Abstract:

This research is aligned with the Social Development perspective, which proposes that, in addition to economic criteria, the development must be measured also from social, political, cultural and emotional concerning to individuals, groups and territories. On this perspective, the happiness studies seek to determine the level of satisfaction of individuals with their own lives, being taken into consideration the analysis of socioeconomic, demographic, environmental, cultural and emotional factors that can present connections with the happiness of the individuals. The question that guided this study was: what are the factors and how they can serve as parameters for the construction of a social development measure based on measuring happiness? The overall objective was to build a theoretical and methodological proposal for social development measure based on happiness, titled Local Happiness Index (LHI) and apply the LHI to a population sample of inhabitants of the municipalities of Cabo de Santo Agostinho and Ipojuca. For this, various statistical procedures were used, especially the multivariate regression analysis technique, correlation tests and tests of mean differences. As a result it turns out that the population has high rates of socioeconomic inequality and social vulnerability, especially with regard to education, the income of workers and positions in the labor market. On this study, the subjective well-being indicators have greater inter-relations with the happiness levels of individuals than objective indicators of social well-being. Among the subjective well-being indicators analyzed, it appears that the indicators regarding satisfaction with the purchasing power/consumption capacity and to meet with the family and friendship relationships are those with greater influence in the happiness of individuals. These results demonstrate that in the context analyzed, happiness tends to be influenced by material aspects and not material living conditions. Finally, it turns out that among the populations investigated, recent immigrants tend to have less level of subjective well-being and happiness in comparison with non-migrants.

Abstract:
This study reviews a series of theoretical models and educational experiences, in order to examine how some of the claims made in the existing literature regarding the role of IT - mainly computer technologies - in writing instruction play out in the case of Italian as a Foreign Language (IFL). With this purpose in mind, this study examines a specific context - three IFL modules taught at the University of Warwick - and uses relevant teaching and learning experiences as a case-study and data sample. By using qualitative analysis supported by some quantitative methodologies, this study triangulates data from questionnaires, semi-structured interviews, focus-groups, field notes, classroom observation rubrics, as well as classroom artefacts, including online resources and educational software used over the course of the academic years 2004-2005 and 2005-2006. The data collected is filtered through a tripartite framework - learning/instructional environment, IFL tutors, and IFL students - designed to address the need expressed in the literature for analysis of multiple dimensions in complex interactions (Abbott, 1997; Athanases and Heath, 1995; Ramanathan and Atkinson, 1999; Snyder, 1997). The salient themes which emerge from the study are the critical roles of IFL tutors' and IFL students' expectations as well as the framework of values underlying these, along with particular features of information technologies themselves, in shaping participants' experiences and practices with respect to IT and writing, sometimes in unanticipated ways. Finally, the study considers the ways in which the results of the present research support, contradict, or expand existing literature, especially in relation to a number of specific factors, such as: the type of IT used in writing instruction; the physical configurations of IT-enhanced classrooms; and students' as well as tutors' approaches to learning and teaching IFL writing with and without technology. While the present work, like many other studies in the field of SLA and L2 writing, does not provide complete answer to the complex questions of language learning, it highlights the importance of both the instructional environment as well as the participants' framework of values. Only then, IT will be able to potentially enhance language instruction and become an integral component of learning. This research raises new questions, providing the basis for further research in the area of SLA theory and pedagogy.

