Abstract:
PAT1 is a recently identified member of the PAT family of proton/amino acid co-transporters with predominant expression in the plasma membrane of enterocytes and in lysosomal membranes of neurons. Previous studies in Xenopus oocytes expressing PAT1 established proton/substrate co-transport associated with positive inward currents for a variety of small neutral amino acids. Here we provide a detailed analysis of the transport mode of the murine PAT1 in oocytes using the two-electrode voltage-clamp technique to measure steady-state and pre-steady-state currents. The GPC (giant patch clamp) technique and efflux studies were employed to characterize the reversed transport mode. Kinetic parameters [Km (Michaelis constant) and Imax (maximum current)] for transport of various substrates revealed a dependence on membrane potential: hyperpolarization increases the substrate affinity and maximal transport velocity. Proton affinity for interaction with PAT1 is almost 100 nM, corresponding to a pH of 7.0 and is independent of substrate. Kinetic analysis revealed that binding of proton most likely occurs before substrate binding and that the proton and substrate are translocated in a simultaneous step. No evidence for a substrate-uncoupled proton shunt was observed. As shown by efflux studies and current measurements by the GPC technique, PAT1 allows bidirectional amino acid transport. Surprisingly, PAT1 exhibits no pre-steady-state currents in the absence of substrate, even at low temperatures, and therefore PAT1 takes an exceptional position among the ion-coupled co-transporters.

Abstract:
Recently, CdZnTe (CZT) detectors have been widely proposed and developed for room-temperature X-ray spectroscopy even at high fluxes, and great efforts have been made on both the device and the crystal growth technologies. In this work, the performance of new travelling-heater-method (THM)-grown CZT detectors, recently developed at IMEM-CNR Parma, Italy, is presented. Thick planar detectors (3 mm thick) with gold electroless contacts were realised, with a planar cathode covering the detector surface (4.1 mm × 4.1 mm) and a central anode (2 mm × 2 mm) surrounded by a guard-ring electrode. The detectors, characterized by low leakage currents at room temperature (4.7 nA cm−2 at 1000 V cm−1), allow good room-temperature operation even at high bias voltages (>7000 V cm−1). At low rates (200 counts s−1), the detectors exhibit an energy resolution around 4% FWHM at 59.5 keV (241Am source) up to 2200 V, by using commercial front-end electronics (A250F/NF charge-sensitive preamplifier, Amptek, USA; nominal equivalent noise charge of 100 electrons RMS). At high rates (1 Mcounts s−1), the detectors, coupled to a custom-designed digital pulse processing electronics developed at DiFC of University of Palermo (Italy), show low spectroscopic degradations: energy resolution values of 8% and 9.7% FWHM at 59.5 keV (241Am source) were measured, with throughputs of 0.4% and 60% at 1 Mcounts s−1, respectively. An energy resolution of 7.7% FWHM at 122.1 keV (57Co source) with a throughput of 50% was obtained at 550 kcounts s−1 (energy resolution of 3.2% at low rate). These activities are in the framework of an Italian research project on the development of energy-resolved photon-counting systems for high-flux energy-resolved X-ray imaging.

