Abstract:
AimsStaphylococcus aureus (S. aureus) is a life-threatening pathogen with high morbidity and mortality rates which causes nosocomial and community-acquired infections. Biofilm, considered to be a common virulence factor for pathogens, plays a significant role in recurrent and untreatable infections. Biofilm formation of S. aureus is mediated by synthesis of either poly-N-acetylglucosamine in an ica-dependent manner or surface proteins in an ica-independent manner. In some cases treatment is impossible and recurrent. In this study, ica-dependent biofilm-producing S. aureus isolates were detected and the anti-biofilm effect of ascorbic acid against biofilm formation of isolates was investigated.MethodsA total of 21 methicillin-sensitive S. aureus (MSSA) clinical isolates stored in our bacterial stock were used to detect ica-dependent biofilm-producing MSSA isolates. The anti-biofilm study was undertaken with three ica-dependent biofilm-producing isolates (MSSA2–4) and ATCC 29213 (MSSA1). Biofilms and the anti-biofilm effect of ascorbic acid were detected using the microtitre plate (MtP) method. 16S-rRNA, nuc, icaA and icaD genes and expression levels of icaA and icaD of isolates were detected by RT-PCR.ResultsThe minimum inhibitory concentrations (MICs) of ascorbic acid prevented biofilm formation of MSSA1 and MSSA3. Also, 1/2 MIC of ascorbic acid prevented biofilm formation of MSSA3. It was observed that biofilm formation decreased with increased concentration. There was no significant increase in ica gene expression of MSSA1 and MSSA2. Expression of icaA and icaD of MSSA3 decreased 13% and 38%, respectively. Expression of icaA in MSSA4 decreased 12%.ConclusionThe results of our study show that ascorbic acid can be used as an anti-biofilm agent to prevent biofilm formation of S. aureus and thus biofilm-related infections.

ABSTRACT The atlC gene (1,485 bp), encoding an autolysin which binds fibronectin, and the ica operon, involved in biofilm formation, were isolated from the chromosome of an infectious isolate of Staphylococcus caprae and sequenced. AtlC (155 kDa) is similar to the staphylococcal autolysins Atl, AtlE, Aas (48 to 72% amino acid identity) and contains a putative signal peptide of 29 amino acids and two enzymatic centers (N-acetylmuramoyl-l-alanine amidase and endo-β-N-acetylglucosaminidase) interconnected by three imperfect fibronectin-binding repeats. The glycine-tryptophan (GW) motif found in the central and end part of each repeat may serve for cell surface anchoring of AtlC as they do in Listeria monocytogenes. The S. caprae ica operon contains four genes closely related to S. epidermidis and S. aureus icaA, icaB, icaC, and icaDgenes (≥ 68% similarity) and is preceded by a gene similar toicaR (≥70% similarity). The polypeptides deduced from theS. caprae ica genes exhibit 67 to 88% amino acid identity to those of S. epidermidis and S. aureus icagenes. The ica operon and icaR gene were analyzed in 14 S. caprae strains from human specimens or goats' milk. Some of the strains produced biofilm, and others did not. All strains carry the ica operon and icaR of the same sizes and in the same relative positions, suggesting that the absence of biofilm formation is not related to the insertion of a mobile element such as an insertion sequence or a transposon.

Abstract:
Aim: This study was designed to assess the antibiofilm activity of quercetin on characterized S. aureus isolates. Methodology: This study evaluated 36 S. aureus isolates, each of which was identified using Gram staining, culture, biochemical, and PCR assays. Isolates were cultured and their biofilm production was evaluated using Congo red agar (CRA) plates, microtiter plate tests and PCR, and the effects of quercetin were examined. Results: The CRA results revealed that eight (22.3%) S. aureus isolates were strongly positive for biofilm production and an additional 18 isolates (50%) showed moderate biofilm capacity. The remaining 10 isolates were negative (27.7%) for biofilm production. S. aureus isolates were divided into strong positive, intermediate, and negative groups, 27.8%, 44.5%, and 27.7%, respectively. Scanning electron microscopy showed that the biofilm-producing isolates appeared as aggregates of cells within a heavy matrix. In addition, PCR assay identified IcaA and IcaD (66.6% for both) biofilm production genes in most isolates and IcaC (61.1%), IcaB, FnbB (33.3% for both), and Fib (22.2%) in several other strains. Quercetin significantly inhibited biofilm activity in biofilm producing S. aureus isolates in a dose-dependent manner, with an inhibition rate of 29.6-87.7%. Interpretation: Biofilm production is dependent on Ica gene phenotype and strains with an IcaABCD or IcaABD phenotype produce more biofilm than strains with IcaAD phenotype. Quercetin significantly inhibited S. aureus biofilm production, irrespective of Ica phenotype.