Abstract:
The present research was carried out at UCL and may be considered an 'action-research', being the result of theoretical reflections and didactic applications. It originated in the observation that learning the Italian language was not perceived by ab initio students as connected to their personal and academic experiences. An initial hypothesis was formulated that an early introduction to literary texts accompanied by tasks could favour the interlanguage development in ab initio University students. Furthermore, this development, mainly as far as written production is concerned, might be facilitated in learners who are specifically guided and trained to elaborate a written input, which might be recognized by them as 'meaningful', thanks to noticing and attention management tasks, and to production tasks, which have a relevant influence on the re-organization of forms and meaning. In other words, this action-research aimed at verifying how written performance in ab initio students was activated by the Task Based Approach and reflected the integration of language and content through the acquisition of written skills in Italian. The method of the three year study (2002-05) can be described as a qualitative approach in any stage: pre-actional, i.e. selection of literary texts and of appropriate tasks actional, i.e. organization of texts and tasks into modules and collection of data post-actional, i.e. systematic analysis of students' productions and feedbacks. In order to carry out this action-research, a course was devised and was called Text and Task Course (TTC), in which literary texts were the starting point and the model necessary to process language through specific tasks. So linguistically easy literary extracts of about 300 words each have been selected to represent different written text types such as letters and dialogues, poems and ballads, narratives and descriptions, each of them dealing with topics familiar to the learners. Each text was accompanied and enhanced by pre-, while- and production tasks. Production tasks in particular favoured the re-elaboration of texts and combined representational structures with controlled attention. Measures for the assessment of written production during TTC were: Holistic Rating, which included fluency and creativity Accuracy Ratios, which included intelligibility index and error index Complexity Ratios, which included dependent and coordinate clauses per t-units ratio and re-elaboration of a model or text type. Each of these parameters have been analytically described and applied. In the light of these considerations, it seems consistent to claim that the positive results in written production, after a relatively short time of instruction, were influenced by the training Texts and Tasks learners (TTL) received in the TTC and their interlanguage development was favoured by the constant re-elaboration of written input. In conclusion, the initial hypotheses were confirmed and the Text and Task approach, under given circumstances, was validated.

The choice of this theme is due to the requirement stipulated by law 11.892 of December 29, 2008, the Federal Institutes of Education, Science and Technology, in offering undergraduate courses, especially in the areas of Science and Mathematics, despite its recognized expertise in professional education. Some research has dealt with the theme of the presence of the degrees in FIs, discussing the profile, the deployment and the challenges that they are consoli dated. However, here we seek teachers' perception of the process. Characterized as a qualitative study we used bibliographical research to survey the changes via legal documents in the institution, as well as research on the site e -MEC for recognizing the quantity of courses offered in this mode before and after the creation of the IFs. For analysis of the survey responses a nd interviews with teachers of Physical area of Goiânia the IFG campus, we used content analysis, according to Bardin (2002). It was identified as perceived by teachers who: (i) changes in the institution provided an expansion and internalization, even disorderly; (Ii) the change in the teaching profession and school mischaracterization with devaluation of technical education; (Iii) the course designed to meet the law, structure and operation in construction, has a focus on specific knowledge at the expense of teaching; (Iv) teachers work almost exclusively in the classroom, with little or no research and extension activities. These aspects lead to understand that there is still a lack of identity in I Fs after implantation of degrees and forward the changes in this institutions.<br>Neste trabalho buscou-se compreender de que forma a Licenciatur a em Física no campus Goiânia do IFG se estrutura e qual a perspectiva dos professores da área , especificamente, na implantação da licenciatura em Física , e frente as mudanças ocorridas na instituição.

Abstract:
In this thesis, a multi-scale and multi-physics coupling computation procedure for a 2D and 3D setting is presented. When modeling the behavior of a structure by a multi-scale method, the macro-scale is used to describe the homogenized response of the structure, and the micro-scale to describe the details of the behavior on the smaller scale of the material where some inelastic mechanisms, like damage or plasticity, can be taken into account. The micro-scale mesh is defined for each macro-scale element in a way to fit entirely inside it. The two scales are coupled by imposing a constraint on the displacement field over their interface. The computation is performed using the operator split solution procedure on both scales, using the standard finite element method. In a 2D setting, an embedded discontinuity is implemented in the Q4 macroscale element to capture the softening behavior happening on the micro-scale. For the micro-scale element, a constant strain triangle (CST) is used. In a 3D setting, a macro-scale tetrahedral and hexahedral elements are developed, while on the micro-scale Timoshenko beam finite elements are used. This multi-scale methodology is extended with a multi-physics functionality, to simulate the behavior of a piezoelectric material. An additional degree of freedom (voltage) is added on the nodes of the 3D macro-scale tetrahedral and hexahedral elements. For the micro-scale element, a Timoshenko beam element with added polarization switching model is used. Also, a multi-scale Hellinger- Reissner formulation for electrostatics has been developed and implemented for a simple electrostatic patch test. For implementing the proposed procedure, Finite Element Analysis Program (FEAP) is used. To simulate the behavior on both macro and micro-scale, FEAP is modified and two different version of FEAP code are implemented – macroFEAP and microFEAP. For coupling, the two codes are exchanging information between them, and Component Template Library (CTL) is used. The capabilities of the proposed multi-scale approach in a 2D and 3D pure mechanics settings, but also multi-physics environment have been shown. The theoretical formulation and algorithmic implementation are described, and the advantages of the multi-scale approach for modeling heterogeneous materials are shown on several numerical examples

