Abstract:
We assessed the sensitivity and specificity of a newly developed DNA PCR kit (Roche Diagnostic Corporation, Indianapolis, Ind.) that incorporates primers for all the group M viruses for the detection of human immunodeficiency virus (HIV) type 1 (HIV-1) infection in Zimbabwe. A total of 202 whole-blood samples from adults whose HIV status was known were studied. This included 100 HIV-1-positive and 102 HIV-1-negative samples selected on the basis of concordant results obtained with two enzyme-linked immunosorbent assay kits. The prototype Roche DNA PCR assay had a 100% sensitivity for the detection of HIV-1 DNA and a specificity of 100%. We conclude that the new Roche DNA PCR kit is accurate for the detection of HIV DNA in Zimbabwean samples, in which HIV-1 subtype C dominates.

NAMI-A ((ImH)[trans-RuCl4(dmso-S)(Im)], Im = imidazole) and KP1019/1339 (KP1019 = (IndH)[trans-RuCl4(Ind)2], Ind = indazole; KP1339 = Na[trans-RuCl4(Ind)2]) are two structurally related ruthenium(III) coordination compounds that have attracted a lot of attention in the medicinal inorganic chemistry scientific community as promising anticancer drug candidates. This has led to a considerable amount of studies on their respective chemico-biological features and to the eventual admission of both to clinical trials. The encouraging pharmacological performances qualified KP1019 mainly as a cytotoxic agent for the treatment of platinum-resistant colorectal cancers, whereas the non-cytotoxic NAMI-A has gained the reputation of being a very effective antimetastatic drug. A critical and strictly comparative analysis of the studies conducted so far on NAMI-A and KP1019 allows us to define the state of the art of these experimental ruthenium drugs in terms of the respective pharmacological profiles and potential clinical applications, and to gain some insight into the inherent molecular mechanisms. Despite their evident structural relatedness, deeply distinct biological and pharmacological profiles do emerge. Overall, these two iconic ruthenium complexes form an exemplary and unique case in the field of medicinal inorganic chemistry.

Abstract Context.—A specific mutation, JAK2V617F, was recently recognized as having diagnostic value for myeloproliferative disorders. No practical assay is currently available for routine use in a clinical laboratory. Objective.—We report the development of a real-time polymerase chain reaction melting curve analysis assay that is appropriate for molecular diagnostics testing. Design.—Specific primers and fluorescence resonance energy transfer probes were designed, and patients with a previously diagnosed myeloproliferative disorder, de novo acute myeloid leukemia, or reactive condition were selected. The DNA was extracted from fresh and archived peripheral blood and bone marrow specimens, and real-time polymerase chain reaction melting curve analysis was performed on the LightCycler platform (Roche Applied Science, Indianapolis, Ind). Results.—The JAK2 region was successfully amplified, and wild-type amplicons were reproducibly discriminated from JAK2V617F amplicons. Titration studies using homozygous wild-type and mutant cell lines showed the relative areas under a melting curve were proportional to allele proportion, and the assay reliably detected one mutant in 20 total cells. JAK2V617F was identified in patients previously diagnosed with a myeloproliferative disorder or acute myeloid leukemia transformed from myeloproliferative disorder, whereas a wild-type genotype was identified in patients with reactive conditions or de novo acute myeloid leukemia. Conclusions.—These findings demonstrate the suitability of this assay for identifying JAK2V617F in a clinical laboratory setting. Furthermore, the semiquantitative detection of JAK2V617F in archived specimens provides a new tool for studying the prognostic significance of this mutation.