Abstract:
Objective:The complications related to implant associated infections in post-angioplasty patients were considered to increase due to biofilm formation.Methods: Genes responsible for the biofilm formation in the target organisms was investigated in the present study. The presence of the intercellular adhesion biofilm genes (icaA, icaB and icaD) was determined by the Polymerase Chain Reaction method. As preliminary investigations, standard tests, exit-site challenge test and microtitre plate method were used to study the biofilm efficiency of five different test organisms.Results: Exit-site challenge test, was used to identify the ability of test organisms to grow on a bio-materials used in the study. Among the five selected test organisms, Staphylococcus aureus showed the highest ability to colonize the stent materials with in 24h to 48h. In microtitre plate method, Staphylococcus aureus and Staphylococcus epidermidis showed high biofilm forming index values of 0.29 and 0.27 respectively. Biofilm gene studies using PCR revealed the presence of all the three ica genes (Ica A, Ica D and, Ica B) in Staphylococcus aureus. The present research finding has great significance in the treatment to implant associated infections.Conclusions: The results suggest that the virulence factors contributing to the development of infections can be revealed by understanding the presence of biofilm expression genes in the target organisms. This would also prevent dissemination of virulent bacteria in the health care centre; method also considered significant to detect healthy carriers of slime-producing staphylococci.

Abstract:
Staphylococcus aureus is an opportunistic human pathogen that is often involved in severe infections such as pneumonia and sepsis in which bacterial virulence factors play a key role. Infections caused by S. aureus are often difficult to eradicate, particularly when they are associated with biofilm. The physiological roles of the Crp/Fnr family regulator ArcR are elusive in S. aureus. In this study, it was found that the deletion of arcR increased the hemolytic ability and biofilm formation in S. aureus. Differential gene expression analysis by RNA-seq and real-time quantitative reverse transcription PCR showed that genes associated with hemolytic ability (hla and hlb) and biofilm formation (icaA, icaB, icaC and icaD) were significantly upregulated compared with those in the wild-type strain. The results revealed that ArcR regulated the expression of the hla and ica operon by binding to their promoter regions, respectively. This study provided new insights into the functional importance of ArcR in regulating the virulence and biofilm of S. aureus.

