Abstract:
Purpose: To investigate the effect of icariin on autophagy and apoptosis of chondrocytes, and the associated mechanisms.Methods: The chondrocytes were randomly divided into control (PBS intervention), TNF-α intervention, icarin +TNF-α, and NF-κB inhibition +TNF-α, with 8 strains in each group. The levels of IL-1, IL-6 and IL-12 were assayed by ELISA. The mRNA and protein expressions of ATG5, ATG7, Bax and Bcl-2 cells were determined by polymerase chain reaction (PCR) and Western blotting, while protein expressions of p-p65 and IκBα were assayed using Western blotting.Results: In the cartilage tissue of rats in the icariin +TNF-α group and NF-κB inhibition +TNF-α group, IL-1, IL-6 and IL-12 levels were significantly lower than those in TNF-α treatment group (p < 0.05). The AATG5 mRNA and protein in cartilage tissues of rats in icariin +TNF-α and NF-κB inhibition +TNF-α groups were significantly higher than those in TNF-α group. Bax mRNA and protein in cartilage tissues of icariin +TNF-α and NF-κB inhibition +TNF-α groups were downregulated, relative to TNF-α group; on the other hand, Bcl-2 mRNA and protein were significantly higher than those of TNF-α group (p < 0.05). In the cartilage tissues of Icarin +TNF-α, NF-κB inhibition +TNF-α groups, P-p65 protein was significantly lower than that of TNF-α (p < 0.05).Conclusion: TNF-α enhances the production of a large number of inflammatory factors by cartilage cells, inhibits autophagy of cartilage cells, and promotes cell apoptosis through regulation of NF-κB signaling pathway. Penelitian yang telah dilakukan ditujukan guna mengembangkan Lembar Kegiatan Peserta.Didik (LKPD) berbasis ICARE.pada.pokok bahasan sistem koloid untuk kelas XI SMA/MA sederajat. LKPD berbasis ICARE sebagai objek penelitian tersebut dirancang dan dikembangkan sesuai dengan tahapan model pembelajaran ICARE. Penelitian pengembangan ini mengacu pada model pengembangan 4-D. Instrumen pengumpulan data berupa lembar validasi dan angket respons pengguna. Luaran penelitian berupa LKPD berbasis ICARE dinyatakan valid berdasarkan aspek kelayakan isi, karakteristik ICARE, kebahasaan, penyajian, dan kegrafisan berturut-turut yaitu 89,81%, 91,67%, 95%, 97,92%, 100%. LKPD berbasis ICARE diuji cobakan kepada guru kimia sebanyak 3 orang dan 20 orang peserta didik dengan perolehan persentase masing-masing sebesar 91,02% dan 80,96% berada pada kriteria sangat baik. LKPD berbasis ICARE yang dikembangkan telah valid dan dapat digunakan pada materi sistem koloid.

Abstract:
Background: The Icare® PRO rebound tonometer is being promoted as a patient-friendly device for glaucoma detection and monitoring, allowing rapid and accurate intraocular pressure measurement avoiding anaesthesia. The aim of this systematic review was to examine the concordance of Icare® PRO and Goldmann applanation tonometry in glaucoma patients. Methods: The German and English literature research was carried out using PubMed and Cochrane Library between January 2010 and March 2018. Method comparison trials in a paired sample study design were identified. Search criteria were ‘Icare PRO’, ‘Rebound’, ‘Goldmann’ and ‘Applanation’. Adult patients with glaucoma of any aetiology were included. The intraindividual intraocular pressure deviation between Icare® PRO and Goldmann applanation tonometry (mm Hg) served as primary endpoint, parameterized by the reported mean differences. For each study, an asymptotic 95% confidence interval was derived. The overall intraocular pressure meta-difference (Icare® PRO-Goldmann applanation tonometry) and 95% confidence interval were estimated using the random effect model. Result: Of 147 publications, 6 studies (672 eyes) were included after reviewing. Four studies showed an intraocular pressure underestimation by the Icare® PRO, two an overestimation. The meta-difference (Icare® PRO-Goldmann applanation tonometry = −0.14 mm Hg; 95% confidence interval: (−0.43 mm Hg; 0.15 mm Hg)) indicated a non-significant, clinically irrelevant mean deviation (p = 0.335). The devices showed good agreement, but there was a strong heterogeneity between the studies (I² = 77%): two studies presented a significant difference (Icare® PRO-Goldmann applanation tonometry), however, with deviations in opposite directions. Conclusion: Although the manufacturer advertises the Icare® PRO as a ‘tonometer with superior accuracy’, the present review could neither confirm nor deny considerable agreement between Goldmann applanation tonometry and Icare® PRO in glaucomatous patients.