Adanya molting death sindrom yang umumnya terjadi pada stadia zoea-5 ke megalopa dan ke krablet-1 pada kepiting bakau, Scylla olivacea, diduga berkaitan dengan ketidakcukupan nutrien yang dikonsumsi larva, sehingga perlu dicobakan penggunaan pakan buatan (mikro) pada stadia tersebut. Penelitian ini bertujuan untuk mendapatkan dosis optimum penggunaan pakan mikro (micro diet, MD) untuk mensubstitusi penggunaan nauplius Artemia (Art) dalam pemeliharaan larva kepiting bakau. Hewan uji yang digunakan adalah larva kepiting bakau stadia zoea-4—5. Hewan uji tersebut dipelihara dalam wadah bak fibre berisi air laut 150 L dengan kepadatan 12 ind./L. Perlakuan yang dicobakan adalah pemberian pakan uji berupa: nauplius Artemia sebanyak 100% (100% Art), nauplius Artemia 75% + pakan mikro 25% (75% Art + 25% MD), nauplius Artemia 50% + pakan mikro 50% (50% Art + 50% MD), nauplius Artemia 25% + pakan mikro 75% (25% Art + 75% MD), dan pakan mikro 100% (100% MD). Pemberian pakan uji dilakukan pada pagi dan sore hari selama 15 hari pemeliharaan (hingga larva mencapai stadia krablet-1). Hasil penelitian menunjukkan bahwa pada penggunaan nauplius Artemia 50% + pakan mikro 50% didapatkan sintasan krabet-1 tertinggi (5,6%) dan berbeda nyata (<0,05) dengan sintasan krablet pada penggunaan 100% nauplius Artemia (sintasan 2,4%) dan 100% pakan mikro (sintasan 2,1%). Bobot badan, lebar karapaks krablet, dan aktivitas enzim pencernaan relatif sama di antara perlakuan. It is suspected that the event could be related to nutrient insufficiency consumed by the larvae which can be supplemented using artificial diet (micro diet). This study aims to obtain an optimum dosage use of the micro diet (MD) to substitute the use of Artemia nauplii (Art) in the crab-larva rearing. Test animals used were mud crab larvae of zoea-4—5 stadia. The test animals were reared in the fiberglass containers, filled with seawater as much as 150 L, and stocked with a density of 12 ind./L. The treatments tested were feeding tests in the form of: Artemia nauplii as much as 100% (100% Art), Artemia nauplii 75% + micro diet 25% (75% Art + 25% MD), Artemia nauplii 50% + micro diet 50% (50% Art + 50% MD), Artemia nauplii 25% + 75% micro diet (25% Art + 75% MD), and micro diet 100% (100% MD). The larvae were fed daily in the morning and afternoon for 15 days until the larvae reach crablet stage. The results showed that the use of Artemia nauplii 50% + 50% micro diets obtained the highest survival rate (5.6%) of crablet-1 and significantly different (<0.05) with the survival rates of crablet fed with 100% of Artemia nauplii (survival rate of 2.4%) and crablet fed with 100% micro diet (survival rate of 2.1%). Body weight, carapace width of crablet, and digestive enzyme activities were relatively similar between the treatments. The use of micro diet could replace 50% of the utilization of Artemia nauplii in larvae (zoea-5 to crablet-1) rearing of mud crab.

Abstract:
Abstract Introduction.—Measurement of hemoglobin A1c (HbA1c) is used as an objective measure of long-term blood glucose control in diabetic patients. Recent improvements in automation combined with new recommendations for precision and accuracy have caused us to reevaluate our methods for measuring HbA1c. Objective.—We evaluated a newly automated high-performance liquid chromatography (HPLC) instrument for measurement of HbA1c (Tosoh A1c 2.2 Plus Glycohemoglobin Analyzer, Tosoh Medics, Foster City, Calif) and compared the results obtained by HPLC to those obtained with an immunoassay (Hitachi 911, Boehringer Mannheim Corporation, Indianapolis, Ind). Results.—The Tosoh analyzer was found to be linear in a range of 5.3% to 17% and had a throughput of 20 samples per hour. HbA1c results for 102 patient samples by the 2 techniques showed good correlation, with a slope of 0.87 and an intercept at 1.27% ± 0.15%. Both the total and within-run coefficients of variation were consistently lower for the HPLC method compared with the immunoassay method. The HPLC method produces a chromatogram that shows the different hemoglobin fractions, allowing identification of abnormal hemoglobin variants. In heterozygous individuals, HbA1c measurements are made with no interference from the hemoglobin variant. In the case of homozygous or doubly heterozygous hemoglobin variants, the Tosoh HPLC identifies the hemoglobin variants as such and correctly does not report a HbA1c value in the presence of a markedly decreased amount of hemoglobin A. Conclusions.—The Tosoh HPLC provides adequate throughput and improved precision, and the method is traceable to the Diabetes Control and Complications Trial.

