We have utilised the copper(I) triflate catalysed intramolecular [2+2] photocycloaddition reaction to cyclise diene 53 to afford the exo product 54 in 60% yield (Chapter 2). (Fig. 15762A) Using choline chloride.2zinc chloride ionic liquid we have carried out a one pot Fischer Indole synthesis. Phenyl hydrazine 149 was reacted with ethylmethyliketone 298 to give 2,3-dimethyl-indole 273 as the only product in 80% yield. Increases in the expression of prostaglandin E2 (PGE2) are widely known to be involved in aberrant growth in the early stage of colon cancer development. We herein demonstrated that the novel indole compound MW-03 reduced PGE2-induced cAMP formation by catalization to an inactive metabolite by inducing 15-hydroxyprostaglandin dehydrogenase through the activation of peroxisome proliferator-activated receptor-γ. Resumo: O pró-fármaco de diclofenaco, 1-(2,6-diclorofenil)indolin-2-ona (DICCIC), apresenta atividade anti-inflamatória comparável ao diclofenaco sem efeitos gastroulcerativos. Essa vantagem torna-o uma possível alternativa terapêutica em quadros de inflamação onde há a indicação de anti-inflamatórios não esteroidais. Part 1: The Direct Arylation of Azine N -Oxides with Aryl Triflates. Two sets of conditions for the palladium-catalyzed direct arylation of azine N-oxides with aryl triflates have been developed. Using palladium(II) acetate, a trialkylphosphine ligand, a carbonate base and pivalic acid, both mono- and diarylated azine N-oxides could be synthesized in good to excellent yields.* The reaction regioselectivity was examined for 3-substituted pyridine N-oxides, with arylation at C2 being favoured.