ISSN 0804–4643 The aim of this work was to characterize both newly diagnosed insulin-dependent diabetic subjects and their siblings with positive tests for islet cell-specific autoantibodies (ICSAA) and to evaluate whether there is an association between the ICSAA levels detected in the diabetic children and siblings. We analysed 781 probands younger than 15 years of age for islet cell antibodies (ICA) and 755 for insulin autoantibodies (IAA) and 610 of their 3–19-year-old non-diabetic siblings for ICA and IAA upon diagnosis of the proband. Islet cell antibodies were observed in 657 of the probands (84.1%) and IAA in 353 (46.8%). The ICA-positive probands were younger in age and had higher IAA levels than the ICA-negative probands, while the IAA-positive probands were younger and had higher levels of ICA than the IAA-negative probands. Islet cell antibodies were detected in 46 (7.5%) and IAA in 16 (2.6%) siblings, and the ICA-positive siblings had higher IAA levels than the ICA-negative siblings. A falling trend was seen in the frequency of ICA [2A7E] 20 Juvenile Diabetes Foundation units in the siblings with decreasing degrees of HLA identity with the index case. Infections during the preceding year, especially respiratory infections, increased the prevalence of both ICA and IAA in the diabetic children at diagnosis and the frequency of IAA in the siblings. There was a significant, although weak, correlation between the IAA levels of the probands and those of their siblings when 594 pairs were tested (rs = 0.15; p < 0.001). No association could be seen between the ICA levels of the probands and those of their siblings, not even when including only HLA-identical proband–sib pairs in the analysis. The lack of any relation between ICA levels in the probands and siblings supports the view that there may be multiple exogenous factors capable of inducing ICA formation or else a common factor but variable responsiveness in the index case and the sibling. Bovine mastitis is mainly caused by bacteria of the genus Staphylococcus spp., which possess different virulence factors, including the capacity for biofilm formation that provides enhanced protection against the action of immune system components and serves as a barrier against the penetration of antimicrobial agents. This study aimed to characterize 181 Staphylococcus spp. Strains—including Staphylococcusaureus and coagulase-negative staphylococci (CoNS) isolated from bovine subclinical mastitis in six Brazilian states—by molecular methods. RT-qPCR was used to verify the expression of genes of the ica operon—mainly responsible for biofilm formation—as well as bap and bhp. Chromosome similarity among the isolates was investigated by pulsed-field gel electrophoresis (PFGE) and multilocus sequence typing (MLST). The icaA gene was detected in 79 (43.6%) isolates, icaB in 24 (13.2%), icaC in 57 (31.4%), and icaD in 127 (70.1%). The bap gene was identified in 66 (36.4%) isolates, while the bhp gene was found in nine (4.9%). RT-qPCR confirmed the expression of the icaA gene in 60 (75.9%) isolates, of icaB in six (25%), of icaC in 26 (45.6%), and of icaD in 80 (63%). Clonal typing of the isolates by PFGE permitted the identification of eight Staphylococcusaureus clusters that simultaneously included ≥3 strains, with a similarity of ≥80%. Regarding the other species studied, three clusters were observed for Staphylococcuschromogenes and four clusters for Staphylococcusepidermidis. Only one cluster each was identified for Staphylococcussaprophyticus and Staphylococcussimulans, while the other species did not form any cluster. With respect to MLST, ST126 and ST1 were the prevalent sequence types in S. aureus, while in S.epidermidis all sequence types were different. These results reveal strains with the same evolutionary origin as other isolates, which might cause infections in humans and animals, suggesting their ability to spread between these species.

Ca2+ current ( ICa) recovery from inactivation is necessary for normal cardiac excitation-contraction coupling. In normal hearts, increased stimulation frequency increases force, but in heart failure (HF) this force-frequency relationship (FFR) is often flattened or reversed. Although reduced sarcoplasmic reticulum Ca2+-ATPase function may be involved, decreased ICa availability may also contribute. Longer action potential duration (APD), slower intracellular Ca2+ concentration ([Ca2+]i) decline, and higher diastolic [Ca2+]i in HF could all slow ICa recovery from inactivation, thereby decreasing ICa availability. We measured the effect of different diastolic [Ca2+]i on ICa inactivation and recovery from inactivation in rabbit cardiac myocytes. Both ICa and Ba2+ current ( IBa) were measured. ICa decay was accelerated only at high diastolic [Ca2+]i (600 nM). IBa inactivation was slower but insensitive to [Ca2+]i. Membrane potential dependence of ICa or IBa availability was not affected by [Ca2+]i <600 nM. Recovery from inactivation was slowed by both depolarization and high [Ca2+]i. We also used perforated patch with action potential (AP)-clamp and normal Ca2+ transients, using various APDs as conditioning pulses for different frequencies (and to simulate HF APD). Recovery of ICa following longer APD was increasingly incomplete, decreasing ICa availability. Trains of long APs caused a larger ICa decrease than short APD at the same frequency. This effect on ICa availability was exacerbated by slowing twitch [Ca2+]i decline by ∼50%. We conclude that long APD and slower [Ca2+]i decline lead to cumulative inactivation limiting ICa at high heart rates and might contribute to the negative FFR in HF, independent of altered Ca2+ channel properties.