Penelitian ini bertujuan untuk meningkatkan hasil belajar siswa pada mata pelajaran Pengolahan Citra Digital (PCD) dengan menerapkan model pembelajaran ICARE (Introduction Connection Application Reflection Extention) dan mengetahui respon siswa terhadap penerapan model pembelajaran ICARE. Penelitian ini dilaksanakan di SMK N 1 Klungkung, dimana melibatkan siswa kelas XI MM2 (29 siswa) tahun pelajaran 2016/2017. Desain penelitian ini adalah penelitian tindakan kelas (PTK) yang terdiri dari 2 siklus, data yang dikumpulkan dalam penelitian ini adalah hasil belajar dan respon siswa terhadap penerapan model pembelajaran ICARE pada mata pelajaran Pengolahan Citra Digital (PCD). Data hasil belajar siswa dikumpulkan melalui tes kognitif dan lembar observasi afektif serta psikomotor, sedangkan data respon siswa dikumpulkan dengan lembar angket tertutup. Hasil penelitian menunjukkan bahwa (1) terdapat peningkatan hasil belajar siswa. Hal ini terlihat dari ketuntasan klasikal siswa yang diperoleh pada siklus I sebesar 17,24%, dan pada siklus II sebesar 100,00%. Hasil belajar ini mengalami peningkatan sebanyak 82,76%. (2) rata-rata respon siswa terhadap terhadap penerapan model pembelajaran ICARE tergolong positif 65,25 peneliti menyimpulkan bahwa penerapan model pembelajaran ICARE pada mata pelajaran Pengolahan Citra Digital (PCD) dapat meningkatkan hasil belajar dan mendapat respon yang positif dari siswa. Kata Kunci : ICARE (Introduction Connection Application Reflection Extention), PCD, Hasil belajar, Respon siswa. This study aimed at improving students’ learning outcomes on the subject of Technology of information by implementing ICARE (Introduction Connection Application Reflection Extention) instructional model and finding out the students’ responses toward the implementation of ICARE instructional model. This research was conducted at SMK N 1 Klungkung which involved the students of class XI MM2 (29 students) in the academic year 2016/2017. This research was designed in the form of Class Action Researh which consisted of 2 cycles. The data which were gathered in this research were the students’ learning outcomes and responses toward the implementation of ICARE instructional model on the subject of Digital Image Processing. The data of the students’ learning outcomes were gathered by means of cognitive test as well as affective and psikomotor observation sheet, while the data of the students’ responses were gathered by closed questionnaire. The result of this study showed that (1) there was an improvement of the students’ learning outcomes. This can be seen from the classical mastery of the students obtained in the fist cycle was 17.24%, and in the second cycle was 100.00%. The learning outcomes improved by 82.76%. (2) The students responses on the average toward the implementation of ICARE instructional model could be classified as positive that was equal to 65.25. This Research aims to develop Student Activity Worksheet (LKPD) based on ICARE of determination of the enthalpy changes subject for class XI SMA/MA valid based on content feasibilities, characteristics ICARE, linguistic, presentation, and graphics. The type of this research is development research (Research and Development) with the process referring to the plomp development model. The object of this research is LKPD based on ICARE. Data collection instruments was a validation sheets filled by 3 validators, the results of the validity data analysis of feasibility aspect of content, characteristics of ICARE, language, presentation and graphics are 92,59%, 96,67%, 95%, 97,22%, and 95,83% with valid category. LKPD that has been valid by validators was tested on 20 students and got response results with score 3,45 with very good criteria, and 3 teachers got response result with score 3,6 with very good criteria. From results of analysis data can conclusion that LKPD based on ICARE developed was valid and can be applied for determination of the enthalpy changes subject.

AbstractVision health is recognized as a critical unmet need in North America. The ocular morbidity associated with glaucoma results from increased intraocular pressure (IOP) and early detection is crucial for the management of glaucoma. Our objective was to find a diagnostically accurate screening tool for intraocular hypertension that can be used in rural communities. We sought to validate the diagnostic accuracy of the iCare rebound tonometer against the gold standard Perkins applanation tonometer (PAT) in measuring IOP.Patients from two rural communities in Ontario, Canada visiting their optometrists for routine appointments had their IOP measured by a non-contact tonometer (NCT), an iCare rebound tonometer, and a Perkins applanation tonometer (PAT). Values of sensitivity, specificity, and likelihood ratios for a positive and negative result were calculated for the iCare and the NCT.Complete data was collected from 209 patients. Overall, the iCare tonometer had high levels of validity, as compared to the gold standard PAT. The iCare tonometer displayed excellent sensitivity of 98.3% (90–99%, 95% CI) and excellent negative likelihood ratio of 0.024 (0.0088–0.066, 95% CI) which is useful for ruling out intraocular hypertension.The iCare tonometer is a reasonably valid tool for detecting elevated IOP. Its ease of use, simplicity, and accessibility makes it a good screening tool to improve eye health in rural areas.

Purpose. To examine the influence of positional misalignments on intraocular pressure (IOP) measurement with a rebound tonometer.Methods. Using the iCare rebound tonometer, IOP readings were taken from the right eye of 36 healthy subjects at the central corneal apex (CC) and compared to IOP measures using the Goldmann applanation tonometer (GAT). Using a bespoke rig, iCare IOP readings were also taken 2 mm laterally from CC, both nasally and temporally, along with angular deviations of 5 and 10 degrees, both nasally and temporally to the visual axis.Results. Mean IOP ± SD, as measured by GAT, was14.7±2.5 mmHgversusiCare tonometer readings of17.4±3.6 mmHg at CC, representing an iCare IOP overestimation of2.7±2.8 mmHg (P<0.001), which increased at higher average IOPs. IOP at CC using the iCare tonometer was not significantly different to values at lateral displacements. IOP was marginally underestimated with angular deviation of the probe but only reaching significance at 10 degrees nasally.Conclusions. As shown previously, the iCare tonometer overestimates IOP compared to GAT. However, IOP measurement in normal, healthy subjects using the iCare rebound tonometer appears insensitive to misalignments. An IOP underestimation of <1 mmHg with the probe deviated 10 degrees nasally reached statistical but not clinical significance levels.