Abstract:
This thesis aims to develop a method for optical localization and tracking of microconveyors moving over a surface in the context of micro-robotics. The principle of the proposed localization method consists of detecting a beam that is reflected on the micro-conveyor and analyzing this optical received signal to obtain the corresponding positions. The main elements of the system are a corner cube reflector, a MEMS mirror and a beam splitter. The corner cube is placed on the micro-conveyor in order to allow the optical signal to be reflected in its incident direction. The MEMS mirror allows the surface to be scanned with the laser. As for the beam splitter, it is an optical component used to guide the reflected light to a photodetector which is in turn used for localization and tracking through the exploitation of the signal that it detects. The important role of localization is to optimize the trajectory followed by the micro-conveyors. This could be achieved by controlling the respective position and speed of each of them in order to be finally able to localize the micro-conveyor with high precision and to avoid possible collisions between them. The surface used is a micro-conveying surface based on an electromagnetic principle, developed at the Roberval laboratory. The surface is used to perform experimental tests and to evaluate the developed sensor. A system calibration, using homography, was carried out in order to ensure the localization and tracking of the micro-conveyor, and then to control its trajectory and speed. Simultaneous localization and tracking of two micro-conveyors using time division multiplexing was also developed and validated in this work. An application of the thesis at the macroscopic scale is presented as well through applying free space optical communication between two mobile systems controlled by closed-loop optical tracking. Each system includes an active transmitter and a receiver (instead of a passive corner cube reflector in the case of the micro-conveyor). The system is driven by two servomotors which permit it to have free movement in three-dimensional space. The two modules must be aligned in order to be able to send and receive information by laser and thus communicate. In this case, a high-precision tracking system which is capable of aligning the two moving modules is required

Abstract:
The modelization of nano-reinforced material requires to take into account the size effect caused by the local phenomena at the interface between the nano-inclusion and the matrix. This size effect is interpreted through an increase in the ratio interface/volume and can be taken into account by introducing a surface elasticity at the interface. Whereas a lot of works have been developed from the analytical point of view, few contributions are related to numerical description and implementation of such surface elasticity in Finite Element Method (FEM). Our studies aim to develop efficient numerical tools based on FEM for the modeling of nanocomposites. Firstly, we evaluate the two existent numerical strategies namely the XFEM approach and the Interface element approach in reproducing the size effect in the homogenization process. Secondly, based on a performance test on the three types of formulations of E-FEM for the case of weak discontinuity, we propose an enhanced SKON formulation allowing to incorporate the effect of a coherent interface. Finally, the numerical modeling on the nonlinear behavior of nanocomposites is investigated. In the first step, a von Mises type elastoplastic law with linear isotropic hardening is considered for the bulk while the interface is considered as linear elastic

Abstract:
They are essential for humans because we are unable to synthesize them in situ. These two fatty acids are the representatives of omega-6 (LA) and omega-3 (ALA). Although they are essential and therefore necessary, the issue of the omega-6 / omega-3 ratio between these two families is also important to take into account in the diet. The consumption habits of the French over the past thirty years have shifted the balance against omega-3s compared to omega-6s. Or a rebalancing of the omega-6 / omega-3 balance tends to reduce cardiovascular risks. Linseed and camelina oils are endemic crops in France extremely rich in ALA. They are the most suitable for supplementation with omega-3 of plant origin. However, the high contents of polyunsaturated fatty acids in these oils are responsible for their low oxidative stabilities. The consumption of oxidized oils or fats is not recommended because they can contribute toxic molecules. The aim of this project was to improve the shelf life of oils rich in polyunsaturated fatty acids by enhancing the antioxidant extracts of the cakes obtained after crushing the seeds. Among the molecules studied, gallic acid, caffeic acid, propyl gallate as well as pinoresinol and lariciresinol purified from flax showed a real interest in slowing down oxidation, while secoisolariciresinol was ineffective

