Full textLassa virus causes a hemorrhagic fever endemic in West Africa and represents a threat for civilians. The pathogenesis and the immune responses associated with the disease are poorly understood. We followed pathological, virological and immunological parameters associated with fatal and non-fatal Lassa virus infection in the cynomolgus monkey. The clinical picture was characterized by depression, anorexia, weight loss and asthenia in survivors whereas the same symptoms were supported by fever, respiratory difficulties and epistaxis in animals infected with the lethal dose. Only fatalities have shown coagulation parameters dysfunction, rhabdomyolysis and an increase of renal function markers. We observed a different viral tropism in a function of the disease severity, with viral dissemination in organs that was more important and faster in fatalities, the appearance of numerous infectious particles number and more severe pathologic changes. Early and robust innate and adaptive immune response has been associated with the control of infection and recovery whereas fatal infections were characterized by a sepsis like inflammatory response, defective immune response as well as uncontrolled viral replication. This study sheds light on the pathogenesis of Lassa fever and reveals infection markers predictive of the disease outcome

Full textEast Asian lacquers are the saps of the species Rhus vernicifera, Rhus succedanea and Melanorrhoea usitate, obtained through an incision in the trunk of the tree. Lacquers have been applied as protective and decorative coatings in the most varied kinds of objects, for thousands of years. Lacquered objects are rare and highly valued artefacts that evidence, in part, the cultural exchange between Europe and Asia, initiated with the Portuguese maritime expansion. This cultural exchange has promoted the emergence of new artistic styles, which combine many decorative influences that complicate the attribution of the provenance of many lacquered artefacts. As lacquer producing species are distributed by relatively well defined regions, the identification of the lacquer applied in a given object can contribute to the determination of its origin. In this work, Py-GC/MS methods that allow discriminating between the three types of oriental lacquers were developed, and the polymerization process of lacquers was characterized by μ-FTIR and Py-GC/MS. These methods allowed the identification of a tree from Tapada da Ajuda’s Botanical Park, through the analysis of its dried sap, as belonging to Rhus vernicifera species, as well as to study the origins and techniques of a set of luso-oriental objects of unknown origin, and two regions for the provenance of these artefacts were recognized

Full textThe environmental movement has as one of its most evident facets a certain mystique or particular spirituality which usually tends to be characterised as \"green\". Thus, as matter of historical association, this distinctive feature is also present in the standards as well as in the theoretical and practical assumptions of some current environmental education (EE) practices, which firstly shaped the Anglo-Saxon context before becoming known in other countries. More specifically, however, with respect to the academic mileu of the Brazilian EE, as a central theme the dimension of spirituality has not received the same attention as other dimensions, such as the political and the social. Motivated by this fact this thesis sought to examine the place of spirituality in environmental education, including its limits and possibilities. To achieve this aim, this thesis is supported by three guiding objectives as follows: (1) the contemporary concept of spirituality; (2) the place of spirituality in education; (3) the ancestral importance of nature as a source for spirituality. In the first objective the aim was to determine the current meaning carried by the term spirituality, focusing on differentiating it from religion and/or religiosity; this is followed by the introduction and discussion of the concept of secular or immanent spirituality as developed by some contemporary philosophers, as well as the horizontal transcendence and sacred immanence, wich without denying the role of spirituality identify it as a meta-dimension of the human condition that can be grown from both secular and mystical-religious perspectives. The second objective sought to discuss one of the dilemmas present in contemporary education, embodied by the contrast between religious proselytism and the impoverished secularism. This section also argues for the possibility of an education of the spirit which, through important secular values for coexistence and human dialogue, recognises the relevance of the individual search of both the secular or religious sacred practices. Within the second objective, this thesis then explores the assumptions of the Holistic Education examining the importance of the master-disciple relationship in any educational process, and in the context of an education of the spirit in particular. The aim of the third objective was to shed light on the dichotomy which opposes sacralised/desecrated facets of the nature in the contemporary world. The third objective also aims to discuss the spirituality of the environmental movement and of the Deep Ecology in particular; to examine current proposals of a secular planetary religiosity; and to further explore the conditions enabling ecological religiosities to be established in the contemporary society. Lastly, in the fourth chapter draws from the earlier theoretical foundations focusing on a more authorial inflection in order to: (1) discuss the \"absence in terms\" of spirituality and the sacred in research in environmental education in Brazil, considering as explanatory hypotheses (a) the indifference from academia to the field of spirituality, partly explained by the latter being often associated with organized religions and neo-esoteric practices commonly known as New Age, and (b) the growing influence of the EE macro-critical tendency, which properly emphasizes the importance of the political and social dimensions in the everyday practice but tend to ignore other dimensions, in particular the spiritual/sacred, which in turn is relevant among the conservative camps; (2) suggest fundamental points of convergence between the notion of lay spirituality and the issues and themes directly related to environmental education, focussing on the discussion of the EE spiritual vocation and its initiatory potential, in that it is able to reveal elements to an immanent transcendence. In conclusion, we discuss the importance of an environmental education that also values the spiritual dimension in the current context of increasing uncertainty which is part of the current dwindling social practices that seems to become the hallmark of the Anthropocene.