Abstract:
Introduction: Contact Goldmann applanation tonometry (GAT) is gold standard for measuring intraocular pressure; however its routine use is limited by its non-portability and the need for a Slit Lamp Microscope. The Portable Perkins tonometer is also considered gold standard because it is based on the same principles as the GAT. The iCare is a newly introduced, portable, non-contact tonometer (NCT) that measures intraocular pressure (IOP) using a thin metallic probe. Objective: To evaluate reliability and accuracy of IOP measurements using iCare and Keeler Pulsair tonometers against Perkins tonometer. Subjects and methods: A comparative, randomized, prospective clinical study conducted on 166 eyes of 83 (n=83) subjects in the age group 14 to 71 years. The pressures were first recorded by iCare and Pulsair and then by Perkins. The SPSS 11.00 version was used for analysis. Results: Mean pressures and standard deviation (+/-SD) for iCare, Pulsair and Perkins were 14.62(+/- 2.47), 14.53(+/-3.36) and 13.06(+/-2.69) and the Standard Error of the Mean (SEM) was 0.27, 0.36 and 0.30 respectively. There was a good correlation between iCare and Perkins with statistically significant difference (r=0.610, p <0.05). Regression analysis was performed. Using the Bland-Altman analysis 95% Limits of Agreement (LoA) for iCare and Pulsair were determined as -6.1 to 2.9 and -4.5 to 7.5 respectively. Conclusion: Although both tonometers overestimated the Perkins values, Pulsair showed a better agreement with Perkins tonometer than iCare tonometer. Abstract:
Purpose. To evaluate the different IOP readings by Goldmann applanation tonometer (GAT), ICare rebound tonometer, and Tono-Pen in keratoconus patients after MyoRing implantation. To assess the influence of central corneal thickness (CCT) and thinnest corneal location (TCL) on IOP measurements by different tonometers. Setting. Prospective observational study was conducted in two private centers in Egypt from February 2015 to November 2016. Methods. Seventeen eyes of 10 patients suffering from keratoconus and who underwent MyoRing implantation were recruited. All subjects underwent GAT, ICare, and Tono-Pen IOP measurements in random order. Central corneal thickness and thinnest corneal location were assessed by Pentacam. Difference in mean in IOP readings was assessed by T-test. Correlation between each pair of devices was evaluated by Pearson correlation coefficient. The Bland–Altman analysis was used to assess intertonometer agreement. Results. Seventeen eyes (10 patients) were evaluated. The mean IOP reading was 13.9 ± 3.68, 12.41 ± 2.87, and 14.29 ± 1.31 mmHg in GAT, ICare, and Tono-Pen group, respectively. There was a significant difference between IOP readings by GAT/ICare and Tono-Pen/ICare (p value: 0.032 and 0.002, respectively) with no significant difference between GAT/Tono-Pen (p value: 0.554). Mean difference in IOP measurements between GAT/ICare was 1.49 ± 2.61 mmHg, Tono-Pen/ICare was 1.89 ± 2.15 mmHg, and GAT/Tono-Pen was −0.39 ± 2.59 mmHg. There was no significant correlation between the difference in IOP readings among any pair of devices and CCC or TCL. The Bland–Altman analysis showed a reasonable agreement between any pair of tonometers.

Abstract Background External validation of risk models is critical for risk-stratified breast cancer prevention. We used the Individualized Coherent Absolute Risk Estimation (iCARE) as a flexible tool for risk model development and comparative model validation and to make projections for population risk stratification. Methods Performance of two recently developed models, one based on the Breast and Prostate Cancer Cohort Consortium analysis (iCARE-BPC3) and another based on a literature review (iCARE-Lit), were compared with two established models (Breast Cancer Risk Assessment Tool and International Breast Cancer Intervention Study Model) based on classical risk factors in a UK-based cohort of 64 874 white non-Hispanic women (863 patients) age 35–74 years. Risk projections in a target population of US white non-Hispanic women age 50–70 years assessed potential improvements in risk stratification by adding mammographic breast density (MD) and polygenic risk score (PRS). Results The best calibrated models were iCARE-Lit (expected to observed number of cases [E/O] = 0.98, 95% confidence interval [CI] = 0.87 to 1.11) for women younger than 50 years, and iCARE-BPC3 (E/O = 1.00, 95% CI = 0.93 to 1.09) for women 50 years or older. Risk projections using iCARE-BPC3 indicated classical risk factors can identify approximately 500 000 women at moderate to high risk (&gt;3% 5-year risk) in the target population. Addition of MD and a 313-variant PRS is expected to increase this number to approximately 3.5 million women, and among them, approximately 153 000 are expected to develop invasive breast cancer within 5 years. Conclusions iCARE models based on classical risk factors perform similarly to or better than BCRAT or IBIS in white non-Hispanic women. Addition of MD and PRS can lead to substantial improvements in risk stratification. However, these integrated models require independent prospective validation before broad clinical applications.

Abstract Background: It has been reported that reusing Icare tonometer probes may not pose a considerable risk of transmission of infection, thereby supporting this practice under extraordinary circumstances, such as mass glaucoma screenings in developing countries. The present study sought to determine whether reusing the probes reduced the validity of pressure readings. Methods: Thousands of measurements were made with the same Icare probe on a purpose-built calibration device. A second set of measurements was made during which the Icare probe head was wiped with an alcohol pad between each reading. Results: Statistically different readings were observed between the no-wipe and wipe groups for most of the settings evaluated. In addition, statistically significant readings were noted for a few of the settings as the number of readings increased for either group. However, these differences would not be considered clinically significant. Conclusion: This study suggests that reusing Icare tonometer probes, even for hundreds of measurements, would not reduce the clinical utility of the pressure readings. For mass screenings in developing countries where cost savings is crucial, considering the reuse of Icare tonometer probes seems reasonable.

