Abstract:
Major isoenzymes of androsterone-sulphating sulphotransferase (AD-ST) were isolated from liver cytosols of weanling and young adult female rats and their isoelectric properties were compared. On chromatofocusing the enzyme activity of young adults was eluted over a wider range of pH than was that of weanling rats. The activity at pH 7.8-7.2 (fraction I) is obvious at both ages, whereas the activity eluted over the pH 6.6-5.5 range (fraction II) is much lower in weanlings than in young adults. The AD-ST activities eluted in fractions I and II were separately purified by 3′-phosphoadenosine 5′-phosphate-agarose affinity chromatography at both ages. Two-dimensional gel electrophoresis of the isolated enzyme revealed several subunits with distinct pI values, but with the same molecular mass, namely 30 kDa. The relative levels of the pI 6.7 and pI 7.2 subunits are high and the relative level of the pI 6.1 is low in fraction I. In fraction II, the levels of pI 6.1 and pI 6.7 subunits are high and the level of the pI 7.2 subunit is low. There is no significant difference in the relative levels of the pI variants in each fraction between weanlings and young adults. The N-terminal amino acid sequences of the pI variants are identical within the area determined, irrespective of animal age or pI values. These results demonstrate that the pI variants of AD-ST are derived from the same precursor by post-translational modification or that they are products of closely related, but distinct, genes. The pI 6.1 and 6.7 subunits presumably increased during the development from the weanling stage to adulthood, resulting in the increase in acidic form(s) of AD-ST (fraction II) in adult females.

Abstract:
We examined the relationship between ventricular isomyosin composition and plasma thyroxine (T4) 5 wk after partial constriction of the abdominal aorta in weanling (21 day) and adult (8 wk) rats. Cardiac enlargement in weanling aorta-constricted animals was associated with a significant (P less than 0.001) decrease in %V1 isomyosin in both left (32%) and right ventricles (25%) with a corresponding increase in the %V3 isomyosin and a reduction in plasma T4 levels. However, the ratio of V1/T4 was similar in weanling control (17.8 +/- 0.8) and aorta-constricted (18.0 +/- 1.4) rats. In adult aorta-constricted animals, there was a significant (P less than 0.001) reduction in the %V1 (16%) isomyosin in the left ventricle and a smaller decrease in the right ventricular V1 (8%) with no change in plasma T4 levels. There was also a significant difference in V1/T4 between control (16.1 +/- 0.4) and aorta-constricted (13.9 +/- 0.7) adult rats in contrast to the maintenance of the V1/T4 in weanling aorta-constricted animals. Thus both increased workload and changes in thyroxine levels contribute to the isomyosin redistribution seen in weanling rats subjected to a pressure overload, whereas, in adult hypertrophied hearts, alterations of the ventricular isomyosin composition appear to be due solely to the increased pressure overload.

Enzyme activities related to aerobic metabolism and cyclic nucleotides were evaluated in muscle and nonmuscle cells of rat heart. The perinatal period from weaning to adult was studied. Malate dehydrogenase, citrate synthase, and 3-hydroxyacyl-CoA dehydrogenase activities of nonmuscle cells equal or exceed muscle cell activities in the weanling heart. Aerobic enzymes remain unchanged in nonmuscle cells during growth; however, muscle cell activities are enhanced. Adenylate cyclase and guanylate cyclase activities are higher in heart homogenates of weanling than adult rats. Despite elevated adenylate cyclase activity, cyclic AMP levels are identical in weanling and adult rats. Cyclic GMP levels are twofold higher in weanling than in adult rats. Muscle cell metabolism and cyclic nucleotide levels are associated with growth-related changes in heart function and cellularity, respectively.

In vitro degradation of 125I-labeled somatostatin-14 (Tyr11) [I-SS-14(Tyr11)] by luminal flushings of rat gastrointestinal segments was studied to characterize the fate of somatostatin in the gastrointestinal lumen. In addition, we evaluated the effect of rat milk as a potential inhibitor of luminal degradation of 125I-SS-14(Tyr11). Degradation of 125I-SS-14(Tyr11) was not detected in stomach flushings from either suckling or weanling rats. Luminal flushings from the small intestine degraded 125I-SS-14(Tyr11), with a gradient increase of activity from duodenum to midjejunum (degradation in suckling rat midjejunum and ileum was about five times lower than that in weanling rat). Degradation of 125I-SS-14(Tyr11) by luminal flushings of suckling rat midjejunum was dose dependently inhibited by rat milk casein and soluble fractions. Inhibitory activity of rat milk soluble fraction was heat labile and several times more potent than that of casein fraction. Casein fraction appeared to be stable at 100 degrees C for up to 30 min of exposure. These studies suggest that somatostatin is stable in the gastric lumen and that milk protects somatostatin from intestinal luminal proteolysis, indicating a possible physiological significance of milk-borne SS-14 for the suckling rat gastrointestinal tract.