Abstract: Nowadays, Brazil is one of the main silicon metal and iron-silicon producer in the world. But on the other hand, the amount of natural quartz that has been extracted for this purpose is up to 2 milliontons/year. The key-point is the large quantity of rejected quartz lascas, approximately 300,000 tons/year, generated during the various stages of extraction, crushing, selection, transportation, and washing. A new environrnentally mendly purification methodology denominated "quench-Ieaching" and "crush-leaching, that only uses aqueous leaching, has been developed. The result shows an effective elimination of major quartz impurities, such as Al, Fe, Na, K, Ca, Mn, ... , that transforms this rejected material into a 99.9% purity SI 'O IND 2'. The quality of this material is as high as the quartz powder commercially available in the intemational market for use as "fillers" and translucent silica glass raw material for semiconductor industries

For Luc Ferry the contemporaneity is characterized fundamentally as the era of love. According to the author, the secularization and the deconstruction process that happened in philosophy did not bequeath to man a society without foundations, not even a nihilistic pessimism. Instead, what is seem is a deification of the human being, through the appreciation of the other as the one who allows the man to come out of himself, and then, to exceed the subjective limits to expand his thoughts. This deification is a clear result of the appreciation of freedom, being this the divine spark in the human being. After all, freedom is characteristically what displaces man beyond all enabling determinisms to become himself then. In this way, what it is seen is a disruption, according to the author, to all the traditional ways of thinking and an overcoming of the religious and its seductions. The God-man is therefore the great accountable for the whole process of secularization: of both moral and love as well as of His own spirituality. In this context, it becomes visible a seemingly secular spirituality because, based on the secularized agape, it presents above all as a Christian spirituality, as it cannot disentangle itself from its own bases of influence of Christian love.

