once appropriate IVM treatment is used. The difference observed between LOPU and slaughterhouse oocytes in terms of response to IVM treatments may be related to FSH stimulation prior to the LOPU session or to postmortem changes in oocyte responsiveness in the slaughterhouse group. Table 1. Effects of oocyte origin [laparoscopic ovum pickukp (LOPU) or slaughterhouse derived] and maturation treatment [epidermal growth factor (EGF) or follicular fluid (FF)-FSH] on in vitro embryo production (6 replicates)

Abstract:
Embryo output in sheep is increased when superovulatory FSH treatments are started in the presence of a high number of small follicles (2–3mm in size) and in absence of large follicles (>6mm, Gonzalez-Bulnes et al., 2002. Theriogenology, 57, 1263–1272). Administration of GnRH antagonists (GnRHa) suppresses large follicles (Cognie et al., 2003. Theriogenology, 59, 171–188), whereas the use of growth hormone (GH) would increase the number of small follicles (Campbell et al., 1995. J Reprod Fertil Suppl, 49, 335–350). Our aim was to evaluate the usefulness of pre-treatments with GH or GH plus GnRH antagonists for sheep embryo production. First, we studied the effects on follicular population by serial ultrasonographies. Thereafter, we determined whether such treatments can affect oocyte developmental competence. In a first trial, a total of 18 Manchega ewes were treated with intravaginal FGA sponges (Chronogest®, Intervet Int., H) during breeding season (beginning of April). Six animals received daily i.m. doses of 15mg of ovine GH (Tuenre, GA) for 6 days, while six females received GH plus two s.c. doses of 1.5mg of GnRHa (Antarelix™, Zentaris, G) on Days 0 and 3 of GH treatment, and six ewes acted as controls receiving saline. Number of follicles >2mm, determined by daily transrectal ultrasonography, increased to reach significant differences on Day 4 in sheep treated with GH/GnRHa (22.7±0.8 v. 16.7±0.5, P<0.001) and on Day 5 in ewes injected with GH (20.3±0.4 v. 17.0±0.6, P<0.05). The second trial involved 18 Manchega ewes treated with progestagen sponges on Day 0 and distributed in three groups at the beginning of breeding season (end of July). In the first group (n=7), sheep were treated with two doses of GnRHa on Days 0 and 3 after sponge insertion and with three doses of 15mg of GH on Days 3, 4, and 5. Thereafter, ewes from this group and from a second experimental group (n=7) were treated with 3 doses of 1.5mL of FSH (Ovagen™, ICP, NZ) every 12h, starting on the afternoon of Day 5. A third group of sheep (n=4) did not receive GH/GnRHa or FSH, acting as controls. On Day 7, follicles were aspirated and the cumulus-oocyte complexes (COC) were cultured for 24h at 38.5°C, 5% CO2, in TCM-199 supplemented with ovine FSH (Ovagen), LH, FCS, 17-βoestradiol, cysteamine, and sodium pyruvate (Sigma Chemical Co., MO, USA). Nuclear maturation was measured by Hoechst 33342 fluorescence. Mean number of COC was higher in GH/GnRHa+FSH group (8.7±0.9 v. 6.8±1.3 in FSH group, NS and 4.5±0.8 in control, P<0.05) due to higher number of follicles with similar recovery rates (45.0±4.5, 40.3±1.4, and 39.1±7.1%, respectively). There were no significant differences on the ability of COC to resume meiosis, although this was higher in FSH group (63.1±9.5% for GH/GnRHa+FSH, 79.5±6.3% for FSH and 60.0±8.8% for control group), which can indicate the necessity of a higher FSH supply to induce final development in follicles/oocytes from ewes treated with GH and GnRHa. In conclusion, the use of GH and GnRHa would help to increase the number of gonadotrophin-responsive follicles prior to gonadotrophin injections;; also, adjustment of FSH treatments improved embryo yields in superovulatory protocols.

Abstract:
In recent years, with the advent of the internet of things (IOT), computers and information technology (IT), small and medium-sized enterprises (SMEs) have been immersed in a cluster of information difficult to analyze in a conventional way which provides value that can ensure its permanence and survival in the market. Therefore, it is important to make use of data mining technology tools and statistical techniques that help analyze and better understand the information generated by sales processes, through a thorough analysis of the data. For this research, the IBM Cognos Analytic tool was used, which provides a predictive data visualization and analysis service that can be used to determine patterns, relationships, associations and meaning of a large set of data quickly and in a timely manner. Likewise, statistical techniques such as correlation and simple linear regression were used to establish the behavior of the company's product demand in the marketing, manufacturing and importation of the cleaning product sector.