<p>This paper emerged from many months of regular participation in the parkour community in Indianapolis, Indiana. First, this study looks at the art of parkour as a bricolent engagement with architecture. Acts of bricolage, a sort of artistic making-do with objects (including one’s body) in the environment, play with(in) the dominant order to “manipulate the mechanisms of discipline and conform to them only in order to evade them” (de Certeau, 1984: xiv). Second, this study investigates architecture’s participation in the production and maintenance of what de Certeau calls, “operational logic” (p. xi). That is, how architecture acts as a communicative mode of space; one, which conveys rationalized or acceptable ways of being in space. This critical ethnography, then, takes to task the investigation of how traceurs, the practitioners of parkour, uncover emancipatory potential in city space through bricolent use of both architecture and the body.</p>

Die Moderne kommt aus Japan. Jedenfalls, wenn man dem Gründungsdirektor des New Yorker Museum of Modern Art Alfred H. Barre folgt, der auf dem vielleicht berühmtesten Schaubild der Kunstgeschichte die Moderne mit dem Jahr 1890 beginnen ließ und daneben kommentarlos notierte: «Japanese Prints». Japanische Kunst gilt nicht erst seit Barrs Entwicklungsgeschichte der abstrakten Kunst, die er 1936 skizzierte, als eine der wichtigsten Quellen der westlichen Moderne. Schon lange vor Barr hatten Kunstkritiker, -historiker und Künstler in der japanischen Kunst eine Tradition für die Moderne gesucht oder in ihr den ästhetischen Ausgleich für die Defizite der Modernisierung gefunden. Und bis heute ist Japan für unzählige Autoren das «Heimatland der Abstraktion», in dem, wie der japanische Maler Saburô Hasegawa 1940 schrieb, «schon vor 400 Jahren» verwirklicht wurde, was der Westen erst ind er Kunst des 20. Jahrhunderts erproben sollte.

AbstractWe present a new approach to construct several leakage-resilient cryptographic primitives, including leakage-resilient public-key encryption (PKE) schemes, authenticated key exchange (AKE) protocols and low-latency key exchange (LLKE) protocols. To this end, we introduce a new primitive called leakage-resilient non-interactive key exchange (LR-NIKE) protocol. We introduce an appropriate security model for LR-NIKE protocols in the bounded memory leakage (BML) settings. We then show a secure construction of the LR-NIKE protocol in the BML setting that achieves an optimal leakage rate, i.e., 1 – o(1). Our construction of LR-NIKE requires a minimal use of a leak-free hardware component. We argue that the use of such a leak-free hardware component seems to be unavoidable in any construction of an LR-NIKE protocol, even in the BML setting. Finally, we show how to construct the aforementioned leakage-resilient primitives from such an LR-NIKE protocol as summarized below. All these primitives also achieve the same (optimal) leakage rate as the underlying LR-NIKE protocol. We show how to construct a leakage-resilient (LR) IND-CCA-2-secure PKE scheme in the BML model generically from a bounded LR-NIKE (BLR-NIKE) protocol. Our construction of LR-IND-CCA-2 secure PKE differs significantly from the state-of-the-art constructions of these primitives, which mainly use hash proof techniques to achieve leakage resilience. Moreover, our transformation preserves the leakage-rate of the underlying BLR-NIKE protocol. We introduce a new leakage model for AKE protocols, in the BML setting, and present a leakage-resilient AKE protocol construction from the LR-NIKE protocol. We introduce the first-ever leakage model for LLKE protocols in the BML setting and the first construction of such a leakage-resilient LLKE from the LR-NIKE protocol.