Abstract:
Aim: to conduct a comparative assessment of the hypotensive effect, as well as the effect on endothelial function, oxidative stress, and pulmonary artery pressure of chlorthalidone and hydrochlorothiazide as part of combined antihypertensive therapy in patients with arterial hypertension (AH) in combination with chronic obstructive pulmonary disease (COPD).Patients and Methods: the prospective study included 66 patients divided into two groups. As the main antihypertensive therapy, group 1 was prescribed with a combination of azilsartan medoxomil 40 mg and chlortalidone 12.5 mg as a fixed combination of Edarbi® CLO. Group 2 received a free combination of azilsartan medoxomil 40 mg (Edarbi®) and hydrochlorothiazide 12.5 mg. All patients underwent 24-hour blood pressure monitoring: (ABPM), echodopplercardiography, endothelium-dependent vasodilation, lipid peroxidation (LPO), nitric oxide metabolites, and endothelin-1 levels at baseline and after 6 months of treatment. Results: target blood pressure values (<130/80 mm Hg) were achieved in 91% of patients in group 1, and 51.5% in group 2 after 1 month of the study. After 6 months of treatment, all patients in both groups reached the target BP values, but in group 2, the dose of hydrochlorothiazide had to be increased to 25 mg. According to the ABPM data, after 6 months of treatment, group 1 showed a decrease in the morning surge in SBP by 7.0±2.1% and DBP by 10±7.3%. There was also an increase in the number of patients with the daily profile of «dipper» type to 78.8%. In group 2, there was a decrease in the morning surge in SBP by 6.3±5.9% and DBP by 4.8±4.6% after 6 months of treatment. There was an increase in the number of patients with the daily profile of «dipper» type to 36.4%. After 6 months of treatment, there was more pronounced improvement in laboratory parameters of group 1 characterizing endothelial dysfunction and oxidative stress. Statistically significant results were obtained for conjugated trienes, NO2, S, Imax, and endothelin-1 when comparing groups 1 and 2.Conclusion: treatment of AH in patients with concomitant COPD in the form of a fixed combination of azilsartan medoxomil and chlorthalidone versus free combination of azilsartan medoxomil with hydrochlorothiazide has a more pronounced antihypertensive effect, positively affecting the daily BP profile, pulmonary artery pressure, endothelial function and lipid peroxidation processes after 6 months of treatment.KEYWORDS: arterial hypertension, chronic obstructive pulmonary disease, endothelial dysfunction, lipid peroxidation, azilsartan medoxomil, chlorthalidone.FOR CITATION: Grigorieva N.Yu., Samolyuk M.O., Sheshina T.V. et al. Abstract. Intraplate regions characterized by low rates of seismicity are challenging for seismic hazard assessment, mainly for two reasons. Firstly, evaluation of historic earthquake catalogues may not reveal all active faults that contribute to regional seismic hazard. Secondly, slip rate determination is limited by sparse geomorphic preservation of slowly moving faults. In the Vienna Basin (Austria), moderate historical seismicity (Imax,obs/Mmax,obs=8/5.2) concentrates along the left-lateral strike-slip Vienna Basin Transfer Fault (VBTF). In contrast, several normal faults branching out from the VBTF show neither historical nor instrumental earthquake records, although geomorphological data indicate Quaternary displacement along those faults. Here, located about 15 km outside of Vienna, the Austrian capital, we present a palaeoseismological dataset of three trenches that cross one of these splay faults, the Markgrafneusiedl Fault (MF), in order to evaluate its seismic potential. Comparing the observations of the different trenches, we found evidence for five to six surface-breaking earthquakes during the last 120 kyr, with the youngest event occurring at around 14 ka. The derived surface displacements lead to magnitude estimates ranging between 6.2±0.5 and 6.8±0.4. Data can be interpreted by two possible slip models, with slip model 1 showing more regular recurrence intervals of about 20–25 kyr between the earthquakes with M≥6.5 and slip model 2 indicating that such earthquakes cluster in two time intervals in the last 120 kyr. Direct correlation between trenches favours slip model 2 as the more plausible option. Trench observations also show that structural and sedimentological records of strong earthquakes with small surface offset have only low preservation potential. Therefore, the earthquake frequency for magnitudes between 6 and 6.5 cannot be constrained by the trenching records. Vertical slip rates of 0.02–0.05 mm a−1 derived from the trenches compare well to geomorphically derived slip rates of 0.02–0.09 mm a−1. Magnitude estimates from fault dimensions suggest that the largest earthquakes observed in the trenches activated the entire fault surface of the MF including the basal detachment that links the normal fault with the VBTF. The most important implications of these palaeoseismological results for seismic hazard assessment are as follows. (1) The MF is an active seismic source, capable of rupturing the surface despite the lack of historical earthquakes. (2) The MF is kinematically and geologically equivalent to a number of other splay faults of the VBTF. It is reasonable to assume that these faults are potential sources of large earthquakes as well. The frequency of strong earthquakes near Vienna is therefore expected to be significantly higher than the earthquake frequency reconstructed for the MF alone. (3) Although rare events, the potential for earthquake magnitudes equal or greater than M=7.0 in the Vienna Basin should be considered in seismic hazard studies.

AbstractWe report a study of proton spin relaxation of an Iridium N-heterocyclic carbene complex [Ir(COD)(IMes)Cl] complex (where COD=1,5-cyclooctadiene, Imes=1,3-bis(2,4,6-trimethylphenyl)imidazol-2-ylidene). This compound is a pre-catalyst of the most efficient complex allowing the signal amplification by reversible exchange (SABRE) effect, relevant for enhancing weak signals in nuclear magnetic resonance (NMR). An important feature of the study is a combination of relaxation measurements over a wide field range with high-resolution NMR detection. As a result, we are able to measure nuclear magnetic relaxation dispersion (NMRD) curves in the field range 0.1 mT–16.4 T (corresponding to the frequency range 4 kHz–700 MHz) for individual protons in the complex under study.