Abstract:
The image processing techniques in transform domain are employed as analysis tools for enhancing the detection of mineral deposits. The process of decomposing the image into important components increases the probability of mineral extraction. In this study, the performance of Principal Component Analysis (PCA) and Independent Component Analysis (ICA) has been evaluated for the visible and near-infrared (VNIR) and Shortwave infrared (SWIR) subsystems of ASTER data. Ardestan is located in part of Central Iranian Volcanic Belt that hosts many well-known porphyry copper deposits. This research investigated the propylitic and argillic alteration zones and outer mineralogy zone in part of Ardestan region. The two mentioned approaches were applied to discriminate alteration zones from igneous bedrock using the major absorption of indicator minerals from alteration and mineralogy zones in spectral rang of ASTER bands. Specialized PC components (PC2, PC3 and PC6) were used to identify pyrite and argillic and propylitic zones that distinguish from igneous bedrock in RGB color composite image. Due to the eigenvalues, the components 2, 3 and 6 account for 4.26% ,0.9% and 0.09% of the total variance of the data for Ardestan scene, respectively. For the purpose of discriminating the alteration and mineralogy zones of porphyry copper deposit from bedrocks, those mentioned percentages of data in ICA independent components of IC2, IC3 and IC6 are more accurately separated than noisy bands of PCA. The results of ICA method conform to location of lithological units of Ardestan region, as well.

Strain 406 PPI prepared by cell fusion of mouse myeloma NS-1 cells with spleen mice BALB/ C, pre-immunized three times at an interval of two weeks, the cells of the cell line of human melanoma melKor. Merging conducted using a solution of PEG/DMSO. For screening received mAb 406 used human melanoma cell lines which differed in the expression of CD117 antigen FSBSI "N.N. Blokhin RCRC" collection. N.N Blokhin. Antigen expression was studied in immunofluorescence and evaluated on a flow cytometer BD FACS CantoTMII. ICA IC0-406 was compared with commercial ICA against antigen CD 117 (Germany). The results indicated the identity of the frequency of antigen-positive cases and the percentage of antigen cells. ICA IC0-406 block binding to cells melKor ICA anti-CD117. Linking ICA IC0-406 antigen-positive cells causes modulation of antigen CD 117.

De handlare som ingick i studien driver butiker i Uppsalaområdet. Utifrån relationsteorier om engagemang och förtroende utformades och genomfördes intervjuer med ICA-handlare. Informationen från intervjuerna användes för att uppfylla studiens syfte och besvara frågeställningen. Studien visar att ICA-handlare som driver butiker inom profilerna Nära- och Supermarkets relation till ICA AB är bra.

Idag finns det begränsade teorier som behandlar det postmoderna konsumtionssamhället utifrån ett medie- och kommunikationsvetenskaligt perspektiv. Syftet med examensarbetet är att undersöka spänningsfältet mellan ICA och högskolestudenter i ett konsumtionssamhälle genom att granska varumärkeskommunikationens betydelse på digitala plattformar. Materialinsamlingen är genomförd i form av kvalitativa intervjuer, med medarbetare på ICA, samt fokusgrupper med studenter. Med studien vill vi ge en inblick i hur kommunikationsprocesser strategiskt utformas för att nå rätt målgrupp, hur relationer vårdas och vad en god dialog innebär och har för framtida betydelser. Studien har analyserat det spänningsfält som ligger i grund till valet av konsumtion gällande matvaruhandel som högskolestudent. Studiens slutsatser visar på att det är en rad olika faktorer som är bidragande till att bygga upp en stark relation med ett företag.
Today there is limited theories considering reasearch on the postmodern consumtion society from a media and communication perspective. The purpose with this thesis is to investigate the tension between ICA and Univeristy students in a consumption society through analysis brandbuilding communications meaning on digital platforms. The material to the thesis is collected by qualitative interviews, both with employees at ICA and focus groups with university students. With this thesis we want to give an insight of how communication processes strategically is designed to reach their target group, how relationships are nurtured and what a good dialogue means for future brand building. The study has analyzed the tension of what includes the choice of consumption when it comes to retail business from university students perspective. The conclusions indicate how there are a number of factors that are contributing to build a strong relationship with an organization.