Abstract:
Une approche utilisant les résidus lissés a posteriori a été développée pour détecter ces changements de carte<br>Localization is an essential basic capability for vehicles to be able to navigate autonomously on the road. This can be achieved through already available sensors and new technologies (Iidars, smart cameras). These sensors combined with highly accurate maps result in greater accuracy. In this work, the benefits of storing and reusing information in memory (in data buffers) are explored. Localization systems need to perform a high-frequency estimation, map matching, calibration and error detection. A framework composed of several processing layers is proposed and studied. A main filtering layer estimates the vehicle pose while other layers address the more complex problems. High-frequency state estimation relies on proprioceptive measurements combined with GNSS observations. Calibration is essential to obtain an accurate pose. By keeping state estimates and observations in a buffer, the observation models of these sensors can be calibrated. This is achieved using smoothed estimates in place of a ground truth. Lidars and smart cameras provide measurements that can be used for localization but raise matching issues with map features. In this work, the matching problem is addressed on a spatio-temporal window, resulting in a more detailed pictur of the environment. The state buffer is adjusted using the observations and all possible matches. Although using mapped features for localization enables to reach greater accuracy, this is only true if the map can be trusted. An approach using the post smoothing residuals has been developed to detect changes and either mitigate or reject the affected features

Abstract:
This research work is exploring the development and management of Product-Service Systems. These new integrated offers go along with specific issues which challenge the current product development strategies. Analyzing the academic literature on this matter, this work focuses on methodological frameworks for the development of PSS offers. The first level of these frameworks is about engineering approaches considers Service Lifecycle Management (SLM) and Product Lifecycle Management (PLM) interactions. The second level is about specific systems or PSS development processes and methods. The third and last level challenges the definitions of a PSS model. Fed by this review, this work provides a coherent proposal for PSS development across these three levels, qualified by 7 analysis criterias. On the first level, a detailed PLM-SLM interaction scheme is proposed. It supports a development process on the second level, alongside a PSS model based on FBS and result-resource-process triplet as a matter for service adequate description. This proposal has been experimented through two study cases. The first one, about an existing and mature PSS, enabled the development of a quality management tool. The second, about the development of a connected diagnostic PSS enabled to strengthen the illustration of the proposal for its adoption

Tancred, Linda. "Mechanisms of the intracellular survival of Francisella tularensis." Doctoral thesis, Umeå universitet, Klinisk bakteriologi, 2011. http://urn.kb.se/resolve?urn=urn:nbn:se:umu:diva-45869.