Abstract:
Purpose: To evaluate and compare the accuracy of self-measurement of intraocular pressure using Icare Home rebound tonometer with Goldmann applanation tonometer and assess acceptability of self-tonometry in patients with glaucoma and ocular hypertension. Methods: In the study, 117 subjects were trained to use Icare Home for self-measurement. Icare Home tonometer readings were compared with Goldmann applanation tonometer, including one eye per patient. Agreement between the two methods of measurement was evaluated by Bland and Altmann analysis. Questionnaire was used to evaluate patients’ perception of self-tonometry. Results: One hundred and three out of 117 patients (88%) were able to measure their own intraocular pressure and 96 (82%) fulfilled the requirements for certification. The mean (SD) difference Goldmann applanation tonometer minus Icare Home was 1.2 (2.4) mmHg (95% limits of agreement, –3.4 to 5.9 mmHg). The magnitude of bias between the two methods depended on central corneal thickness, with greater bias at central corneal thickness <500 µm. In 65 out of 96 subjects (67.7%), Icare Home results were within 2 mmHg of the Goldmann applanation tonometer. Seventy-three out of 93 (78.5%) felt that self-tonometry was easy to use and 75 patients (80.6%) responded that they would use the device at home. Conclusion: Icare Home tonometry tends to slightly underestimate intraocular pressure compared to Goldmann applanation tonometer. Most patients were able to perform self-tonometry and found it acceptable for home use. Measurements using rebound self-tonometry could improve the quality of intraocular pressure data and optimize treatment regimen.

Modern scholars have linked this one-line fragment with a Hellenistic theory that placed the origin of drama in the village of Icaria, in the Attic countryside. It is likely that the fragment did mention the word ‘billy-goat’ (τράγος), but there are no definite elements in favour of the hypothesis that Eratosthenes devised a theory on the origin of tragedy in Icaria.

Abstract:
In this study, we used LS174T cells as a model to investigate the protective effects of icariin and phosphorylated icariin on LPS-induced goblet cell dysfunction. Our results indicated that icariin and phosphorylated icariin increased the cell viability and decreased lactate dehydrogenase activity in LPS-treated LS174T cells. Icariin and phosphorylated icariin attenuated LPS-induced changes in mucin 2 synthesis and secretion. Besides, Icariin and phosphorylated icariin reduced the levels of ROS, MDA, and H2O2 and increased the activity of SOD, GPx, CAT, and T-AOC in LPS-treated LS174T cells. Moreover, the levels of IL-1β, IL-6, IL-8, and TNF-α were reduced in the Icariin and phosphorylated icariin group. Furthermore, Icariin and phosphorylated icariin decreased gene abundance or enzyme activity of Bip, XBP1, GRP78, CHOP, caspase-3, and caspase-4 in LPS-treated LS174T cells. Our data suggest that Icariin and phosphorylated icariin effectively attenuate LPS-induced intestinal goblet cell function damage through regulating oxidative stress, inflammation, apoptosis, and mucin expression.

This article describes the prevalence of violent victimization and injuries among U.S. adults and examines how these estimates differ by individual- and household-level characteristics using the second nationally representative Injury Control and Risk Survey (ICARIS-2). The ICARIS-2 was administered to 9,684 adults using a computer assisted random-digit-dial telephone survey. These data suggest that 5.4% of the U.S. adult population, approximately 11.66 million people, experienced at least one violent victimization in the past 12 months. Most victims (57%) believed that the person who struck them intended to injure them, and one in three victims reported that they were physically injured on at least one occasion. Effective violence prevention strategies require the collection of valid data to understand the scope of the problem, the consequences, and the groups most impacted. The results from ICARIS-2 indicate that the prevalence of violent victimization in the United States far exceeds the estimates derived from crime surveys.

Jenis penelitian ini adalah eksperimen semu dengan desain penelitian adalah Post Test Only Control Group Design. Populasi penelitian ini adalah seluruh siswa kelas VIII SMP Laboratorium Undiksha Singaraja tahun ajaran 2018/2019 yang berjumlah 98 orang danterdistribusi ke dalam 4 kelas. Sampel dalam penelitian ini adalah siswa kelas VIII-1 dan VIII-4 yang diambil menggunakkan teknik cluster random sampling. Data kemampuan pemecahan masalah matematika dikumpulkan dengan tes kemampuan pemecahan masalah matematika yang diberikan kepada siswa dalam bentuk tes uraian serta diberikan di akhir penelitian. Data hasil tes kemampuan pemecahan masalah matematika dianalisis menggunakan uji-t satu ekor dengan taraf siginifikansi 5%. Hasil penelitian menunjukkan bahwa thitun=3,3996>ttabel=2,0117sehingga H0 ditolak. Ini berarti, kemampuan pemecahan masalah matematika siswa yang dibelajarkan dengan model pembelajaran ICARE berbantuan masalah matematika terbuka lebih baik daripada kemampuan pemecahan masalah matematika siswa yang dibelajarkan dengan model pembelajaran konvensional. Background. Stroke patients with mild-moderate upper extremity motor impairments and minimal sensory and cognitive deficits provide a useful model to study recovery and improve rehabilitation. Laboratory-based investigators use lesioning techniques for similar goals. Objective. To determine whether stroke lesions in an upper extremity rehabilitation trial cohort match lesions from the preclinical stroke recovery models used to drive translational research. Methods. Clinical neuroimages from 297 participants enrolled in the Interdisciplinary Comprehensive Arm Rehabilitation Evaluation (ICARE) study were reviewed. Images were characterized based on lesion type (ischemic or hemorrhagic), volume, vascular territory, depth (cortical gray matter, cortical white matter, subcortical), old strokes, and leukoaraiosis. Lesions were compared with those of preclinical stroke models commonly used to study upper limb recovery. Results. Among the ischemic stroke participants, median infarct volume was 1.8 mL, with most lesions confined to subcortical structures (61%) including the anterior choroidal artery territory (30%) and the pons (23%). Of ICARE participants, <1% had lesions resembling proximal middle cerebral artery or surface vessel occlusion models. Preclinical models of subcortical white matter injury best resembled the ICARE population (33%). Intracranial hemorrhage participants had small (median 12.5 mL) lesions that best matched the capsular hematoma preclinical model. Conclusions. ICARE subjects are not representative of all stroke patients, but they represent a clinically and scientifically important subgroup. Compared with lesions in general stroke populations and widely studied animal models of recovery, ICARE participants had smaller, more subcortically based strokes. Improved preclinical-clinical translational efforts may require better alignment of lesions between preclinical and human stroke recovery models.

