Abstract:
Background and aim: In situ gelling formulations are liquids that undergo a phase transition to form semi-solid gel structures within the physiological environment. Sustained release of drug products can therefore be delivered via injectable in situ gelling formulations where the gel formed in vivo acts as a drug reservoir, releasing drug via diffusion and/or degradation of the gel. Due to limitations of previously described in situ gelling formulations, the current research project aimed to address these issues and develop a biocompatible injectable in situ gelling formulation/s containing a

In situ forming implants (ISIs) are popular due to their ease of use and local drug delivery potential, but they suffer from high initial drug burst, and release behavior is tied closely to solvent exchange and polymer properties. Additionally, such systems are traditionally viewed purely as drug delivery devices rather than potential scaffold materials due to their poor mechanical properties and minimal porosity. The aim of this research was to develop an injectable ISI with drug release, mechanical, and microstructural properties controlled by micro- and nanoparticle additives. First, an inj

Corneal perforations are medical emergencies in which the cornea is partially or completely ruptured, resulting in the loss of stability of the whole eye. Such situations can be caused by bacterial or fungal keratitis, autoimmune, or ocular-surface related disorders. Corneal perforations, if left untreated, can cause partial or total blindness. Therefore, immediate treatment is necessary. The best treatment available is corneal transplantation; however, due to donor limitation, this treatment is non-feasible. Bacterial biofilms are involved in most device-associated infections. This thesis has developed an in-situ method to treat biofilm and a coculture for testing new antimicrobial biomaterials. We used surface immobilisation to promote Alpha-amylase and silver ion’s disruption of S. aureus biofilms. The treatment also helped coating the surface with silver particles to prevent recurrence. We also formed a coculture of S. aureus and pre-osteoblastic cells and showed that bacteria preferentially attached to the osteoblasts and became more resistant to antibiotic treatment.