Full text
Abstract:
Francisella tularensis is a gram-negative, highly virulent, intracellular bacterium which causes the zoonotic disease tularemia. The subspecies tularensis and holarctica are clinically important, and the former is the more virulent. The intracellular lifestyle of F. tularensis is not completely understood, but after uptake in monocytes, the bacterium escapes from the phagosome within hours and replicates massively in the cytosol. The escape is dependent on factors encoded by the Intracellular Growth Locus (igl) operon, located in the Francisella Pathogenicity Island, FPI. The thesis was aimed to clarify and understand the interaction of F. tularensis strains with the endosomal pathway of monocytic cells in general and the roles of the Igl proteins and the global regulator MglA for this interaction in particular. A focus has also been to elucidate the roles of reactive oxygen and nitrogen species for the intracellular host-parasite interaction. We show that mutants in the IglB, IglC, or IglD proteins or their regulator MglA of the live vaccine strain, LVS (subspecies holarctica), all demonstrated reduced replication rates and lowered cytopathogenicity compared to the wild type in a J774 mouse macrophage cell model. Colocalization with LAMP-1 was significantly increased for the IglC, IglD and MglA mutants compared to LVS. This indicated an impaired ability to escape into the cytoplasm, while at the same time they, like LVS, partly prevented fusion with lysosomes. IFN-γ activation of the J774 host cells prior to infection had a bactericidal effect on LVS and all of the mutants, though the cidal effect was significantly more pronounced for the mutants. Following IFN-γ activation, a majority of the mutant-containing phagosomesfused with lysosomeswhile LVS remained localized in the cytosol without significantly increased interactions with the endosomal pathway. Previous studies have revealed that IFN-γ activation of F. tularensis-infected macrophages leads to control of infection but conclusions about the importance of reactive nitrogen and oxygen species on bacterial killing are inconsistent. We found that the growth inhibition resulting from IFN-γ activation could not be attributed to an increased oxidative burst since PMA-induced superoxide production was still inhibited by LVS to the same extent as in non-activated macrophages. On the other hand, reactive nitrogen species may in part have contributed to the cidal effect. To further assess the role of reactive nitrogen species to the killing of F. tularensis, nitric oxide was administrated exogenously to J774 cells infected with LVS. This led to significant killing of intracellular LVS with a concomitant increased phagosomal localization and downregulation of the virulence gene regulator mglA. These effects were reversed by addition of a peroxynitrite decomposition catalyst. A spontaneous avirulent mutant of subspecies tularensis, strain FSC043, was previously demonstrated to provide protective immunity in mice. Here, microscopic analyses of the strain revealed an unusual intracellular localization with a delayed phagosomal escape. This may account for the low virulence, while at the same time FSC043 remains immunogenic and thereby confers protection. The igl operon is intact in strain FCS043 and we hypothesize that a defect in the FPI gene pdpC contributed to the observed phenotype. Altogether, this thesis work demonstrates the importance of the mglA and igl genes for the virulence of F. tularensis and specifically their important roles for a functional phagosomal escape and inhibition of the host cell oxidative burst. Also, addition of exogenous nitric oxide likely leads to formation of peroxynitrite intracellularly, a reactive molecule which confines the bacterium to the phagosome and confers a significant bactericidal effect on intracellular F. tularensis.

Chen, Shu. "Studies on interferon (IFN) induction and isolation of IFN-inducing mutant viruses." Thesis, University of St Andrews, 2011. http://hdl.handle.net/10023/1678.

Full text
Abstract:
The interferon (IFN) system is a powerful antiviral defense system. Host cell pattern recognition receptors (PRRs) recognise pathogen-associated molecule patterns (PAMPs) which when activated, lead to the transcription of the IFN-β gene. As a consequence IFN is secreted from the cell and activates the JAK-STAT pathway to up-regulate the transcription of IFN-stimulated genes (ISGs). The products of many ISGs inhibit viral replication and cell proliferation. Viruses encode IFN antagonists that dampen down the IFN response, making it less effective. However, within a virus population, there are always likely to be naturally occurring mutant viruses that have lost the ability to circumvent the host IFN response, and if isolated, these viruses would be unlikely to cause severe disease in the host and may therefore be developed as live attenuated virus vaccine candidates. To develop a methodology to rapidly isolate IFN-inducing mutant viruses, we generated an A549 reporter cell-line in which expression of GFP was driven by the IFN-β promoter. Using this cell-line, we show that the number of cells that became positive for GFP correlated with the amount of IFN secreted by the infected cells and the number of defective interfering (DI) particles within the virus preparations. However, we were unable to isolate IFN-inducing mutant viruses using the A549/pr(IFN-β).GFP cell-line(s). Possible reasons for this may be either that, in cells infected by IFN-inducing mutant viruses, an antiviral state was established independent of IFN that prevented virus replication in the reporter cells in which the IFN-β promoter was activated; or the viruses that activated the IFN-β promoter were DIs only which were not be able to replicate without non-defective helper viruses. A549/pr(IFN-β).GFP cells are also being used for high throughput assays to screen chemical libraries for compounds that block IFN induction. Such compounds may be potential candidates for anti-inflammatory drugs.

Cai, Di. "IfD - information for discrimination." Thesis, University of Glasgow, 2004. http://theses.gla.ac.uk/3972/.