Keterampilan berpikir kreatif merupakan salah satu cara berpikir yang dibutuhkan dalam kehidupan kerja dan masyarakat di abad 21, yang harus dilatihkan kepad apeserta didik Ketika proses pembelajaran. Namun kenyataannya, keterampilan berpikir kreatif ini jarang sekali dilatihkan kepada peserta didik dalam proses pembelajaran. Maka penelitian ini bertujuan untuk menerapkan model pembelajaran ICARE untuk meningkatkan keterampilan berpikir kreatif peserta didik pada materi alat optik. Digunakan metode penelitian semu (quasi experiment) dengan desain one group pretest posttest. Instrumen penelitian berupa soal uraian sebanyak 12 soal yang sudah diuji validitas dan reliabilitasnya. Partisipan dalam penelitian ini adalah 24 peserta didik, dengan mengunakan teknik analisis data gain ternormalisasi untuk melihat peningkatan keterampilan berpikir kreatif dan uji effect size untuk mengetahui efek dari model pembelajaran ICARE. Hasil penelitian berupa nilai gain ternormalisasi sebesar 0,44 yang termasuk kategori sedang dengan taraf signifikansi 0,05. Selanjutnya diperoleh hasil uji effect size untuk keterampilan berpikir kreatif sebesar 3,47 yang termasuk pada kategori tinggi. Hasil ini menunjukan bahwa model pembelajaran ICARE efektif dalam meningkatkan keterampilan berpikir kreatif peserta didik pada materi alat optik. One of the skills needed to solve problems is creative thinking skills. Creative thinking skills are one of the ways of thinking that a person needs in work life and society in the 21st century. However, in reality, these creative thinking skills are not trained to students in the learning process. So, this study aims to apply the ICARE learning model to improve the creative thinking skills of class XI SMA students on optical instrument material. A quasi-experimental research method was used with a one-group pretest-posttest research design. The instrument in this study was 12 questions that were reliable and valid in terms of descriptions of creative thinking skills and were given a score of 1-4 in accordance with the assessment rubric. Participants in this study were 24 students, with data analysis techniques using N-Gain and the effect size to determine the effect of the ICARE learning model. The result of this research is a normalized gain value of 0.44 which is included in the medium category with a significance level of 0.05. Furthermore, the effect size test results obtained for creative thinking skills of 3.47 which are included in the high category. These results that the ICARE learning model is effective in improving students' creative thinking skills in optical instrument material. This study investigated icariin (ICA) nanoparticles on angiogenesis in rats with pulmonary fibrosis and its mechanism. First, icariin solid nanoliposomes (ICA-SLN) were prepared. The in vitrorelease of icariin nanoparticles was determined using a UV-Vis spectrophotometer, after which the plasma concentration of icariin nanoparticles in rats was determined. The bioavailability of icariin nanoparticles was investigated, and the effect of icariin on angiogenesis of pulmonary fibrosis rats was re-observed. The results showed that the bioavailability of icariin in vivo was enhanced after nanomodification, which indicated that icariin solid nanoliposome was a good choice for oral sustained-release nanocarrier materials. in vivo experiments showed that icariin could significantly inhibit angiogenesis in rats with pulmonary fibrosis, and the inhibitory effect was related to the dose and time of action. Most importantly, this study provides the possibility of icariin as a targeted agent for future-targeted therapy.

Impaired endothelial nitric oxide synthase (eNOS)/nitric oxide (NO) pathway induces atherogenesis. The present study examined whether icariin improves the eNOS/NO pathway to prohibit the atherogenesis of apolipoprotein E-null (ApoE−/−) mice. In vitro, primary human umbilical vein endothelial cells (HUVECs) were randomly divided into 7 groups: control; vehicle; icariin 10; lyphosphatidylcholine (LPC) group; LPC + icariin 1; LPC + icariin 3; and LPC + icariin 10. In vivo, 80 mice were separated randomly into 4 groups (n = 20): control, ApoE−/−, ApoE−/− + icariin 10, and ApoE−/− + icariin 30. ApoE−/− mice had significantly more atherosclerosis in the aortic root together with increased aortic ROS production, body mass, plasma triglyceride (TG) and total cholesterol (TC) concentration, decreased aortic eNOS expression, and plasma NO concentration. LPC (10 μg/mL) treatment induced a big decline in NO level in the conditioned medium and eNOS expression, and an increase in intracellular reactive oxygen species (ROS) production in HUVECs. Icariin treatment decreased atherogenesis, ROS production, body mass, plasma TG concentration, and plasma TC concentration, and increased NO concentration and eNOS expression. These findings suggested icariin could improve eNOS/NO-pathway to prohibit the atherogenesis of apolipoprotein E-null mice by restraining oxidative stress.

