All the chlorophyll in higher plants is bound to specific proteins. These chlorophyll-proteins absorb light energy which is used by the electron transport processes of the photosynthetic apparatus. The synthesis and assembly of the photosynthetic apparatus and chlorophyll proteins is regulated by light and other factors which are incompletely understood. Antibodies were used to investigate the accumulation of chlorophyll proteins in plant systems which were completely lacking one or more of the factors involved in chlorophyll protein synthesis or degradation. Abstract:
The staphylococcal superantigen-like proteins (SSLs) are a family of polymorphic paralogs encoded within the Staphylococcus aureus genome, whose function remains unknown. The ability of SSL7, and a closely related paralog SSL9, to interact with cells of the immune system was investigated. Within the populations of human white blood cells, both SSLs interact selectively with monocytes, via specific but separate binding sites, leading to rapid uptake of SSLs. In addition, SSLs are rapidly taken up by dendritic cells (DC), but not macrophages, and target the mannose-receptor dependent endosomal a

The studies presented in this thesis focus on human and canine models for autoimmune disease, with the main aim to gain new knowledge about disease mechanisms and to further evaluate the dog as a model for autoimmune disease. Autoimmune Polyendocrine Syndrome type 1 (APS-1) is a hereditary human multiorgan disease caused by mutations in the autoimmune regulator (AIRE) gene. Hallmarks of APS-1 are chronic mucocutaneous candidiasis caused by Candida albicans, together with the autoimmune endocrine disorders hypoparathyroidism and adrenocortical failure. I have also developed clinically applicable objective measures of different aspects of gut inflammation so that the interface between gut inflammation and nutritional abnormalities may be explored. Chronic myeloid leukaemia (CML) is characterised by the constitutively active tyrosine kinase BCR-ABL. Standard treatment with tyrosine kinase inhibitors (TKI) in the chronic phase (CP) of CML conveys excellent long-term prognosis but is associated with side effects and costs. Treatment free remission (TFR) is possible in a proportion of patients discontinuing treatment after obtaining deep treatment responses but it is not fully known how to select the right patients for stopping attempts. Background: Type 1 diabetes (T1D) is a chronic, autoimmune disease caused by a T cell mediated destruction of ß-cells in pancreas. The development of T1D is determined by a combination of genetic susceptibility genes and environmental factors involved in the pathogenesis of T1D. This thesis aimed to investigate diverse environmental and immunological risk factors associated with the development of T1D. This was accomplished by comparing autoantibody development, T cell responses and the function of CD4+CD25+ regulatory T cells between healthy children, children at risk of T1D and T1D patients.

A modified Ellman method using acetylthiocholine as substrate has been used to reliably measure erythrocyte AChE activity (CV=7%). Reliable immunological quantitation of this enzyme (CV=7%) was achieved using monoclonal antisera to AChE (AE-1 and AE-2) in a fluid phase homogeneous enzyme-linked antiglobulin test (ELAT). AChE activity and antigen levels were similar for both adult male and female samples. Cord samples at term measured 60% of the adult value and reached 100% within 5 months. An overall increase in AChE activity was observed as pregnancy progresses with patients 1 to 3 days post

This thesis presents genetic studies aiming at enlarging our knowledge regarding the genetic factors underlying two immune-mediated diseases, hypothyroidism and autoimmune Addison’s disease (AAD), in dogs and humans, respectively. Genetic and environmental factors are indicated to contribute to canine hypothyroidism, which can be considered a model for human Hashimoto’s thyroiditis (HT). In Paper I we performed the first genome-wide association (GWA) study of this disease in three high-risk dog breeds (Gordon Setter, Hovawart and Rhodesian Ridgeback). Malondialdehyde (MDA) is a highly reactive three carbon dialdehyde produced as a byproduct of polyunsaturated fatty acid peroxidation and arachidonic acid metabolism. MDA readily forms intermolecular or intramolecular cross-linkages with chemical groups on various molecules and alters both their physicochemical properties and immunogenicity. In vivo these changes may generate neoantigens that consequently can elicit an immune response involving antibody production and immune complex formation. The overall aim of this thesis is to improve knowledge about the prevention of infectious and bleeding complications in patients with hematological malignancies, primarily in those with chronic lymphocytic leukemia (CLL) and myelodysplatic syndrome (MDS). Hypogammaglobulinemia, impaired production of immunoglobulins (Ig), is an established risk factor for infection, but the impact of IgG pure subclass deficiency (IgG subclass deficiency with adequate production of IgG, IgA, and IgM) has been debated.