Full textLassa fever, an endemic disease with seasonal epidemic recrudescence, is considered to be responsible for 300 000 cases and 5000 death each year throughout Western Africa. As such, it has been listed as a priority for research. Over the past few years, Nigeria reported the highest and ever increasing number of cases, now exceeding a thousand per year. However, fifty years after the discovery of the disease, our knowledge is still limited and clinical research capacities remained poorly developed. The efficacy of the only available treatment, ribavirin, is still to be assessed. Hence, there is a need for up to date and reliable data on the disease course, management, mortality and prognostic factors. It is also of paramount importance that the patients suffering from Lassa fever could gain access to an optimised standard of care. Lastly, clinical research capacities need to be developed urgently in endemic areas. In this thesis, we review the knowledge gathered on Lassa fever from its discovery in 1969 till date. Then, we explain the reasons that led us to set up a response and research program on Lassa fever, as well as our approach to reinforce the standard of care and build a clinical research platform in collaboration with one of the main Lassa treatment centres in Nigeria. Next, we present the first results of the LASCOPE cohort study which represent the cornerstone of our program. From April 2018 to March 2020, 534 patients with RT-PCR proven Lassa fever were followed-up of whom 77 died (14.4%). 510 participants were enrolled in the cohort: 258 men and 252 women, 84 being children. The median delay between the symptoms onset and admission was 8 days (IQR 7-13). On admission, 37.8% had a Lassa RT-PCR Ct value <30. From admission to end of follow-up, 120 (26.5%) had a NEWS2 ≥7, 67(13.5%) had a KDIGO ≥2, and 41 (8.0%) underwent dialysis. All of them received ribavirin therapy with a median duration of 10 days (IQR 9-13). 62 (12.2%) died (57 [13.4%] adults, 5 [6.0%] children. The median delay between admission and death was 3 days (IQR 1-6). The following characteristics on admission were independently associated to mortality: age ≥ 45 years (adjusted Odds Ratio [aOR] 16.30 [95%CI 5.31-50.30]), NEWS2 ≥7 (aOR 4.79 [95%CI 1.75-13.10]), KDIGO ≥2 (aOR 7.52 [95%CI 2.66-21.20]), ALAT ≥ 3 times the upper limit of normal range (aOR 4.96 [95%CI 1.69- 14.60]), a Lassa RT-PCR Ct value <30 (aOR 4.65 [95%CI 1.50-14.50]). Those results will be helpful for the conception of future therapeutic clinical trials as well as for updating existing management guidelines. The framework of this cohort also gave us the opportunity to describe an unusual presentation of the disease, namely delayed paraparesis. This original observation add to the evolving knowledge regarding the spectrum of Lassa fever-related neurological manifestations. Two complementary studies are nested in the LASCOPE cohort, concerning the pharmacokinetics of ribavirin, as it is used for the treatment of Lassa fever, as well as the pathophysiology of cardiovascular dysfunction that can occur during Lassa fever. Inclusions in these nested studies is still ongoing. Finally, an upcoming phase II clinical trial evaluating the pharmacokinetics, security and tolerability of a novel antiviral drug, favipiravir, for the treatment of Lassa fever is currently being prepared in two Nigerian sites, including the one we are collaborating with

Full textThe cattle-raid (boēlasía) theme is a constant in the preserved texts of the tradition of Greek archaic epic, wherein it appears either in the form of brief narratives or of allusive references. Nonetheless, little has been written about this theme, and the few studies there are have focused only in the consideration of the recurrence of the boēlasía as a reflex of the importance of cattle in the honour economy typical of the homeric poems or in its explanation as a derivation of mythical structures inherited from the proto-indoeuropeans. This dissertation aims precisely at such blind spot, recovering and comparing part of the homerical references to this activity and examinig them via theoretical and methodological insights originated in the oralist critical tradition of the archaic épos, particularly those theme-based analytical methods that take into consideration the reception of the poems.

Full textLassa virus (LASV) is the causative agent of Lassa Fever (LF), an acute and occasionally fatal disease with hemorrhagic features. Current treatments for LF are limited to ribavirin and supportive care. The objective of this dissertation project was to characterize a human monoclonal antibody (mAb) with the potential to treat LASV infections. This antibody, mAb 8.9F, was isolated from the serum of a convalescent patient and has been shown to protect guinea pigs from lethal LASV challenge. Our approach focused on confirming that mAb 8.9F bound a quaternary neutralizing epitope (QNE), determining which residues comprised the epitope, and examining how mutations to critical residues would impact viral fitness. Our results showed that mAb 8.9F shared a number of features similar to previously characterized QNEs including broad neutralization of viral subtypes and high sensitivity to epitope disruption by detergents. We found that mAb 8.9F recognizes sites on both the GP1 and GP2 subunits of the LASV glycoprotein. Both subunits dissociate from the antibody at similar rates under chaotropic conditions, suggesting that these subunits have equal binding affinity with mAb 8.9F. To identify amino acids important to the epitope, we used a method of knockout site-directed mutagenesis that replaced sequences of LASV glycoprotein with homologous sequences from lymphocytic choriomeningitis virus. We created single mutations for those regions unreactive with mAb 8.9F and identified H124F, P145R, and F147N as mutations capable of abrogating mAb 8.9F reactivity. Viral fitness of these mutants was evaluated by measuring glycoprotein processing and viral infectivity. Our results showed that mutations abrogating mAb 8.9F binding did not significantly inhibit glycoprotein production, processing, or surface transport; neither was pseudovirus formation affected. However, these mutations did significantly reduce pseudovirus infectivity suggesting that mutants escaping mAb 8.9F neutralization may be less fit. The role of N-linked glycosylation in mAb 8.9F recognition was also examined but results proved inconclusive. In summary, this work provides a detailed analysis of the first documented QNE found on the Lassa virus glycoprotein. This work will help direct rational vaccine design and post-exposure antibody therapy.