The activities of two mitochondrial enzymes concerned in the utilization of acetoacetate, namely 3-oxoacid CoA-transferase and acetoacetyl-CoA thiolase, were high throughout the suckling and weanling period in brown adipose tissue of the rat. In contrast, 3-hydroxybutyrate dehydrogenase activity was comparatively low during this period. The activity of cytosolic acetoacetyl-CoA synthetase (involved in lipogenesis) declined after birth and remained low until the pups were weaned. Experiments with brown-adipose-tissue slices from weanling rats indicated that 70% of the [3-14C]acetoacetate utilized was oxidized to 14CO2, and this value was not altered appreciably by the addition of glucose and insulin.

Developmental aspects of the intestinal transport of 5-methyltetrahydrofolate (5-CH3H4-PteGlu) were studied in suckling (14-day-old), weanling (22-day-old), and adult (90-day-old) rats by use of the intestinal everted-sac technique. Mucosal-to-serosal transport of 0.5 microM 5-CH3H4PteGlu was linear with time for 40-min incubation and occurred at a rate of 0.035, 0.032, and 0.010 nmol X g initial tissue wet wt-1 X min-1 for suckling, weanling, and adult rats, respectively. The transport of 5-CH3H4PteGlu in all age groups was pH dependent (maximal at pH 6) and was higher in the jejunum than in the ileum. In all age groups the transport of 5-CH3H4PteGlu occurred by an active carrier-mediated system. The system was saturable; energy, temperature, and Na dependent; inhibited by structural analogues; and capable of accumulating the substrate against a concentration gradient. Kinetic parameters of the transport process, however, showed some difference. A progressive decrease in Vmax was observed from suckling to weanling to adult rats (5.1, 3.7, and 0.8 nmol X g initial tissue wet wt-1 X 30 min-1, respectively), while apparent Kt was similar (2.2, 1.73, and 1.79 microM, respectively). This study demonstrates that the transport system of 5-CH3H4PteGlu in the rat is fully developed at the suckling age. The results also suggest that the activity and/or the number, but not the affinity, of the transport carriers decrease with maturation.

Abstract:
Folate is an essential micronutrient that in mammals must be obtained from exogenous sources via intestinal absorption. Previous studies from our laboratory and others have demonstrated that folate absorption from the small intestine is mediated via the reduced-folate carrier (RFC). The goal of this study was to determine whether the initial step of folate uptake by intestinal epithelial cells, i.e., transport across the brush-border membrane (BBM) of the polarized enterocytes, is ontogenically regulated, and if so, to determine the molecular mechanism involved. Purified BBM vesicles (BBMV) isolated from suckling, weanling, and adult rats were used in this study. The initial rate of carrier-mediated uptake of a physiological concentration of folic acid (0.1 μM) by jejunal BBMV was found to be significantly ( P < 0.01) higher in suckling compared with weanling rats, which was, in turn, significantly ( P < 0.01) higher than that in adult rats. This decline in carrier-mediated folate uptake with maturation was found to be mediated via a decrease in the maximum velocity of the folate uptake process (6.55 ± 0.87, 2.16 ± 0.10, 0.90 ± 0.16 pmol·mg protein-1·10 s-1 for suckling, weanling, and adult rats, respectively), with no changes in its apparent Km. Western blot analysis of BBM protein and real-time PCR showed RFC protein and mRNA levels, respectively, to be significantly ( P < 0.01 for both) higher in suckling compared with weanling rats, which were in turn significantly ( P < 0.01 for both) higher than that in adult rats. These changes were found by nuclear run-on assay to be associated with a parallel decline in the RFC transcriptional rate in jejunal epithelia with maturation. In situ hybridization showed a similar pattern of RFC message distribution along crypt/villus axis in suckling and adult rat jejunum. These results demonstrate for the first time that folate transport across the intestinal BBM is under ontogenic regulation during early stages of life and that this regulation involves a transcriptional regulatory mechanism(s).

