Full textMICA molecules interact with the NKG2D activating receptor on CD8 T cells. We investigated the role of MICA/NKG2D interactions in the maturation of the T cell repertoire within the thymus and in the destruction of intestinal epithelium by intraepithelial T lymphocytes in celiac disease. MICA tetramers identified a small population of late stage CD8 single-positive thymocytes ready to emigrate to the periphery as naive cells. MICA molecules were expressed in the Hassal's corpuscules of normal thymus. MICA overexpression in thymona was associated with a decreased percentage of NKG2D positive thymocytes wich expressed a less mature phenotype. MICA is strongly expressed a less mature phenotype. MICA is strongly expressed at epithelial cell surface in patients with active celiac desease, and is induced by gliadin or its p31-49.

Full textL-glutamate, the major excitatory neurotransmitter, also plays a role in peripheral tissues. Immune T cells express glutamate receptors and communicate through immunological synapses. T cell receptor signaling precedes mature immunological synapse formation, raising questions about the precise role of the synaptic structure. We show here that L-glutamate acts as an immunotransmitter at synapses between dendritic cells and thymic developing T cells. DCs express glutamate-specific vesicular VGLUT transporters and release glutamte through Ca2+- dependent exocytosis. Thymic T cells express functional NMDA receptors modulating the Ca2+ signal and apoptosis induced by T cell-DC interaction. NMDAR is corecruited with PSD-95 to contact zones, suggesting a role for TCR signaling in shaping a functional glutamatergic synapse. We propose that the glutamate release by DCs may elicit focal responses mediated by GluR signaling in developing T cells

Full textThe drug hypersensitivity syndrome or DRESS (Drug Rash With eosinophilia and Systemic Symptoms) is a severe skin reaction secondary to drug intake associated with multiple organ involvement. The immune response in DRESS syndrome is characterized by activated CD8 T lymphocytes, cytotoxic, with a cutaneous homing phenotype and directed against viral antigens such as EBV. Expended populations of CD8+ T lymphocyte are found in the peripheral blood System and in the skin, liver and lungs. We demonstrated that DRESS is characterized by the presence of numerous viral reactivation such as EBV, HHV-6, HHV-7; virus whose production is closely linked to drug taking. The presence of viral reactivation in DRESS syndrome is correlated with a significant increase in serum IL-10. During DRESS, regulatory B lymphocytes are thé principal source of IL-10 secretion. In addition this cytokine secretion is induced by drugs in vitro. Analysis of clinical and biological characteristics of patients with a prolonged course of disease allowed us to demonstrate that the essential component to this prolonged course would be the ethnicity of the patient. In conclusion, we evaluated the effectiveness of treatment of DRESS with intravenous immunoglobulin. We show that this treatment is neither safe nor effective and induces many undesirable side effects. Full textProstate cancer is the most common cancer and the third leading cause of death from cancer among Canadian men. According to Cancer Canada, in 2020 23 300 men will be diagnosed and 4 200 men will die from prostate cancer. Prostate cancer is one of the rarest cancers to have a slow evolution. However, there is a considerable variation in disease recurrence, treatment response and disease-specific death between individuals showing similar clinico-pathological characteristics. There is therefore a great need for the development of more precise predictive tools for the evolution and the classification of the disease. In recent years, there is a growing evidence suggesting that a comprehensive analysis of tumor-infiltrating immune cells could help to predict cancer progression. We therefore hypothesized that the study of immune cell infiltration in the prostate cancer microenvironment in patients treated by radical prostatectomy can predict the evolution of the disease. We used two techniques to analyze the immune context in prostate cancer. First, we profiled the immune microenvironment of the prostate cancer by immunohistochemistry. By focusing not only on the type, but also the quantity and the localization of immune-infiltrating cells in the various compartments of the tumor microenvironment. Our experiments showed that antigen-presenting cells infiltrating the center of the tumor does not predict the evolution of prostate cancer. On the other hand, infiltration into the normal-like peritumoral epithelium and the tumor margin are associated with the evolutions of the disease either by increasing the risks of progressing to biochemical recurrence for high infiltration by immature CD209+ dendritic cells in the tumor margin and higher risks of lethal PCa for high infiltration by CD163+ M2 macrophages in the tumor margin and normal-like peritumoral epithelium. Or by reducing the risks of definitive androgen deprivation therapy (ADT) or lethal PCa for a high infiltration by mature CD83+ dendritic cells in the normal-like peritumoral epithelium and the tumor margin. Subsequently, our analyzes showed that the predictive value of lymphocyte infiltration is mainly concentrated in the tumor center, as demonstrated by the increased risks of biochemical recurrence by intra-tumor low ratios of CD45RO/CD3 and high ratio of FoxP3/CD45RO. In addition, a lower risk of definitive ADT for low tumor ratios of FoxP3/CD3 and FoxP3/CD45RO and a decreased risk of lethal PCa for a high CD45RO/CD3 ratio in the tumor epithelium was also observed. Secondly, we performed gene expression profile analysis targeting a series of genes related to immune function using RT-qPCR experiments to complement the immunohistochemistry experiments. These experiments allowed us to identify a subgroup of genes related to the function of antigen presenting cells in which, overexpression is associated with decreased rates of lethal prostate cancer-free survival. In addition, we identified TIM-3 as being an important immunecheckpoint in the progression of prostate cancer since high expression levels of TIM-3 are associated with shorter survival to definitive ADT. The findings of our work have highlighted the importance of tumor compartments in a comprehensive study of immune infiltration in prostate cancer. Indeed, our data suggest that the prognostic value of immune infiltration depends on the type and density of the infiltrated cells but also on their location in the tumor microenvironment. In addition, we were able to identify important players in the evolution of the disease, such as cells expressing FoxP3, CD45RO, CD163, CD209 and the immune checkpoint TIM-3. The methodology of this study could be adapted to clinical reality with the analysis of preoperative biopsies. The use of such a prognostic tool in combination with routine clinico-pathologic tests could help identify patients who might benefit from intra prostatic immunotherapies prior to surgery to reverse this immunosuppressive condition in a context of precision medicine.