Full text
Abstract:
The problem of term mismatch and ambiguity has long been serious and outstanding in IR. The problem can result in the system formulating an incomplete and imprecise query representation, leading to a failure of retrieval. Many query reformulation methods have been proposed to address the problem. These methods employ term classes which are considered as related to individual query terms. They are hindered by the computational cost of term classification, and by the fact that the terms in some class are generally related to some specific query term belonging to the class rather than relevant to the context of the query. In this thesis we propose a series of methods for automatic query reformulation (AQR). The methods constitute a formal model called IfD, standing for Information for Discrimination. In IfD, each discrimination measure is modelled as information contained in terms supporting one of two opposite hypotheses. The extent of association of terms with the query can thus be defined based directly on the discrimination. The strength of association of candidate terms with the query can then be computed, and good terms can be selected to enhance the query. Justifications for IfD are presented from several aspects: formal interpretations of information for discrimination are introduced to show its soundness; criteria are put forward to show its rationality; properties of discrimination measures are analysed to show its appropriateness; examples are examined to show its usability; extension is discussed to show its potential; implementation is described to show its feasibility; comparisons with other methods are made to show its flexibility; improvements in retrieval performance are exhibited to show its powerful capability. Our conclusion is that the advantage and promise IfD should make it an indispensable methodology for AQR, which we believe can be an effective technique for improvement in retrieval performance.

In this paper, we study ill-posedness concepts of nonlinear and linear inverse problems in a Hilbert space setting. We define local ill-posedness of a nonlinear operator equation $F(x) = y_0$ in a solution point $x_0$ and the interplay between the nonlinear problem and its linearization using the Frechet derivative $F\acent(x_0)$ . To find an appropriate ill-posedness concept for the linarized equation we define intrinsic ill-posedness for linear operator equations $Ax = y$ and compare this approach with the ill-posedness definitions due to Hadamard and Nashed.

Tranter, Rhys Edward. "Ill seen Ill said : trauma, representation and subjectivity in Samuel Beckett's post-war writing." Thesis, Cardiff University, 2014. http://orca.cf.ac.uk/59995/.

Full text
Abstract:
Over the last two decades, our understanding of Samuel Beckett’s life and work has been expanded by an unprecedented number of biographies, memoirs and personal correspondence published for the first time. As a result, academic research has been able to plot a series of connections between the writer’s literary work and the cultural and historical moments that shaped it. Beckett has been hailed as a poet whose work engages like no other with the atrocities of the Second World War. This thesis takes as its starting point an issue that often arises in evaluations of the writer, but which has never before been explored in detail: the theme of trauma. With reference to the work of prominent contemporary theorists, this project elucidates what we mean by the term trauma, and why it can be useful to our understanding of Samuel Beckett’s work. Drawing on the writings of Sigmund Freud, Cathy Caruth, and others, this thesis diagnoses traumatic symptoms and gestures in Beckett’s post-war writing. It identifies the role that ‘acting out’ and ‘working through’ plays in some of his late theatrical texts. And, moreover, the thesis begins to trace the role that trauma can play in our understanding of language and meaning. Adopting a broadly poststructuralist view, this study engages with texts by Maurice Blanchot and Jacques Derrida to ask how trauma challenges the status of language and the Western humanist subject. It will demonstrate how how trauma problematises our understanding of walking and thinking in Beckett’s post-war prose; where the presence of live theatrical production is unsettled by traumatic repetition; and why Beckett’s plays for radio undermine our expectations of twentieth-century modernity. While charting the way that Beckett uses and adapts traumatic themes and ideas, the thesis observes how the term signals a broader crisis in Western humanist understandings of time, place and identity.

Abstract:
All the work in this portfolio is concerned with making sense of ill health. In particular it primarily focuses on person centred approaches to working with people that have either been affected directly or indirectly by ill health. The research focuses on person centred therapists’ subjective experiences of working with older adults who have cancer. The aim is to draw the focus to Counselling Psychologists and other mental health professionals to the experiences of person centred therapists of working with older adults with cancer, as no research has been undertaken in this area previously. It is hoped that the research can illuminate an area in which little research has been undertaken previously. The Critical Literature Review focuses on the literature, which evaluates the effectiveness of psychological interventions for older adults with chronic illness. In particular it outlines the paucity that exists in this research and the possible reasons for this. It is hoped that this can focus mental health professionals on the need for more research to inform future interventions and indeed future debate on the effectiveness of different psychotherapeutic models in order provide clarity for professionals working with this client group. The Case Study provides an example of using a person centred approach with a client whose father has cancer. It provides an insight in to how the focus of the therapeutic relationship can help a client focus on underlying feelings and help see his situation with more clarity. It also provides an example of seeing a client’s lived experience descriptively as opposed to being seen as an expert trying to explain or analyse these experiences.