The aim of the present work was to explore the anti-hepatoma effects of icariin both in vitro and in vivo and to elucidate its potential mechanism of action. The MTT assay was applied to test the anti-proliferative effects of icariin in vitro. HepG2 bearing NMRI nu/nu mice were used to test the anticancer effects of icariin in vivo. Immunohistochemical assay and flow cytometry assay (FACS) were applied to detect the possible mechanisms of action of icariin. MTT assay illustrated that icariin inhibited the proliferation of HepG2 cells in a concentration dependent manner; meanwhile, icariin inhibited the tumor growth in HepG2 bearing NMRI nu/nu mice. The tumor weight was inhibited by 55.6% and tumor volume was inhibited by 47.2%. Icariin did not influence the spleen and body weights or blood parameters. Immunohistochemical analysis indicated that the expressions of both CD31 and Ki67 in the icariin treated group were significantly lower than those in the control group (p < 0.01). FACS assay showed that icariin dramatically decreased the percentage of CD4+ and CD8+ cells in bone marrow and CD19+ cells in blood on day 8. On day 17, the percentage of CD8+ cells in blood was lower than those in the control group. CD4/CD8 ratio in icariin group was significantly elevated in bone marrow on day 17. Icariin showed anticancer efficacy both in vitro and in vivo. The possible mechanism of action could be related to its anti-angiogenesis and anti-proliferative effects in tumors.

Background: Natural products often have novel frameworks and unique mechanisms, It is an important way to develop new anti-tumor drugs.The paper explored the effect of Icariin on HepG2 cell apoptosis and cell cycle arrest. Material and methods: Human liver cancer HepG2 cells were studied. The biological activity of Icariin on HepG2 cells was comprehensively investigated. The mechanism was preliminarily explored from the aspects of proliferation, cell cycle, and apoptosis. Results: Icariin showed significant inhibitory effect on cell proliferation after administration, and the effect was time-and-concentration-dependent. Annexin V-PI detection showed that, after 48 hours of administration of Icariin at different concentrations, the apoptosis rate of HepG2 cells increased in a concentration-dependent manner. The results of Hoechst 33342 staining showed that, after 48 hours of intervention with Icariin at different concentrations, HepG2 cells appeared densely stained and granular fluorescence, characterizing apoptosis. In the JC-1 mitochondrial membrane potential experiment, Icariin was found to destroy cell mitochondrial membrane potential and induce HepG2 cell apoptosis. After 48 h administration of Icariin at different concentrations, Bcl-2 and survivin proteins were down-regulated, and Bax was up-regulated, both in a concentration-dependent manner. PI single staining combined with flow cytometry to detect cell cycle results showed that, Icariin can induce G2/M phase arrest of HepG2 cells, and is time-and-concentration-dependent. Western Blot detection revealed that Icariin can down-regulate the cycle-related proteins Cyclin B and CDK1 in a concentration-dependent manner, and also significantly down-regulate the expressions of p-AKT, AKT, p-ERK and ERK proteins. Conclusion: Icariin is a selectively potential active compound that can treat liver cancer. The paper provided theoretical basis and experimental support for the clinical application of Icariin in the drug treatment of liver cancer. It is necessary to further study the antitumor effect of Icariin, explore and find the target, and provide higher selectivity for the treatment of liver cancer by Icariin.

Icariin, the main effective component extracted from epimedium, has been shown to stimulate osteogenic differentiation and bone formation and to increase synthesis of the cartilage extracellular matrix. However, there has been little study on the effects of icariin on osteoarthritis. In this study, we loaded icariin onto poly(lactic-co-glycolic acid) (PLGA) electrospinning. The aim of this study was to explore a composite scaffold and to inhibit the progression of osteoarthritis. Our main experimental results demonstrated that the PLGA/icariin composite spinning scaffold had higher hydrophilicity, and icariin was released slowly and steadily from the scaffold. According to the results of an MTT test, immunofluorescence staining, an alkaline phosphate activating assay and a real-time polymerase chain reaction (RT-PCR) assay, the PLGA/icariin composite scaffold had good biocompatibility. In models of osteoarthritis, the results of a RT-PCR assay indicated that the PLGA/icariin scaffold promoted the synthesis of the extracellular matrix. The results of X-ray microtomography and histological evaluation demonstrated that the PLGA/icariin scaffold maintained the functional morphology of articular cartilage and inhibited the resorption of subchondral bone trabeculae. These findings indicated that the PLGA and icariin composite scaffold has therapeutic potential for use in the treatment of osteoarthritis.

Icariin has been mostly reported to enhance bone fracture healing and treat postmenopausal osteoporosis in ovariectomized animal model. As another novel animal model of osteoporosis, there is few publication about the effect of Icariin on osteoprotegerin-deficient mice. Therefore, the goal of this study is to find the effect on bone formation and underlying mechanisms of Icariin in osteoprotegerin (OPG) knockout (KO) mice. We found that Icariin significantly stimulated new bone formation after local injection over the surface of calvaria at the dose of 5 mg/kg per day. With this dose, Icariin was also capable of significantly reversing OPG-deficient-induced bone loss and bone strength reduction. Real-time PCR analysis showed that Icariin significantly upregulated the expression of BMP2, BMP4, RUNX2, OC, Wnt1, and Wnt3a in OPG KO mice. Icariin also significantly increased the expression of AXIN2, DKK1, TCF1, and LEF1, which are the direct target genes ofβ-catenin signaling. The in vitro studies showed that Icariin induced osteoblast differentiation through the activation of Wnt/β-catenin-BMP signaling by in vitro deletion of theβ-catenin gene usingβ-cateninfx/fxmice. Together, our findings demonstrate that Icariin significantly reverses the phenotypes of OPG-deficient mice through the activation of Wnt/β-catenin-BMP signaling.