Abstract:
Background: Business intelligence (BI) tools are generally associated with organisations that have resources to purchase and implement these tools. Evidences abound regarding the correlation between BI tools and improved business decision-making. This study’s unit of analysis is affordability as a feature of IBM Cognos making it suitable for small-and medium-sized enterprises (SMEs).Objective: The research aim was to identify the fundamental features of IBM Cognos which would address decision-making needs of SMEs. The objective was to determine the significance of BI tool features by means of identifying affordable features suitable for SMEs’ decision-making.Method: Quantitative research design and a deductive approach were best suited for assessing the fundamental features of IBM Cognos for SMEs’ decision-making needs. The signification framework variables, such as presumed, prized and perceived value of BI tool features, werequantified and measured using statistical analysis tools. A non-probability convenience sampling technique was used with a sample size of 200, that is, 80 BI consultants, 60 SME BI developers and 60 SME managers. Results: Affordable key features of BI tools in the context of SMEs’ business decision-making include consistency and comfort, intuitive interface, avoiding impulsivity, cost effectiveness, availability of information, best programmed visualisations, reporting quickly and easily and financial decision-making.Conclusion: The signification framework’s presumed, prized and perceived value indicators link affordable BI tool features to the consistency of the decision-making process and present an alternative view of affordability. An intuitive interface relates to convenience and ease of authoring content, designing, building and securing reports to the SME, which helps inimproving consistent decision-making.

Abstract:
Dramatic increasing of amount of data in running a business has been consireded a typical phenomenon of the last decade. This fact, however, has its impact on the business planning and decision making. The area, which deals with this problematics, is called Business Intelligence (BI). The problem lies in the implementation of applications directing to the BI, because it is often oriented only on isolated projects, so they cause only partial effect. In such cases, BI cannot fulfil the expected role, but still preserves the negatives instead. By saying negatives, it is meant: complicated searching for analytical outputs, significant dete-rioration of their output data, cost increase in administation of BI systems, and others. The solution can represented by integration of partial BI systems and by transfer of their BI metadata into a single informational system, serving as an "entering gate" for all the business workers. From the whole company perspective, there exist also benefits in the metadata managament, such as: determination of quality and usage of reports, prolonging of the life cycle, cooperative forming of the knowledge basis, consolidation of the terms in the business, more efficient way of browsing in the system and comprehensibility of analytical system's outputs. The aim of this diploma thesis is formation of a metadata presentation and its realization into a preselected information system, together with meeting of the advantages resulting from applying business metadata.

Abstract:
Thesis is devoted to current trends in approaching cost allocation developed in IBM Cognos TM1 software. Concept, which was originally elaborated in my bachelor thesis, has recently experienced restrictions caused by increasing requirements on analytical tools and information they provide. Goal of the thesis is therefore to analyse causalities of emerging weaknesses, design and develop optimalized and reengineered solution answering current demands. Reqarding the quantitative evaluation of attained results, the thesis is extended with analysis of frameworks and standards dedicated to benchmarking of OLAP tools and their synthesis into own complex model. Proposed model specializes on measuring multiple OLAP applications across four main perspectives including performance, development, usability and financial benefits. Attained results prove, that reengineered model is faster, data richer, easier to use and appropriate for any organization with structured and algorithmic approach to cost allocation. Second half of the thesis focuses on extending the presentation layer to web browser, designing and developing of custom visualizations for most usual analytic tasks. Considering the absence of advanced application interface in IBM Cognos TM1, the thesis also includes theoretical analysis of current trends in API development and design of concept allowing communication and data transportation between applications and TM1 server. In the concluding section of the thesis, proposed concept is materialized into universal library developed in PHP and applied to novel allocation model. Leveraging the library, two exemplary interfaces for allocator operation and data consumption are implemented. Gained knowledge can serve as basis for development of additional components communicationg with TM1 in variety of projects or theoretical framework for API implementation in general.

Abstract:
A big data warehouse enables the analysis of large amounts of information that typically comes from the organization's transactional systems (OLTP). However, today's data warehouse systems do not have the capacity to handle the massive amount of data that is currently produced. Business intelligence (BI) is a collection of decision support technologies that enable executives, managers, and analysts to make better and faster decisions. Organizations must make good use of business intelligence platforms to quickly acquire desirable information from the huge volume of data to reduce the time and increase the efficiency of decision-making processes. In this chapter, the authors present a comparative analysis of commercial and open source BI tools capabilities, in order to aid organizations in the selection process of the most suitable BI platform. They also evaluated and compared six major open source BI platforms: Actuate, Jaspersoft, Jedox/Palo, Pentaho, SpagoBI, and Vanilla; and six major commercial BI platforms: IBM Cognos, Microsoft BI, MicroStrategy, Oracle BI, SAP BI, and SAS BI & Analytics.