Full text
Abstract:
Viral myocarditis is an important human disease associated with a wide variety of viruses. The cardiac damage and inflammation associated with viral myocarditis can be immune mediated and/or due to direct cytopathic effect. Reovirus-induced myocarditis reflects direct virus-mediated apoptosis of cardiac cells, providing an excellent model to study direct cytopathic effect in the heart. Previous work has found interferon-beta (IFN) to be an important determinant for protection against viral myocarditis. Importantly, IFN signals through the Jak-STAT pathway to induce the expression of interferon-stimulated genes (ISGs) and establish an antiviral state. First, we investigated the underlying mechanisms of the IFN response in these cardiac cells. We found that high basal IFN-beta expression in cardiac myocytes protects this vulnerable, nonreplenishable cardiac cell type, while high basal expression of interferon alpha receptor 1 and latent Jak-STAT components in adjoining cardiac fibroblasts renders these cells more responsive to IFN and prevents them from inadvertently serving as a virus reservoir. This provides the first evidence for an integrated network of cell-type-specific innate immune components for organ protection. Next, we investigated whether particular reovirus strains were able to repress IFN signaling. We found that the mildly myocarditic reovirus, T1L, but not the nonmyocarditic reovirus, T3D, could repress IFN induction of ISGs and that this repressor function is determined by the M1 gene of T1L. Infection with T1L or reassortant or recombinant viruses containing the T1L M1 gene results in accumulation of IRF9 in the nucleus; an effect not previously described for any virus. The M1 gene has also previously been identified as a determinant of virus strain-specific differences in the IFN response, and the M1 gene and the IFN response have been identified as determinants of virus strain-specific differences in induction of murine myocarditis. Here, we find that virus-induced myocarditis is associated with repression of IFN function, providing new insights into the pathophysiology of this disease. Together, these data provide the first report of an increase in IRF9 nuclear accumulation associated with viral subversion of the IFN response, and provide evidence that virus strain-specific differences in IFN antagonism are a determinant of disease.

Abstract:
Mobile phones are here to stay, and the interaction with them is an evolving process. Designing software for mobile devices poses new challenges not seen in desktop computer environments. Screen size, performance and inputs, are just a few areas where they significantly differ. Although, intriguingly the gap between them is narrowing. Devices are now able to do things previously only restricted to PCs. The graphics chip integrated with high-end devices of today and tomorrow has capabilities worth exploring. With more power and advanced shader support, techniques such as image based lighting becomes worth exploring. To solve the problem described in this thesis I built a framework for image processing. This framework is further described indirectly throughout the report and is available upon request. The framework is never the focus of the report since any framework should suffice and the developed framework should not hinder the reader’s own experimentations. The purpose of this thesis is to see whether image based lighting (IBL) and image based rendering (IBR) is feasible with a mobile phone, as well as explore which applications it would suit for. Also, examining if it can be combined with high dynamic range imaging (HDRI) to enhance the user experience. Both IBL/IBR and HDRI might be new terms for the reader; if so I encourage you to read 2.2 for a deeper understanding.

Abstract:
The genomic RNA encoding the structural proteins of louping ill, a tickborne flavivirus, was cloned and sequenced. Sequence comparisons of louping ill envelope protein showed greater homology with tick-borne than mosquito-borne flaviviruses and greater homology with the western than the far eastern subtype of tick-borne encephalitis virus. Louping ill and tick-borne encephalitis viruses are probably varieties of a common tick-borne ancestral virus. The average amino acid sequence diversity between members of the tick-borne serogroup was significantly lower than that of mosquito-borne serogroups, suggesting that tick-borne flaviviruses have been subjected to different evolutionary immune selection pressure from the mosquito-borne viruses. Using the published model of tick-borne encephalitis envelope protein and the derived sequence data on louping ill virus, three discontinuous peptides (amino acids 81-88, 207-212 and 230-234) which may represent critical molecular determinants within the receptor binding site of tick-borne flaviviruses, were identified. These peptides may provide a specific genetic marker for these viruses. Recombinant baculoviruses and vaccinia viruses containing cloned DNA, encoding either the envelope protein or the structural proteins of louping ill virus, were constructed. Glycosylated envelope protein, presented both inside and on the surface of insect and mammalian cells, was expressed by all four recombinant viruses. Differences in antigenic presentation of envelope protein were observed between envelope protein and structural protein constructs as well as between insect cell and mammalian cell expression systems. Despite the expression of epitopes known to elicit neutralizing and protective antibodies when present in authentic antigen, the recombinant envelope protein expressed by either baculovirus or vaccinia virus failed to induce, under the experimental conditions employed, either neutralizing or protective antibodies in both mice and rabbits against louping ill virus. Hence, louping ill envelope protein expressed by baculoviruses and vaccinia viruses was antigenically reactive but immunogenically inert.