Viral myocarditis (VM) is an inflammatory lesion of the myocardium caused by a viral infection. Icariin has anti-viral effects, but its therapeutic effect and mechanism of VM action are still not clear. This study evaluated icariin’s intervention effect on the myocardial cAMP-PKA signaling pathway and explored the possible mechanism of icariin’s effect. The results showed that a high dose of icariin could improve VM rats’ quality of life. Icariin-treated rats had lower mortality, insignificant weight loss, and decreased CK-MB and LDH. Hematoxylin-Eosin (HE) staining to observe the pathological changes of rats. The myocardium of VM rats is severely damaged, and myocardial cells have dissolution, necrosis, degeneration, and inflammatory cell infiltration. The degree of myocardial pathology in rats after icariin treatment is less than that in VM model rats. ELISA detected the serum IL-6 and IL-10 levels, and it was found that the levels of IL-6 and IL-10 in rats after icariin treatment were lower than those in model rats. Prove that icariin plays a protective effect on cardiomyocytes. The positive expression of Fas in the myocardium was measured using the immunohistochemical method. The results demonstrated that the positive expression of Fas in rats was reduced after icariin treatment. The results proved that icariin reduced cardiomyocyte apoptosis. Western Blot detected myocardial cAMP and PKA protein content. In contrast with model rats, icariin reduced the cAMP and PKA levels of VM rats, showing that icariin reduces myocardial cell damage.

This research explored the use of the ICARE Model to studying the writing ability at the Senior high school in Parepare. ICARE includes five essential elements of a good learning experience, can be applied to children, teenagers, and parents, namely Introduction, Connection, Application, Reflection, and Extension. This research involved a population from Senior high school in Parepare in the academic year 2015/2016. The students consist of two classes, an experimental class, and a control class. Each class consists of 36 students. This research utilized a cluster random sampling technique to select the sample. The instrument used in this research is the writing test. The data were analyzed by using t-test. The research reveals that there was a significant increase in students writing ability. This finding indicates that the mean score of the post-test of the experimental group is getting much better than the control class. It meant that using ICARE developed the students' ability in writing, and the writing component increased the highest is in the vocabulary aspect.

Neurosurgeons performing intracerebral hemorrhage evacuation procedures have limited options for monitoring hematoma evacuation and assessing residual hematoma burden intraoperatively. Here, we report the successful neuroendoscopic adaptation of intravascular ultrasound, referred to here as intracavitary ultrasound (ICARUS), in two patients. Pre-evacuation ICARUS demonstrated dense hematomas in both patients. Post-evacuation ICARUS in patient 1 demonstrated significant reduction in clot burden and two focal hyperechoic regions consistent with pockets of hematoma not previously seen with the endoscope or burr hole ultrasound. These areas were directly targeted and resected with the endoscope and suction device. Post-evacuation ICARUS in patient 2 showed significant reduction of hematoma volume without indication of residual blood. ICARUS findings were confirmed on intraoperative DynaCT and postoperative CT 24 hours later. ICARUS is feasibly performed in a hematoma cavity both before and after hematoma aspiration. ICARUS may provide additional information to the operating surgeon and assist in maximizing hematoma removal.

In this study, we aimed to investigate the effects of icariin and its intestinal metabolites on the activities of human UDP-glucuronosyltransferase (UGT) activities. Using a panel of recombinant human UGT isoforms, we found that icariin exhibited potent inhibition against UGT1A3. It is interesting that the intestinal metabolites of icariin exhibited a different inhibition profile compared with icariin. Different from icariin, icariside II was a potent inhibitor of UGT1A4, UGT1A7, UGT1A9, and UGT2B7, and icaritin was a potent inhibitor of UGT1A7 and UGT1A9. The potential for drug interactionsin vivowas also quantitatively predicted and compared. The quantitative prediction of risks indicated thatin vivoinhibition against intestinal UGT1A3, UGT1A4, and UGT1A7 would likely occur after oral administration of icariin products.