Abstract:
The metabolism of a mixture of [2-14C] and [3',5',7,9-3H] folic acid was studied in female weanling rats. Intact folates and folate catabolites were excreted in the urine. Folate polyglutamates were found in the tissues. Rats treated with the oestrogen diethylstilbestrol and 17 -ethynyloestradiol exhibited marked changes in the metabolic handling of folic acid and folate catabolism was greatly increased compared to controls. Allopurinol treatment gave greater label retention in the gut, with a substantial increase in catabolism compared to normals. A dose response relationship was illustrated between allopurinol dose and folate catabolism. After lead acetate dosing there was little radioactivity in the urine and tissues over 72h and more radioactivity was retained in the faeces compared to normals. Excretion of intact folates was depressed, especially 5MeTHF and 10CHOTHF. A tenfold increase in both lead and folic acid dosage resulted in an even further decrease of radioactivity in the tissues and urine over 72h. Excretion in the faeces was further elevated. Ferrous sulphate administration resulted in increased catabolism. The retention of radioactivity in the liver, kidney and gut was greatly reduced. A new method of folate analysis; Sephadex LH-20 was introduced. In vitro superoxide anion formation was illustrated using an allopurinol/xanthine oxidase system. Histological studies were employed to qualitatively and quantitatively illustrate the oxidative status in livers and brains of allopurinol and ferrous sulphate dosed rats. Increased dose related formazan deposition was observed when livers of pretreated animals were incubated with nitroblue tetrazolium. Formazan deposition was reduced in pretreated animals also treated with the anti-oxidants vitamin E, mannitol or 4-hydroxy-methyl-4,6-ditertiary-butylphenol. A possible route of folate catabolism is scission by a non-enzymic oxidation involving active oxygen species.

Abstract:
This thesis characterises the head direction system in pre-weanling rats around the time of eye-opening. Head direction (HD) cells are neurons in a distributed network of brain areas that fire action potentials as a function of the animal’s directional orientation relative to its environment. Environmental sensory information and self-motion cues are used to update the directional signal, with visual landmarks being particularly salient in controlling the preferred firing directions of HD cells. HD cells have previously been shown to be adult-like as early as postnatal day 16 (P16) in the rat pup (Wills et al 2010, Langston et al 2010), just after its eyelids come unfused and coinciding with the first spontaneous exploration of its environment. To determine if the HD cell circuit can organise before the onset of patterned vision, I recorded from the postsubiculum (PoS) before eye-opening in pre-weanling rats. The earliest instance of HD cells is found at P12, three days before eye opening. This early HD signal carries low directional information content and lacks stability both within and across trials. However, once the eyes of the rats are open, the HD system matures rapidly, with a dramatic increase in the number of directional cells and the quality and reliability of their directional signal. A prominent visual landmark is also able to exert control over HD responses within 24 hr of eye-opening. The data suggest that the directional circuit can be organised in the absence of visual spatial information, while patterned vision is rapidly integrated once it becomes available, for accurate and reliable orientation in space.

L'encapsulation peut affecter la digestion et la bioaccessibilité des composés encapsulés, ce qui peut ensuite affecter leur métabolisme. Le but de ce projet était d'étudier les effets de l'encapsulation sur la bioaccessibilité et le métabolisme du DHA, en étudiant une huile de DHA non-encapsulée ou encapsulée, apportée sous forme d'omelette comme matrice alimentaire.L'huile de DHA, composée de triacylglycérols riches en DHA, a été préparée sous forme d'émulsion de Pickering, stabilisée par des isolats de protéines de lactosérum dénaturés par la chaleur. De l'huile pure ou sous forme d’émulsion a ensuite été ajoutée à l’œuf liquide pour obtenir de l’omelette. Les effets de l'encapsulation sur la digestion ont été étudiés à l'aide d'un modèle de digestion statique in vitro pour adulte, puis l’impact sur le métabolisme du DHA a été mesuré sur un modèle rat pris au sevrage.Les résultats ont montré in vitro que l'encapsulation peut augmenter la surface de contact entre l'huile de DHA et les enzymes de digestion, favorisant l'hydrolyse lipasique de l’huile de DHA et améliorant ainsi la bioaccessibilité du DHA. In vivo, l'encapsulation n'a pas impacté le profil global des acides gras, et particulièrement l’accrétion du DHA dans le cerveau. En revanche, le profil des oxylipines, dérivés oxydatifs d’acides gras, a été fortement modifié dans le plasma, le cœur et même le cerveau. Les métabolites dérivés du DHA ont globalement été augmentés tandis que ceux issus des acides gras de la famille n-6 ont été essentiellement atténués.Par conséquent, l'encapsulation de l'huile de DHA pourrait non seulement améliorer la bioaccessibilité du DHA, mais constitue également un facteur clé dans le métabolisme du DHA pour produire des précurseurs de protectines et de maresines, améliorant ainsi le statut de santé global<br>Encapsulation may affect the digestion and bioaccessibility of the encapsulated bioactive compounds, which in turn affects their metabolism. The purpose of this project was to study the effects of encapsulation on DHA bioaccessibility and metabolism, based on omelet as a food matrix, which contains DHA oil as encapsulated or unencapsulated form.DHA oil composed of DHA-rich triacylglycerols was prepared as a Pickering emulsion, which is stabilized by heat-denatured whey protein isolates. Pure oil or emulsion was then incorporated into eggs and cooked in an omelet. The effects of encapsulation on the digestion and metabolism of DHA were studied by using INFOGEST static in vitro digestion model for adults and in a weanling rat model, respectively.The results showed that encapsulation can increase the contact surface between DHA oil and lipase during the in vitro digestion, thereby promoting the hydrolysis of DHA oil and improving DHA bioaccessibility. In vivo, encapsulation of DHA oil did not modulate the fatty acid profile in tissues, but remarkably modified the oxylipin pattern in plasma, heart and even brain. Specific oxidized metabolites derived from DHA were upgraded while those from n-6 fatty acids were essentially mitigated.Therefore, encapsulation of DHA oil could not only improve the bioaccessibility of DHA, but is also a key factor in the metabolism of DHA to produce protectins and maresins precursors, thereby improving global health status