Abstract:
Abstrakt Syftet med vår undersökning var att studera ICAs reklamfilmer ur ett genusperspektiv. Vi har specificerat oss på om det manliga eller kvinnliga könet framkommer mer eller mindre i filmerna, om det har skett någon förändring kring genusperspektivet från år 2006 till 2016, samt om varorna som finns med i filmerna är mer eller mindre riktade till något av könen.   Frågeställning:
    Hur framställs kvinnor respektive män i Icas reklamfilmer? Har det skett någon förändring från år 2006 till 2016? Är varorna som visas i reklamfilmerna avsedda för kvinnor, män eller båda?   Vi har utgått från teorier som vi anser är relevanta för vår undersökning. Vi har delat in teorierna i tre olika områden: Genus Medier och identitet Reklam och media. För att genomföra undersökningen har vi gjort en kvantitativ innehållsanalys på 15 olika reklamfilmer. Vi har avgränsat oss till reklamfilmer som har visats på TV under tre olika högtider i Sverige: jul, midsommar och påsk. Vi har sedan gjort en kvalitativ innehållsanalys på fem filmer som vi anser lockar fram olika stereotyper.   Resultatet vi fann i vår undersökning var att männen dominerar i ICAs reklamfilmer, och med det menar vi att det bland annat är fler manliga karaktärer som är med och har en slags ledarroll. Vårt resultat skiljer sig något mot vad teorierna kring ämnet påstår, nämligen att kvinnor ofta dominerar reklambranschen.
    Abstract The purpose of our survey was to study the gender perspective in ICA's commercials. We have specified whether the male or female gender appears more or less in the movies, if there has been any change in gender perspective from 2006 to 2016 and if the items featured in the movies are more or less directed to any of the sexes. We have limited ourselves to commercials that have been shown on television during three different festivals in Sweden: Christmas, Midsummer and Easter.   Question:
      How are women and men produced in Ica's commercials? Has there been any change from 2006 to 2016? Are the items shown in the commercials intended for women, men or both?   We have assumed theories that we consider relevant to our survey. We have divided the theories into three different areas: Gender Media and identity Advertising and the media. In order to conduct the survey, we have made a quantitative content analysis of 15 different commercials. We have limited ourselves to commercials that have been shown on television during three different festivals in Sweden: Christmas, Midsummer and Easter. We have then made a qualitative content analysis on five films that we consider attracting different stereotypes.   The result we found in our survey was that the men dominate ICA's commercials, and we mean that among other things there are more male characters that are in and have a sort of leadership role. Our results differ slightly from the theory of the subject, namely that women often dominate the advertising industry.

Abstract:
In the midst of stiff and fierce competition and increased number of grocery retail outlets providing a variety of products, customers have become accustomed to patronizing multiple outlets. Retailers have recognized this trend and are of the view that customer satisfaction plays a role in the success of business strategies. Therefore it has become important for grocery retail stores to try and manage customer satisfaction. This paper was thus developed to investigate the satisfaction levels of customers in supermarkets. Data was collected from three supermarkets in Visby, Gotland. The study examined the importance of overall dimensions and specific elements of customer satisfaction towards the measurement of satisfaction levels. In addition, comparative analysis was conducted between the three relative grocery stores. Customer satisfaction levels were measured. The highest percentage of customers of the whole sample belonged to medium level of satisfaction. The statistical result concluded that customers of ICA Nära and customers of ICA were higher satisfied than those of Coop Forum. The results showed that customers felt satisfied with Location, Staff courtesy and Reliability of supermarkets as the top aspects that drive satisfaction while additional services were ranked lowest. Due to these results, the impact of these attributes on Customer satisfaction must not be underestimated.