Critical thinking skill plays an important role in achieving maximum learning outcomes. The lack of involvement in critical thinking skills activities impacts students’ learning outcomes. This study aims to analyze whether the ICARE learning model application combined with constructivism-based learning modules can improve students' critical thinking skills and learning outcomes. The research method is an experiment with a pretest-posttest control group design. This study was conducted in MAN 4 Aceh Selatan and MAS Ashhabul Yamin Aceh Selatan during the even semester (2020/2021 Academic Year) with the population of 196 students of science class XI. The research sample was 119 students, namely the experimental group 58 and the control group 60. The parameters measured are critical thinking skills and learning outcomes. Data on critical thinking skills and learning outcomes were analyzed by parametric statistics using an independent sample t test, while to observe the correlation of critical thinking skills with learning outcomes was conducted through correlation and regression analysis. The results showed that there was an increase in critical thinking skills and student learning outcomes after being taught using the application of the ICARE learning model combined with constructivism-based learning modules on the respiratory system material. The correlation coefficient obtained is 0.832 which indicates there is a positive relationship between critical thinking skills and learning outcomes with strong interpretations. The conclusion of this study is that the application of the ICARE learning model combined with constructivism-based learning modules can improve students’ critical thinking skills and learning outcomes on respiratory system material. Abstract Background Online interventions to prevent mental health problems have proven to be effective. However, knowledge about their implementation in real-world practice as well as for dissemination to the target groups in different settings is scarce. The goal of the ‘ICare’ network is to establish a comprehensive model of eMental-health service delivery in and across different European countries. Since implementation and dissemination are influenced by many contextual factors, in the first phase of ICare a stakeholder survey was conducted. The survey aim was to explore stakeholders’ experiences, needs and attitudes regarding Internet-based prevention of mental health problems and hindering and fostering factors for implementation and dissemination. This article is part of a supplement and describes the design of the stakeholder survey. Survey results are published in separate articles in the same supplement. Methods Based on a literature review and the individual characteristics of the ICare interventions, stakeholder groups were identified in different settings across six European countries. The RE-AIM framework guided the development of the research questions and survey instruments. A concurrent mixed methods design was applied comprising focus groups with the intended target groups of ICare interventions, an online questionnaire with potential facilitators/delivery staff and semi-structured interviews with policy makers. Conclusion The challenge was to develop a design that allowed flexibility but at the same did not jeopardize the validity of the study. Implications drawn from this survey are not restricted to specific preventive interventions but will provide general information on how online mental illness prevention can be best implemented in various settings.

Rheumatoid arthritis (RA) is a chronic autoimmune inflammatory disease that causes deformity of the joints and physical disability. Icariin, a natural flavonoid glucoside isolated from plants in theEpimediumfamily, has been proven to have various pharmacological activities. A recent study showed that icariin suppressed cartilage and bone degradation in mice of collagen-induced arthritis. However, the mechanism needs to be further investigated. In our current study, we found that icariin reduced the arthritis score and the incidence of arthritis compared with that in mice treated with water. Icariin inhibits the expression of various osteoclastogenic markers, such asβ3 integrin, cathepsin K, and MMP9in vitro. Icariin treatment in mice with CIA also resulted in less number of Th17 cells and decreased ratio of CD4+IL-17+cells. The alleviated arthritis score and incidence of arthritis and reduced serum levels of IgG2a induced by icariin were abolished with additional IL-17 administration. Furthermore, icariin inhibited STAT3 activation in T cells and STAT3 inhibitor resulted in decreased IL-17 production and alleviated RA. In conclusion, icariin decreases Th17 cells and suppresses the production of IL-17, which contributes to the alleviated rheumatoid arthritis, through the inhibition of STAT3 activation.

Preclinical literature suggests that icariin, a flavonoid found in Epimedium, may have potential for medical and psychiatric conditions. The objective of this study was to examine the safety, tolerability, and pharmacokinetics of orally administered icariin at doses of 100 to 1,680 mg/day in 24 healthy adult participants. Cognition, mood, and side effects were assessed over 5 days. Multiple blood samples were obtained over 24 hours to assess bioavailability and pharmacokinetics. Data were analyzed using a Wilcoxon signed rank test and Mann-Whitney U test. At all doses, either very low or undetectable blood levels of icariin were observed, demonstrating the low bioavailability of the oral formulation and preventing a determination of pharmacokinetic properties. No significant between-group differences were observed on side effect scales, either by self-report, or on cognitive assessments. A statistically significant, but not clinically significant, increase in self-reported depressive symptom severity was observed with icariin relative to placebo. Tolerability of icariin was good except at the highest dose. Two participants receiving 1,680 mg of icariin discontinued the study drug due to gastrointestinal symptoms. Bioavailability of oral icariin appears to be low at all doses tested. Although icariin appears generally to have a favorable tolerability profile, the highest doses may be associated with gastrointestinal distress. Different drug formulation and delivery method may be needed to assess the pharmacokinetic profile of icariin adequately.

Diabetic Osteoporosis (DOP) is a common metabolic bone disease, characterized by decreased bone mineral density (BMD) and destruction of bone microstructure. It has been reported that icariin is beneficial for estrogen deficiency-induced osteoporosis, and alcohol-induced osteoporosis; whether icariin has protective effects on diabetes-induced osteoporosis has not been reported. In this study, a rat model of diabetic osteoporosis was established by streptozotocin injection, the bone protective effects and potential mechanism of icariin on diabetes-induced bone loss was observed. Thirty 8-week-old female Sprague Dawley rats were divided into control group (vehicle treatment), T1DM (diabetic) group and T1DM-icariin (ICA) group (diabetic rats treated with icariin), 10 rats in each group. The bone histomorphometry parameters, bone mineral density (BMD), serum bone turnover markers, and bone marrow adipogenesis were analyzed after 8 weeks of icariin administration. The results showed consumption of icariin at a doses of 100 mg kg−1 decreased blood glucose, and increased the BMD of diabetic rats. Icariin effectively decreased serum bone turnover marker levels, including CTX-1, ALP, TRACP 5b, osteocalcin, and PINP. Meanwhile, the bone histomorphometry parameters, the number of osteoclasts per bone perimeter were turned to be normal level, and the icariin treatment suppressed bone marrow adipogenesis. The runt-related transcription factor 2 (RUNX 2), as well as the osteoprotegerin (OPG)/receptor activator of nuclear factor-κ B ligand (RANKL) ratio in serum and bone tissues were increased significantly after icariin treatment in diabetic rats. All of the above indicate that oral administration of icariin can prevent diabetic osteoporosis; the effect is mainly related to its ability to reduce blood glucose, inhibit bone turnover and bone marrow adipogenesis, as well as up-regulate bone RUNX 2, and OPG expression.