Full textThe most important political fault line in American politics today is marked by the postwar liberal consensus itself. What is often overlooked, however, is that both liberals and anti-liberals assume a modern, progressive view of history in which the world is growing up to become more secular, technologically advanced, and egalitarian. Liberals celebrate this trajectory as they see themselves “on the right side of history.” They consider their opponents backward holdouts or, more generously, those not yet enjoying the goods of modern life. Anti- liberals on the right see the world according to liberalism proceeding apace to undo traditional morality, globalize economies, automate jobs, replace the nation-state, and undermine cultural norms. A nostalgic politics of reaction aspires to reverse the course of history and return to an unmolested golden age. In the words of one recent variation on this theme, only such a reversal can “Make America Great Again.” This dissertation offers intellectual portraits of three American social critics: Christopher Lasch, Paul Goodman, and Jane Jacobs. Each was a critic of progressive habits of mind in different ways, but all three offer an alternative to the progressive optimism and nostalgia for the past at work in today’s debates. If, then, these thinkers were reactionaries in resisting progressive programs of their times, they were enlightened reactionaries insofar as they rationally resisted the deeper assumption of inevitable progress that animates both left and right. While I address a specific concern in the work of each writer, I draw out three points common to their thought. First, each thinker dissolves the dichotomy between past and future that is central to progressive history. The progressive view of history shared by liberal and anti-liberal alike points toward, alternatively, an inevitably improved future or a past that is slipping away. Lasch, Goodman, and Jacobs, however, point to the continuity of past and future and resist subsuming the present in a deterministic account of history. Second, the thought of each embodies a defense of tradition – historically conditioned ways of knowing, as opposed to supposedly trans-historical universal reason. That defense is expressed not only in each thinker’s view of the past as a resource for the present, but in his or her resistance to the very idea of an Archimedean point that is assumed by claims to have seen the end of history. Indeed, each thinker’s arguments are presented explicitly as part of a tradition, and the work of each points to the importance of tradition as an indispensable lens on the world. Each author shows how the assumption of progress, despite progressives’ claims to have escaped tradition, does not reflect an inescapable law of history but is itself part of a modern tradition that we are free to modify. This in turn points to the political possibilities of recovering tradition as the basis of common discourse. To the extent we are conscious of the decisive role of tradition, we will be aware of the degree to which we are responsible agents: responsible for the contingent way we see the world, and for the contingent choices made by the light of our traditions. Finally, I argue that Lasch, Goodman, and Jacobs’s use of tradition stands in contrast not only to transcendent, objective reason but also to an understanding of traditions as closed language games, coherent in themselves but rationally inaccessible to one another. Lasch, Goodman, and Jacobs present a view in which traditions are dynamic, self-correcting, ongoing arguments within and between themselves. Their use of tradition-bound arguments to develop counter-traditions against dominant progressive perspectives exemplifies the way in which traditions might confront and correct one another