Full textJusqu'à présent, peu ou pas d'études ont été réalisées pour évaluer l'activité de l'IL-16 sur les macrophages et les cellules dendritiques, bien qu'elles expriment la molécule CD4. Nous démontrons que l'IL-16 module, dans les macrophages et les cellules dendritiques, l'expression de récepteurs de surface (CD25, CD80, CD86, CD83), l'expression des (co)récepteurs du VIH (CD4, CCR5, CXCR4), et la sécrétion de cytokines (IL-10, IL1-RA, TNF-[Alpha] IL-6) et de chimiokines (MIP-1[Alpha], RANTES). Dans un second temps, nous démontrons que l'IL-16 est capable d'inhiber la multiplication du VIH dans les macrophages et les cellules dendritiques. Ses résultats suggèrent que l'IL-16 est une molécule intéressante dans le cadre d'une immunothérapie anti-VIH. La seconde partie de notre travail concerne le monoxyde d'azote (NO°), une molécule qui possède, entre autres, des activités antivirales. Notre objectif était d'établir une corrélation entre la libération de NO°, l'apoptose des cellules T et la multiplication virale dans les macrophages. Malgré une faible régulation de l'apoptose spontanée des PBMC, le NO° ne module pas la mort cellulaire des PBMC induite par les anticorps anti-CD3 ou la phytohémagglutinine. Par ailleurs, ni les PBMC issus de patients, ni les macrophages infectés par le VIH, ne libèrent du NO°, ce qui est confirmé par l'absence de régulation de l'ARN messager de la NO-synthase. Enfin, le NO° ne module pas la multiplication virale dans les macrophages confirmant un rôle mineur du NO° dans la pathogenèse du VIH

Full textAllergy is an increasing public health problem in industrialized countries. It is a multifactorial disease influenced by the nature and characteristics of the allergen. The objective of this thesis was to characterize recombinant or purified forms of wheat grain allergens, and to identify the molecular structures responsible for sensitization: the epitopes. The reduction of disulfide bonds in the prolamin superfamily decreases, and sometimes suppresses, their IgE reactivity. The repetitive and non-repetitive domains of wheat storage proteins interact, which enhances their immunogenicity. There is a clear effect of the structure on the wheat allergens immunoreactivity. In addition to continuous epitopes, we showed the existence of conformational epitopes in gliadins, glutenins, LTP1, and Serpin-Z2B. Serpin-Z2B could be considered as a marker of large sensitization to wheat proteins. Several recombinant wheat allergens are now characterized and available for diagnosis of respiratory allergy and food allergy, and could be used for specific immunotherapy

Full textThe aim of my PhD thesis, conducted as part of a collaboration between the laboratory of protein separation sciences and nanotechnology (Paris-Sud Galen Institute) and the Micro and Nano System (basic electronics institute) group was to study the influence of self-assembled monolayers on the biological activity of bioreceptor toward biosensor development. In this project, we choose the organosilanes that can bind covalently to the silicon. Two silanes (7-octenyl trichlorosilane(OTS) and 3- aminopropyltriethoxysilane (APTES)) were studied., Their impact on the stability and the functionality of bio- receptor , model mouse immunoglobulin G (IgG), were evaluated. Spectroscopic characterization by XPS and infra- red Fourier transformed (FTIR) was first carried out to assess that the silanized surface exhibit carboxylic groups. The homogeneity of the surfaces was measured by AFM. Then, IgG were immobilized on these supports, covalently and a topographic AFM study was conducted to measure the distribution of these antibodies. The orientation of the grafted antibody was investigated by immune-enzymatic assays. We have also evaluated the binding capacity of the IgG immobilized on both surfaces. Then, the impact of aging on APTES surface was evaluated by spectroscopics and biological methods