Abstract:
‘Place cells’ are hippocampal pyramidal neurons which fire only when an animal visits a particular location in an environment (‘place fields’). Their location-specific firing is supported by configurations of multi-modal sensory cues. Almost all work on place cells focused on the properties of these cells in adult rats. Recent work (Langston et al., 2010; Scott et al., 2011; Wills et al., 2010) however could identify place cells in the hippocampus of very young rat pups (ca. 2 weeks old), and furthermore show that these cells undergo a strong functional maturation in terms of their location-specific firing properties. In this thesis, we investigated when the configural integration of sensory information first emerges during the postnatal development of the hippocampus, by probing the response of place cells to manipulations of sensory cues in a familiar environment in pre- (aged 2-3 weeks) and post-weanling (aged 3-4 weeks) rat pups as well as in adult controls. These included changing certain parts of the local olfactory/tactile cues as well as removing visual cues. Recordings were also undertaken in a completely novel environment. These experiments will further our understanding about how the brain’s system for the representation of space develops and in particular will shed light on the question whether place cells in young rat pups are driven by single cues or already have configural properties as in adult rats. The results described in this thesis are compatible with the view that place cell responses recorded in very young rat pups already integrate multimodal cues, as in adults, suggesting that hippocampal spatial responses are inherently configural. However, some evidence points out to a stronger influence of local non-visual intra-maze cues early in development, while the influence of vision seems to increase across development. This could be due to the concurrent maturation of the sensory systems. In a novel environment, animals of all ages form a novel spatial representation, showing that even animals as old as 2 weeks can already distinguish between two distinct environments.

碩士<br>國立陽明大學<br>解剖學研究所<br>87<br>Abstract Folic acid (folate) is an essential water-soluble vitamin and it undergoes a number of metabolic changes which involves the one-carbon groups transferation in organisms. Folate is needed for the synthesis of purines, pyrimidines and DNA; it also associates with the formation of the sperms and proteins. Recent studies had indicated that folate deficiency changed the concentration of monoamines in hypothalamus and caudate nucleus, and it also induced the behavior of spilling food in the mice. The objective of the study was to correlate the possible role of folate deficiency in the activity of monoaminergic neuron activity. Weanling Spraque-Dawley male rats were supplemented with either 0 or 4.0 mg of folic acid per kg diet lasting 5 weeks. The group of rats which fed with the folic acid-deficient diet had the tendency of spilling more food and consumed less water than the group with normal feeding, and produced less waste during the study. The concentration of DOPAC and DA were reduced in nigra-striatum, median eminence and paraventricular nucleus; but not in nucleus accumbens. There was a significant increased in 5-HIAA and 5-HT levels in paraventricular nucleus and nucleus accumbens of folate depleted rats. The moving distances were reduced in folate-deficient rats with auto-track system (ATS). The concentration of prolactin in serum was higher in deficient than control rats with radioimmunoassay (RIA). At least, after 5 weeks of feeding, the construction of fore-limb and hind-limb skeletal muscle by folate-depletion were different in two groups. In conclusion, folate deficiency affected the moving and feeding behavior may act on the changes of the dopaminergic and serotoninergic neuron activity. Abstract:
Mycotoxins are natural contaminants of livestock feeds that can impair production efficiency. Specialized feed additives known as mycotoxin adsorbents may reduce absorption of mycotoxins in animals. Many additives are thought to use nonspecific binding to sequester mycotoxins in the lumen of the gastrointestinal tract, but there is a lack of in vivo mechanistic evidence. The current research sought to quantify the adsorptive capabilities of a yeast cell wall-derived mycotoxin adsorbent and confirm whether its mode of action is by adsorption of the mycotoxin. Using radioactive aflatoxin B2, it was found that there was a significant reduction in urine radioactivity when the adsorbent was present and observed over time. No other significant effects of diet were noticed in the liver, kidneys or muscle. Without feces and tissue data in agreement with urine, it was inconclusive whether this adsorbent reduced absorption of aflatoxin in rats.<br>OMAFRA, Alltech