Tidningar och reklamfilmer som innehöll information om ICA I love eco-sortimentet under perioden mars 2008 - oktober 2011 valdes ut för textanalys. I textanalysen utgick vi ifrån semiotiska och retoriska begrepp för att se meningen som texterna skapar. Resultat visar att ICA använder samhällsengagemang som ett kommunikationsverktyg och strategi. Resultat visar även att ICA använder fem olika dimensioner av nytta, positiv och negativ emotionell nytta, värde för pengarna, sociala livet samt funktionaliteten, i marknadsföringen av det ekologiska sortimentet.

Abstract:
The aim with this essay is to map down how the language plays a part in the persuasion carried out by ICA-Stig – one of the most beloved characters from ICA’s commercial soap opera. Through a discourse analysis with a linguistic focus this study can provide one explanation to why the audience trust ICA-Stig and how he persuades them to do their grocery shopping at ICA. The study takes off from the rhetorical means of persuasion, ethos, logos and pathos, along with how ICA-Stig commits a linguistic intimisation and exploits intertextual strategies in order to approach the audience and their reality. The result shows that Stig uses personal pronouns in a way that enables him to commence intimisation in the sense of addressing his co-workers and the audience in a family-like manner. Regarding intertextuality, Stig uses a composition of different discourses in favour of his character since he is able to reach a large audience whilst nourishing the personal touch, combined with proving himself credible. Through his language, ICA-Stig reduces the gap between non-fiction and fiction, and in his transcendent role he becomes a factive character – neither real nor fictive.  This role allows him to become self-aware, which gives him an outside perspective on commercials in general. From a rhetorical perspective, the intimisation and intertextuality function as to have ICA-Stig relate to the reality, habits, and experiences of the audience. This creates the situation that Daddy-ICA has something to say to his ICA-family, which becomes the premise on which the commercial is founded. The intimisation enhances ICA-Stig’s ethos since he can prove to be a caring father and a knowledgeable shop manager. The appeals to pathos lie within his caring language. Appeals to logos can be found in the product placement of the groceries in an environment very much reminiscent of reality and everyday-life. Finally, the essay problematises the role commercial has and plays in today’s society due to the fact that this discourse is approaching the discourse of private life and communication, which, in turn,  paves the way for new ways of persuading the costumers.
Syftet med detta arbete är att genom en diskursanalys kartlägga hur språkbruket fungerar i övertalningen hos ICA-Stig ‒ en av de mest folkkära karaktärerna från ICA:s reklamsåpa. Med språket som utgångspunkt kan denna analys bidra till förklaringen varför tv-tittarna litar på ICA-Stig och hur han får dem att vilja handla på just ICA. Studien tar avstamp i tre utgångspunkter där retorikens medel för övertalning, ethos, logos och pathos, tas i beaktande jämte hur ICA-Stig begår en språklig intimisering med tittarna, samt utnyttjar intertextuella strategier för att närma sig deras verklighet. Resultatet visar att ICA-Stig tillämpar intimisering genom att bruka pronomen på ett sådant sätt att såväl tittare som medarbetare tilltalas familjärt. Intertextuellt kan ICA-Stig sägas utnyttja olika diskursmöten till sin fördel för att samtidigt nå en bred kundkrets och värna det intima, samt visa på sin trovärdighet. Genom språket tänjer han på gränserna mellan fiktion och fakta, och kan i sin diegesöverskridande roll sägas vara en faktiv karaktär ‒ varken overklig eller verklig. Faktionen gör det möjligt för ICA-Stig att visa självmedvetenhet, vilket ger distans till reklambranschen. Retoriskt sett, har intimiseringen och intertextualiteten funktionen av att låta Stig anknyta till publikens verklighet, vanor och erfarenheter. Det skapar förutsättningar så att reklamen grundas på premissen att Pappa ICA-Stig har något att berätta för sin ICA-familj. Intimiseringen ger ICA-Stig ethos eftersom han bevisar att han är en trygg och vårdande fadersgestalt, såväl som kunnig butikschef. Anspelningar till pathos finns i hans vårdande språkbruk. Logosargumenten ligger i varornas produktplacering i en verklighetsnära vardagsmiljö. Avslutningsvis problematiserar uppsatsen reklamens plats och funktion i samhället utifrån det faktum att reklamens diskurs närmar sig privatlivets diskurs och på så sätt dels kan påverka konsumenterna på helt andra sätt, dels bli en än större del i samhället och dess vardag.