The Laúca Dam is located at km 307.5 of the Kwanza River (measured from its mouth), about 47 km downstream from Capanda Hydroelectric Project, near the village of Ya N hangue Pepe in the province of Kwanza Norte - Angola. Usually dam foundations are designed and constructed along the eroded channel rivers and supported on the rocks, and many of these rocks are fractured, and generate some discontinuities with a high degree of hydraulic conductivity of the dam foundations .This dissertation aims to assess and to evaluate the seepage conditions throughout the foundations of hydroelectric Laúca. This evaluation is normally made using the tests of loss of water with pressure and interpreted using the proposal of Maurice Lugeon, i.e., the well-known in the practice of dam engineering as the Lugeon test. This test is performed in rock masses through boreholes and different stages, aims to determine the hydraulic conductivity and the behavior of the fractured rock mass, taking into the consideration the water flow through the discontinuities and fractures. Using the Lugeon set of results, it can be estimated important engineering characteristics such as: the volume and the type of flow and representatives values of the equivalent mass of the hydraulic conductivity. All this information is useful to establish the grouting procedure and also to verify the performance of the treatment carried out to reduce leakage through the dam foundation.

Sequencing followed by metagenomic analysis is an extremely promising method for broad, unbiased disease profiling of patients for disease surveillance and diagnosis. Here, we use two popular metagenomics tools in union, k-mer-based Kraken with reads and BLAST- and LCAbased MEGAN with assembled contiguous sequence. We analyze sequence from 463 febrile and afebrile patients from Sierra Leone before and during the 2014 Ebola virus outbreak. We find that co-infection with malaria is correlated with increased survival of Ebola virus patients, from 18% survival rate to 53%. We also explore the utility of and emphasize the need for both positive and negative controls to distinguish and remove noise and contaminants from real signal, especially to keep up with increasing sensitivity in sequencing.

Full textAndesitic volcanoes in subduction zones are characterized by complex eruptive dynamics. Ground deformations are among less understood precursor signals of their explosive activity. They are related to internal dynamics as well as local tectonics and magma emplacement interactions. This study, focused on Lascar volcano (North Chile) and regional basement had two main objectives : 1) to better understand its recent activity from ground deformations and 2) to investigate its internal structure and relationships with local tectonics. We carried for that purposes a multidisciplinary study using different satellite and ground observations. Lascar is the most active volcano in Central Andes, one of the most adapted regions to investigate tectono-volcanic interactions. Furthermore, Lascar cyclic and historical activity makes it a very good candidate to study andesitic volcanoes dynamics. We improved Lascar recent activity understanding by showing and quantifying ground deformations from spatial geodesy methods and thermal variations using satellite observations. We also contributed with first detailed geophysical information (gravimetry) on Lascar volcanic complex internal structure as well as its links to previously fractured basement. Finally, we confirmed radar interferometry potentialities combined with ground geodetic observations (microgravity, GPS) to study and monitor this kind of edifices

The subject of this study is the self-theorization in the novel Fanny Owen (1979) by Portuguese writer Agustina Bessa-Luís. The self-theorization occurs when literature looks at itself in a movement of self-reflexivity. Fanny Owen herself is entirely a self-theorization exercise. Her work is a fictional treatment of biographical facts related to historical personalities – mainly to the writer Camilo Castelo Branco, to his friend José Augusto Pinto de Magalhães, and to Fanny Owen, daughter of the British Colonel Hugh Owen , who had a leading role in the Portuguese Civil War (1828 -1834) . These three characters are involved in a love triangle. The narrative presents as a background the cultural context dominated by the Romantic movement, which has notable influence on the characters, especially on Fanny and Jose Augusto, readers of literature – mainly of Lord Byron. Given the importance that the theme of the literature appears in Fanny Owen, it seemed appropriate to allocate it in the subgenre "novel of reading", concept created by German theorist Volker Rollof. The novel of reading is that work whose reading of literature by its characters featured prominently in the plot. Such a condition of the work herein studied favors our approach of the self-theorization theme, since this category of novels, when addressing the relationship between reader and literature reading, necessarily establishes a discussion on the literary phenomenon. However, self-theorization also appears at other levels in this novel. It occurs through the narrator, who at several moments uses strategies in an attempt to be confused with the empirical author and that, moreover, mind-wanders about the art of writing. It also occurs when it portrays the writer Camilo Castelo Branco as a writer in training. Chapter I addresses specifically the self-theorization theme and reflections supported by theoretical contributions from Jonathan Culler, Antoine Compagnon, David Lodge, Umberto Eco, Lelia Pereira Duarte and Karin Volobuef. In Chapter II, it begins the study of the novel Fanny Owen discussing its main themes: the romantic culture, which appears portrayed in panorama along the plot, and the love triangle, which I analyze in accordance with the ideas of Denis de Rougemont and René Girard. The third chapter deals specifically with the self-theoretical mechanisms in the novel Fanny Owen.