Abstract:
Thesis: M. Eng., Massachusetts Institute of Technology, Department of Electrical Engineering and Computer Science, 2017.<br>This electronic version was submitted by the student author. The certified thesis is available in the Institute Archives and Special Collections.<br>Cataloged from student-submitted PDF version of thesis.<br>Includes bibliographical references (page 26).<br>Severity of illness scores are useful in quality improvement for benchmarking care in the ICU and in research for risk-adjustment. [1] However, these scores lack generalizability beyond the patients on whom the scores were trained on. An international consortium, Global Open Source Severity of Illness Score (GOSSIS) Project, has put together various databases to create a definition of critical care severity independent of geographic and cultural data. [2] In order to combine the databases efficiently, 200 variables of interest were identified and extracted from the databases, when possible. Some variables showed different distributions throughout the databases that can be attributed to differences in medical practices and selection bias of hospitals. Thus, the predictive models that were trained in one database scored poorly when tested on a different database. Thus, a combination of the extracted variables from different databases with some necessary data manipulation should create a more generalizable severity of illness score.<br>by Maria I. Fabre.<br>M. Abstract:
Multiple sclerosis is the most common chronic disease of the central nervous system and is characterized by the destruction of myelin. Nowadays is subject of numerous studies to establish accurately the causes and define an effective treatment. The different susceptibility to disease between men and women, blacks and whites, young and old and unequal geographical distribution of MS appears to suggest an effective interaction between genetic and environmental factors. Although it is an autoimmune disease, treatment with immunosuppressive agents is still poor and it was initiated the use of immunomodulators such as interferon beta (IFN-β) to reduce the frequency and severity of the attacks.

Abstract:
Flertalet av studiens deltagare var positivt inställda till att krav ställs på att IFC måste användas i byggprojekt, när formatet utvecklas ytterligare.<br>Building information modeling (BIM) can be carried out in various software applications that are used by the different actors in a construction project. However, interoperability problems are one of the biggest challenges for BIM users today. The purpose of this study was to investigate how IFC format management is experienced by different actors in a construction project. The study also examines development opportunities of IFC format from the user’s perspectives to ensure smooth and quality-assured information flow in a project. In this research, a literature study was carried out to gain an understanding of the management of IFC format at present. A qualitative and a quantitative study was also carried out, by conducting interviews with actors from different companies and collecting data through a survey. The conclusion is that IFC format is very common in construction projects. IFC format is a tool that enables and facilitates communication and coordination between models and disciplines. However, the majority of IFC users consider that the IFC is still a defective format. The participants were primarily optimistic of the fact of IFC being a requirement within the construction industry. However, believes that further development is needed before being implemented.

Abstract:
<p> Since deinstitutionalization, the responsibility for mentally ill members of society has shifted to the criminal justice system in a process of trans-institutionalization or &ldquo;criminalization of mental illness&rdquo; (Slate &amp; Johnson, 2013, p. 28). Though various groups have been studied to ascertain their perception of mentally ill individuals and offenders, previous research focuses largely on students of psychology, social work, and medicine. Little research has been conducted regarding the perceptions of criminal justice students toward mental illness, despite the increasing involvement of the criminal justice system in treating and handling mentally ill individuals in the past thirty years. This exploratory research serves as a replication to a study which was conducted by Thompson, Paulson, Valgardson, Nored, and Johnson (2014). </p>