Abstract:
The bacteria Staphylococcus aureus has been discovered to be a major source of community and hospital-acquired infections. The production of ica-dependent biofilms is critical in the persistence of infections in hospitalized patients. Between November 2017 &April 2018, the current study was conducted at Teaching Baquba Hospital's Bacteriology Laboratory in Baquba City and the laboratory of microbiology and polymerase chain reaction (PCR )unit in the Biology Department / College of Science/ Diyala University (2018). Materials and methods: We obtained 13(17.3%) Staph.aureus isolates from 100 clinical specimens (burns, wounds, urine, and blood) after identified them. Following by employed Congo Red Agar(CRA) and tissue culture plate method (TCP)to detect Biofilm development in isolates, as well as a PCR assay and particular primers to determine the presence of the icaA &icaD genes. The results showed ica A/D were found in 69 % (9/13) of cases, icaA gene is present at 7 (53.8%) and the icaD gene at 2(15 .3%) in Staph.aureus isolates. CRA method found biofilm generation in 6 (46%) of thirteen Staph. aureus isolates, while TCP detected biofilm creation in 10 (76%) isolates. When phenotypic approaches compared to the detection of the icaA and icaD genes, only 5 (71%) of the icaA genes were found to be positive by TCP, while only 2 (1% ) of the icaD genes were found to be positive by TCP. In short: The findings show the significance of S. aureus' virulence factors in clinical samples for the icaA and icaD genes and the phenotypic biofilm formation variety. The creation of in vitro slime using the CRA approach is not necessarily consistent even when the icaA and icaD genes exist. Although certain isolates lack the genes icaA & icaD, the ability to generate biofilms highlights the importance of the further gene research, and the absence of the icaA and icaD genes, the capability from certain isolates to create biopolymes emphasises the need for continuous genetic study into icas caused by variations in the number of genes associated with biofilms. When comparing phenotypic techniques, TCP is still the best tool for the screening of biofilms. The aim of this research though is that the biofilm forming potential should be actually linked to the presence of icaA and icaD genes in S. aureus isolates