Full textThis work is about the teacher Leolinda Daltro s proposal of laity catechesis, at the early republican period in Brasil. It intends to give a panoramic history of Indian education in Brasil, since the Imperial period until today, showing the changes occurred since the implementation of the religious catechesis system until the last discussions about the Indian population s needs and conquest in the multicultural and bi-language fields, according each people needs. The work also tells the teacher s trajectory, the beginning of her actuation in the Indian cause, her excursion at Goiás and Mato Grosso s inlands, her attempt to found an association of Indian protection, and the moment she changes her focus at the feminine cause. Finally, it shows the book Da catequese dos índios do Brasil (notícias e documentos para a história) 1896-1911, that contain documents selected and edited by the teacher Leolinda Daltro in 1920. The book can be seeing as a mix of trip journal and scrapbook, telling a non-linear or chronological trajectory of this teacher, and it is a certification of the difficulties facing by a woman who decided to cross the Brazilian inland

Full textThis research aims to discuss the relevance of religious education in public schools and secular Brazil. At first develops an analysis of the historical-critical trajectory and evolution of religious education in public schools in the country, highlighting especially the relationship Church and State, National Legislation and lines underlying the pedagogical process. In the second chapter we work with João Décio Passos e Afonso Soares, we suggest three models of Religious Education present in Brazil, the Catechism, the Theological and Religious Studies, which examines the proposal to adopt the model of the Sciences of Religion as the best option to sustain pedagogical and epistemological autonomy of religious education in public schools in the country today. Finally, it is argued the need for a more comprehensive model for the study of religious phenomena through the transdisciplinary approach of Edgar Morin and Basarab Nicolescu, aimed at training the students a perspective of wholeness and citizenship, as expressed in Article 33 of LDB No. 9.394/96, with the new wording of Article 33.

Full textBiodiesel is an alternative fuel, renewable, biodegradable and nontoxic. The transesterification of vegetable oils or animal fat with alcohol is most common form of production of this fuel. The procedure for production of biodiesel occurs most commonly through the transesterification reaction in which catalysts are used to accelerate and increase their income and may be basic, acid or enzyme. The use of homogeneous catalysis requires specific conditions and purification steps of the reaction products (alkyl ester and glycerol) and removal of the catalyst at the end of the reaction. As an alternative to improve the yield of the transesterification reaction, minimize the cost of production is that many studies are being conducted with the application of heterogeneous catalysis. The use of nano-structured materials as catalysts in the production of biodiesel is a biofuel alternative for a similar to mineral diesel. Although slower, can esterify transesterified triglycerides and free fatty acids and suffer little influence of water, which may be present in the raw material. This study aimed at the synthesis, characterization and application of nano-structured materials as catalysts in the transesterification reaction of soybean oil to produce biodiesel by ethylic route. The type material containing SBA-15 mesoporous lanthanum embedded within rightly Si / La = 50 was used catalyst. Solid samples were characterized by X-ray diffraction, thermogravimetric analysis, infrared spectroscopy, nitrogen adsorption and desorption. For the transesterification process, we used a molar ratio of 20:1 alcohol and oil with 0.250 g of catalyst at 60?C and times of 6 hours of reaction. It was determined the content of ethyl esters by H-NMR analysis and gas chromatography. It was found that the variable of conversion obtained was 80%, showing a good catalytic activity LaSBA-15 in the transesterification of vegetable oils via ethylic route