Abstract:
The project sought to understand factors and mechanisms involved in the hardening of potato tubers. This syndrome inhibits heat softening due to intercellular adhesion (ICA) strengthening, compromising the marketing of industrially processed potatoes, particularly fresh peeled-cut or frozen tubers. However, ICA strengthening occurs under conditions which are inconsistent with the current ideas that relate it to Ca-pectate following pectin methyl esterase (PME) activity or to formation of rhamnogalacturonan (RG)-II-borate. First, it was necessary to induce strengthening of the middle lamellar complex (MLX) and the ICA as a stress response in some plant parenchyma. As normally this syndrome does not occur uniformly enough to study it, we devised an efficient model in which ICA-strengthening is induced consistently under simulated stress by short-chain, linear, mono-carboxylic acid molecules (OAM), at 65 oC [appendix 1 (Shomer&Kaaber, 2006)]. This rapid strengthening was insufficient for allowing the involved agents assembly to be identifiable; but it enabled us to develop an efficient in vitro system on potato tuber parenchyma slices at 25 ºC for 7 days, whereas unified stress was reliably simulated by OAMs in all the tissue cells. Such consistent ICA-strengthening in vitro was found to be induced according to the unique physicochemical features of each OAM as related to its lipophilicity (Ko/w), pKa, protonated proportion, and carbon chain length by the following parameters: OAM dissociation constant (Kdiss), adsorption affinity constant (KA), number of adsorbed OAMs required for ICA response (cooperativity factor) and the water-induced ICA (ICAwater). Notably, ICA-strengthening is accompanied by cell sap leakage, reflecting cell membrane rupture. In vitro, stress simulation by OAMs at pH<pKa facilitated the consistent assembly of ICAstrengthening agents, which we were able to characterize for the first time at the molecular level within purified insoluble cell wall of ICA-strengthened tissue. (a) With solid-state NMR, we established the chemical structure and covalent binding to cell walls of suberin-like agents associated exclusively with ICA strengthening [appendix 3 (Yu et al., 2006)]; (b) Using proteomics, 8 isoforms of cell wall-bound patatin (a soluble vacuolar 42-kDa protein) were identified exclusively in ICA-strengthened tissue; (c) With light/electron microscopy, ultrastructural characterization, histochemistry and immunolabeling, we co-localized patatin and pectin in the primary cell wall and prominently in the MLX; (d) determination of cell wall composition (pectin, neutral sugars, Ca-pectate) yielded similar results in both controls and ICA-strengthened tissue, implicating factors other than PME activity, Ca2+ or borate ions; (e) X-ray powder diffraction experiments revealed that the cellulose crystallinity in the cell wall is masked by pectin and neutral sugars (mainly galactan), whereas heat or enzymatic pectin degradation exposed the crystalline cellulose structure. Thus, we found that exclusively in ICA-strengthened tissue, heat-resistant pectin is evident in the presence of patatin and suberinlike agents, where the cellulose crystallinity was more hidden than in fresh control tissue. Conclusions: Stress response ICA-strengthening is simulated consistently by OAMs at pH< pKa, although PME and formation of Ca-pectate and RG-II-borate are inhibited. By contrast, at pH>pKa and particularly at pH 7, ICA-strengthening is mostly inhibited, although PME activity and formation of Ca-pectate or RG-II-borate are known to be facilitated. We found that upon stress, vacuolar patatin is released with cell sap leakage, allowing the patatin to associate with the pectin in both the primary cell wall and the MLX. The stress response also includes formation of covalently bound suberin-like polyesters within the insoluble cell wall. The experiments validated the hypotheses, thus led to a novel picture of the structural and molecular alterations responsible for the textural behavior of potato tuber. These findings represent a breakthrough towards understanding of the hardening syndrome, laying the groundwork for potato-handling strategies that assure textural quality of industrially processed particularly in fresh peeled cut tubers, ready-to-prepare and frozen preserved products.

En Colombia, las colectas de especies con potencial agrícola iniciaron en el año 1942 con el fin de crear colecciones de trabajo para que los mejoradores del país pudieran hacer nuevos cruzamientos y crear nuevas variedades. Posteriormente, entre 1962 hasta 1994, estas colecciones fueron administradas por el ICA. En 1994, se funda la Corporación Colombiana de Investigación Agropecuaria (Corpoica, ahora AGROSAVIA) con funciones de investigación separadas del ICA. A partir de ese momento, se establece el Sistema Nacional de Bancos de Germoplasma de la Nación Colombiana (SNBGNC) animal y vegetal utilizando como base las colecciones de trabajo.

En el año 2017, la Fundación Telefónica Movistar y el Banco Interamericano de Desarrollo (BID), mediante su laboratorio de innovación BID Lab, empezaron un proyecto denominado Integración del impacto social en actividades de aceleración corporativas, que buscaba desarrollar el emprendimiento tecnológico social en dos regiones del Perú. Esta iniciativa se desplegó durante tres años en Piura e Ica junto a las socias impulsoras Universidad de Piura (UDEP) y la Cámara de Comercio, Industria y Turismo de Ica, sensibilizando a casi 5,000 personas, fortaleciendo a más de 400 emprendedores e incubando más de 70 emprendimientos en estas dos regiones. En la presente publicación Emprendimientos tecnológicos peruanos: descentralizando el impacto social, documentamos y difundimos los resultados logrados compartiendo la experiencia adquirida durante el desarrollo del proyecto, animando a seguir impulsando los ecosistemas de emprendimiento tecnológico con impacto social desde las regiones.
